The Mediterranean world is the cradle for the diversification of a large number of plant species, including legumes belonging to the Tribe Genisteae. Nodule bacteria from three species of Genista legumes indigenous to northwestern Africa (G. ferox, G. numidica, G. tricuspidata) were sampled across a 150 km region of Algeria in order to investigate symbiotic relationships. Partial 23S rRNA sequences from 107 isolates indicated that Bradyrhizobium was the predominant symbiont genus (96% of isolates), with the remainder belonging to Rhizobium or Mesorhizobium. A multilocus sequence analysis on 46 Bradyrhizobium strains using seven housekeeping (HK) genes showed that strains were differentiated into multiple clades with affinities to seven species: B. canariense (17 isolates), B. japonicum (2), B. ottawaense (2), B. cytisi/B. rifense (9), ‘B. valentinum’ (5), and B. algeriense (11). Extensive discordance between the HK gene phylogeny and a tree for four loci in the symbiosis island (SI) region implied that horizontal transfer of SI loci has been common. Cases of close symbiont relationship across pairs of legumes hosts were evident, with 33% of isolates having as their closest relative a strain sampled from a different Genista species. Nevertheless, tree permutation tests also showed that there was substantial host-related phylogenetic clustering. Thus, each of the three Genista hosts utilized a measurably different array of bacterial lineages.  相似文献

Soil erosion is a serious problem of land degradation in many parts of the world, and particularly in mountain rangelands. To understand this process it is necessary to develop methods to assess soil erosion rate in a quick, economic and accurate manner. Based on the analysis of exposed Polylepis australis roots, we tested a dendrogeomorphological method for determining soil loss rate in rills and gullies. Few studies considered non-coniferous tree rings in soil erosion analysis and we used, for the first time, an experimental procedure of root exposure and provided a comparison with roots exposed by gully erosion. Our main results showed that as a consequence of soil erosion, exposed roots changed from root-like to a more stem-like wood anatomical structure. The percentage of vessel area per tree-ring area decreases by an average of 22% to 43% during the first and second year after exposure, respectively. Moreover, and during the same time interval, the mean vessel area decreased 32% and 65%, and the number of vessels increased 7% and 48%, respectively. Scars formed at the upper side of the exposed roots are coincident with changes in wood anatomy, and both evidences may be applied to reconstruct an erosion process. This study confirms that the wood anatomy analysis of partially exposed roots can be used to determine the year in which roots are exposed and provides a useful tool to monitor soil erosion rates with a high accuracy.  相似文献

Summary An in vitro regeneration technique using root callus has been combined with colchicine application to produce hexaploid plants from the sterile triploid cherry rootstock Colt. Colchicine (50 mg/l) was applied together with auxin (IBA) (3 mg/l) either before or after root primordium formation in single excised micropropagating shoots. In both cases shoots were regenerated in vitro from the putative hexaploid roots. Both methods produced hexaploid plants but more than 70% of the regenerated clones were still triploid. The hexaploid plants were distinguishable from the triploids on morphological as well as cytogenetic grounds. Quantitative characters such as leaf length/breadth ratio, stomatal length and density in the hexaploids were significantly different from both regenerated and non-regenerated triploids.  相似文献

Summary Nuclear staining with acridine orange was used to assess cell viability in the cortex of wheat and barley seminal roots from glasshouse and field experiments. Results from this method correlated well with nuclear assessments made in unstained or Feulgen-stained roots, and other evidence is presented to support the validity of the method. The pattern of root cortex death (RCD) was similar in wheat and barley and consistent over a wide range of conditions. Behind the extending root tip and zone of nucleate root hairs, nuclei disappeared progressively from the outer five (of six) cortical cell layers of the root axes, starting in the epidermis. Stainable nuclei remained in the sixth cell layer, next to the endodermis, and in most cell layers around the bases of root laterals and in a small region immediately below the grain. The onset of cell death was apparently related more to the age of a root region than to its distance behind the root tip, and it was not closely correlated with endodermal or stelar development assessed by staining with phloroglucinol/HCl. The rate of RCD was much faster in wheat than barley in both glasshouse and field conditions, and faster in some spring wheat cultivars than in others in the glasshouse. RCD occurred in sterile vermiculite and perlite and was not enhanced by the presence of soil microorganisms; nor was it enhanced in soil by the addition of the non-pathogenic fungal parasitesPhialophora radicicola var..graminicola orMicrodochium bolleyi. RCD is suggested to be endogenously controlled by the amount of photosynthate reaching the cortex. Its implications for growth of soil microorganisms and especially for growth and biological control of root-infecting fungi are discussed.  相似文献

Management practices over the past 100 years, especially fire exclusion and suppression, in Yosemite Valley, Yosemite National Park, CA resulted in forest stands with basal areas in excess of 110 m2 ha-1. Accelerated attack by several species of bark beetles (Scolytidae) followed a severe drought in 1976–77. Pinus ponderosa, already under attack by root rot (Heterobasidion annosus) was affected severely, and mortality exceeded 70%. Calocedrus decurrens did not suffer comparable mortality and has replaced P. ponderosa as the dominant species. Size class distributions as well as comparisons of relative density and relative basal area from 1961, 1976 and 1983–85 also demonstrate a major change in forest composition. This change is most pronounced on alluvial landforms.  相似文献

Abstract: Canada goose (Branta canadensis) harvest management depends on reliable estimates of harvest composition, and established genetic methods provide an alternative to traditional methods. We expanded upon previous genetic studies by comparing the utility of 6 nuclear microsatellite loci and mitochondrial (mtDNA) control region sequences for discriminating among giant (B. c. maxima) and interior (B. c. interior) populations in Ohio (USA) Canada goose harvests at both individual and population levels. Subspecies and populations exhibited greater differentiation in mtDNA (FST = 0.202) than microsatellites (FST = 0.021), as would be expected based on differences in effective population size. Neither microsatellites nor mtDNA alone were sufficient for estimating harvest composition at the subspecies or population level in simulations and empirical blind tests using individuals of known origin; however, a combined microsatellite + mtDNA dataset yielded accurate and precise harvest derivations at the subspecies level. Both population-level mixed stock analysis and individual-level assignment tests provided accurate results, but a large proportion of birds could not be assigned with confidence at the individual level. We applied mixed stock analysis and the combined microsatellite + mtDNA dataset to Ohio's 2003–2004 harvest and found that interior populations accounted for 4.9% (95% CI = 1.7–8.0%) of the statewide early season and 9.3% (95% CI = 6.9–11.6%) of the regular and late-season harvested sample. These results suggest that maximum likelihood harvest derivations are highly dependent on the choice of genetic markers. Studies should only employ markers that exhibit sufficient variation and have been shown through simulations and empirical testing to accurately discriminate among the subspecies or management populations of interest.  相似文献

Background and aimHertwig’s Εpithelial Root Sheath (HΕRS) has a major function in the developing tooth roots. Earlier research revealed that it undergoes epithelial–mesenchymal transition, a vital process for the morphogenesis and complete development of the tooth and its surrounding periodontium. Few studies have demonstrated the role of HERS in cementogenesis through ΕMΤ. The background of this in-silico system biology approach is to find a hub protein and gene involved in the EMT of HERS that may uncover novel insights in periodontal regenerative drug targets.Materials and methodsThe protein and gene list involved in epithelial–mesenchymal transition were obtained from literature sources. The protein interaction was constructed using STRING software and the protein interaction network was analyzed. Molecular docking simulation checks the binding energy and stability of protein-ligand complex.ResultsResults revealed the hub gene to be DYRK1A(Hepcidin), and the ligand was identified as isoetharine. SΤRIΝG results showed a confidence cutoff of 0.9 in sensitivity analysis with a condensed protein interaction network. Overall, 98 nodes from 163 nodes of expected edges were found with an average node degree of 11.9. Docking results show binding energy of ?4.70, and simulation results show an RMSD value of 5.6 Å at 50 ns.ConclusionIsoetharine could be a potential drug for periodontal regeneration.  相似文献

The effect of Al stress on H2O2 production of rice (Oryza sativa L.) seedlings and difference in responses of antioxidant enzymes between Al-tolerant variety (Azucena) and Al-sensitive rice one (IR 64) were investigated. Aluminum-induced H2O2 production and malondialdehyde (MDA) content were more pronounced for IR 64 than for Azucena. In the presence of 2 mM Al, addition of 10 mM imidazole (inhibitor of NADPH oxidase) and 1 mM azide (inhibitor of peroxidase) significantly decreased H2O2 production by 16% and 43% for Azucena, and 21% and 68% for IR 64, respectively. Under Al treatment, the Al-tolerant variety Azucena had significantly higher activities of catalase, ascorbate peroxidase, dehydroascorbate reducase, glutathione peroxidase and glutathione reductase, and higher concentrations of reduced glutathione than the Al-sensitive one IR 64. Treatment with buthionine sulfoximine, a specific inhibitor of GSH synthesis, significantly increased H2O2 production in both varieties in the presence and absence of Al. In contrast, the treatment with GSH significantly decreased the production of H2O2 induced by Al stress. Results suggest that GSH may play an important role in scavenging H2O2 caused by Al stress.  相似文献

Partial rootzone drying (PRD) is widely investigated as an effective irrigation technique, resulting in higher water use efficiency and yield for plants growing under mild water deficit. Nutrition is another important factor affecting plant yield, but nutrient acquisition has only rarely been considered in conjunction with PRD. Here we investigate the interaction between water and fertilizer supply in a pot experiment with oilseed rape (Brassica napus L.). Eight treatments were set up for the experiment, a factorial combination of four watering regimes (100% control watering at both sides of the plants; 50% control watering at both sides of the plants; 50% fixed watering applied only to one side of the plants; 50% alternate watering applied alternately to both sides of the plant) and two fertilizer placement levels (uniform over the entire pot, and patchy supplied to one side). For the 50% watering treatments, the total amount of water supplied to the plants was the same, only the pattern of application differed between treatments. Also the total fertilizer applied was the same for all treatments. Oilseed rape roots foraged effectively for water and nutrients resulting in relatively small differences in nutrient uptake and above-ground growth among the water-deficit treatments. Placing fertilizer at one side of the plants increased nutrient uptake, but there were differences between the water treatments and interactions with water uptake. Alternate watering resulted in the highest growth, as a result of the largest nitrogen and phosphorus uptake with the smallest root investment among the three water deficit treatments. Fixed watering resulted in poorest performance when fertilizer was uniformly spread throughout the pot, because the plants were unable to acquire the nutrients on the dry side. Our results show that PRD can be well combined with patchy fertilizer supply, but that reduced nutrient uptake may be expected when nutrients are supplied in parts of the soil volume that remain too dry.
