Crystalline bis(N,N-di-iso-butyldithiocarbamato-S,S′)(pyridine)cadmium(II) - adduct 1 was prepared and studied by means of multinuclear 13C, 15N, 113Cd CP/MAS NMR spectroscopy, single-crystal X-ray diffraction and simultaneous thermal analysis (STA). In molecular structure 1, the cadmium atom coordinates with four sulphur atoms and one nitrogen atom of pyridine, forming a coordination polyhedron [CdS4N], whose geometry is an almost ideal tetragonal pyramidal (C4v). The coordinated py molecule is in the apical position, while two structurally non-equivalent di-iso-butyldithiocarbamate ligands, playing the same terminal S,S′-chelating function, define the basal plane. To characterise additionally the structural state of the cadmium atom in this fivefold coordination, 113Cd chemical shift anisotropy (CSA) parameters, δaniso and η, were calculated from experimental MAS NMR spectra that revealed an almost axially symmetric 113Cd chemical shift tensor. From a combination of TG and DSC measurements taken under an argon atmosphere, we found that the mass of adduct 1 is lost in two steps involving initial desorption of coordinated py molecules with subsequent thermal destruction of liberated cadmium(II) di-iso-butyldithiocarbamate, with yellow-orange, fine-powdered solid CdS as the final product. 相似文献

Microalgae have the ability to mitigate CO2 emission and produce oil with a high productivity, thereby having the potential for applications in producing the third-generation of biofuels. The key technologies for producing microalgal biofuels include identification of preferable culture conditions for high oil productivity, development of effective and economical microalgae cultivation systems, as well as separation and harvesting of microalgal biomass and oil. This review presents recent advances in microalgal cultivation, photobioreactor design, and harvesting technologies with a focus on microalgal oil (mainly triglycerides) production. The effects of different microalgal metabolisms (i.e., phototrophic, heterotrophic, mixotrophic, and photoheterotrophic growth), cultivation systems (emphasizing the effect of light sources), and biomass harvesting methods (chemical/physical methods) on microalgal biomass and oil production are compared and critically discussed. Tumor cells release NKG2D ligands to evade NKG2D-mediated immune surveillance. The purpose of our investigation was to explore the cellular mechanisms of release used by various members of the ULBP family. Using biochemical and cellular approaches in both transfectant systems and tumor cell lines, this paper shows that ULBP1, ULBP2, and ULBP3 are released from cells with different kinetics and by distinct mechanisms. Whereas ULBP2 is mainly shed by metalloproteases, ULBP3 is abundantly released as part of membrane vesicles known as exosomes. Interestingly, exosomal ULBP3 protein is much more potent for down-modulation of the NKG2D receptor than soluble ULBP2 protein. This is the first report showing functionally relevant differences in the biochemistry of the three members of the ULBP family and confirms that in depth study of the biochemical features of individual NKG2D ligands will be necessary to understand and manipulate the biology of these proteins for therapy. 相似文献

对潮棕壤不同土地利用方式下0~100 cm土体中土壤有机碳含量的剖面分布、有机碳储量及C/N进行了研究.结果表明:不同土地利用方式下土壤有机碳含量的剖面分布差异明显,林地、割草地、荒地及裸地各土层有机碳含量高于农田生态系统;不同土地利用方式下的土壤有机碳与全氮呈极显著的正相关;土壤C/N随剖面土层深度的增加呈下降趋势,林地土壤的C/N相对较高,割草地、荒地和裸地次之,农田生态系统的土壤C/N较低.在0~100cm深度土壤,荒地每年截获的土壤有机碳分别比农田不施肥、农田循环猪圈肥处理、农田化肥NPK处理、农田化肥NPK 循环猪圈肥处理高4.52、4.25、4.46和3.58 t.hm-2.说明荒地在增加土壤有机碳储量方面有很大潜力. 相似文献

目的:研究Na^+/H^+交换抑制剂阿米洛利对低氧刺激的大鼠肺动脉平滑肌细胞(PASMCs)增殖的影响,以及Na^+/H^+交挟体-l(NHE-1)活性和表达的变化.方法:常氧(21%O2)或低氧(2%O2)条件下培养PASMCs,并分别给予浓度为1.653、3.125、6.25、12.5、25和50μmol/L.等不同浓度的阿米洛利,培养24h,采用MTT比色实验和免疫组化检测PCNA阳性细胞率的方法反映细胞增殖情况,同时采用激光共聚焦检测细胞内pH以反映Na^+/H^+交换体-1活性,RT—PCR法检测Na^+/H^+交换体-1mRNA的表达量.结果:低氧培养的PASMCs细胞内pH升高,NHE—1mRNA的表达增多,而阿米洛利可以降低细胞内pH,减少NHE—1mRNA的表达量。同时低氧较常氧培养MTT光吸收值较常氧培养明显升高。PCNA阳性细胞率明显增高,而给予阿米洛利时上述两个指标随药物浓度增加而逐渐下降。结论:低氧可以激活PASMCs细胞膜上的E—1,增加其mRNA水平表达量,使细胞内碱化,促进细胞增殖,而Na^+/H^+交换抑制剂阿米洛利可以抑制其活性,减少mRNA水平的表达,导致细胞内酸化,从而抑制细胞增殖,并且此抑制作用在3.125~50μmol/L.浓度范围内呈现明显的浓度依赖性。 相似文献

Oxidized human neuroglobin (Ngb), a heme protein expressed in the brain, has been proposed to act as a guanine nucleotide dissociation inhibitor (GDI) for the GDP-bound form of the heterotrimeric G protein alpha-subunit (Galpha(i)). Here, to elucidate the molecular mechanism underlying the GDI activity of Ngb, we used an glutathione-S-transferase pull-down assay to confirm that Ngb competes with G-protein betagamma-subunits (Gbetagamma) for binding to Galpha(i), and identified the Galpha(i)-binding site in Ngb by chemical cross-linking with 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride and sulfo-N-hydroxysuccinimide, coupled with mass spectrometry (MS). Matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) MS analysis for tryptic peptides derived from the cross-linked Ngb-Galpha(i) complex revealed several binding regions in Ngb. Furthermore, MALDI-TOF/TOF MS analysis of the cross-linked Ngb and Galpha(i) peptides, together with the MS/MS scoring method, predicted cross-linking between Glu60 (Ngb) and Ser206 (Galpha(i)), and between Glu53 (Ngb) and Ser44 (Galpha(i)). Because Ser206 of Galpha(i) is located in the region that contacts Gbetagamma, binding of Ngb could facilitate the release of Gbetagamma from Galpha(i). Binding of Ngb to Galpha(i) would also inhibit the exchange of GDP for GTP, because Ser44 (Galpha(i)) is adjacent to the GDP-binding site and Glu53 (Ngb), which is cross-linked to Ser44 (Galpha(i)), could be located close to GDP. Thus, we have identified, for the first time, the sites of interaction between Ngb and Galpha(i), enabling us to discuss the functional significance of this binding on the GDI activity of Ngb. 相似文献

Measles virus-specific T cells and the production of cytokines play a critical role in the immune response following measles immunization. To understand the genetic factors that influence variation in IFN- and IL-4 responses following measles immunization and to provide insight into the factors influencing both cellular and humoral immunity to measles, we assessed associations between human leukocyte antigen (HLA) class II genes and measles-specific Th1 and Th2-type cytokine responses in peripheral blood lymphocytes from 339 children previously vaccinated with two doses of measles-mumps-rubella vaccine (MMR-II). Median values for measles-specific IFN- and IL-4 secretion levels were 40.73 and 9.71 pg/ml, respectively. The global tests suggested associations between measles-specific IFN- response and alleles of the DRB1 and DQB1 loci (P=0.07 and P=0.02, respectively). Specifically, DRB1*0301, *0901, and *1501 alleles were significantly associated with IFN- secretion. The alleles that suggested evidence of an HLA association with IL-4 secretion were DRB1*0103, *0701, and *1101. Th1 cytokine responses and DQB1 allele associations revealed that the alleles with the strongest association with IFN- secretion were DQB1*0201, *0303, *0402, and *0602. Specific alleles with a suggestive association with low measles-specific Th2 cytokine responses were DQB1*0202 and *0503. In addition, DPB1*0101, *0201, and *0601 alleles provided suggestive evidence of an HLA association with measles-induced IFN- response, while DPB1*0501 was associated with an IL-4 response. These data suggest that IFN- and IL-4 cytokine responses to measles may be genetically restricted in part by HLA class II genes, which in turn can restrict the cellular immune response to measles vaccine. 相似文献
