Kinetic regularities of the enzymatic acyl group transfer reactions have been studied using ampicillin synthesis catalyzed by E. coli penicillin acylase as an example. It was shown that ampicillin synthesis proceeds through the formation of an acylenzyme–nucleophile complex capable of undergoing hydrolysis. The relative nucleophile reactivity of 6-aminopenicillanic acid (6-APA) is a complex parameter dependent on the nucleophile concentration. The kinetic analysis showed that the maximum yield of antibiotic being synthesized depended only on the nucleophile reactivity of 6-APA, the ratio between the enzyme reactivities with respect to the target product and acyl donor, and the initial concentrations of reagents. The parameters characterizing the nucleophile reactivity of 6-APA have been determined. The algorithm of modeling the enzymatic synthesis has been elaborated. The proposed algorithm allows the kinetics of the process not only in homogeneous, but also in heterogeneous (aqueous solution–precipitate) systems to be quantitatively predicted and described based on experimental values of parameters of the reaction. It was shown that in heterogeneous aqueous solution–precipitate systems PA-catalyzed ampicillin synthesis proceeds much more efficiently compared to the homogeneous solution.  相似文献

This study examined the influence of birth date, salivary testosterone [sT] concentration, sexual maturity status, and general strength on the selection process of an elite Brazilian soccer club during a 12-month period, which was a 2nd phase of a 24-month selection process. The stature, body mass, sT, sexual maturity status [Tanner scale], and hand grip strength were assessed for 143 players during 2 weeks. From these 143 players, 100 players were dismissed [DIS] and 43 players were selected to integrate the club’s under-14 squad. Following 1-year training period, the under-14 team was assembled with 9 players designated as starters [STA], and then, comparisons were conducted taking into account a group of non-starters (selected; SEL = 34 players) and STA (n = 09). The DIS, SEL, STA players, and reference population, were compared for birth distribution. A greater proportion of players was born in the first trimester in the STA [75.0%], SEL [57.1%] and DIS [50.0%] groups compared to the reference population [25.8%]. One-way ANOVA showed a higher sT for STA group [516.0 ± 129.9 pmol·L-1], compared to SEL [415.5 ± 117.9 pmol·L-1] and DIS groups [390.9 ± 84.9 pmol·L-1; p = 0.003), and Kruskall-Wallis test showed a higher gonadal development for STA compared to DIS [p = 0.001]. The current findings suggest a strong influence of birth date and biological maturation on young soccer players selection process. Soccer coaches should consider these influences when making decisions about player selection of elite youth players.  相似文献

摘要 目的:探讨与分析慢性阻塞性肺疾病(COPD)患者运动负荷气道反应性与T细胞亚群的关系。方法:2020年1月到2022年4月选择在本院诊治的慢阻肺患者88例作为慢阻肺组,同期选择在本院进行健康体检者88例作为健康组,检测两组T细胞亚群含量,判定两组的运动负荷气道反应性情况并进行相关性分析。结果:慢阻肺组的CD8+T淋巴细胞比例明显高于健康组,CD3+T淋巴细胞、CD4+T淋巴细胞比例明显低于健康组(P<0.05)。慢阻肺组的运动负荷气道反应性发生率为20.9 %,明显高于健康组的1.2 %(P<0.05)。在慢阻肺中,Spearsman分析显示运动负荷气道反应性发生率与CD3+T淋巴细胞、CD4+T淋巴细胞、CD8+T淋巴细胞比例存在相关性(P<0.05)。logistic回归分析显示CD3+T淋巴细胞、CD4+T淋巴细胞、CD8+T淋巴细胞比例都为影响运动负荷气道反应性发生的重要危险因素(P<0.05)。结论:慢阻肺患者多伴随有T细胞亚群异常,也多伴随有运动负荷气道反应性,运动负荷气道反应性与T细胞亚群存在相关性,也表明T细胞亚群紊乱是导致运动负荷气道反应性发生的重要因素。  相似文献

Streptococcus uberis is an environmental pathogen associated with subclinical and clinical IMI in both lactating and non-lactating cows. RC19 strain was isolated from a cow with subclinical mastitis, qualitatively classified as moderate biofilm producer in Todd Hewitt medium (THB), and it showed a high value of the adhered bacteria (CFU/ml). Hence, the aims of this study were (a) to determine ability to adhere to and internalize into epithelial cells MAC-T for 1, 2 and 3 h, (b) to evaluate the relative expression of adherence-associated genes from co-cultures of S. uberis with MAC-T cells at 1, 2 and 3 h. We hypothesized that upon contact with bovine mammary epithelial cells, S. uberis upregulates adherence-associated genes encoding adhesins, which enable it a higher adherence to and/or internalization into host cells. Four to six genes increased their R with regard to the control after initial contact with MAC-T cells (group 1) at 1, 2 and 3 h. The highest value of R was observed at 2 h after co-culture between RC19 and MAC-T cells.  相似文献

ObjectivesThe aim of the current study is to evaluate the accuracy and the precision of raw-data-based relative electron density (REDraw) and the calibration-based RED (REDcal) at a range of low-RED to high-RED for tissue-equivalent phantom materials by comparing them with reference RED (REDref) and to present the difference of REDraw and REDcal for the contrast medium using dual-energy CT (DECT).MethodsThe REDraw images were reconstructed by raw-data-based decomposition using DECT. For evaluation of the accuracy of the REDraw, REDref was calculated for the tissue-equivalent phantom materials based on their specified density and elemental composition. The REDcal images were calculated using three models: Lung-Bone model, Lung-Ti model and Lung-Ti (SEMAR) model which used single-energy metal artifact reduction (SEMAR). The difference between REDraw and REDcal was calculated.ResultsIn the titanium rod core, the deviations of REDraw and REDcal (Lung-Bone model, Lung-Ti model and Lung-Ti model with SEMAR) from REDref were 0.45%, 50.8%, 15.4% and 15.0%, respectively. The largest differences between REDraw and REDcal (Lung-Bone model, Lung-Ti model and Lung-Ti model with SEMAR) in the contrast medium phantom were 8.2%, ?23.7%, and 28.7%, respectively. However, the differences between REDraw and REDcal values were within 10% at 20 mg/ml. The standard deviation of the REDraw was significantly smaller than the REDcal with three models in the titanium and the materials that had low CT numbers.ConclusionThe REDcal values could be affected by beam hardening artifacts and the REDcal was less accurate than REDraw for high-Z materials as titanium.Advances in knowledgeThe raw-data-based reconstruction method could reduce the beam hardening artifact compared with image-based reconstruction and increase the accuracy for the RED estimation in high-Z materials, such as titanium and iodinated contrast medium.  相似文献

Gentle handling seems to elicit positive states in sheep. The study investigated whether spatial distance alters sheep responses to brushing and whether spatial distance is influenced by reactivity. Twenty Romane ewes were assessed in three sessions: in Sessions 1 and 3, one grid separated the test animal from pen mates, with no distance between them, and in Session 2 two grids separated the test animal from pen mates by a distance of about 1.7 m. Ewes had been genetically selected for low (R−) or high (R+) behavioural reactivity to social isolation. Body postures, head orientation, ear postures, closed and half-closed eyes, tail wagging and feeding behaviour, in addition to heart rate (HR) and HR variability, as the root mean square of successive differences (RMSSD), standard deviation of all normal-to-normal (NN) intervals (SDNN), RMSSD/SDNN ratio and ratio between low-frequency (LF) and high-frequency (HF) powers (LF/HF) were assessed. Data were analysed using generalized linear models and linear mixed models. Session, genetic line and phase (pre-, brushing and post-brushing) were considered fixed effects. Increased distance in Session 2 might not have influenced ewes’ responses. Fewer changes in ear postures were noted in Session 3 than 1 (P<0.01), suggesting that ewes were more relaxed in Session 3. The RMSSD/SDNN ratio was higher mainly during brushing in Sessions 1 and 3 (P<0.05), indicating that ewes were more relaxed during brushing, and at no distance between pen mates. However, spatial distance influenced R− and R+ ewes’ responses; R+ ewes performed more asymmetric ear postures in Session 2 than 1 and 3 (P<0.01), and in Session 3 than 1 (P<0.01), indicating that spatial distance had a negative effect on R+ ewes. Low reactive ewes spent less time on horizontal ear postures in Session 2 than 1 and 3 (P<0.01), and R+ ewes spent more time on horizontal postures in Session 1 than 3 (P<0.01). Curiously, R− ewes spent more time eating and ruminating in Session 3 than 1 (P<0.01), and in Session 2 than 1 and 3 (P<0.01), whereas R+ ewes ate and ruminated more in Session 1 than 3 (P<0.05). Higher HR was found among R− ewes in Session 2 than 1 and 3, and in Session 3 than 1 (P<0.01). High reactive ewes showed higher HR in Session 1 than 3 (P<0.01). The findings suggest that the social context might influence sheep responses to gentle handling, and the effects depend on their reactivity traits.  相似文献

Standard presentations of epidemiological results focus on incidence-ratio estimates derived from regression models fit to specialized study data. These data are often highly nonrepresentative of populations for which public-health impacts must be evaluated. Basic methods are provided for interval estimation of attributable fractions from model-based incidence-ratio estimates combined with independent survey estimates of the exposure distribution in the target population of interest. These methods are illustrated in estimation of the potential impact of magnetic-field exposures on childhood leukemia in the United States, based on pooled data from 11 case-control studies and a U.S. sample survey of magnetic-field exposures.  相似文献

In Bayesian and empirical Bayes analyses of epidemiologic data, the most easily implemented prior specifications use a multivariate normal distribution for the log relative risks or a conjugate distribution for the discrete response vector. This article describes problems in translating background information about relative risks into conjugate priors and a solution. Traditionally, conjugate priors have been specified through flattening constants, an approach that leads to conflicts with the true prior covariance structure for the log relative risks. One can, however, derive a conjugate prior consistent with that structure by using a data-augmentation approximation to the true log relative-risk prior, although a rescaling step is needed to ensure the accuracy of the approximation. These points are illustrated with a logistic regression analysis of neonatal-death risk.  相似文献

The traditional definition of a confidence interval requires the coverage probability at any value of the parameter to be at least the nominal confidence level. In constructing such intervals for parameters in discrete distributions, less conservative behavior results from inverting a single two-sided test than inverting two separate one-sided tests of half the nominal level each. We illustrate for a variety of discrete problems, including interval estimation of a binomial parameter, the difference and the ratio of two binomial parameters for independent samples, and the odds ratio.  相似文献
