Most dentate vertebrates, from fish to humans, replace their teeth and yet the molecular basis of tooth replacement is poorly understood. Canonical Wnt signaling regulates tooth number in mice and humans, but it is unclear what role it plays in tooth replacement as it naturally occurs. To clarify this, we characterized Wnt signaling activity in the dental tissues of the ball python Python regius. This species replaces teeth throughout life (polyphyodonty) and in the same manner as in humans, i.e., sequential budding of teeth from the tip of the dental lamina. From initiation stage onwards, canonical Wnt read-out genes (Lef1 and Axin2) are persistently expressed by cells in the dental lamina tip and surrounding mesenchyme. This implies that molecular signaling at work during dental initiation carries over to tooth replacement. We show that canonical Wnt signaling promotes cell proliferation in python dental tissues and that by confining Wnt activity in the dental lamina the structure extends instead of thickens. Presumably, lamina extension creates space between successive tooth buds, ensuring that tooth replacement occurs in an ordered manner. We suggest that hedgehog signaling confines Wnt activity in the dental epithelium by direct planar repression and, during tooth replacement stages, by negatively regulating BMP levels in the dental mesenchyme. Finally, we propose that Wnt-active cells at the extending tip of the python dental lamina represent the immediate descendents of putative stem cells housed in the lingual face of the lamina, similar to what we have recently described for another polyphyodont squamate species.  相似文献

Fractionation of the β-endorphin-sized material from freshly dissected reptile intermediate pituitaries by ion exchange chromatography on sulfopropyl Sephadex (SP) revealed at least three distinct forms of immunoreactive β-endorphin. These forms eluted at 0.25 M NaCl, 0.28 M NaCl, and 0.32 M NaCl and represent respectively, 6%, 65% and 29% of the total immunoreactivity. Only the 0.28 M NaCl peak and the 0.32 M NaCl peak exhibited naloxone reversible opiate bioactivity when tested in the isolated guinea pig ileum bioassay system; taking into account the molar amount of immunoreactive peptides the 0.32 M NaCl peak was 6 fold more potent than the 0.28 M NaCl peak. Intermediate pituitaries in culture were incubated with either [3H]tyrosine, [3H]arginine, or [35S]methionine for periods up to 24 hours and β-endorphin-sized peptides were prepared by immunoprecipitation and gel filtration. Fractionation of the labeled β-endorphin-sized peptides by ion exchange chromatography yielded profiles nearly identical to the immunoassay analyses of freshly dissected tissue. Further analysis of the major labeled forms of reptile β-endorphin by chromatography on Sephadex G-50 equilibrated in 6 M guanidine HCl indicated that the 0.32 M NaCl peak had an apparent molecular weight of 3500±100 and the 0.28 M NaCl peak had an apparent molecular weight of 3200±100. Furthermore, pulse/chase experiments showed that the 0.32 M NaCl peak was the precursor for the 0.28 M NaCl peak. These results coupled with the relative opiate bioactivities of the major forms argue that the principal post-translational modification of reptile β-endorphin is COOH-terminal proteolytic cleavage.  相似文献

Since the beginning of this century evidence has accumulated which demonstrates that non-mammalian vertebrates possess photoreceptors situated deep within the brain. While many attempts have been made to localize these sensory cells, studies have either failed or been inconclusive. In this report we have used several experimental approaches to localize the deep brain photoreceptors of the lizard Anolis carolinensis. Using 3 antibodies that bind vertebrate cone opsins, we have immunolabelled cerebrospinal fluid (CSF)-contacting neurons located at the ventricular border within the nucleus ventromedialis of the septum. Western blot analysis indicates that these antibodies recognized a single 40 kD protein in ocular, anterior brain, and pineal extracts. Immunoblots of rodent brain did not show a similar protein band. We have also identified specific retinoids associated with phototransduction (11-cis and all-trans-3,4-didehydroretinaldehyde) within anterior brain extracts. This combined data provides the most detailed analysis of deep brain photoreceptors in any vertebrate. In the viviparous skink Niveoscincus metallicus clutch size appears to be determined before vitellogenesis, and is not altered later by follicular atresia or embryonic loss. This suggests that the number of follicles recruited is determined by the endocrine environment early in the vitellogenic period. Through a series of experiments in which we manipulated gonadotropin concentrations by administering exogenous FSH, we aimed to investigate this hypothesis. Pre-vitellogenic females showed no response to exogenous ovine FSH. In early vitellogenic females, FSH induced follicular recruitment: follicles were enlarged and clutch size increased by recruitment of a second cohort of follicles; some females also ovulated. Females treated with FSH in mid-vitellogenesis had elevated mean plasma estradiol concentrations compared to controls; no follicular recruitment was observed, but most of these animals ovulated. Females treated with a range of doses of FSH in late vitellogenesis ovulated at least one month before natural ovulation, again without recruitment of extra follicles. It appears therefore that in Niveoscincus metallicus exogenous FSH can induce recruitment of additional follicles only if administered during early vitellogenesis. We conclude that in this species clutch size is determined by proximate environmental factors influencing gonadotropin levels early in follicular recruitment, and cannot be increased even if conditions become more favourable once vitellogenesis is established.  相似文献

Summary By using various staining techniques, two acidophilic and three basophilic cell-types have been distinguished in the pars distalis of Trionyx sinensis. Seasonal fluctuations in their cytoplasmic granulation, cell and nuclear size have been compared with seasonal patterns of activity in the gonads, adrenals and thyroid. On the basis of the results, specific functions have been ascribed to each cell-type. Basophils 1 activity closely parallels the seasonal thyroid cycle. Basophils 2 are probably FSH-secreting gonadotropes and their activity correlates with the seasonal cycle of spermatogenesis in the seminiferous tubules. The functional activity of the testicular interstitial cells correlates with the secretory pattern of the basophils 3. That these are LH-secreting cells is also suggested by the epididymidal cycle, which indicates a high level of androgen secretion coincident with the rapid degranulation of these cells, at a time of no spermatogenetic activity. Adrenal activity shows no relationship with the basophils 3 cycle, and chromophobic cells may be the locus of ACTH secretion. Acidophils 2 remain stable throughout the year and are probably somatotropes, but acidophils 1 increase their activity in autumn and are likely to be lactotropes.  相似文献

Summary This study examined whether the mating or feeding success of females of two tick species, Amblyomma limbatum and Aponomma hydrosauri, is influenced by their site of attachment on a host. Marked interspecific differences were detected in the preferred sites of attachment of females to their common host, the sleepy lizard Tiliqua rugosa, with Amb. limbatum found predominantly in the ears and on the lower back, while Ap. hydrosauri preferred to attach under the forelimbs and on the midback. Data from field surveys suggested that higher proportions of females were mated in the preferred sites for Ap. hydrosauri, and in one of the preferred sites for Amb. limbatum. However, laboratory and field experiments showed that for both species, the mean time before females were mated, mean engorgement time and mean feeding rate were independent of where they were attached to a host. Four hypotheses were considered as explanations of the observed niche segregation; intespecific competition, avoidance of pheromone jamming or adaptations, to maximize mating and feeding, and minimize physical disturbance. These could not totally explain the observed site specificity in either tick species. Site specificity may represent adaptations by each species to other host species encountered within their distributional range.  相似文献

1The decrease of temperatures along an elevation gradient imposes physiological constraints on reptiles that ultimately determine their distribution ranges. Forest patterns are likely to interact with this process, but very few studies have examined their contribution in determining distribution limits.2We examined the role played by thermal physiology and forest cover in determining the elevational ranges of a lizard, Eutropis longicaudata. We integrated this species’ thermal traits in simulating its maximum activity time under different conditions of forest cover and elevation using a NicheMapR model. In addition, we evaluated the influence of winter temperatures on the range limit by examining the simulated soil temperatures at the occurrence sites.3Laboratory experiments showed that E. longicaudata has a high preferred body temperature and low cold tolerance. The model predicts that maximum activity time decreases with elevation and forest cover. Although unforested areas may provide longer active time in all simulated elevations, mountain areas in Taiwan are heavily forested and are predicted to allow only a very short period of activity above 1000 m elevation.4All sightings were indeed located in areas below 1000 m elevation, in which the predicted average soil temperature is above 10 °C in January in cold years.5Our results show that reptile physiological response does respond strongly to the change of microclimate induced by forest cover and elevation. Overall, this suggests that forest cover is a major determinant of some reptiles’ elevational range.  相似文献

Summary Aponomma hydrosauri and Amblyomma limbatum are tick species with the same major host species, the sleepy lizard Trachydosaurus rugosus, but parapatric distributions in South Australia. Near Mt. Mary one species is abruptly replaced by the other over one kilometre. There is substantial overlap in feeding sites on the hosts. This suggested that interspecific competition, during feeding, may be an important mechanism preventing further overlap of the distributions of the two species. However, in the overlap region there was no resource shortage, and each species infested hosts independently of the other species. Also in laboratory experiments at the highest densities encountered in the field, larvae and nymphs of each species attached and engorged equally well whether alone or mixed with the other species. These data refuted the, hypothesis of interspecific competition between the two species while feeding.  相似文献

Egg whites of three species of tortoise and turtle have been compared by gel chromatography for inhibitory activity against proteases. The egg white of Geomda trijuga trijuga Schariggar contains trypsin/subtilisin inhibitor while the egg white of Caretta caretta Linn. contains both trypsin and chymotrypsin inhibitors. No protease inhibitory activity has been detected in the egg white of Trionyx gangeticus Cuvier. An acidic trypsin/subtilisin inhibitor has been purified to homogeneity from the egg white of tortoise (G. trijuga trijuga). It is a single polypeptide chain of 100 amino acid residues, having a molecular weight of 11 700. It contains six disulphide bonds and is devoid of methionine and carbohydrate moiety. Its isoelectric point is at pH 5.95 and is stable at 100°C for 4 h at neutral pH. The inhibitor inhibits both trypsin and subtilisin by forming enzyme-inhibitor complexes at a molar ratio close to unity. Their dissociation contants are 7.2·10?9 M for bovine trypsin adn 5.5·10?7 M for subtilisin. Chemical modification of amino groups with trinitrobenzene sulfonate has reduced its inhibitory activities against both trypsin and subtilisin, but the loss of its trypsin inhibitory activity is faster than of its subtilisin inhibitory activity. It has independent binding sites for inhibition of trypsin and subtilisin.  相似文献

Blood pythons in northeastern Sumatra display a series of discrete colour morphs, even among hatchlings within a single clutch. The first step towards understanding the maintenance of this polymorphism is to test the null hypothesis that colour variation in this species has no major biological correlates. Data on >2,000 blood pythons killed for the commercial leather industry enabled us to test, and reject, this hypothesis. The four colour morphs differed significantly in most of the traits that we measured, including temporal and spatial abundances, sex ratios, age structures, mean adult body sizes, body shapes (tail length and body mass relative to snout-vent length), energy stores, numbers of gut parasites, prey types, feeding frequencies and clutch sizes. The causal basis for these associations remains unclear, but is likely to involve three processes: direct effects of colour, linkages between genes for colour and other traits, and correlated spatial heterogeneity in colour, morphology and ecology. The colour polymorphism may be maintained by frequency-dependent selection and genotype-specific habitat selection, because these sedentary ambush predators are under strong selection for effective camouflage to hide them from both predators and potential prey. In support of this hypothesis, similar colour polymorphisms have evolved independently in several other snake taxa that rely upon ambush predation.
