Primary structure effects on peptide group hydrogen exchange

The rate of exchange of peptide group NH hydrogens with the hydrogens of aqueous solvent is sensitive to neighboring side chains. To evaluate the effects of protein side chains, all 20 naturally occurring amino acids were studied using dipeptide models. Both inductive and steric blocking effects are apparent. The additivity of nearest-neighbor blocking and inductive effects was tested in oligo-and polypeptides and, suprisingly, confirmed. Reference rates for alanine-containing peptides were determined and effects of temperature considered. These results provide the information necessary to evaluate measured protein NH to ND exchange rates by comparing them with rates to be expected for the same amino acid sequence is unstructured aligo- and polypeptides. Improved pulse sequences for measuring coupling constants in 13C, 15N-labeled proteins

Summary NMR pulse sequences for measuring coupling constants in ¹³C, ¹⁵N-labeled proteins are presented. These pulse sequences represent improvements over earlier experiments with respect to resolution and number of radiofrequency pulses. The experiments are useful for measuring J_NH , J_NCO, J_NC , J_H ^N _CO and J_H ^N _H . Applications to chymotrypsin inhibitor 2 (CI-2) are shown.     

3D 13C/1H NMR-based assignments for side-chain resonances of Lactobacillus casei dihydrofolate reductase. Evidence for similarities between the solution and crystal structures of the enzyme

Summary ¹³C-based three-dimensional ¹H–¹H correlation experiments have been used to determine essentially complete ¹³C and ¹H resonance assignments for the amino acid side chains of uniformly ¹³C/¹⁵N labelled L. casei dihydrofolate reductase in a complex with the drug methotrexate. Excellent agreement is observed between these assignments and an earlier set of partial assignments made on the basis of correlating nuclear Overhauser effect and crystal structure data, indicating that the tertiary structure of the enzyme is similar in solution and in the crystal state.To whom correspondence should be addressed.     

Nutritional properties of four marine microalgae for albino rats

The nutritive value of the marine microalgaeTetraselmis suecica, Isochrysis galbana, Dunaliella tertiolecta andChlorella stigmatophora was studied in diets given to rats. Biological assays were carried out in order to determine the Protein Efficiency Ratio (PER) and the Food Conversion Efficiency (FCE). Each dried microalga was fed to weaning Wistar albino rats as the sole protein source at a protein level of 12%. Control rats were given diets containing 12 % casein. Food consumption was similar in all groups. PER values obtained were 1.14 withT. suecica diet, 1.13 withI. galbana diet, 2.07 withD. tertiolecta diet and 1.13 withC. stigmatophora diet (casein, 2.50). FCE values followed a similar pattern. The data showed that the marine microalgaD. tertiolecta is a source of protein of good quality. Its PER is quite high, compared to vegetable and cereal proteins, and compares favourably with other microbial protein sources, such as yeasts or different freshwater microalgae. Haematological tests showed no significant differences among the groups in haemoglobin levels, red and white blood cell counts, differential count and mean corpuscular volume. Different blood parameters were also determined and a significant decrease in triglycerides levels appeared with all the microalgal diets, whereas a significant decrease in cholesterol appeared inD. tertiolecta andC. stigmatophora diets.     

非洲猪瘟病毒I226R蛋白抑制cGAS-STING通路介导的天然免疫应答

本研究旨在探究非洲猪瘟病毒(African swine fever virus, ASFV) I226R蛋白(I226R protein, pI226R)抑制cGAS-STING信号通路的作用机制。利用双荧光素酶报告系统和实时荧光定量PCR (real-time quantitative PCR, qPCR)证明pI226R显著抑制cGAS-STING通路介导的I型干扰素及干扰素刺激相关基因的产生。免疫共沉淀及激光共聚焦显微镜试验发现pI226R与cGAS蛋白相互作用。免疫印迹分析证明pI226R通过自噬-溶酶体途径促进cGAS蛋白的降解。同时,pI226R阻碍了cGAS与E3泛素连接酶三基序蛋白56 (tripartite motif protein 56, TRIM56)的结合,导致cGAS的单泛素化减弱,从而抑制了cGAS的活化和cGAS-STING通路的激活。总之,本研究证明ASFV pI226R通过拮抗cGAS进而抑制宿主的抗病毒天然免疫反应,进一步增加了对研究ASFV免疫逃逸机制的理解,为疫苗的研发提供了理论基础。     

基于N蛋白单克隆抗体的小反刍兽疫病毒抗体检测胶体金试纸条的研制

本研究旨在建立一种简便、快捷、可直观检测小反刍兽疫病毒(peste des petits ruminants virus,PPRV)抗体的检测方法。将pET-32a-N重组质粒转化至大肠杆菌(Escherichia coli) Rosetta(DE3)感受态细胞中进行诱导表达,以纯化的PPRVN蛋白免疫8周龄BALB/c小鼠,取其脾细胞与SP2/0骨髓瘤细胞进行融合,间接酶联免疫吸附试验(enzyme-linked immunosorbent assays, ELISA)筛选及亚克隆,获得了抗PPRV N蛋白的单克隆抗体。将PPRV N蛋白分别作为金标抗原及检测线(T线)包被抗原、单克隆抗体作为质控线(C线)包被抗体,组装成检测PPRVN蛋白抗体的胶体金免疫层析试纸条。结果显示：成功获得1株能稳定分泌抗N蛋白抗体的杂交瘤细胞株,命名为1F1;间接ELISA检测1F1腹水效价为1:128000;亚类鉴定结果为IgG1,轻链为kappa链。Westernblotting结果显示,1F1能与PPRV N蛋白特异性结合;间接免疫荧光(indirect immunofluorescent ass...     

儿童最大有氧活动能力的发展特征

核纤层蛋白B1通过影响端粒酶活性调控肝癌细胞生长

核纤层蛋白B1(nuclear lamina protein B1,LMNB1)高表达于肝癌组织中,通过敲低LMNB1探讨其对肝癌细胞增殖的影响及其机制。利用siRNA在肝癌细胞中敲低LMNB1,Western blotting检测敲低效果,使用端粒重复序列扩增法(telomeric repeat amplification protocol assay,TRAP)检测其端粒酶活性变化。利用实时定量聚合酶链反应(quantitative real-time polymerase chain reaction,qPCR)检测其端粒长度变化。并通过CCK-8、克隆形成、Transwell、划痕实验检测其生长,侵袭和迁移能力变化。利用慢病毒系统构建稳定敲低LMNB1的HepG2细胞,检测其端粒长度及端粒酶活性变化,采用SA-β-gal衰老染色检测细胞衰老情况,通过裸鼠皮下成瘤实验及对肿瘤后续的组化染色,SA-β-gal衰老染色,端粒荧光原位杂交(fluorescence in situ hybridization,FISH)检测其对成瘤性的影响。最后利用生物信息分析的方法寻找LMNB1在临床肝癌组织中的表达情况,及其与临床分期、病人生存期的关系。HepG2和Hep3B中敲低LMNB1后端粒酶活性显著降低,细胞增殖、迁移和侵袭能力显著降低,细胞和裸鼠成瘤实验证明稳定敲低LMNB1后端粒酶活性降低的同时端粒长度缩短,细胞发生衰老,此外细胞成瘤性降低,Ki-67表达降低,生物信息分析结果显示,LMNB1高表达于肝癌组织,且与肿瘤分期和患者生存相关。LMNB1在肝癌细胞中过表达,其有望成为评估肝癌患者临床预后的指标和精准治疗的靶点。     

糜子黄、黑种皮遗传及转录组学分析

同源异型域-亮氨酸拉链(homedomain-leucine zipper,HD-Zip)转录因子广泛参与植物的生长发育和抗胁迫过程。该研究通过生物信息学方法对青稞HD-Zip基因家族进行全基因组分析鉴定,并采用qRT-PCR技术分析非生物胁迫下该基因的表达特性,为深入探讨青稞HD-Zip转录因子的生物学功能及其在高原作物抗逆育种中的应用奠定基础。结果表明：(1)成功从青稞基因组中共鉴定出41个HD-Zip基因家族成员,依次命名为i>HvvHD-ZipⅠ-1~Ⅳ-13,且这些基因在7条染色体上呈不均匀分布。(2)理化性质分析发现,HvvHD-Zip蛋白包含197～885个不等的氨基酸残基;分子量范围在19 914.36～94 014.87 Da;亚细胞定位表明HvvHD-Zip蛋白都位于细胞核。(3)根据多序列比对、系统进化、基因结构和保守基序差异将其聚为4个亚家族,各亚家族分类特征与系统聚类结果一致。(4)顺式作用元件预测分析发现,i>HvvHD-Zip基因启动子中含有11种植物激素和胁迫响应元件。(5)qRT-PCR结果显示,HvvHD-Zip Ⅰ、Ⅱ、Ⅳ亚家族基因对各胁迫响应明显;与根组织相比,多数i>HvvHD-Zip基因在叶组织中响应明显(上调或下调);与冷和盐胁迫相比,i>HvvHD-Zip各基因对旱胁迫响应较强。     

鸡冠花种子蛋白质的提纯及分析

鸡冠花种子干燥后用石油醚脱酯,用凯氏定氮法测定蛋白质含量。按Osbern系统分别提取白蛋白、球蛋白、醇溶蛋白、谷蛋白;用Folin一酚试剂法测定各自相对含量,并用单向和双向SDS—PAGE方法分析种子总蛋白和4类不同溶性蛋白质的组成成分及这些组份的热稳定性。实验发现:鸡冠花种子蛋白质含量达26.04％,白蛋白、球蛋白、醇溶蛋白、谷蛋白的相对含量分别为11.5％、72％、4.6％、12％。SDS—PAGE表明;其种子蛋白中20KD球蛋白成份含量最高,其它一些次要组分为63KD、61KD、15KD、13KD白蛋白成份,58KD、37KD、23KD球蛋白成份,39KD、34KD、25KD谷蛋白成份及26KD醇溶蛋白成份,其中58KD由37KD和20KD两亚基经二硫键连接而成,39KD组份由24.5KD和18KD两亚基通过二硫键连接而成,23KD球蛋白组份遇热沉淀。
