Depolarized light-scattering studies of bilayer structures in phospholipid vesicles

Depolarized light scattering has been used to investigate the hydrocarbon chain packing of phospholipids in vesicles below the phase transition and ordering of their chains above the phase transition. The chain packing and ordering have been demonstrated for vesicles of l-α-dipalmitoylphosphatidylethanolamine and some phosphatidylcholines of different hydrocarbon chain lenghts. Anisotropy ratios for phospholipid vesicles could be determined by measuring depolarization ratios for several vesicle sizes at low concentrations of the lipids. The following results were obtained. Hydrocarbon chains of l-α-dimyristoyl and distearoylphosphatidylcholines below their phase transitions pack at tilting angles in good agreement with X-ray diffraction data. On the other hand, hydrocarbon chains of dipalmitoylphosphatidylethanolamine pack perpendicular to the bilayer surface. Values of the averaged order parameter for dimyristoyl, dipalmitoyl and distearoylphosphatidylcholines at 2.5°C above their phase transition are all the same and the value for dipalmitoylphosphatidylcholine is in agreement with results from ²H-NMR experiments. The value of the order parameter for dipalmitoylphosphatidylethanolamine is slightly larger than that for dipalmitoylphosphatidylcholine.     

Analysis of ribosomal DNA restriction patterns in the genusKluyveromyces

Riboprinting was used to determine the relationships among strains belonging to 15 species of the genusKluyveromyces. The small subunit ribosomal RNA gene (SSU rDNA) was amplified using the Polymerase Chain Reaction (PCR) and subjected to a battery of nine restriction enzymes. Similarity coefficients between strains were calculated based on shared and unique restriction fragments. Cluster analysis revealed three major groups that generally correlated with previously reported relationships based on other molecular data. Variations in SSU rDNA restriction fragments may be used for differentiation of theKluyveromyces strains included in this study.The U.S. Government right to retain a non-exclusive, royalty free licence in and to any copyright is acknowledged.     

加替沙星注射液治疗肺癌患者呼吸道感染临床评价

评价加替沙星注射液治疗肺癌患者呼吸道感染临床疗效和药物不良反应。回顾性分析2007年1-12月非随机选择45例肺癌患者医院外获得性呼吸道感染入院治疗的临床资料进行统计分析。应用加替沙星组与应用注射用头孢哌酮钠／舒巴坦组治疗呼吸道感染的总有效率分别为92．31％和94．74％（P〉0．05）;用药3d后咳嗽咳痰、发热、湿哆音、血象好转率分别为84．62％、95．45％、85．71％、88．OO％和57．89％、71．43％、64．29％、82．35％（P〈0．05）。不良反应发生率分别11．54％和10．53％（P〉0．05）。加替沙星治疗肺癌患者呼吸道感染疗效好,见效快,是较理想的药物。     

盐度和CO2倍增环境下碱蓬幼苗呼吸酶活性的变化

研究了生长在正常大气ＣＯ２和ＣＯ２倍增环境中的盐生植物碱蓬（Ｓｕａｅｄａｓａｌｓａ）幼苗呼吸酶活性对ＫＣｌ和ＮａＣｌ的反应．结果表明,在ＣＯ２倍增（７００μｌ·Ｌ－１）和正常大气ＣＯ２（３５０μｌ·Ｌ－１）下,３００ｍｍｏｌ·Ｌ－１ＫＣｌ和ＮａＣｌ均能抑制琥珀酸脱氢酶（ＳＤＨ）和苹果酸脱氢酶（ＭＤＨ）活性,而异柠檬酸脱氢酶（ＩＤＨ）活性为ＮａＣｌ抑制、ＫＣｌ促进;ＮａＣｌ和ＫＣｌ明显抑制细胞色素氧化酶（ＣＯ）和光呼吸中乙醇酸氧化酶（ＧＯ）、羟基丙酮酸还原酶（ＨＰＲ）活性;并指出在ＫＣｌ胁迫下,ＣＯ２使三羧酸循环（ＴＣＡＣ）的运行变慢,ＮａＣｌ胁迫下使其加快,ＴＣＡＣ运行限速步骤与ＭＤＨ无关,ＣＯ为盐对呼吸代谢影响的重要位点．另外,Ｋ＋、Ｎａ＋对蛋白表达的影响有差异,ＣＯ２可使盐胁迫下的碱蓬幼苗蛋白表达降低．     

miR-455-5p靶向SOCS3抑制RSV感染致气道上皮炎症反应的机制研究

摘要 目的：揭示miR-455-5p对呼吸道合胞病毒（RSV）感染致气道上皮细胞炎症反应的作用机制。方法：qRT-PCR检测30例健康体检儿童（健康组）、RSV感染轻症组（n=41）和重症组（n=31）患儿血清miR-455-5p水平。将16HBE细胞分为Control组、NC-agomir组、miR-455-5p-agomir组、NC-antagomir组、miR-455-5p-antagomir组。使用Lipofectamine 3000转染16HBE细胞后培养48 h后,分为Blank组、NC组（转染了NC-agomir的细胞）、RSV+NC-agomir组、RSV+miR-455-5p-agomir组,用RSV病毒液感染RSV+NC-agomir组和RSV+miR-455-5p-agomir组16HBE细胞,Blank组和NC组16HBE细胞正常培养。用CCK-8法和EdU法检测细胞增殖、TUNEL法检测细胞凋亡、ELISA法检测上清液中TNF-α、IL-6、IL-8水平,qRT-PCR检测miR-455-5p和SOCS3的mRNA水平,Western blot检测SOCS3、IFN-α、STAT1、STAT2、p-STAT1和p-STAT2的蛋白水平。结果：与健康组相比,轻症组和重症组患儿的血清miR-455-5p水平降低（P<0.05）。与轻症组相比,重症组的血清miR-455-5p水平降低（P<0.05）。与Control组和NC-agomir组相比,miR-455-5p-agomir组16HBE细胞的miR-455-5p水平、相对细胞活力和EdU阳性率升高（P<0.05）,TUNEL阳性率降低（P<0.05）,上清液中的TNF-α、IL-6和IL-8水平降低（P<0.05）,SOCS3 mRNA和蛋白水平降低（P<0.05）,IFN-α蛋白、STAT1和STAT2磷酸化水平升高（P<0.05）。与Control组和NC-antagomir组相比,miR-455-5p-antagomir组16HBE细胞的miR-455-5p水平、相对细胞活力和EdU阳性率降低（P<0.05）,TUNEL阳性率升高（P<0.05）,上清液中的TNF-α、IL-6和IL-8水平升高（P<0.05）,SOCS3 mRNA和蛋白水平升高（P<0.05）,IFN-α蛋白、STAT1和STAT2磷酸化水平降低（P<0.05）。与Blank组和NC组相比,RSV+NC-agomir组16HBE细胞的miR-455-5p水平、相对细胞活力和EdU阳性率降低（P<0.05）,TUNEL阳性率升高（P<0.05）,上清液中的TNF-α、IL-6和IL-8水平升高（P<0.05）,SOCS3 mRNA和蛋白水平升高（P<0.05）,IFN-α蛋白、STAT1和STAT2磷酸化水平降低（P<0.05）。与RSV+NC-agomir组相比,RSV+miR-455-5p-agomir组的miR-455-5p水平、相对细胞活力和EdU阳性率升高（P<0.05）,TUNEL阳性率降低（P<0.05）,上清液中的TNF-α、IL-6和IL-8水平降低（P<0.05）,SOCS3 mRNA和蛋白水平降低（P<0.05）,IFN-α蛋白、STAT1和STAT2磷酸化水平升高（P<0.05）。结论：miR-455-5p在RSV感染患儿血清中下调,上调miR-455-5p通过抑制SOCS3的转录和表达从而激活RSV感染的16HBE细胞中IFN-α介导的抗病毒反应。     

Control of Redox Balance by the Stringent Response Regulatory Protein Promotes Antioxidant Defenses of Salmonella

We report herein a critical role for the stringent response regulatory DnaK suppressor protein (DksA) in the coordination of antioxidant defenses. DksA helps fine-tune the expression of glutathione biosynthetic genes and discrete steps in the pentose phosphate pathway and tricarboxylic acid cycle that are associated with the generation of reducing power. Control of NAD(P)H/NAD(P)⁺ redox balance by DksA fuels downstream antioxidant enzymatic systems in nutritionally starving Salmonella. Conditional expression of the glucose-6-phosphate dehydrogenase-encoding gene zwf, shown here to be under DksA control, increases both the NADPH pool and antioxidant defenses of dksA mutant Salmonella. The DksA-mediated coordination of redox balance boosts the antioxidant defenses of stationary phase bacteria. Not only does DksA increase resistance of Salmonella against hydrogen peroxide (H₂O₂), but it also promotes fitness of this intracellular pathogen when exposed to oxyradicals produced by the NADPH phagocyte oxidase in an acute model of infection. Given the role of DksA in the adjustment of gene expression in most bacteria undergoing nutritional deprivation, our findings raise the possibility that the control of central metabolic pathways by this regulatory protein maintains redox homeostasis essential for antioxidant defenses in phylogenetically diverse bacterial species.     

Effect of some extrinsic factors on the respiratory metabolism of the freshwater Leech,Poecilobdella viridis: Oxygen tension,pH and salt concentrations

The effect of oxygen tension, pH and salt concentrations on the respiratory metabolism of the freshwater leech, Poecilobdella viridis has been studied. The respiratory rate increased significantly (p < 0.05) with progressive elevation of the oxygen tension of the medium and decreased significantly (p < 0.05) in both acidic and alkaline media. The respiratory rate was enhanced significantly (p < 0.05) as the salt concentration of the medium increased. Functional significance of these trends to the leech in counteracting the environmental eddies is briefly discussed.
