Abstract. Vegetation and its correlation with environment has been traditionally studied at a single scale of observation. If different ecological processes are dominant at different spatial and temporal scales, the results obtained from such observations will be specific to the single scale of observation employed and will lack generality. Consequently, it is important to assess whether the processes that determine community structure and function are similar at different scales, or whether, how rapidly, and under what circumstances the dominant processes change with scale of observation. Indeed, early work by Greig-Smith and associates (Greig-Smith 1952; Austin & Greig-Smith 1968; see Greig-Smith 1979; Kershaw & Looney 1985; Austin & Nicholls 1988) suggested that plant-plant interactions are typically important at small scales, but that the physical environment dominates at large scales. Using a gridded and mapped 6.6 ha portion of the Duke Forest on the North Carolina piedmont for a case study, we examined the importance of scale in vegetation studies by testing four hypotheses. First, we hypothesized that the correlation between vegetation composition and environment should increase with increasing grain (quadrat) size. Our results support this hypothesis. Second, we hypothesized that the environmental factors most highly correlated with species composition should be similar at all grain sizes within the 6.6-ha study area, and should be among the environmental factors strongly correlated with species composition over the much larger extent of the ca. 3500 ha Duke Forest. Our data are not consistent with either portion of this hypothesis. Third, we hypothesized that at the smaller grain sizes employed in this study (< 256 m2), the composition of the tree canopy should contribute significantly to the vegetation pattern in the under-story. Our results do not support this hypothesis. Finally, we predicted that with increased extent of sampling, the correlation between environment and vegetation should increase. Our data suggest the opposite may be true. This study confirms that results of vegetation analyses can depend greatly on the grain and extent of the samples employed. Whenever possible, sampling should include a variety of grain sizes and a carefully selected sample extent so as to ensure that the results obtained are robust. Application of the methods used here to a variety of vegetation types could lead to a better understanding of whether different ecological processes typically dominate at different spatial scales.  相似文献

We summarize some of the population-dynamic consequences of the mosaic structure of plant populations for the evolution of seed dispersal. A fairly elaborated set of theoretical ideas exist regarding the evolution of dispersal and we have synthesized some of them in an attempt to make them more accessible to field ecologists. We consider the relationship of these general theoretical ideas to our understanding of fruit and seed dispersal.We develop three related models to describe the similarities and differences in how dispersal functions for risk reduction (bet hedging), escaping the negative consequences of crowding, and escaping high concentrations of relatives. We also briefly discuss directed dispersal as a fourth population-dynamic aspect of dispersal. Dispersal can have a risk-reducing function only when there is global (metapopulation) temporal variance in success. Dispersal to escape the negative consequences of crowding requires only spatial and local temporal environmental variation. Dispersal for escaping high concentrations of relatives requires no environmental variation, but does require genetic population structure. Directed dispersal, defined as non-random into particular patch types contingent on the expectation of local success, is always valuable when possible and represents an advantage independent the others which can occur with random dispersal.In an effort to accommodate for the differences between simple mathematical models and the behavior of complex natural fruit and seed dispersal systems we have discussed the following issues: actual patterns of patch structure and dispersal distance; the implications of plant cosexuality, perenniality, and allocation costs of dispersal structures; and the impact of the detailed nature of density dependence, breeding systems, and genetic structure. We briefly compare the population-dynamic functions of dispersal presented here with the widely cited functions of colonization, escape, and directed dispersal. Finally, we suggest how the theoretical models can be used with field data to estimate the fitness consequences of dispersal.  相似文献

Temporal density‐dependent parasitism appears to explain the development of some pest‐ and pathogen‐suppressive soils. In many examples, an enrichment of hosts (soil‐borne pests or pathogens) precedes and supports a build up of beneficial parasites in perennial crops or in monocultures, and the population dynamics of hosts and parasites may be described in epidemiological terms. Although the examples support the concept of a balance of nature based on density‐dependent regulation, we lack detailed understanding of the processes that drive these beneficial epidemics. In addition, the epidemics develop slowly and often do not provide the control expected by farmers. Current research, therefore, emphasizes inundative release of beneficial agents. Nevertheless, we should study epidemics in soil in order to learn how they might be enhanced; to learn how better to utilize inoculum that is inundatively added to soil; and to contribute to the general discussion of population biology and regulation.  相似文献

Abstract: The amyloid β peptide (Aβ) of Alzheimer disease is derived from the proteolytic processing of the amyloid precursor proteins (APPs), which are considered type I transmembrane proteins. Here we report that the soluble fraction of isolated adrenal medullary chromaffin granules (CG), a model neuronal secretory vesicle system, contains an antigen that immunochemically and on sodium dodecyl sulfate-polyacrylamide gel electrophoresis was indistinguishable from full-length APP. A truncated APP fragment with intact Aβ sequence was also detected in the soluble fraction of CG. In vitro experiments showed that full-length APP was solubilized from CG membranes at 37°C as a function of pH, with a peak of activity between pH 8.5 and pH 9.0. Solubilization of full-length APP was inhibited by several protease inhibitors, including aprotinin, cystatin, and iodoacetamide, by the divalent cations Ca2+ and Zn2+, and by preheating of the membranes. These results are consistent with and suggest the involvement of an enzymatic mechanism in the solubilization of potentially amyloidogenic full-length APP. Production of Aβ from a transmembrane APP predicts a proteolytic cleavage within the lipid bilayer, a site relatively inaccessible to proteases. Thus, the detected soluble, potentially amyloidogenic, full-length APP may be a substrate for the proteases producing Aβ. The detection of soluble APP with intact Aβ sequence in secretory vesicles is consistent with the extracellular topology of amyloid depositions.  相似文献

We have investigated the electrophysiological basis of potassium inward rectification of the KAT1 gene product from Arabidopsis thaliana expressed in Xenopus oocytes and of functionally related K+ channels in the plasma membrane of guard and root cells from Vicia faba and Zea mays. The whole-cell currents passed by these channels activate, following steps to membrane potentials more negative than –100 mV, with half activation times of tens of milliseconds. This voltage dependence was unaffected by the removal of cytoplasmic magnesium. Consequently, unlike inward rectifier channels of animals, inward rectification of plant potassium channels is an intrinsic property of the channel protein itself. We also found that the activation kinetics of KAT1 were modulated by external pH. Decreasing the pH in the range 8.5 to 4.5 hastened activation and shifted the steady state activation curve by 19 mV per pH unit. This indicates that the activity of these K+ channels and the activity of the plasma membrane H+-ATPase may not only be coordinated by membrane potential but also by pH. The instantaneous current-voltage relationship, on the other hand, did not depend on pH, indicating that H+ do not block the channel. In addition to sensitivity towards protons, the channels showed a high affinity voltage dependent block in the presence of cesium, but were less sensitive to barium. Recordings from membrane patches of KAT1 injected oocytes in symmetric, Mg2+-free, 100 mM-K+, solutions allowed measurements of the current-voltage relation of single open KAT1 channels with a unitary conductance of 5 pS. We conclude that the inward rectification of the currents mediated by the KAT1 gene product, or the related endogenous channels of plant cells, results from voltage-modulated structural changes within the channel proteins. Elementary K+ currents through isolated ATP-sensitive K+ channels from neonatal rat cardiocytes were recorded to study their temperature dependence between 9°C and 39°C. Elementary current size and, thus, K+ permeation through the open pore varied monotonically with temperature with a Q10 of 1.25 corresponding to a low activation energy of 3.9 kcal/mol. Open-state kinetics showed a complicated temperature dependence with Q10 values of up to 2.94. Arrhenius anomalies of open(1) and open(2) indicate the occurrence of thermallyinduced perturbations with a dominating influence on channel portions that are involved in gating but are obviously ineffective in altering pore-forming segments. At 39°C, open-state exit reactions were associated with the highest activation energy (O2 exit reaction: 12.1 kcal/ mol) and the largest amount of entropy. A transition from 19°C to 9°C elucidated a paradoxical kinetic response, shortening of both O-states, irrespective of the absence or presence of cAMP-dependent phosphorylation. Another member of the K+ channel family and also a constituent of neonatal rat cardiocyte membranes, 66 pS outwardly-rectifying channels, was found to react predictably since open increased on cooling. Obviously, cardiac K (ATP) + channels do not share this exceptional kinetic responsiveness to a temperature transition from 19°C to 9°C with other K+ channels and have a unique sensitivity to thermally-induced perturbations.  相似文献

Alternaria infectoria E . G . Simmons was isolated from naturally infected eggs of the fig wax scale insect , Ceroplastes rusci L . (Homoptera: Coccidae) , that were showing different degrees of shriveling . This fungus proved to be pathogenic to eggs , nymphs and adults of this pest . The fungus had significant effects on both the mortality and hatchability of the eggs ( P = 0 . 0001) . Three days after exposure to the fungus , nearly one - third (31 . 5%) of the fungus - treated eggs became discolored and 17% showed moderate shrinking but no mortality . Fifteen days post - application , 91% of the fungus - treated eggs were diseased (74 . 8% of them were dead) due to A. infectoria, while no disease developed on the untreated eggs . Hatching decreased by 39 . 6% over a 15 - day period in the fungus - treated eggs compared to the control . Additionally , crawlers hatching from fungus - treated eggs became infected . The fungus seemed to induce crawlers to enter the settling stage . Seventy - two per cent of the fungus - treated crawlers versus 9% of the untreated controls entered the settling stage 6 days after exposure to the fungal inoculum . The highest level of nymphal mortality attributed to A. infectoria occurred when 30% (wet w:v blended mycelium in water containing 0 . 5% w:v Metamucil) inoculum was applied , and a high relative humidity was maintained for the following 48 h . This is the first record of A. infectoria on the fig wax scale and for the genus Alternaria as an entomopatho genic fungus .  相似文献

The binding of sodium n-dodecyl sulphate to lysozyme has been measured by equilibrium dialysis at 25°C and pH 3.2 over a range of ionic strengts from 0.0119 to 0.2119. Binding isotherms in the region corresponding to ionic binding between the surfactant anions and cationic amino acid residues on the protein have been interpreted in terms of the Hill equation and exhibit positive cooperativity with Hill coefficients in the region of 7–11. The Gibbs energies of binding have been calculated from the Hill binding constants and from the Wyman binding potentials. The stability of the surfactant-protein complexes is discussed in relation to the stability of surfactant micelles. Ionic binding of the surfactant is weakened and hydrophobic binding strengthened by increasing ionic strength.  相似文献
