本文报道了我国产的5种和14个品种的蔷薇属花卉的染色体数目和核型,结果如下:小花型为二倍体,2n=2x=14,少数为混倍体;中花型为三倍体,2n=3x=21;大花型为四倍体,2n=4x=28。大部分种的核型均由其中部和近中部着丝点染色体组成,少数种具近端着丝点染色体。它们可以区分为3种核型类型,即1A、2A和1B。  相似文献

为了探析磷对刺梨(Rosa roxburghii Tratt.)幼苗生长、养分含量及其相关生理的影响,揭示刺梨适应低磷胁迫的生理机制,该研究采用石英砂和蛭石混合基质培育的方法,分析不同供磷水平对刺梨幼苗生长和根系形态特征、根和叶中的酸性磷酸酶(APase)、硝酸还原酶(NR)和根中的分泌性磷酸酶活性(SAPase)、根中小分子有机酸组分及含量和植株中营养元素含量的影响。结果表明:(1)供磷水平高于或低于45mg·L-1均会抑制刺梨幼苗的生长,明显降低刺梨幼苗生物量和根总长度、总表面积、总体积、平均直径以及总根尖数;在5mg·L-1的低磷水平下,刺梨幼苗的根冠比值、根毛长度和密度最大,随着供磷水平的增加这3个指标明显降低。(2)根和叶片的APase及根的SAPase活性随供磷水平的增大而减弱,根和叶片中NR活性均于45mg·L-1供磷水平时最高,小于或大于45mg·L-1磷水平后NR活性明显减弱;根中的草酸、酒石酸、柠檬酸、苹果酸、乙酸和琥珀酸的含量均随着供磷水平的增大而降低。(3)刺梨幼苗中P、Ca和Mg含量均随着供磷水平的提高而增大,Zn含量随供磷水平的增大而减小;在45mg·L-1供磷水平下刺梨幼苗中的N、P、K、Mn、Cu、B含量最高,降低或提高供磷水平后这些元素的含量都明显降低。研究认为,供磷水平过低或过高对刺梨幼苗的生长和营养元素吸收均有不利影响;根毛长度和密度的增大、叶片和根中APase及根中SAPase的活性增强、根中小分子有机酸含量的增加是刺梨幼苗应对低磷胁迫采取的重要适应策略。  相似文献

The effects of oryzalin, a dinitroaniline herbicide, on chromosome behavior and on cellular microtubules (MTs) were examined by light microscopy and immunogold staining, respectively, in endosperm cells from Haemanthus katherinae Bak. Brief treatments with 1.0·10-8 M oryzalin reduced markedly the migration rate of anaphase chromosomes and 1.0·10-7 M oryzalin stopped migration abruptly. Oryzalin (1.0·10-7 M) depolymerized MTs and prevented the polymerization of new MTs at all stages of the mitotic cycle. The chromosome condensation cycle was unaffected by oryzalin. Endothelial cells from the heart of Xenopus leavis showed no chromosomal or microtubular rearrangements after oryzalin treatment. The inhibition by oryzalin of the polymerization of tubulin isolated from cultured cells of Rosa sp. cv. Paul's scarlet was examined in vitro by turbidimetry, electron microscopy and polymer sedimentation analysis. Oryzalin inhibited the rapid phase of taxol-induced polymerization of rose MTs at 24°C with an apparent inhibition constant (K i ) of 2.59·106 M. Shorter and fewer MTs were formed with increasing oryzalin concentrations, and maximum inhibition of taxol-induced polymerization occurred at approx. 1:1 molar ratios of oryzalin and tubulin. Oryzalin partially depolymerized taxol-stabilized rose MTs. Ligand-binding experiments with [14C]oryzalin demonstrated the formation of a tubulin-oryzalin complex that was time- and pH-dependent. The tubulin-oryzalin interaction (24°C, pH 7.1) had an apparent affinity constant (K app) of 1.19·105 M-1. Oryzalin did not inhibit taxol-induced polymerization of bovinebrain MTs and no appreciable binding of oryzalin to brain tubulin or other proteins was detected. The results demonstrate pharmacological differences between plant and animal tubulins and indicate that the most sensitive mode of action of the dinitroaniline herbicides is the direct poisoning of MT dynamics in cells of higher plants.Abbreviations MT microtubule - SIB sucrose isolation buffer - TO tubulin-oryzalin complex  相似文献

本文对‘香槟’月季(80sachinensis‘Xiangbin’)的组织培养技术和诱导试管开花进行了研究。结果表明:以茎段为外植体能诱导获得无菌苗,适宜的启动培养基为MS+6-BA1.0mg-L-1+IBA0.1mg·L-1,幼芽继代增殖的最佳培养基是MS+6.BA1.0mg·L-1。+IBA0.1~0.2mg·L-1,诱导生根的适宜培养基为1/2MS+NAA0.3mg·L-1,生根率达80.0%。诱导试管开花的适宜培养基为MS+6.BA0.5mg·L-1+NAA0.1mg·L-1最适宜的诱导试管开花的蔗糖含量是30g·L-1;在三角瓶中培养,试管花可以正常开放,在培养瓶中培养花芽不能正常开放;MS培养基中增加2倍磷的含量,可以提高花芽诱导率,为25.O%;诱导试管开花的最适培养条件为温度21℃,光照强度80~100μmol·m-2.s-1,光照时间16h—d-1。  相似文献

A structural study of the water-soluble dextran made by Leuconostoc mesenteroides strain C (NRRL B-1298) was conducted by enzymic degradation and subsequent 13C-NMR analysis of the native dextran and its limit dextrins. The α-l,2-debranching enzyme removed almost all of the branched D-glucose residues, and gave a limit dextrin having a much longer sequence of the internal chain length (degree of linearity: n = 24.5 compared with the value of n = 3.3 for the native dextran). The degree of hydrolysis with debranching enzyme corresponded to the content of α-1,2-linkages determined by chemical methods, which suggested that most of the α-l,2-linkages in the dextran B-1298 constituted branch points of a single D-glucose residue. A synergistic increase of susceptibility of the dextran B-1299 was observed by simultaneous use of debranching enzyme and endodex-tranase. 13C-NMR spectral analysis indicated the similarity of structure of dextran B-1298 to that of B-1396, rather than that of B-1299. Occurrence of α-l,3-linkages in the limit dextrin was supported by a newly visualized chemical shift at 83.7 ppm.  相似文献

It has long been recognised that polyploid species do not always neatly fall into the categories of auto‐ or allopolyploid, leading to the term ‘segmental allopolyploid’ to describe everything in between. The meiotic behaviour of such intermediate species is not fully understood, nor is there consensus as to how to model their inheritance patterns. In this study we used a tetraploid cut rose (Rosa hybrida) population, genotyped using the 68K WagRhSNP array, to construct an ultra‐high‐density linkage map of all homologous chromosomes using methods previously developed for autotetraploids. Using the predicted bivalent configurations in this population we quantified differences in pairing behaviour among and along homologous chromosomes, leading us to correct our estimates of recombination frequency to account for this behaviour. This resulted in the re‐mapping of 25 695 SNP markers across all homologues of the seven rose chromosomes, tailored to the pairing behaviour of each chromosome in each parent. We confirmed the inferred differences in pairing behaviour among chromosomes by examining repulsion‐phase linkage estimates, which also carry information about preferential pairing and recombination. Currently, the closest sequenced relative to rose is Fragaria vesca. Aligning the integrated ultra‐dense rose map with the strawberry genome sequence provided a detailed picture of the synteny, confirming overall co‐linearity but also revealing new genomic rearrangements. Our results suggest that pairing affinities may vary along chromosome arms, which broadens our current understanding of segmental allopolyploidy.  相似文献

The quantity of seeds dispersed is considered one of several means to determine the dispersal effectiveness of an animal. However, there is little consistency in the manner in which quantities are measured or presented. Here, we quantify seed dispersal by white-faced capuchin monkeys (Cebus capucinus) in Santa Rosa National Park, Costa Rica by measuring: degree of frugivory, number of plant species consumed, the number of seeds consumed per unit time, the number of seeds dispersed intact per unit time, and the number of seeds dispersed intact per unit space. Forty-nine percent of C. capucinus diet is composed of the fruit of 39 species, 4 of which constitute 82% of the frugivory. Seventy-four percent of consumed fruits contain seeds that pass intact through the capuchin digestive system. Capuchins pass a mean of 15.7 seeds of a mean of 1.3 species per defecation, and defecate 25.4 times per 12-h day. These numbers are compared with extant data for C. capucinus, and possible reasons for discrepancies among results between studies are discussed. We propose a standardization of quantitative measures of seed dispersal so that quantifications of seed dispersal can be compared within species, and eventually across species.  相似文献

Neural crest (NC) cells may be involved in kidney organogenesis by providing inductive signals and contributing to cells of the renal stroma. We show here that the lumbo-sacral NC cells fate mapped with the aid of Wnt-1 promoter in the mouse migrate close to the metanephros at the initiation of organogenesis but these cells remain superficial to the condensed Pax2-expressing mesenchymal cells. NC-derived cells enter later into the kidney proper from the midline region. The NC cells contribute also to development of the extra-adrenal para-aortic bodies, Zuckerkandl's bodies and the nerve cord of the sympathetic nervous system. Splotch (Sp2H/Sp2H) embryos, having a NC defect in the lumbo-sacral region, develop a normal metanephros even though the kidney does not express the NC markers Sox10, Phox2b and tyrosine hydroxylase. Consistent with the histological findings, the kidneys of Sp2H/Sp2H embryos also express the stromal genes Foxd1, Hoxa10 and RARβ normally. Wnt-1 promoter-marked wild-type LacZ NC cells migrate intensely from the heterologous inducer tissue of the embryonic dorsal spinal cord (SPC) to the kidney mesenchyme, but tubule induction does not depend on NC migration, since the Sp2H/Sp2H SPC also induces tubulogenesis. The Sp2H/Sp2H mesenchyme also remains competent for tubulogenesis. We conclude that the NC cells fate mapped with the aid of Wnt-1 promoter migrate to the close to the metanephros and form later derivatives integrating with the kidney, but they may not be essential to the development of the stromal cells nor they may provide critical morphogenetic signals to regulate early kidney development in vivo.  相似文献

分析了蔷薇(Rosa L.)种子休眠原因、解除休眠方法以及环境条件对休眠与萌发的影响.蔷薇种子休眠的主要原因有瘦果果皮和种皮的限制作用,胚生理休眠以及果肉、瘦果果皮、种皮和胚中的抑制物质.解除休眠的方法包括去除瘦果果皮限制、解除胚的生理休眠、去除抑制物质等.种子发育过程中及成熟后,环境因子,如温度、水分和光照,对种子休眠和萌发有影响.此外,微生物、果实采集时间也对种子休眠及萌发有较大影响.蔷薇种子的休眠机制复杂,且种间差异很大.  相似文献
