Exercise, specifically voluntary wheel running, is a potent stimulator of hippocampal neurogenesis in adult mice. In addition, exercise induces behavioral changes in numerous measures of anxiety in rodents. However, the physiological underpinnings of these changes are poorly understood. To investigate the role of neurogenesis in exercise-mediated anxiety, we examined the cellular and behavioral effects of voluntary wheel running in mice with a reduction in hippocampal neurogenesis, achieved through conditional deletion of ataxia telangiectasia-mutated and rad-3-related protein (ATR), a cell cycle checkpoint kinase necessary for normal levels of neurogenesis. Following hippocampal microinjection of an adeno-associated virus expressing Cre recombinase to delete ATR, mice were exposed to 4 weeks of voluntary wheel running and subsequently evaluated for anxiety-like behavior. Wheel running resulted in increased cell proliferation and neurogenesis, as measured by bromodeoxyuridine and doublecortin, respectively. Wheel running also resulted in heightened anxiety in the novelty-induced hypophagia, open field and light-dark box tests. However, both the neurogenic and anxiogenic effects of wheel running were attenuated following hippocampal ATR deletion, suggesting that increased neurogenesis is an important mediator of exercise-induced anxiety. 相似文献

High and Low Activity strains of mice (displaying low and high anxiety-like behavior, respectively) with 7.8–20 fold differences in open-field activity were selected and subsequently inbred to use as a genetic model for studying anxiety-like behavior in mice (DeFries et al., 1978, Behavior Genetics, 8:3-13). These strains exhibited differences in other anxiety-related behaviors as assessed using the light–dark box, elevated plus-maze, mirror chamber, and elevated square-maze tests (Henderson et al., 2004, Behavior Genetics, 34: 267-293). The purpose of these experiments was three-fold. First, we repeated a 6-day behavioral battery using updated equipment and software to confirm the extreme differences in anxiety-like behaviors. Second, we tested novel object exploration, a measure of anxiety-like behavior that does not rely heavily on locomotion. Third, we conducted a home cage wheel running experiment to determine whether these strains differ in locomotor activity in a familiar, home cage environment. Our behavioral test battery confirmed extreme differences in multiple measures of anxiety-like behaviors. Furthermore, the novel object test demonstrated that the High Activity mice exhibited decreased anxiety-like behaviors (increased nose pokes) compared to Low Activity mice. Finally, male Low Activity mice ran nearly twice as far each day on running wheels compared to High Activity mice, while female High and Low Activity mice did not differ in wheel running. These results support the idea that the behavioral differences between High and Low Activity mice are likely to be due to anxiety-related factors and not simply generalized differences in locomotor activity. 相似文献

The risk of cardiovascular disease (CVD) increases in post-menopausal women, yet, the role of exercise, as a preventative measure for CVD risk in post-menopausal women has not been adequately studied. Accordingly, we investigated the impact of voluntary cage-wheel exercise and forced treadmill exercise on cardiac adaptation in menopausal mice. The most commonly used inducible model for mimicking menopause in women is the ovariectomized (OVX) rodent. However, the OVX model has a few dissimilarities from menopause in humans. In this study, we administered 4-vinylcyclohexene diepoxide (VCD) to female mice, which accelerates ovarian failure as an alternative menopause model to study the impact of exercise in menopausal mice. VCD selectively accelerates the loss of primary and primordial follicles resulting in an endocrine state that closely mimics the natural progression from pre- to peri- to post-menopause in humans. To determine the impact of exercise on exercise capacity and cardiac adaptation in VCD-treated female mice, two methods were used. First, we exposed a group of VCD-treated and untreated mice to a voluntary cage wheel. During running, muscles of the lower limb act like a linear spring bouncing on the ground. When approaching an obstacle, the overall stiffness of this leg-spring system (kleg) is modified during the two steps preceding the jump to enhance the movement of the center of mass of the body while leaping the obstacle. The aim of the present study is to understand how kleg is modified during the running steps preceding the jump. Since kleg depends on the joint torsional stiffness and on the leg geometry, we analyzed the changes in these two parameters in eight subjects approaching and leaping a 0.65 m-high barrier at 15 km h−1. Ground reaction force (F) was measured during 5–6 steps preceding the obstacle using force platform and the lower limb movements were recorded by camera. From these data, the net muscular moment (Mj), the angular displacement (θj) and the lever arm of F were evaluated at the hip, knee and ankle. At the level of the hip, the Mj–θj relation shows that muscles are not acting like torsional springs. At the level of the knee and ankle, the Mj–θj relation shows that muscles are acting like torsional springs: as compared to steady-state running, the torsional stiffness kj decreases from ~1/3 two contacts before the obstacle, and increases from ~2/3 during the last contact. Brown adipose tissue (BAT) is a major site of nonshivering thermogenesis (NST) during cold acclimation for most mammals. Repetitive nonthermal stress such as immobilization has been shown to enhance the capacity of NST as cold acclimation. In the present study, the effects of running training, another type of nonthermal stress, were investigated on in vitro thermogenesis and the cellularity of interscapular BAT in rats. The rats were subjected to treadmill running for 30 min daily at 30 m/min under 8° inclination for 4–5 weeks. In vitro thermogenesis was then measured in minced tissue blocks incubated in a Krebs-Ringer phosphate buffer containing glucose and albumin at 37° C, using a Clark type oxygen electrode. The trained rats showed less body weight gain during the experiment. The weights of BAT and epididymal white adipose tissue were smaller in the trained rats. Noradrenaline- and glucagon-stimulated oxygen consumption were also significantly smaller in the trained rats. The tissue DNA level was greater in the trained rats, but the DNA content per tissue pad did not significantly differ. The results indicate that running training reduces BAT thermogenesis, possibly as an adaptation to conserve energy substrates for physical work. 相似文献

This study aimed to determine the minimum time required for assessing spatiotemporal variability during continuous running at different submaximal velocities and, thereby, the number of steps required. Nineteen trained endurance runners performed an incremental running protocol, with a 3-min recording period at 10, 12, 14 and 16 km/h. Spatiotemporal parameters (contact and flight times, step length and step frequency) were measured using the OptoGait system and step variability was considered for each parameter, in terms of within-participants standard deviation (SD) and coefficient of variation (CV%). Step variability was considered over six different durations at every velocity tested: 0–10 s, 0–20 s, 0–30 s, 0–60 s, 0–120 s and 0–180 s. The repeated measures ANOVA revealed no significant differences in the magnitude of the four spatiotemporal parameters between the recording intervals at each running velocity tested (p ≥ 0.05, ICC > 0.90). The post-hoc analysis confirmed no significant differences in step variability (SD and CV% of each spatiotemporal parameter at any velocity tested) between measurements. The Bland-Altman limits of agreement method showed that longer recording intervals yield smaller systematic bias, random errors, and narrower limits of agreement, regardless of running velocity. The results suggest that the duration of the recording period required to estimate spatiotemporal variability plays an important role in the accuracy of the measurement, regardless of running velocity (10–16 km/h). 相似文献

We sampled benthic macroinvertebrates and recorded oxygen and temperature regime during 2-day periods in 12 stream sites at
an altitude of 3,800–4,000 m a.s.l. on the Bolivian Altiplano, during low flow conditions at the end of the dry season. Eight
of the sites were relatively unpolluted, while the remaining four sites were affected by domestic/industrial sewage. Compared
to other Andean streams, the fauna was poor with a total of 28 and a mean of 11 taxa (mostly families) in the unpolluted sites.
The entire EPT group was represented by just five families. Of these, only Baetidae and Hydroptilidae were common. At all
sites, the dominant taxa were found among just four taxa (Elmidae, Chironomidae, Oligocheata and Hyallelidae). Mean water
temperature was 12.9°C, while mean diel amplitude was 13.4°C and the maximum range 17.4°C. Ten richness and biotic indices
were used to analyse for effects of temperature and oxygen on the fauna. Most measures of fauna richness were negatively correlated
with mean and max temperature (even excluding the four polluted sites), while biotic indices were mostly uncorrelated with
temperature. Thus, the large fluctuations in temperature seemed to exclude taxa, thereby reducing overall diversity of Altiplano
streams. Oxygen saturation also varied considerably, with a mean diel range of 48% and a maximum range of 93%. Richness measures
were uncorrelated with oxygen %, while all biotic indices were positively correlated with either mean or min oxygen %. Most
measures of faunal composition showed a marked shift at levels of 10–30% min oxygen saturation. Earlier studies of high Andean
streams in Ecuador have shown a major shift in faunal composition at 50–60% oxygen saturation. The fauna in the Altiplano
streams may be adapted to the large fluctuations in oxygen saturation, and therefore more robust towards low oxygen saturations.
The implications for biotic assessment of Altiplano streams are discussed. 相似文献

It was the purpose of this investigation to: 1) compare the ratings of perceived exertion (RPEs) in high and low fit individuals when walking and running at comparable exercise intensities and 2) to determine if ventilation (VE) provides a central signal for RPEs. Nine high fit and nine low fit male subjects completed two exercise bouts on a treadmill, one uphill walking and the other level running. Workloads for each bout were set at 90% of each subject's ventilatory threshold (VT) as determined from a graded exercise test. Oxygen consumption (Vo2), heart rate (HR), and VE were all similar between the walk and run trials for the low fit subjects (P greater than 0.05). HR were found to be significantly greater during the walk trial vs. the run trial (P less than 0.05) for the high fit subjects, whereas, VE was significantly greater during the run trial. Oxygen consumption was similar for the high fit subjects during both trials (P greater than 0.05). During the walk and run trials, central (12.1 +/- 1.6 vs. 11.4 +/- 1.5), local (14.0 +/- 1.3 vs. 13.9 +/- 1.1) and overall (12.8 +/- 1.2 vs. 12.4 +/- 1.4) RPEs were not found to be significantly different for the low fit group (P greater than 0.05). In contrast, during the walk vs. the run trial there was a significant increase in central (10.7 +/- 2.0 vs. 9.2 +/- 1.9), local (11.5 +/- 2.0 vs. 9.8 +/- 1.8) and overall (11.2 +/- 2.4 vs. 9.6 +/- 2.3) RPEs for the high fit group (P less than 0.05).(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献
