The suborder Notothenioidei dominates the Antarctic ichthyofauna. The non-Antarctic monotypic family Pseudaphritidae is one of the most primitive families. The characterization of the oxygen-transport system of euryhaline Pseudaphritis urvillii is herewith reported. Similar to most Antarctic notothenioids, this temperate species has a single major hemoglobin (Hb 1, over 95% of the total). Hb 1 has strong Bohr and Root effects. It shows two very uncommon features in oxygen binding: At high pH values, the oxygen affinity is exceptionally high compared to other notothenioids, and subunit cooperativity is modulated by pH in an unusual way, namely the curve of the Hill coefficient is bell-shaped, with values approaching 1 at both extremes of pH. Molecular modeling, electronic absorption and resonance Raman spectra have been used to characterize the heme environment of Hb 1 in an attempt to explain these features, particularly in view of some potentially important nonconservative replacements found in the primary structure. Compared to human HbA, no major changes were found in the structure of the proximal cavity of the alpha-chain of Hb 1, although an altered distal histidyl and heme position was identified in the models of the beta-chain, possibly facilitated by a more open heme pocket due to reduced steric constraints on the vinyl substituent groups. This conformation may lead to the hemichrome form identified by spectroscopy in the Met state, which likely fulfils a potentially important physiological role. 相似文献

In common bean (Phaseolus vulgaris L.), Fusarium root rot (caused by Fusarium solani f. sp. phaseoli) disease severity is increased by environmental factors that stress the plant. The current study used reciprocal grafting
techniques with the resistant cultivar FR266 and the susceptible cultivar Montcalm to determine if the genetic control of
resistance is conferred by the rootstock (root genotype) or the scion (shoot genotype) and if root vigor played a role in
resistance. The influence of a compacted layer on root and shoot genotype response and root rot resistance was studied. Root
rot resistance was found to be controlled by the root genotype, such that on a scale of 1 to 7 (severe disease) the FR266
root had an average score of 2.3 and the Montcalm root had an average score of 4.4. However, when grafted plants were grown
in the presence of a compacted layer, the FR266 root and/or shoot genotype in any graft combination with the susceptible Montcalm
had reduced root rot (score = 2.4 average) than the Montcalm self graft (score = 4.5). Root mass was shown to be controlled
by the root genotype in the absence of compaction such that the FR266 root was 26% larger that the Montcalm root when grafted
onto a FR266 shoot or a Montcalm shoot. When a compacted layer was present the root and shoot genotype both contributed to
root mass. Average root diameter was controlled by the shoot genotype, as the FR266 shoot grafted to Montcalm or FR266 roots
had thicker roots (average diameter 0.455 mm) than the Montcalm shoot (average diameter 0.418 mm). This study shows evidence
that root vigor in the presence of Fusarium disease pressure should be evaluated to effectively develop common bean lines

Rearrangement of cellulose microfibrils within cell-wall matrices is considered one of the most critical steps in the regulation of both the orientation and extent of cell expansion in plants. Xyloglucan endotransglucosylase/hydrolases (XTHs) are a family of enzymes that mediate the construction and restructuring of load-bearing cross links among cellulose microfibrils. The Arabidopsis thaliana XTH genes AtXTH17, 18, 19, and 20 are phylogenetically closely related to one another and are preferentially expressed in the roots. However, they exhibit different expression profiles within the root and respond to hormonal signals differently. To investigate their functions in root growth, we examined phenotypes of loss-of-function mutants for these genes using T-DNA insertion lines and RNAi plants. These functional analyses disclosed a principal role for the AtXTH18 gene in primary root elongation. Of the four XTH genes, AtXTH18 exhibits the highest level of mRNA expression. We also determined auxin-signaling pathways for these genes using a mutant with a defect in the AXR2/IAA7 gene and found that the expression of AtXTH19 in the elongation/maturation region of the root is under the control of the AXR2/IAA7 signaling pathway. 相似文献

采用两两互作实验并以尿标记来确定共居至成年的2雄性根田鼠同胞在行为竞争上的差异,之后在行为选择箱中分别观察它们对来自熟悉和陌生同性个体的新鲜尿气味的行为反应,以确定气味识别差异。结果表明:①雄性根田鼠两两互作时,尿标记的多少可以作为判断其社会等级的标准;②从属鼠对熟悉气味存在明显偏好,其对熟悉气味的访问时间、接近频次和自我修饰频次都显著高于陌生气味;③优势鼠则优先访问陌生气味,其对陌生气味的访问时间、嗅舔频次、嗅舔时间、自我修饰及反标记均显著高于熟悉气味;④对于陌生气味,优势鼠和从属鼠之间存在明显不同的行为反应模式。本实验结果提示,雄性根田鼠对同性尿气味识别的差异以及对陌生气味的行为反应,可能暗含着其领域防卫的信息。 相似文献

The dynamics of networks of sparsely connected excitatory and inhibitory integrate-and-fire neurons are studied analytically. The analysis reveals a rich repertoire of states, including synchronous states in which neurons fire regularly; asynchronous states with stationary global activity and very irregular individual cell activity; and states in which the global activity oscillates but individual cells fire irregularly, typically at rates lower than the global oscillation frequency. The network can switch between these states, provided the external frequency, or the balance between excitation and inhibition, is varied. Two types of network oscillations are observed. In the fast oscillatory state, the network frequency is almost fully controlled by the synaptic time scale. In the slow oscillatory state, the network frequency depends mostly on the membrane time constant. Finite size effects in the asynchronous state are also discussed. 相似文献

Aims Fine root decomposition is the major pathway of carbon and nutrient input to the soil in forest ecosystems. However, the patterns and controlling factors of the decomposition of these roots, especially the finest roots, are poorly understood. Methods Using a root branch-order classification, we separated the first four orders of fine root systems of Pinus koraiensis, Larix gmelinii, Fraxinus mandschurica and Betula platyphylla into two classes: first- and second-order roots combined into lower-order; third- and fourth-order roots combined into higher-order. We conducted a four-year field litterbag study on decomposition of these four root orders of four temperate tree species in northeast China. Important findings The results showed that the lower-order and higher-order roots had a decomposition rate constant of 0.342 and 0.461 for Pinus koraiensis, 0.304 and 0.436 for Larix gmelinii, 0.450 and 0.555 for Fraxinus mandschurica, and 0.441 and 0.579 for Betula platyphylla, respectively. We observed slower decay rates in lower-order than in higher-order roots in all four studied tree species. The root decay constants (k) was significantly correlated with both acid-unhydrolyzable fraction (AUF) and total non-structural carbohydrate (TNC). 目的:观察新型激光光敏根管消毒剂、甲苯胺蓝和亚甲基蓝在牙本质中的渗透效果,评价三种光敏剂在牙本质中的渗透性。方法:收集新鲜拔除的离体单根管牙90颗,根管预备后随机分为三组。每组30颗。A组为新型激光光敏根管消毒剂组;B组为甲苯胺蓝组;c组为亚甲基蓝组。A、B、C三组实验根管内分别用浸有饱和的新型激光光敏根管消毒剂、甲苯胺蓝和亚甲基蓝的棉捻在根管内停留60秒。沿牙体长轴颊舌纵向劈开,倒置荧光显微镜下观察并拍照,用Photoshop8.01软件测量三种光敏剂渗透入牙本质的平均深度。结果:新型激光光敏根管消毒剂在牙本质内的渗透平均深度为553.25μm,甲苯胺蓝在牙本质内的渗透平均深度为350.75μm,亚甲基蓝在牙本质内的渗透平均深度为168.25μm。结论:新型激光光敏根管消毒剂在牙本质渗透性明显优于甲苯胺蓝和亚甲基蓝,具有良好的牙本质渗透性。 相似文献

The loading of amino acids and nitrate into the xylem was investigated by collection and analysis of root-pressure exudate from the cut hypocotyl stumps of seedlings of Ricinus communis L. Glutamine was found to be the dominant amino acid in the exudate and also to be the amino acid which is transferred to the xylem most rapidly and accumulated to the greatest extent. The comparison between uptake and xylem loading showed significant differences in specificity between these two transport reactions, indicating a different set of transport systems. Nitrate is transferred to the xylem at a higher relative rate than any amino acid despite the great nitrate-storage capacity of the root system. Thus the supply of nitrate to Ricinus plants leads to enhanced nitrogen allocation to the shoots. 相似文献
