The relative growth rate of the brown alga Sargassum horneri (Turner) C. Agardh in the sea at Maizuru Bay, Japan, was investigated from summer to winter to examine the impact of declining solar radiation and temperature. The observed change in the growth rate between summer and winter was insignificant, in spite of a major decline in solar radiation and water temperature. The mean relative growth rate was 4.6% per day, equivalent to the daily net photosynthetic production previously reported for this species.  相似文献

The importance of annual recruitment to the structure of adult stands of Sargassum was determined for a mixed species Sargassum bed at Rottnest Island, Western Australia. The morphologically similar species Sargassum spinuligerum Sonder, S. distichum Sonder, and S. podacanthum Sonder grew together in the shallow subtidal (6 m). Positive species determinations were only possible when thalli were reproductive, so recruits, bases, and vegetative annuals for all species were grouped together. Densities of recruits, perennial bases, vegetative annuals, and reproductive annuals were determined at monthly intervals from 20 randomly placed 0.25-m?2 quadrats. Recruitment and mortality for recruits and adults were further determined at three monthly intervals from 6-×-1-m?2 permanent quadrats. The density of adults varied little with season (between 32 and 58 m?2). Growth of annuals was initiated in April, thalli became reproductive by late August–early September, and senescence occurred in December–January. Density of recruits was highly variable (1.6–210 individuals-m?2) and peaked seasonally during late summer (January–February) and then declined rapidly. Adults showed a complete turnover of thalli in the bed over 25–27 months. Adult mortality was compensated by annual recruitment from propagules (43%) and vegetative regeneration from fragments of holdfasts left on the reef (57%). A seasonal pattern in survivorship was observed for adults that grew from recruits with higher initial numbers and lower mortalities for August and November cohorts. Little seasonally was observed in survivorship of adults that grew vegetatively from remnant crusts. Although initial cohort sizes were smaller for adults grown from recruits than from remnant crusts, mortality was lower, resulting in similar contributions to adult density from both recruits and remnant crusts. Recruitment from propagules and vegetative regeneration played an important role in buffering the adult stand from high rates of mortality and reducing seasonal variation in adult density and contributed to the persistence and seasonal structure of Sargassum beds at Rottnest Island.  相似文献

A method for isolating high‐quality DNA is presented for the green algae Caulerpa sp. (C. racemosa, C. prolifera, and C. taxifolia) and the brown alga Sargassum muticum. These are introduced, and invasive species in Europe, except for the native C. prolifera. Previous methods of extraction, using cetyl trimethyl ammonium bromide or various commercial kits, were used to isolate genomic DNA but either no DNA or DNA of very low quality was obtained. Genomic libraries were attempted with Caulerpa sp. on three occasions and either the restriction enzyme, the Taq polymerase, or the T4 ligase was inhibited, probably by the large amount of polysaccharides in these algae. The method presented here consists of the rapid isolation of stable nuclei, followed by DNA extraction. Yields of 6–10 μ g genomic DNA from 1 g fresh blades were obtained. After genomic DNA was isolated from fresh material, the quality was checked by agarose gel. Quantification of DNA concentration was performed using UV spectrophotometric measurement of the A 260/ A 280 ratio. The DNA was suitable for PCR, cloning, and hybridization. The DNA isolated using this method allowed successful construction of microsatellite libraries for Caulerpa species and S. muticum . The technique is inexpensive and appropriate for the isolation of multiple samples of DNA from a small amount of fresh material.  相似文献

Measuring qualitative traits of plant tissue is important to understand how plants respond to environmental change and biotic interactions. Near infrared reflectance spectrometry (NIRS) is a cost‐, time‐, and sample‐effective method of measuring chemical components in organic samples commonly used in the agricultural and pharmaceutical industries. To assess the applicability of NIRS to measure the ecologically important tissue traits of carbon, nitrogen, and phlorotannins (secondary metabolites) in brown algae, we developed NIRS calibration models for these constituents in dried Sargassum flavicans (F. K. Mertens) C. Agardh tissue. We then tested if the developed NIRS models could detect changes in the tissue composition of S. flavicans induced by experimental manipulation of temperature and nutrient availability. To develop the NIRS models, we used partial least squares regression to determine the statistical relationship between trait values determined in laboratory assays and the NIRS spectral data of S. flavicans calibration samples. Models with high predictive power were developed for all three constituents that successfully detected changes in carbon, nitrogen, and phlorotannin content in the experimentally manipulated S. flavicans tissue. Phlorotannin content in S. flavicans was inversely related to nitrogen availability, and nitrogen, temperature, and tissue age interacted to significantly affect phlorotannin content, demonstrating the importance of studies that investigate these three variables simultaneously. Given the speed of analysis, accuracy, small tissue requirements, and ability to measure multiple traits simultaneously without consuming the sample tissue, NIRS is a valuable alternative to traditional methods for determining algal tissue traits, especially in studies where tissue is limited.  相似文献

Immunostimulants are valuable for control of shrimp diseases and the immunostimulatory effects of some polysaccharide additives for shrimp have been reported. In this study, the Sargassum fusiforme polysaccharide extract (SFPSE) was assessed as a feed additive when supplemented in the diet (0%, 0.5%, 1.0%, and 2.0%) for juvenile shrimp, Fenneropenaeus chinensis, in order to study the effects of SFPSE on vibriosis resistance and immune activity. Shrimp were cultured in the same pond with cages. The body weight, survival, the cumulative mortality after injection with Vibrio harveyi (30 microl V. harveyi suspension at 9.3 x 10(7) CFU ml(-1) per shrimp), the total haemocyte counts (THCs), the protein concentration and the phenoloxidase (PO) activity in supernatant of haemolymph, the lysozyme (LSZ) and superoxide dismutase (SOD) activity in muscle of the shrimp were assayed after 14 days feeding period. The results indicated that shrimp survival under the stress of V. harveyi was affected by the dietary SFPSE. The shrimp treated with 1.0% and 0.5% SFPSE displayed significantly lower cumulative mortalities after being injected with V. harveyi suspension 24 and 30 h later, respectively, compared with that of the control. However, cumulative mortality of 2.0% SFPSE treatment was not significantly different from that of the control. There was no significant difference of cumulative mortality between 0.5% and 1.0% SFPSE treatment groups. The immune activities of the shrimp also were affected by dosage of dietary SFPSE. The THCs of the shrimp rose with increasing SFPSE dosage. The protein concentration and PO activity in supernatant of haemolymph as well as muscular LSZ activity first rose then dropped with increasing SFPSE dosage. The protein concentration in supernatant of haemolymph appeared a maximum of 167.46 mg ml(-1) in 1.0% SFPSE treatment. The PO activity and LSZ activity reached the peaks as 13.20 U and 3.21 U mgprot(-1) in 0.5% SFPSE treatment, respectively. SOD activity of the shrimp was not significantly affected by dietary SFPSE. It is therefore suggested that oral administration of SFPSE at an optimal level of 0.5% and 1.0% for 14 days effectively improved vibriosis resistance and enhanced immune activity of the shrimp in general.  相似文献

In the present study, we evaluated the allelopathic effects of three macroalgae, namely Ulva pertusa Kjellml, Corallina pilufifera Postl et Ruprl, and Sargassum thunbergii Mertl O. Kuntze, on the growth of the microalga Skeletonema costaturn (Grev.) Creve using culture systems in which the algae coexisted. The effects of the macroalgal culture medium filtrate on S. costatum were also investigated. Moreover, isolated co-culture systems were built to confirm the existence of allelochemicals and preclude growth inhibition by direct contact. The coexistence assay data demonstrated that the growth of S. costaturn was strongly inhibited when fresh tissues, dry powder and aqueous extracts were used; the allelochemicals were lethal to S. costatum at relatively higher concentrations. The effects of the macroalgal culture medium filtrate on the microalga showed both species specificity and complexity. The inhibitory effect of fresh macroalgal tissue and culture medium filtrate on the microalga was due to the alleochemicals released by the macroalgae. The results of the present study show that the allelopathic effects of macroalgae on the microalga are complex. The present study could shed light onto the basis of the interaction between macro- and microalgae.  相似文献

The occurrence and toxicity of Amphidinium carterae Hulburt is hereby reported for the first time from the North Arabian Sea on the coast of Pakistan. The concentrations of 1.2 × 104 cells ml−1 were found in intertidal pools that were also inhabited by the brown macroalga Sargassum wightii. Both wild and cultured A. carterae cells were tested for ciguatera toxicity through exposure to brine shrimp nauplii (Artemia salina) and albino mice. Although the brine shrimp did not appear to be affected mortalities in mice ranged between 13 and 16% at doses of 7.2 × 104 and 2.5 × 105 cells ml−1, respectively. When mice were affected pharmacological effects such as muscle contraction in lower back area, increased respiration, immobility and paralysis in hind limbs were observed for 2 h. These effects appeared to be reversible and gradually disappeared within 24 h.  相似文献

Sargassum muticum (Yendo) Fensholt is an invasive species that is firmly established on intertidal and subtidal rocky shores of Europe and the Pacific coast of North America. Local success and spread of S. muticum is thought to rely on its reproductive potential that seems dependent on exogenous factors like tidal and lunar cycles. This study is the first to compare the reproductive patterns (periodicity of egg expulsion and embryo settlement) of this invader in two different habitats: the middle and low intertidal. The combination of monthly, daily, and tidal samples at triplicate sites within each habitat showed a semilunar periodicity of egg expulsion and embryo settlement coincident with increasing tidal amplitude just before full and new moons. In both habitats, duration of each egg expulsion event was ~1 week, and embryo settlement occurred during the first daily low tide and with the incoming high tide during spring tides. However, both expulsion and settlement started 1–2 d earlier, expulsion saturation was faster, and settlement was higher in the mid‐ compared to the low intertidal. Our results suggest that the exact timing of gamete expulsion and embryo release of S. muticum responds to local factors, including tidal cues, which result in differences between mid‐ and low‐intertidal habitats.  相似文献

Sargassum muticum (Yendo) Fensholt is one of the most well‐known invasive species in the world. There have, however, been few genetic investigations on both its introduced and native populations. There are also some questions about the taxonomic status of this species. This study is the first to assess the genetic diversity of S. muticum on a global scale, by utilizing one marker each from the extranuclear genomes, namely, plastidial RUBISCO and mitochondrial TrnW_I spacers, as well as the nuclear internal transcribed spacer 2 (ITS2). Based on the markers investigated, both the invasive as well as the native populations of this species appeared very homogenous, when compared with other invasive and brown macroalgae. No variation in ITS2 and RUBISCO spacer was revealed in S. muticum populations, including those from its native ranges in Asia and the introduced ranges in Europe and North America. Two TrnW_I spacer haplotypes with a fixed two‐nucleotide difference were found between the populations of eastern Japan and the other 15 populations examined. This study confirms that there is no cryptic diversity in the introduced range of this species. All the materials collected globally are indeed S. muticum. Results depicting the distribution range of the two TrnW_I spacer haplotypes also support the earlier suggestion that the source of the introduced S. muticum populations is most likely western and central Japan (Seto Inland Sea), where the germlings of S. muticum were likely to have been transported with the Pacific oysters previously introduced for farming in Canada, UK, and France in earlier years.  相似文献

A new species, Sargassum boreale Yoshida et Horiguchi is described. It belongs to the subgenus Bactrophycus section Teretia, with cylindrical receptacles and is distinct from Sargassum confusum C. Agardh, S. pallidum (Turner) C. Agardh and Sargassum microceratium (Turner) C. Agardh in having a rather elongated stem with smooth surface and distantly issuing main branches, with narrow leaves. The distinction between S. boreale and these species is also revealed by a difference in internal transcribed spacer 2 (ITS‐2) sequences. In addition to the base substitutions, the existence of a large gap in S. boreale distinguishes this species from others. Sargassum boreale is distributed around Hokkaido and Saghalien to 50°N latitude. A key to the species of section Teretia is provided.  相似文献
