miR-145 has been found to be significantly downregulated in gliomas, and overexpression of miR-145 increases glioma cell apoptosis and enhances chemosensitivity or herpes simplex virus thymidine kinase gene therapy. However, the correlation between miR-145 and the clinical prognosis of glioblastomas has never been explored. In this study, a retrospective study was conducted in 86 cases of patients with glioblastoma after neurosurgery combined with chemoradiotherapy, and 36 cases with traumatic brain injury. Our results showed that miR-145 was significantly lower in glioblastoma tissues than that in normal brain tissue (P < 0.05). Furthermore, miR-145 was lower in patients with lower Karnofsky Performance Scale (KPS) scores than in patients with higher KPS scores ( P < 0.05). Cox Regression analysis showed that low miR-145 expression was associated with poor patient survival ( P < 0.05). These data suggested that patients with glioblastoma with lower miR-145 expression are prone to shorter overall survival.  相似文献

Stavudine (d4T, 2′,3′‐didehydro‐2′,3′‐dideoxythymidine) was one of the first chain‐terminating nucleoside analogs used to treat HIV infection. We present the first structure of the active, triphosphate form of d4T (d4TTP) bound to a catalytic complex of HIV‐1 RT/dsDNA template‐primer. We also present a new strategy for disulfide (S–S) chemical cross‐linking between N6 of a modified adenine at the second overhang base to I63C in the fingers subdomain of RT. The cross‐link site is upstream of the duplex‐binding region of RT, however, the structure is very similar to published RT structures with cross‐linking to Q258C in the thumb, which suggests that cross‐linking at either site does not appreciably perturb the RT/DNA structures. RT has a catalytic maximum at pH 7.5. We determined the X‐ray structures of the I63C‐RT/dsDNA/d4TTP cross‐linked complexes at pH 7, 7.5, 8, 8.5, 9, and 9.5. We found small (~0.5 Å), pH‐dependent motions of the fingers subdomain that folds in to form the dNTP‐binding pocket. We propose that the pH‐activity profile of RT relates to this motion of the fingers. Due to side effects of neuropathy and lipodystrophy, use of d4T has been stopped in most countries, however, chemical modification of d4T might lead to the development of a new class of nucleoside analogs targeting RNA and DNA polymerases.  相似文献

Effective interactions between plants and pollinators are essential for the reproduction of plant species. Pollinator exclusion experiments and pollen supplementation experiments quantify the degree to which plants depend on animal pollinators and the degree to which plant reproduction is pollen limited. Pollen supplementation experiments have been conducted across the globe, but are rare in high latitude regions. To fill this knowledge gap, we experimentally investigated the dependence on animal pollinators and magnitude of pollen limitation in eight plant species north of the Arctic Circle in Lapland, Finland. Our findings show that all plant species were pollinator dependent, but not pollen limited. We discuss several mechanisms that might buffer our focal plants from pollen limitation, including plant and pollinator generalization, and attractive plant traits. Our results demonstrate that many plant species north of the Arctic Circle are currently receiving adequate pollinator service and provide a baseline for future comparisons of pollinator dependence and pollen limitation in the Arctic across space and time.  相似文献

The land use and land cover pattern of landscapes are key elements of basic landscape structure; accordingly, this pattern has an important role in landscape management, nature conservation and preservation. In Hungary, the naturalness of the vegetation was surveyed between 2003 and 2006, and the vegetation-based Natural Capital Index (NCI) was calculated for almost the entire area of the country. This field-based database gave us the unique opportunity to analyse the statistical connection between the naturalness of the vegetation and the landscape (land cover) pattern on a regional scale. In our study, we analysed the efficiency of the regional-level CORINE Land Cover (CLC) database for the estimation of the naturalness of the vegetation. This connection was analysed at the country scale using every (2272) Flora Mapping Unit (FMU), or 5.5 × 6.5 km quadrate, of Hungary. We calculated the shape-, edge- and size-related landscape indices for all FMUs on a landscape level (including all CLC patches) and a class level (the land cover polygons were classified according to their land cover characteristics and their level of hemeroby). We determined the Spearman’s correlations to reveal the statistical connections between the landscape metric parameters and the NCI values. All of the investigated area-weighted landscape indices: Main Patch Size, (MPS), Main Fractal Dimension Index, (MFDI), Total Edge (TE), Main Shape Index (MSI) and Number of Shape Characteristic Points (NSCP) on the landscape level showed a significant statistical connection with the NCI, but the sign of its correlation with the NCI contrasted with the findings from previous studies on a larger scale. Our study shows that scale has a strong impact on the sign of the correlation between the naturalness of the vegetation and the landscape structure. On a class level, particularly the shape-related landscape indices of the “Forest and semi-natural areas” showed statistically significant correlations with the NCI. The correlation strongly depended on the method of classification of the CLC polygons. Furthermore, the spatial pattern of the land-cover-type-based CLC polygon categories showed higher correlation values with the NCI than CLC polygon classes, which were categorized according to their hemeroby state. These results show that although the sign of the spatial pattern change in the main land cover classes is scale-dependent, they can be used to estimate the increase or decrease in the naturalness of the vegetation better than the spatial changes of the hemeroby-level-based landscape pattern. We can predict the change in the naturalness of vegetation based on the spatial changes in the land cover pattern.  相似文献

Cell disruption is crucial during recovery of biopharmaceuticals overexpressed in E. coli, which tend to be produced intracellularly as insoluble inclusion bodies. Miniaturized high-throughput systems can accelerate the laborious downstream protocol for such biopharmaceuticals and enable integrated process-development. A fast and robust cell disruption method reflecting the protein and impurity profile of homogenates obtained by large-scale methods is required for such an approach. We established a miniaturized bead mill for parallel mechanical cell disruption at the microscale. Its total protein and impurity release, protein pattern, and particle size distribution were compared to results from microscale enzymatic digestion and referred to laboratory-scale high-pressure homogenization. Bead mill disruption led to equivalent protein and impurity release as well as to the same particle size profile as the large-scale reference. In contrast, lysates obtained by enzymatic digestion contained only 30–47% of overall protein, 17% of dsDNA, and 7–10% of endotoxin compared to those obtained by high-pressure homogenization; also larger debris was present in lysates after enzymatic digestion. The established method is fast, efficient, robust and comparable to current large-scale standards, allowing for parallelization of experiments. Thus, it is the method of choice for rapid integrated process development at the microscale.  相似文献

On the basis of high-resolution melting, a high-throughput approach to measure melting temperatures (Tms) of short DNA hairpins was developed. With this method, Tms of thousands of triloop, tetraloop, and pentaloop hairpins involving various loop sequences and various closing base pairs (cbp) were obtained in hours. The stability of triloop hairpins decreased with the change of cbp (5′–3′) in the order of c-g > g-c > t-a ≥ a-t, showing that the cbp of 5′-Pyr-Pur-3′ (Pyr = pyrimidine, Pur = purine) contributed more stability than 5′-Pur-Pyr-3′. For tetraloop hairpins, GNNA, GNAB, and CNNG (N = A, G, C, or T; B = G, C, or T) were found to be highly stable irrespective of the cbp type. TNNA was also stable in both g-c and a-t families, while CGNA only in the c-g family. Pentaloop hairpins of cTGNAGg, cGNYNAg (Y = T or C) and cCGNNAg were exceptionally stable motifs. In most cases, pyrimidine-rich loops were more favorable to stabilize the whole structure than purine-rich ones. The present approach showed a good performance in assessing the thermal stability of large amounts of DNA hairpins comprehensively. These data are useful to understand the sequence dependence of the stability of DNA secondary structures and promising to improve the structure simulation by consummating basic databases.  相似文献

Changes in serotonin (5-HT) and 5-hydroxy indole acetic acid (5-HIAA), its major metabolite, in cerebral cortex, corpus striatum and hippocampus were investigated at 10th and 21st days of chronic ethanol ingestion in Wistar rats. Ethanol (7.2% v/v) was given to rats in a modified liquid diet. Biochemical analysis was performed in two groups of ethanol-treated and control rats (n = 6 for each group). Rats in each group were decapitated at the 10th and 21st days of ethanol consumption. Brains were removed and cerebral cortex, corpus striatum and hippocampus were dissected. 5-HT and 5-HIAA levels were measured in respective brain regions by using high performance liquid chromatography. In cerebral cortex and corpus striatum, 5-HT levels were significantly lower than control at the 10th day of ethanol consumption. At the 21st day, the levels tended to remain low, but not significantly different statistically. In hippocampus, 5-HIAA levels were significantly higher than control at 10th day of ethanol consumption. Increased 5-HIAA level returned to control values at the 21st day of ethanol consumption. Our results suggest that, 5-HT clearly seems to play a critical role in the brain at the 10th day of chronic ethanol consumption.  相似文献

Sexual selection is often assumed to be strong and consistent, yet increasing research shows it can fluctuate over space and time. Few experimental studies have examined changes in sexual selection in response to natural environmental variation. Here, we use a difference in resource quality to test for the influence of past environmental conditions and current environmental conditions on male and female mate choice and resulting selection gradients for leaf‐footed cactus bugs, Narnia femorata. We raised juveniles on natural high‐ and low‐quality diets, cactus pads with and without ripe cactus fruits. New adults were again assigned a cactus pad with or without fruit, paired with a potential mate, and observed for mating behaviors. We found developmental and adult encounter environments affected mating decisions and the resulting patterns of sexual selection for both males and females. Males were not choosy in the low‐quality encounter environment, cactus without fruit, but they avoided mating with small females in the high‐quality encounter environment. Females were choosy in both encounter environments, avoiding mating with small males. However, they were the choosiest when they were in the low‐quality encounter environment. Female mate choice was also context dependent by male developmental environment. Females were more likely to mate with males that had developed on cactus with fruit when they were currently in the cactus with fruit environment. This pattern disappeared when females were in the cactus without fruit environment. Altogether, these results experimentally demonstrate context‐dependent mate choice by both males and females. Furthermore, we demonstrate that simple, seasonal changes in resources can lead to fluctuations in sexual selection.  相似文献
