 Plant responses to saturation vapour pressure deficit (SVPD) were studied by subjecting black spruce [Picea mariana (Mill) B.S.P.] and jack pine seedlings (Pinus banksiana Lamb.) to humid (0.3 – 0.8 kPa) or dry (2.0 – 2.5 kPa SVPD) regimes for 4 weeks using a computer-controlled environmental system to control diurnal variation in SVPD. Dry matter accumulation in needles was not altered by increasing SVPD. However, root growth declined by 60% which increased shoot to root ratio and reduced total seedling dry weight in both black spruce and jack pine. Relative growth rate of jack pine also declined to about half the rate of plants grown under humid conditions. In situ root marking studies showed that the decline in root growth of jack pine under the high SVPD was the result of reduced lateral root initiation, whereas root elongation was unaffected by humidity. A 4-week exposure to dry air increased abscisic acid (ABA) levels in needles, but not roots, of jack pine whereas ABA levels in black spruce were not altered. A short (3-day) exposure failed to increase needle ABA levels in either species. These results suggest that the responses of conifers to dry air were not the result of ABA accumulation. Saturation mapping of a very small genomic region is indispensable for map-based cloning. We applied a method based on sub-cloning and the Southern-hybridization technique for generating RFLP markers directly from yeast artificial chromosomes (YACs). Two YACs overlapping each other and covering the locus of the rice blast resistance gene, Pi-b, were used to construct a plasmid sub-library. Rice-specific and single-copy clones suitable as probes for RFLP analysis were selected from this sub-library by hybridization to the blots of digested DNAs of rice, YACs, and yeast. As a result, 22 markers were produced within a small chromosomal region including Pi-b. This case study shows that overlapping YACs known to cover the gene of interest are very useful in fine-scale physical mapping leading to map-based cloning of the target gene. Methanobacterium thermoautotrophicum was grown on a mineral salts medium in a fermenter gassed with H2 and CO2, which were the sole carbon and energy sources. Under the conditions used the bacterium grew exponentially. The dependence of the growth rate () on the concentration of H2 and CO2 in the incoming gas and the dependence of the growth yield ( ) on the growth rate were determined at pH 7 (the pH optimum) and 65° C (the temperature optimum).The curves relating growth rate to the H2 and CO2 concentration were hyperbolic. From reciprocal plots apparent K s values for H2 and CO2 and max were obtained: app. = 20%; app. = 11%; = 0.69 h-1; t (max)=1 h. was 1.6 g mol-1 and almost independent of the growth rate, when the rate of methane formation was not limited by the supply of either H2 or CO2. The yield increased to near 3 g mol-1 when H2 or CO2 were limiting. These findings indicate that methane formation and growth are less tightly coupled at high concentrations of H2 or CO2 in the medium than at low concentrations. The physiological significance of these findings is discussed. Alzheimer's disease (AD) is associated with plaque deposition in the brain of AD patients. The major component of the aggregate is a 39-42 long peptide termed beta-amyloid (Abeta). Except for Abeta, plaques contain several other components which co-precipitate together with Abeta. One such component is the small heat shock protein (sHSP) alphaB-crystallin. Instead of preventing the cell from the neurotoxicity of Abeta, alphaB-crystallin induces an increased neurotoxicity. We find - using solution state NMR spectroscopy - that alphaB-crystallin competes efficiently for Abeta monomer-monomer interactions. Interactions between Abeta and alphaB-crystallin involve the hydrophobic core residues 17-21 as well as residues 31-32 of Abeta, and thus the same chemical groups which are important for Abeta aggregation. In the presence of alphaB-crystallin, Met35 in Abeta becomes efficiently oxidized. In order to quantify the redox properties of the different complexes consisting of Abeta/alphaB-crystallin/copper, we suggest an NMR assay which allows to estimate the electrochemical properties indirectly by monitoring the rate of glutathion (GSH) auto-oxidation. The oxidation of the side chain Met35 in Abeta might account for the increased neurotoxicity and the inability of Abeta to form fibrillar structures, which has been observed previously in the presence of alphaB-crystallin [Stege, G.J. et al. α-d-Mannopyranosyl units were attached to an aromatic scaffold through disulfide linkages to obtain mono- to trivalent glycosylated ligands for lectin binding studies. Isothermal titration calorimetric (ITC) measurements indicated that binding affinities of these derivatives to Concanavalin A (Con A) were comparable to or slightly higher than that of methyl α-d-mannopyranoside (Ka values in the range of 104 M−1). The stoichiometries of the lectin-ligand complexes were in agreement with the formal valencies (1–3) of the respective ligands indicating cross-linking in interactions with the di- and trivalent derivatives. Multivalency effects could not, however, be observed with the latter. These ligands were shown to bind to the carbohydrate binding site of Con A using saturation transfer difference (STD) NMR competition experiments.  相似文献

The sweet receptor is a member of the G-protein coupled receptor family C that detects a wide variety of chemically and structurally diverse sweet-tasting molecules. We recently used saturation transfer difference spectroscopy (STD) to monitor the direct binding of a set of sweet agonists and antagonists to the human taste receptor in membranes prepared from human embryonic kidney (HEK293) cells transfected with and expressing the sweet receptor [F.M. Assadi-Porter, M. Tonelli, E. Maillet, K. Hallenga, O. Benard, M. Max, J.L. Markley, J. Am. Chem. Soc. 130 (2008) 7212-7213]. Here we review this work and related studies, discuss the procedures involved, and expand on their potential for identifying specific binding interactions of ligands to the membrane spanning and extracellular regions of the full heterodimeric sweet taste receptor. Whereas activity assays are unable to distinguish mutations that alter ligand-binding sites from those that alter signal transduction downstream of the binding site, STD NMR now allows us to make this distinction.  相似文献

Ion channels are proteins with a hole down their middle that control a vast range of biological function in health and disease. Selectivity is an important biological function determined by the open channel, which does not change conformation on the biological time scale. The challenge is to predict the function—the current of ions of different types and concentrations through a variety of channels—from structure, given fundamental physical laws. Walls of ion channels, like active sites of enzymes, often contain several fixed charges. Those fixed charges demand counter ions nearby, and the density of those counter ions is very high, greater than 5 molar, because of the tiny volumes of the channel's pore. Physical chemists can now calculate the free energy per mole of salt solutions (e.g. the activity coefficient) from infinite dilution to saturation, even in ionic melts. Such calculations of a model of the L-type calcium channel show that the large energies needed to crowd charges into the channel can account for the substantial selectivity and complex properties found experimentally. The properties of such crowded charge are likely to be an important determinant of the properties of proteins in general because channels are nearly enzymes.  相似文献

Residue Phe375 of cephalosporin acylase has been identified as one of the residues that is involved in substrate specificity. A complete mutational analysis was performed by substituting Phe375 with the 19 other amino acids and characterising all purified mutant enzymes. Several mutations cause a substrate specificity shift from the preferred substrate of the enzyme, glutaryl-7-ACA, towards the desired substrate, adipyl-7-ADCA. The catalytic efficiency ( [Formula: see text] (cat)/ [Formula: see text] (m)) of mutant SY-77(F375C) towards adipyl-7-ADCA was increased 6-fold with respect to the wild-type enzyme, due to a strong decrease of [Formula: see text] (m). The [Formula: see text] (cat) of mutant SY-77(F375H) towards adipyl-7-ADCA was increased 2.4-fold. The mutational effects point at two possible mechanisms by which residue 375 accommodates the long side chain of adipyl-7-ADCA, either by a widening of a hydrophobic ring-like structure that positions the aliphatic part of the side chain of the substrate, or by hydrogen bonding to the carboxylate head of the side chain.  相似文献

Species richness in communities varies with habitat area, productivity, disturbance level, intensity of species interactions, and regional/historical effects. All of these factors influence coral richness but their effects vary with spatial scale, position on the reef, and regional location. Species richness of corals along depth gradients shows a unimodal, hump-shaped curve that peaks at intermediate depths. Moreover, the peak of the curve is higher in regions with larger species pools. This “regional enrichment” of the local community appears in line transect samples as small as 10 m in length. The pattern suggests that ecological factors operating over scales of tens of meters and regional/historical factors operating over thousands of kilometers can both affect local richness. Regional factors probably include differences in speciation relative to extinction rates among regions and proximity of local sites to richness hotspots. Plausible factors operating at the local scale are species interactions, disturbance, and productivity which combine in different ways to produce the unimodal pattern. Shallow areas support few species because extinction rates are high due to frequent disturbance or because of environmental extremes. In addition, high productivity encourages rapid growth and thus the potential for intense interspecific competition. In areas where branching acroporids are abundant, exclusion by these dominant competitors is possible. Deep areas may be depauperate because few species can tolerate the low light levels found there. Areas of intermediate depth have the richest communities because they are open for colonization by many species and because extinction rates are low. Several theories may explain this “openness” and species persistence: 1. Occasional disturbance coupled with low growth rates results in glacially slow exclusion by the dominant competitor. 2. Aggregation of corals creates spatial variation in the intensity of competition and thus refuges from competition within a spatial landscape. Inferior competitors persist because they are superior at dispersal and refuge colonization. 3. Specialist predators focus on high-density juvenile populations near the parent, creating ecological space for colonization by non-prey. 4. Coral competitive abilities are roughly equal and recruitment into the community is a probabilistic event. The community thus exhibits random drift and exclusion is an extremely lengthy process.
