External zeitgebers synchronize the human circadian rhythm of sleep and wakefulness. Humans adapt their chronotype to the day-night cycle, the strongest external zeitgeber. The human circadian rhythm shifts to evening-type orientation when daylight is prolonged into the evening and night hours by artificial light sources. Data from a survey of 1507 German adolescents covering questions about chronotype and electronic screen media use combined with nocturnal satellite image data suggest a relationship between chronotype and artificial nocturnal light. Adolescents living in brightly illuminated urban districts had a stronger evening-type orientation than adolescents living in darker and more rural municipalities. This result persisted when controlling for time use of electronic screen media, intake of stimulants, type of school, age, puberty status, time of sunrise, sex, and population density. Time spent on electronic screen media use—a source of indoor light at night—is also correlated with eveningness, as well as intake of stimulants, age, and puberty status, and, to a lesser degree, type of school and time of sunrise. Adequate urban development design and parents limiting adolescents' electronic screen media use in the evening could help to adjust adolescents' zeitgeber to early school schedules when they provide appropriate lighting conditions for daytime and for nighttime. Land-based bivalve aquaculture depends on large-scale cultures of live microalgae for food. The intensity of large-scale microalgal production is important for cost-effectiveness. Using Walne’s medium as the control, simplified media containing nitrogen, phosphorus, silica, iron, manganese and vitamins were designed to determine the impact of nitrogen source and molar N:P ratio (sodium nitrate, NO3 9:1, ammonium chloride, NH4 9:1 and NH4 25:1) on growth, dry-weight biomass, culture longevity and lipid content of Chaetoceros muelleri, a diatom commonly used in shellfish aquaculture. During the exponential phase (day 6), dry-weight production in simplified media was similar to controls, indicating that this microalga can grow successfully on simplified media and use ammonium as the nitrogen source. The cultures grown on nitrate or ammonium differed in their time-course. Low nitrogen concentration in cultures grown with nitrate caused the collapse of these cultures within 11–13 days, after a short stationary phase. Cultures grown with ammonium had a longer stationary phase and were still alive on day 20, in spite of the low nitrogen concentrations observed after day 13 in cultures grown with NH4 9:1. During stationary phase (day 18) there was an increase in lipid content of algae under conditions of low nitrogen availability (NH4 9:1) and extended low phosphorus availability (NH4 25:1). Considering dry weight production, culture longevity, nutrient efficiency and lipid composition, simplified media containing ammonium, phosphorus, silica, iron, manganese and vitamins are a viable and profitable choice for batch culture of C. muelleri. In the exponential phase, the simplified medium NH4 9:1 was as effective as the control. Overall, both of the simplified media using ammonium are effective and suitable, depending on the purpose of the cultures and whether lipid contents (NH4 9:1), dry weight biomass (NH4 25:1) or nitrogen input and output (NH4 9:1) are desired.  相似文献

Almond β-glucosidase was used to catalyze the transglycosylation of p-nitrophenyl-β-d-glucoside to alkyl glucosides, with hydrolysis to glucose as a side reaction. The conversions were carried out in alcohols with varying water contents below water saturation. Both the total reaction rate and the ratio between the transglycosylation and hydrolysis increased with increasing water activity, and at a fixed water activity in the different alcohols, rate and transglycosylation/hydrolysis ratio increased in the following order: 1-octanol<1-hexanol<1-butanol. Synthesis of alkyl glucosides by transglycosylation in monophasic alcohol media is thus most favorable for short chain alcohols, and should be carried out at high water content.  相似文献

Typical preparation of seed samples for infrared (IR) microspectroscopy involves imbibition of the seed for varying time periods followed by cryosectioning. Imbibition, however, may initiate germination even at 4° C with associated changes in the chemistry of the sample. We have found that it is possible to section seeds that are sufficiently hard, such as soybeans, on a standard laboratory microtome without imbibition. The use of dry sectioning of unimbibed seeds is reported here, as well as a comparison of different mounting media and modes of analysis. Glycerol, Tissue-Tek, and ethanol were used as mounting media, and the quality of the resulting spectra was assessed. Ethanol was the preferred mountant, because it dried quickly with no residue and thus did not interfere with the spectrum of interest. Analysis in transmission mode using barium fluoride windows to hold the samples was compared with transmission-reflection analysis with sections mounted on special infrared-reflecting slides. The two modes of analysis performed well in different regions of the spectrum. The mode of analysis (transmission vs. transmission-reflection) should be based on the components of greatest interest in the sample.  相似文献

Synthesis of D-alanine oligopeptides from D-alanine methylester hydrochloride has been demonstrated by use of immobilized D-aminopeptidase from Ochrobactrum anthropi (Achromobacter sp.) in non-aqueous media. D-Alanine dimer and trimer were obtained in 56% and 6% yield, respectively, when 250 mM of the substrate was incubated for 3 hours with urethane-prepolymer immobilized D-aminopeptidase (1.5 U/ml) and 3 equivalents of triethylamine in water-saturated toluene. The kcat of this reaction was calculated to be 19,500 (min?1), which is several ten thousand times greater than that of the known enzymatic syntheses of amino acid oligomers.  相似文献

NAD+-dependent formate dehydrogenase (FDH, EC 1.2.1.2) is of use in the regeneration of NAD(P)H coenzymes, and therefore has strong potential for practical application in chemical and medical industries. A low-cost production of recombinant Escherichia coli (E. coli) containing FDH from Candida methylica (cmFDH) was optimized in molasses-based medium by using response surface methodology (RSM) based on central composite design (CCD). The beet molasses as a sole carbon source, (NH4)2HPO4 as a nitrogen and phosphorus source, KH2PO4 as a buffer agent, and Mg2SO4 · 7H2O as a magnesium and sulfur source were used as variables in the medium. The optimum medium composition was found to be 34.694 g L?1 of reducing sugar (equivalent to molasses solution), 8.536 g L?1 of (NH4)2HPO4, 3.073 g L?1 of KH2PO4, and 1.707 g L?1 of Mg2SO4 · 7H2O. Molasses-based culture medium increased the yield of cmFDH about three times compared to LB medium. The currently developed media has the potential to be used in industrial bioprocesses with low-cost production.  相似文献

A rapid and simple method for the detection of drug-resistant Mycobacterium tuberculosis is critical for the efficient treatment and control of this pathogen in developing country. Here we developed a single multiplex amplification refractory mutation system (M-ARMS) PCR, in which chimeric-primer and temperature switch PCR (TSP) strategy were included. Using this method, we detected rifampin resistance-associated mutations at codons 511, 516, 526 and 531 in the rifampin resistance-determining region of rpoB gene. The performance of M-ARMS-PCR assay was evaluated with 135 cultured isolates of M. tuberculosis. The sensitivity and specificity were 94.2% and 100%, respectively, compared with direct DNA sequencing, and 86.67% and 89.71%, respectively, compared with culture-based phenotypic drug susceptibility testing. Therefore, this newly-developed M-ARMS-PCR method is useful and efficient with an intended application in provincial Centers for Disease Control and Prevention for rapid detection of rifampin resistance-associated mutations.  相似文献

Tissue‐depolarization and linear‐retardance are the main polarization characteristics of interest for bulk tissue characterization, and are normally interpreted from Mueller polarimetry. Stokes polarimetry can be conducted using simpler instrumentation and in a shorter time. Here, we use Stokes polarimetric imaging with circularly polarized illumination to assess the circular‐depolarization and linear‐retardance properties of tissue. Results obtained were compared with Mueller polarimetry in transmission and reflection geometry, respectively. It is found that circular‐depolarization obtained from these 2 methods is very similar in both geometries, and that linear‐retardance is highly quantitatively similar for transmission geometry and qualitatively similar for reflection geometry. The majority of tissue circular‐depolarization and linear‐retardance image information (represented by local image contrast features) obtained from Mueller polarimetry is well preserved from Stokes polarimetry in both geometries. These findings can be referred to for further understanding tissue Stokes polarimetric data, and for further application of Stokes polarimetry under the circumstances where short acquisition time or low optical system complexity is a priority, such as polarimetric endoscopy and microscopy.   相似文献
