Sargassum muticum is important in maintaining the structure and function of littoral ecosystems, and is used in aquaculture and alginate production, however, little is known about its population genetic attributes. In this study, random amplified polymorphic DNA (RAPD) and inter-simple sequence repeat (ISSR) markers were used to investigate the genetic structure of four populations of S. muticum and one outgroup of S. fusiforme (Harv.) Setchell from Shandong peninsula of China. The selected 24 RAPD primers and 19 ISSR primers amplified 164 loci and 122 loci, respectively. Estimates of genetic diversity with different indicators (P%, percentage of polymorphic loci; H, the expected heterozygosity; I, Shannon’s information index) revealed low or moderate level of genetic variations within each S. muticum population, and a high level of genetic differentiations were determined with pairwise unbiased genetic distance (D) and fixation index (F ST ) among the populations. The Mantel test showed that two types of matrices of D and F ST were highly correlated whether from RAPD (r = 0.9706, P = 0.009) or ISSR data (r = 0.9161, P = 0.009). Analysis of molecular variance (AMOVA) was conducted to apportion the variations among and within the S. muticum populations. It indicated that variations among populations were higher than those within populations, being 55.82% verse 44.18% by RAPD and 55.21% verse 44.79% by ISSR, respectively. Furthermore, the Mantel test suggested that genetic differentiations among populations were related to the geographical distances (r > 0.6), namely, conformed to the IBD (isolation by distance) model, as expected from UPGMA (unweighted pair group method with arithmetic averages) cluster analysis. On the whole, the high genetic structuring among the four S. muticum populations along the distant locations was clearly indicated in RAPD and ISSR analyses (r > 0.9, P < 0.05) in our study.  相似文献

Mechanisms of biosorption of different heavy metals by brown marine macroalgae   总被引:11,自引:0,他引:11  
The biosorption mechanisms of different heavy metallic cations (Cd, Ni, Pb) to active chemical groups on the cell wall matrix of the nonliving brown marine macroalga, Sargassum vulgaris in its natural form, were examined by the following instrumental and chemical techniques: Fourier-transform infrared (FTIR) analysis, X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM), and extraction of alginic acid and sulfated polysaccharides, which act as metal-binding moieties present in cell wall. From the different techniques used and the known chemical composition of the algal cell wall, it was observed that biosorption of the metallic cations to the algal cell wall component was a surface process. The binding capacities of the different metal cations were between 1 and 1.2 mmol metal/g on a dry weight basis. The main chemical groups involved in the metallic cation biosorption were apparently carboxyl, amino, sulfhydryl, and sulfonate. These groups were part of the algal cell wall structural polymers, namely, polysaccharides (alginic acid, sulfated polysaccharides), proteins, and peptidoglycans. The main cadmium cation sequestration mechanism by the algal biomass was apparently chelation, while the nickel cation sequestration mechanism was mainly ion exchange. Lead cations exhibit higher affinity to the algal biomass, and their binding mechanism included a combination of ion exchange, chelation, and reduction reactions, accompanied by metallic lead precipitation on the cell wall matrix. During the ion exchange process, calcium, magnesium, hydrogen cations, and probably other cations (sodium and potassium) in the algal cell wall matrix were replaced by the tested heavy metals.  相似文献

This innovative study investigates 18 Turbinaria and Sargassum brown seaweed samples from three archipelagos of the South West Pacific Ocean. The phenolic content of crude and size‐fractionated extracts was determined by the Folin‐Ciocalteu method; fucoxanthin was measured using high performance liquid chromatography. Although the phenolic content proved to be low for the species of both genera tested, the levels in the species of Turbinaria were higher than those of Sargassum tested, except in the Fiji islands where both species produced similar contents. These investigations also highlighted variations of total phenolic content with spatial, reef morphology and depth. A large number of small phenolic compounds (<2000 Da) were observed irrespective of the genus analyzed. Nevertheless, the composition of the phenolic pool varied in relation with the genus, the archipelago from which the material was collected, geomorphological features and the depth of each collection field site. A similar observation was for fucoxanthin content and in general, the measured constituents of the Turbinaria were less than those of the Sargassum species. The results are discussed in terms of inter‐ and intra‐specific variability. Variations in both phenolic and fucoxanthin content were noted in tropical members of the Sargassaceae; such differences could be a result of specific chemical defense mechanisms adopted by members of each genus.  相似文献

The construction of artificial seaweed beds in the intertidal zone is a challenge due to extreme levels of physical stress. In order to provide a basis for the construction using the dispersal of microscopic juveniles, a three-way factorial experimental design was used to evaluate the tolerance of Sargassum thunbergii germlings shortly released from fertile thalli to temperature, salinity and desiccation in this study. Results revealed that temperature, salinity and desiccation significantly affected the growth and survival of germlings. Germlings showed rapid growth with relative growth rate (RGR, % day−1) over 16% when cultured at 25 °C and full immersion in normal seawater. Although growths of germlings subjected to moderate conditions were significantly inhibited, RGRs over 13% were obtained. The RGRs of germlings below 10% were observed only at 35 °C and 9 h desiccation treatments. In comparison to growth, survival was less affected by physical stress. Germlings showed low mortalities below 10% under appropriate conditions (25 °C and 30 °C combined with full immersion), and below 60% under moderate conditions, by the end of experiment. However, the mortality rates increased to over 90% under extreme conditions (9 h desiccation and 35 °C combined with full immersion in salinity of 12). These results showed that S. thunbergii germlings had high tolerance to physical stresses. In addition to the main effects, both two-way and three-way interactions between temperature, salinity and desiccation were significant. Based on the magnitude of effect, desiccation was the predominant factor affecting both growth and survival. According to the results, construction of artificial tanks in natural habitat to minimize desiccation may be an effective strategy for S. thunbergii restoration using germlings.  相似文献

We developed a clonal culture of Sargassum horneri to investigate the effect of photoperiod on reproduction in this species. Regenerated vegetative thalli were obtained using lateral branches excised from a thallus grown from a single embryo under short‐day conditions (SD = 10:14 h light : dark cycle). Lateral branches excised from the SD‐regenerated thallus became vegetative thalli that remained in that phase as long as they were cultured under SD. When an excised lateral branch was cultured under long‐day conditions (LD = 14:10 h light : dark cycle), it began to enter the reproductive phase while still less than 50 mm long. Induction of the reproductive phase was accompanied by a distinctive morphological change – suppression of blade formation at the apical region of the branch; elongation of branches without blades was then followed by differentiation of receptacles bearing conceptacles on their surface. Apices of receptacles were able to interconvert between reproductive and vegetative phases, as blades resprouted upon transfer from LD to SD. The critical day length for induction of receptacle formation was between 13 and 14 h; receptacle formation was also induced under SD conditions with night breaks (NBs). These results strongly suggest that reproductive regulation of S. horneri is a photoperiodic long‐day response. NBs with blue and green light were effective for reproductive induction but not with red light. This suggests that blue‐ and/or green‐light photoreceptors are involved in the photoperiodic reproductive response of S. horneri.  相似文献

Fucoxanthin (Fx) and fucosterol (Fs) are characteristic lipid components of brown seaweeds that afford several health benefits to humans. This article describes the quantitative evaluation of lipids of 15 species of brown seaweeds with specific reference to Fx, Fs, and functional long‐chain omega‐6/omega‐3 polyunsaturated fatty acids (PUFAs). In addition, fatty‐acid composition of selected species was also accomplished in the study. Major omega‐3 PUFAs in the brown seaweeds analyzed were α‐linolenic acid (18:3n‐3), octadecatetraenoic acid (18:4n‐3), arachidonic acid (20:4n‐6), and eicosapentaenoic acid (20:5n‐3). Both Fx (mg · g?1 dry weight [dwt]) and Fs (mg · g?1 dwt) were determined to be relatively abundant in Sargassum horneri (Turner) C. Agardh (Fx, 3.7 ± 1.6; Fs, 13.4 ± 4.4) and Cystoseira hakodatensis (Yendo) Fensholt (Fx, 2.4 ± 0.9; Fs, 8.9 ± 2.0), as compared with other brown seaweed species. Studies related to seasonal variation in Fx, Fs, and total lipids of six brown algae [S. horneri, C. hakodatensis, Sargassum fusiforme (Harv.) Setch., Sargassum thunbergii (Mertens ex Roth) Kuntze, Analipus japonicus (Harv.) M. J. Wynne, and Melanosiphon intestinalis (D. A. Saunders) M. J. Wynne] indicated that these functional lipid components reached maximum during the period between January and March. The functional lipid components present in these seaweeds have the potential for application as nutraceuticals and novel functional ingredients after their recovery.  相似文献

A sulphated polysaccharide (SP-2a) from the brown alga Sargassum patens (Kütz.) Agardh (Sargassaceae) was found to significantly inhibit the in vitro replication of both the acyclovir (ACV)-sensitive and -resistant strains of Herpes simplex virus type 1 (HSV-1), in dose-dependent manners, with 50% inhibitions occurring with 1.5–5.3 μg/ml of the polysaccharide. SP-2a exhibited extracellular virucidal activity only against the ACV-sensitive strains, but not the resistant strain, at the concentration of 100 μg/ml. The strongest antiviral activities against the different strains of HSV-1 were observed when this polysaccharide was present during and after adsorption of the virus to host cells. The inhibitory effect of SP-2a on virus adsorption occurred dose-dependently in all the HSV-1 strains tested, and the adsorption of the ACV-resistant DM2.1 strain was reduced by 81.9% (relative to control) with 4 μg/ml of the polysaccharide. This study clearly demonstrated that the antiviral mode of action of SP-2a is mediated mainly by inhibiting virus attachment to host cells, and this sulphated polysaccharide might have different modes of action against the ACV-sensitive and -resistant strains of HSV-1.  相似文献

以枸杞岛铜藻垂直分布特征为基础,并通过现场实验分析了环境因子(沉积物、暴波强度(wave exposure)、光照强度)对铜藻幼苗阶段垂直分布及生长的影响。结果表明:铜藻的主要繁殖群体集中分布在潮间带低潮区121—240 cm的水深范围内,水深大于480 cm没有发现成藻植株;光照对铜藻的生长起重要作用,低光照强度可能不会导致铜藻的大量死亡,但明显抑制了铜藻的生长,248μmol m-2s-1的光照强度为铜藻快速生长提供了有利条件,而并没有导致铜藻幼苗大量死亡;低暴波强度虽降低了铜藻被移除的风险,但为沉积物和附生生物在铜藻附着基上的积累创造了条件,进而影响了铜藻早期定居阶段的存活;随着水深的增加,沉积物在附着基上的沉积水平显著增多(P0.01),沉积物长时间的覆盖导致了铜藻幼苗大量死亡。因此说,沉积物可能是影响铜藻垂直分布格局的主要因子。  相似文献

为探究铜藻多糖(Sargassum horneri polysaccharides, SHP)对H2O2诱导的人角质形成细胞(HaCaT)氧化应激损伤的保护作用,测定了SHP对总抗氧化能力(T-AOC)、DPPH自由基、羟自由基(·OH)和超氧阴离子自由基(O2·-)的清除作用,以评价SHP的体外抗氧化能力,并建立H2O2诱导HaCaT细胞氧化损伤模型;通过测定细胞存活率、细胞活性氧以及酶活,评价SHP对HaCaT细胞氧化损伤的保护作用。结果表明,当SHP为1 mg/mL时,DPPH的清除率为68%、·OH清除能力65.48 U/mL;在SHP为3 mg/mL时,O2·-清除能力为84.86 U/mL,T-AOC为33.55。SHP能显著提高H2O2诱导氧化损伤的HaCaT细胞活力,其中经100μg/mL SHP处理后,HaCaT细胞存活率由56.85%提高到80.57...  相似文献
