The appearance of two electric potentials in the aorta is explained by the blood pulse and the remote steps of atherosclerosis are elucidated from the physical point of view. When the first C-C-2W is deposited in the intimamedia, the primary cause of their retention is the stress-induced polarization of the membrane. C-C-2W possesses a permanent dipole moment which may be reversed by the field produced by the radial expansion/contraction of the arteries.The initial C-C-2W increases the polarization of the aorta walls, favoring accumulation of more oriented material on top of the first. C-C-2W is also ferroelectric and constitutes a polar continuum with the membranes. By the time fatty streaks appear, the entire aorta wall is ferroelectric and energy has been stored in the walls under the form of residual polarization. This storage gradually destroys the wall, leading to aneurism. The biological significance of the new theory is summarized in Table 2.  相似文献

The central concern of this paper is to re-evaluate Rosen's replicating (M,R)-systems, presented in his book 'Life Itself ', where M and R signify metabolism and repair, respectively. We look anew at Rosen's model of an organism in the light of extensive research into natural hierarchical systems, and the paper presents conclusions drawn from a comparison between Rosen's relational model and that of a birational complementary natural hierarchy. We accept that Rosen's relational model provides a useful stepping stone to understanding the nature of life, but also suggest that it induces potentially digressive conclusions. We conclude that a binary segregation of relational assemblies into mechanisms and organisms is insufficient, and indicate how a threefold segregation throws new light on Rosen's model. An organism is not 'the complement of a mechanism': the complement of a mechanism is its ecosystem. An organism is the 'complex interface' between mechanism and ecosystem.  相似文献

I have the pleasure to present a number of personal experiences that I had with Robert Rosen, both as his student and as a research colleague, and I will describe how this affected my academic career over the past decades. As a matter of fact, Rosen's work with (M,R)-systems as well as his continuing mentorship guided me into my own research in gerontology and geriatrics. Amazingly, this still continues to affect my work in complexity theory after 30 years.  相似文献

In order to study the membrane function of tolerant B antigen-binding cells, tolerance to the trinitrophenyl (TNP) determinant was induced in mice by injecting the reactive form of the hapten, trinitrobenzene sulfonic acid (TNBS). By appropriate transfer experiments, Fidler and Golub (J. Immunol.112, 1891, 1974) had previously shown that this form of tolerance is a B-cell property, induced and expressed in the absence of T cells. Hapten inhibition demonstrated the TNP-specificity of receptors on TNP-donkey erythrocyte(TNP-D)-binding cells in tolerant and nontolerant mice. About 88% of these cells were B cells by immunofluorescence, and the remainder were T cells. In the tolerant mice, challenge with TNP-sheep erythrocytes failed to expand the TNP-binding population, but sheep erythrocyte binders and anti-sheep plaque-forming cells expanded normally. Despite little or no change in TNP-binding cell numbers after tolerance induction, the TNP-binding cells of tolerant animals could not cap their receptors, in contrast to the sheep erythrocyte-binding cells from the same animals which capped normally. Although there is no anti-TNP plaque-forming cell response when tolerogen and immunogen are given simultaneously, capping failure is not evident until 2–4 days after tolerogen exposure. By Day 7, substantial recovery of immune responsiveness had occurred, yet even 12 months after a single dose of tolerogen there was no restoration of capping. Thus despite the association of both capping failure and unresponsiveness with tolerogen exposure, these lymphocyte functional defects appeared not to be causally related.  相似文献

Dantrolene sodium, administered orally (about 100 mg/kg, twice daily), produces reversible alterations in pigeon hepatocyte morphology. The drug initially causes weight loss, and discoloration and enlargement of the liver. After about 4 weeks of treatment, weight loss subsides and stabilizes. The pigeons can survive this prolonged drug treatment (26 weeks in this study) after this period. Ultrastructural changes in hepatocytes associated with this drug include loss of rough endoplasmic reticulum (RER), formation of large smooth membrane whorls, and decrease in mitochondrial and increase in lipid volume percentages (as measured morphometrically). Mitochondrial volume percentage decreases by a half after 4 weeks of treatment and remains at this level at 8 and 12 weeks. Increase in lipid is greatest at 4 weeks, and tapers off at 8 and 12 weeks, although remaining significantly above control levels. The morphometric information is related to histochemical succinic dehydrogenase activity. Hepatocytes from treated birds show less SDH activity than controls, suggesting lower mitochondrial activity. The Ca-ATPase activity associated with bile canaliculi in control cells is absent in treated cells. Membrane material accumulated in the whorls of the treated cells is substantial, many of the whorls having diameters as large or larger than the nucleus.On cessation of drug treatment, hepatocytes from dantrolene-treated birds regain a more normal morphology. RER is not produced by ribosomes attaching to membrane whorl material, since the whorls disappear prior to RER proliferation. Hepatocytes are normal in appearance and histochemical properties after 2–4 weeks of recuperation.  相似文献

We have evaluated the effect of purified human α-fetoprotein (AFP) on several in vitro correlates of cell-mediated immunity. AFP (100 μg/ml) had no effect on antigen-induced migration inhibitory factor (MIF) production or on the ability of T cells to bind sheep erythrocytes. In contrast, AFP had a twofold effect on mitogen-and antigen-induced lymphocyte proliferation. In a dose-dependent fashion (1–100 μg/ml), AFP was mitogenic to lymphocyte cultures and also suppressed tritiated thymidine incorporation by PHA- and SK-SD-stimulated cultures. Albumin had somewhat similar effects on lymphocyte proliferation but only at 100 μg/ml. The reason for the latter was not clear since the albumin preparation was free of any detectable AFP. Our studies suggest that human AFP may not have any biologically significant immunosuppressive function.  相似文献

As a boy, I read Sinclair Lewis's Arrowsmithand dreamed of doing research of potential benefit to society. I describe the paths of my scientific career that followed. Several distinguished scientists served as my mentors and I present their profiles. Much of my career was in a small department at a small institution where independent researchers collaborated informally. I describe the unique method of carrying on research there. My curiosity about glycolate metabolism led to unraveling the enzymatic mechanism of the glycolate oxidase reaction and showing the importance of H2O2 as a byproduct. I discovered enzymes catalyzing the reduction of glyoxylate and hydroxypyruvate. I found α-hydroxysulfonates were useful competitive inhibitors of glycolate oxidase. In a moment of revelation, I realized that glycolate metabolism was an essential part of photorespiration, a process that lowers net photosynthesis in C3 plants. I added inhibitors of glycolate oxidase to leaves and showed: (1) glycolate was synthesized only in light as an early product of photosynthetic CO2 assimilation, (2) the rate of glycolate oxidation consumed a sizable fraction of net photosynthesis in C3 but not in C4 plants, and (3) that glycolate metabolism increased greatly at higher temperatures. For a while I studied the control of stomatal opening in leaves, and this led to the finding that potassium ions are a key solute in guard cells. I describe experiments that show that when photorespiration rates are high, as occurs at higher temperatures, genetically increasing leaf catalase activity reduces photorespiration and increases net photosythetic CO2 assimilation. This paper begins with a discussion of the scientia sexualis/ars erotica distinction, which Foucault first advances in History of Sexuality Vol. 1, and which has been employed by many scholars to do a variety of analytical work. Though Foucault has expressed his doubts regarding his conceptualization of the differences between Western and Eastern discourses of desire, he never entirely disowns the distinction. In fact, Foucault remains convinced that China must have an ars erotica. I will explore Foucault’s sources of authority. To this end, I introduce the work of famous Dutch sinologist Robert Hans van Gulik, who published the tremendously influential Sexual Life in Ancient China in 1961, and also explore Joseph Needham’s view on Chinese sex. I argue that, Foucault, in his fierce polemic against the “Repressive Hypothesis”, himself imagined a utopian Other where pleasure and desire were organised differently. I end on a discuss on Orientalism and the project of “Sinography” of comparative literature scholars Haun Saussy, Eric Hayot and others.  相似文献

ABSTRACT

Research on the relationship between humans and animals has identified some links between companion animals and physiological, psychological, and social benefits for the human. Adopting Robert Weiss's (1974) Theory of Social Provisions as a framework, this qualitative study explores the role of the human-animal relationship amongst 30 people living with the Human Immunodeficiency Virus (HIV) in Australia. Despite the transition of HIV from a terminal to chronic condition in many developed nations, there can still be personal and social challenges accompanying an HIV-diagnosis. Thematic analysis of the 30 interviews identified themes of Attachment, Opportunity for Nurturance, Reassurance of Worth, Reliable Alliance, Obtaining of Guidance/Emotional Support, and Social Integration. Extracts coded to these themes indicated that many participants believed their companion animal motivated them to remain socially and physically active; provided an outlet for love and attachment; remained non-judgmental irrespective of the human's physical or social status; and was capable of providing both day-to-day comfort through their reliable presence, and episode-specific supportive responses during periods of heightened stress. It was proposed that for people living with a chronic and/or stigmatized condition like HIV, these aspects of the human-animal relationship may play an important part in their overall wellbeing. In conclusion, this study contributes to a greater understanding of the lived experience of HIV and provides a conceptually sound mechanism for validating the love and support that some HIV-positive people perceive in their relationship with a companion animal. This knowledge draws attention to the need to normalize, validate, and support the human-animal relationship throughout the animal's life, and death.  相似文献
