Administration of low amounts of ethanol for a prolonged period increases rat brain synaptosomal (Na+–K+)-ATPase activity, the increase being less in the protein deficient rats. The adaptive mechanism to offset the stress imposed by the continued presence of ethanol seems to be depressed by low plane of nutrition. In vivo and in vitro effects of ethanol on (Na+–K+)ATPase seems to be different.  相似文献

Abstract. Seedlings of Pinus radiata D. Don were grown in growth chambers for 22 weeks with two levels of phosphorus, under either well-watered or water-stressed conditions at CO2 concentrations of either 330 or 660mm3 dm?3. Plant growth, water use efficiency and conductance were measured and the relationship between these and needle photosynthetic capacity, water use efficiency and conductance was determined by gas exchange at week 22. Phosphorus deficiency decreased growth and foliar surface area at both CO2concentrations; however, it only reduced the maximum photosynthetic rates of the needles at 660 mm3 CO2 dm?3 (plants grown and measured at the same CO2 concentration). Water stress reduced growth and foliar surface area at both CO2 concentrations. Increases in needle photosynthetic rates appeared to be partly responsible for the increased growth at high CO2 where phosphorus was adequate. This effect was amplified by accompanying increases in needle production. Phosphorus deficiency inhibited these responses because it severely impaired needle photosynthetic function. The relative increase in growth in response to high CO2 was higher in the periodically water-stressed plants. This was not due to the maintenance of cell volume during drought. Plant water use efficiency was increased by CO2 enrichment due to an increase in dry weight rather than a decrease in shoot conductance and, therefore, transpirational water loss. Changes in needle conductance and water use efficiency in response to high CO2 were generally in the same direction as those at the whole plant level. If the atmospheric CO2 level reaches the predicted concentration of 660 mm3 dm?3 by the end of next Century, then the growth of P. radiata will only be increased in areas where phosphorus nutrition is adequate. Growth will be increased in drought-affected regions but total water use is unlikely to be reduced.  相似文献

Summary The purpose of this study was to develop a nonenzymatic method of isolating adult islets using atrophied pancreata from copper-deficient rats and to analyze their morphologic characteristics and behavior in culture. This unusual model of isolation was studied because islets remain intact in the course of dietary copper deficiency while the acinar glandular component of the pancreas undergoes selective atrophy and lipomatosis. Small fragments containing islets were readily microdissected from atrophied glands and placed in culture. Within 24 h the fragments congealed into small irregular- to spherical-shaped masses within which the darker profile of islets could be distinguished. Within a period of 3 to 5 d, islet tissue began to bud from the lipocytic mass until by Day 7 spherical aggregates of intact islet tissue separated from the residual fragments. Subsequent to further in vitro treatment, these islets could be maintained as free viable spherical masses if periodically agitated, as attached stationary islets which developed monolayer growth if left undisturbed and as aggregated masses of islet tissue forming megaislets if combined in small groups. Grouped islets treated with actinomycin D and cycloheximide did not exhibit aggregation when incubated with these inhibitors. This suggests that megaislet formation was an active process requiring protein-RNA synthesis rather than passive clumping or aggregation that can accompany metabolically altered or dying islets undergoing cellular shedding and adhesion. Immunohistochemical localization demonstrated that insulin, glucagon, somatostatin, and pancreatic polypeptide-immunoreactive cell types were present within the islets derived from this technique. The cellular topography of these islets was not unlike that described by others for islets cultured from enzymatic isolation. This culture model may serve as a resource for mature, viable islets isolated without mechanical or enzymatic disaggregation which can have attenuating effects on islet function. Summary Aberrations of photoreceptor ultrastructure resulting from carotenoid/retinoid (vitamin A) deprivation were studied in the retina of Manduca sexta. The syndrome of chromophore deficiency included hypertrophy of smooth endoplasmic reticulum, variable dilation of rhabdomeric microvilli, the insertion of endomembrane fingers into such enlarged microvilli, and the formation of rhabdomeric vacuoles, intracellular compartments containing microvilli similar to those of the rhabdomere. Retinas were processed either with conventional procedures employing preliminary aldehyde fixation followed by heavy metal postfixation, or by fixation and incubation in unbuffered OsO4. The latter method deposits osmium throughout the endomembrane system, within the rhabdomeric vacuoles, and in the extracellular space of the rhabdom. However, the intravillous fingers were rarely impregnated with osmium, despite their continuity with densely stained cisternae of the smooth endoplasmic reticulum. We suggest that the insertion of endomembrane fingers into dilated microvilli results from a cytoskeleton-mediated link between cisternae of the smooth endoplasmic reticulum and the rhabdomeric membrane, an association that may be important in the turnover of photoreceptor membrane. We interpret endomembrane hypertrophy and development of rhabdomeric vacuoles as symptoms of disturbance in the pathway leading to the assembly of the rhabdomere resulting from reduced synthesis of visual pigment.  相似文献

Early iron deficiency in rat does not affect the weight or the protein, DNA, and RNA content but results in a slight reduction in gamma-aminobutyric acid (GABA) (13%, p less than 0.01) and glutamic acid (20%, p less than 0.001) content of the brain. The activities of the two GABA shunt enzymes, glutamate dehydrogenase and GABA-transaminase, and of the NAD+-linked isocitrate dehydrogenase (ICDH) were inhibited whereas the glutamic acid decarboxylase, mitochondrial NADP+-linked ICDH, and succinic dehydrogenase activities remained unaltered in brain. On rehabilitation with the iron-supplemented diet for 1 week, these decreased enzyme activities in brain attained the corresponding control values. However, the hepatic nonheme iron content increased to about 80% of the control, after rehabilitation for 2 weeks. A prolonged iron deficiency resulting in decreased levels of glutamate and GABA may lead to endocrinological, neurological, and behavioral alterations.  相似文献

Summary As a result of air pollution, considerable deposition of ammonium sulphate occurs on vegetation and soil in the vicinity of chicken farms and fields dressed with animal slurry. A clear relation exists between this ammonium sulphate deposition and the distance to certain agricultural activities. Field investigations and ecophysiological experiments both show that the needles ofPinus nigra var.maritima (Ait.) Melville take up ammonium and excrete potassium, magnesium and calcium. This often results in potassium and/or magnesium deficiencies and may lead to premature shedding of needles. The high levels of nitrogen in the needles are strongly correlated to fungal diseases.Whether the observed cation leaching will result in disturbed nutrient budgets depends mainly on soil conditions. Leaching of K, Mg and Ca from the soil, caused by ammonium sulphate, may further inhibit nutrient uptake.Field investigations show a clear correlation between increased ratios of NH4 to K, Mg and Ca in the soil solution and the damage to pine forests.  相似文献

There is considerable debate on the role of folate in CNS function. Recent work indicates that folate deficiency may affect CNS serotonin metabolism, and clinical studies describe many consequences of such a deficiency. On the other hand some workers maintain that folate deficiency alone causes CNS abnormalities. We maintained rats, through dietary deprivation, at folate levels below 4 ng/ml for more than 6 weeks and showed that at that time both their liver and brain folate levels were significantly reduced. We then studied their local cerebral glucose utilization (LCGU) using the [14C]deoxyglucose technique. This method assesses cerebral function by measuring regional metabolic activity. We also determined LCGU in rats given the same diet but replenished with folate (folate control) and in others given free access to commercially available food (normal controls). Our results show that this degree of folate deficiency has no effect on cerebral function. This contrasts with the focal suppression of LCGU we previously reported in a model of vitamin B12 deficiency.  相似文献

The concept of adaptation has been used differently in studies of biological and cultural evolution, and this divergence raises the question of whether genetic and cultural adaptations are truly comparable. This paper compares genetic and cultural traits associated with endemic malaria in Sardinia, Italy. Thalassemia and G-6-Pd deficiency, two genetic traits of the Island's population, are believed to enhance fitness against malaria, despite increased risk for the diseases of thalassemia major and favism. Two cultural traits, a pastoral pattern of inverse transhumance and rules limiting the geographical mobility of lowland women, limited exposure to the malaria vector, Anopheles labranchiae; these are used as examples of cultural adaptations. The distribution, costs, and benefits of the adaptive cultural and genetic traits are compared, and the theoretical difficulties of finding a common measure of adaptive value are discussed.  相似文献

DCCD activates the respiratory burst in guinea pig peritoneal neutrophils. The onset of the superoxide producing activity is preceeded by a lag, inversely proportional to the dose of the stimulant and to the temperature. Initial rates of superoxide formation exhibit different dependencies on the concentrations of DCCD and on temperature. Activation of NAD(P)H oxidase is inhibited by preincubation of neutrophils with 2-deoxyglucose and does not require the presence of extra cellular Ca2+.  相似文献

Anaerobic potentiometric titrations of b cytochromes have been carried out in beef heart submitochondrial particles in the presence of several specific inhibitors of electron transfer through the b-c1-site of the respiratory chain. Whereas antimycin shows no significant effect on the titration curve of cytochrome b-562, NoHOQnO is found to shift the Em of b-562 by 20-30 mV to the positive. Funiculosin raises the Em of b-562 by greater than 100 mV and also appears to bring about a minor shift of b-566 midpoint potential. In the presence of myxothiazol, both b cytochromes titrate with Em values 15-30 mV more positive than in the control.  相似文献
