Activity periods and diurnal behaviour of southern white rhino (Ceratotherium simum simum) were studied in Matobo National Park, Zimbabwe, from August 2016 to July 2017 using camera trapping and focal group sampling, respectively. Camera trap data showed that diurnal and nocturnal movements of white rhino varied significantly with moon phase, with more frequent nocturnal photographic captures during full- and third-quarter moon phases than during new and crescent moon phases. Permutational MANOVA and canonical correspondence analysis of focal group observation data were used to test the effects of season, time of day, habitat type, distance to roads, moon phase, presence/absence of other mammals, age/sex category, individual identity and herd composition on daytime behaviour. Weather conditions accounted for the majority of variation in rhino behaviour, with more time spent feeding during cloudy and rainy weather than sunny weather. Time of day and moon phase also accounted for high variation in behaviour, followed by habitat attributes and age/sex category. Variance partitioning by sets of explanatory variables showed that weather conditions had the greatest explanatory power in the variation of rhino behaviour, followed by temporal attributes, habitat attributes and age/sex category. Our findings provide insights for white rhino population management strategies aimed at increasing the effectiveness of security patrols, minimising disturbance and improving sightings for tourists.  相似文献

Background

Campylobacter jejuni is an important food-borne and zoonotic pathogen with a worldwide distribution. Humans and chickens are hosts of this pathogen. At present, there is no ideal vaccine for controlling human campylobacteriosis or the carriage of C. jejuni by chickens. Bacterial in vivo-induced antigens are useful as potential vaccine candidates and biomarkers of virulence.

Methods

In this study, we developed a novel systematic immunoproteomics approach to identify in vivo-induced antigens among the total cell proteins of C. jejuni using pre-adsorbed sera from patients infected with C. jejuni.

Results

Overall, 14 immunoreactive spots were probed on a PVDF membrane using pre-adsorbed human sera against C. jejuni. Then, we excised these protein spots from a duplicate gel and identified using MALDI–TOF MS. In total, 14 in vivo-induced antigens were identified using PMF and BLAST analysis. The identified proteins include CadF (CadF-1 and CadF-2), CheW, TufB, DnaK, MetK, LpxB, HslU, DmsA, PorA, ProS, CJBH_0976, CSU_0396 and hypothetical protein cje135_05017. Real-time RT-PCR was performed on 9 genes to compare their expression levels in vivo and in vitro. The data showed that 8 of the 9 analyzed genes were significantly upregulated in vivo relative to in vitro.

Conclusion

We successfully developed a novel immunoproteomics method for identifying in vivo-induced Campylobacter jejuni antigens by using pre-adsorbed sera from infected patients.

General significance

This new analysis method may prove to be useful for identifying in vivo-induced antigens within any host infected by bacteria and will contribute to the development of new subunit vaccines.  相似文献

Termites from the genus Odontotermes are known to contain numerous species complexes that are difficult to tell apart morphologically or with mitochondrial DNA sequences. We developed markers for one such cryptic species complex, that is, Odontotermes srinakarinensis sp. nov. from Maxwell Hill Forest Reserve (Perak, Malaysia), and characterised them using a sample of 41 termite workers from three voucher samples from the same area. We then genotyped 150 termite individuals from 23 voucher samples/colonies of this species complex from several sites in Peninsular Malaysia. We analysed their population by constructing dendograms from the proportion of shared-alleles between individuals and genetic distances between colonies; additionally, we examined the Bayesian clustering pattern of their genotype data. All methods of analysis indicated that there were two distinct clusters within our data set. After the morphologies of specimens from each cluster were reexamined, we were able to separate the two species morphologically and found that a single diagnostic character found on the mandibles of its soldiers could be used to separate the two species quite accurately. The additional species in the clade was identified as Odontotermes denticulatus after it was matched to type specimens at the NHM London and Cambridge Museum of Zoology.  相似文献

Ethnoprimatology is an important and growing discipline, studying the diverse relationships between humans and primates. However there is a danger that too great a focus on primates as important to humans may obscure the importance of other animal groups to local people. The Waorani of Amazonian Ecuador were described by Sponsel [Sponsel (1997) New World Primates: Ecology, evolution and behavior. New York: Aldine de Gruyter. p 143–165] as the “natural place” for ethnoprimatology, because of their close relationship to primates, including primates forming a substantial part of their diet. Therefore they are an ideal group in which to examine contemporary perceptions of primates in comparison to other types of animal. We examine how Waorani living in Yasuní National Park name and categorize primates and other common mammals. Although there is some evidence that the Waorani consider primates a unique group, the non‐primate kinkajou and olingo are also included as part of the group “monkeys,” and no evidence was found that primates were more important than other mammals to Waorani culture. Instead, a small number of key species, in particular the woolly monkey (Lagothrix poeppigii) and white‐lipped peccary (Tayassu pecari), were found to be both important in the diet and highly culturally salient. These results have implications for both ethnoprimatologists and those working with local communities towards broader conservation goals. Firstly, researchers should ensure that they and local communities are referring to the same animals when they use broad terms such as “monkey,” and secondly the results caution ethnoprimatologists against imposing western taxonomic groups on indigenous peoples, rather than allowing them to define themselves which species are important. The paper ‘Bird diversity in the savanna habitats of Akagera National Park, Rwanda, in the post-war recovery period’ by Gatali and Wallin (Ostrich 86(3): 267–276, 2015) makes several claims for new species records for Akagera National Park and Rwanda. We found that Gatali and Wallin recorded several species considered very rare in Rwanda and new discoveries; these findings often contradicted the existing literature. We doubt the validity of 34 species records and we aim to demonstrate that these claims are mostly the result of misidentification or human error and consider that future researchers should use their findings with caution. Some findings around more common species may be validated through further studies and we encourage others to conduct fieldwork in Akagera National Park.  相似文献

Les bourdons constituent l’un des groupes de pollinisateurs les plus importants dans les écosystèmes montagnards. Cependant, la faune des bourdons du Parc National des Pyrénées occidentales (PNPO) est encore peu connue. Pendant trois ans, la faune des bourdons du Parc National des Pyrénées occidentales a fait l’objet d’une surveillance. Les inventaires effectués en juillet- août 2002, 2003 et 2005 ont permis l’observation de 5889 spécimens de bourdons de 29 espèces. Si l’on tient compte des observations des cinquante dernières années, la diversité spécifique du parc s’élève à 30 espèces de bourdons. Une telle diversité spécifique est remarquable et comparable à celle observée dans d’autres secteurs du massif pyrénéen. La faible différence entre les faunes du Parc et des réserves naturelles d’Eyne et de Nohèdes (Pyrénées-Orientales) rend compte du caractère exceptionnellement diversifié de la faune des bourdons du massif pyrénéen en général.  相似文献

An investigation of population structure and ecology of the African buffalo (Syncerus caffer Sparrman, 1779) was carried out in the Chebera Churchura National Park, Ethiopia during the wet and dry seasons of 2005–2006. Sample counts of African buffaloes were carried out in an area of 1,215 km2. The estimated population of buffalo was 2,617 individuals. Males comprised 35.0%, while females 52.8% of the population. The remaining 12% of the population was young of both sexes and of all ages of unknown sex. It was difficult to categorize the young into male and female in the field, as their primary sexual characteristics were not easily visible. Male to female sex ratio was 1.00 : 1.51. Age structure was dominated by adults, which constituted 72.06% of the total population. Subadults comprised 22.02%, and young accounted for 5.9% of the population. Larger herds of up to 27 individuals were observed during the wet season and smaller herds of eight individuals during the dry season. The mean herd size during wet and dry seasons was 24.81 and 7.77, respectively. The African buffaloes were distributed in four habitat types such as grasslands, woodlands, montane forests and riverine habitat in the study area. They were observed more in the riverine vegetation types during the dry season. Relative abundance of food resources, green vegetation cover and water availability in the area were the major factors governing their distribution in the present study area. They spent a greater proportion of the time in feeding and resting/ruminating activities. On the average, 49.7% of the daytime was spent in feeding, and 38.75% of the day on resting (lying down and standing). Morning and the late afternoon activity peaks were more pronounced during the dry season than the wet season.  相似文献

In the year 2013, the authors, who had already visited some Japanese aquariums and zoos in 2012 (Lange & Tai, 2012), revisited several Japanese aquariums and zoos. They were accompanied by two colleagues from Basle Zoo and the Monterey Bay Aquarium, who joined the tour.Their goals of the tour were focused on new innovative husbandry methods, on new aquarium buildings and new zoo enclosures as well as on rare Japanese endemic animals, which are never or seldom seen outside of Japan.The perfect new aquariums in Kyoto and Kitami City (Hokkaido), but also the very specialized aquariums like the Kamo Aquarium (for jellyfish) in Yamagata, the Chitose Salmon Aquarium (for salmons), the Otaru Aquarium (for sea mammals) and the Umi Kirara Kujukushima Aquarium (for the fauna of the Kujukushima island National Park) have been destinations for this trip. Besides the aquariums also the Asahiyama Zoo (new enclosures for endemic animals of Hokkaido), Kobe Oji Zoo, Kushiro Zoo (many endemic Hokkaido species), Nagasaki Bio Park, Sapporo Maruyama Zoo (new area for South East Asian animals) and the Sasebo Zoological & Botanical Garden were visited.During this tour it was obvious that neither the colleagues in Japan nor the staff in the zoos outside of Japan know sufficient enough about the conception and the husbandry methods in the different institutions in this part of the world. Therefore it is recommendable to organize such tours often for more people, thus both sides can benefit and learn from each other. This again will influence to help to keep the animals better, to attract more visitors in order to inspire them for the conservation of nature and environment.  相似文献

National parks are an important tool for conserving biodiversity, particularly in areas of high biodiversity and endemism such as the tropical Andes. However, national parks often face a variety of stressors related to recreation, road construction and illegal extraction of natural resources. Unfortunately, the influence of these stressors for biodiversity is rarely well documented. Cajas National Park in Ecuador is no exception. Despite being traversed by the Cuenca-Molleturo-Naranjal road, effects of the road construction on biodiversity have not been determined. We therefore assessed the influence of road proximity on bird species richness and abundance as well as composition of bird habitat groups in Cajas National Park using transect walks at 25 m and 250 m distance to the road (overall 18 transects, each 1 km length). In total, we recorded 1110 individuals of 28 páramo bird species. Overall species richness did not differ between transects near and far from the road. Nevertheless, the average abundance of shrubby páramo species was significantly higher far from the road than near the road (Far = 36, Near = 25). Moreover, we found a tendency towards differences in the composition of bird habitat groups between transects near and far from the road. One aspect potentially driving the observed patterns was the increasing proportion of planted non-native woody tree species within páramo grassland near the road, which may have caused reduced abundances of shrubby páramo bird species there. While roads represented a clear impact on the composition of bird species in the páramo, the major effect seems to be driven by the introduction of non-native plant species along the roadside. In order to reduce the impact of roads to a minimum, we suggest that park managers should control the introduction of such plant species.  相似文献
