PI3K/Akt signaling plays an important role in the regulation of cardiomyocyte death machinery, which can cause stress-induced cardiac dysfunction. Here, we report that apoptosis regulator through modulating IAP expression (ARIA), a recently identified transmembrane protein, regulates the cardiac PI3K/Akt signaling and thus modifies the progression of doxorubicin (DOX)-induced cardiomyopathy. ARIA is highly expressed in the mouse heart relative to other tissues, and it is also expressed in isolated rat cardiomyocytes. The stable expression of ARIA in H9c2 cardiac muscle cells increased the levels of membrane-associated PTEN and subsequently reduced the PI3K/Akt signaling and the downstream phosphorylation of Bad, a proapoptotic BH3-only protein. When challenged with DOX, ARIA-expressing H9c2 cells exhibited enhanced apoptosis, which was reversed by the siRNA-mediated silencing of Bad. ARIA-deficient mice exhibited normal heart morphology and function. However, DOX-induced cardiac dysfunction was significantly ameliorated in conjunction with reduced cardiomyocyte death and cardiac fibrosis in ARIA-deficient mice. Phosphorylation of Akt and Bad was substantially enhanced in the heart of ARIA-deficient mice even after treatment with DOX. Moreover, repressing the PI3K by cardiomyocyte-specific expression of dominant-negative PI3K (p110α) abolished the cardioprotective effects of ARIA deletion. Notably, targeted activation of ARIA in cardiomyocytes but not in endothelial cells reduced the cardiac PI3K/Akt signaling and exacerbated the DOX-induced cardiac dysfunction. These studies, therefore, revealed a previously undescribed mode of manipulating cardiac PI3K/Akt signaling by ARIA, thus identifying ARIA as an attractive new target for the prevention of stress-induced myocardial dysfunction.  相似文献

嗜热厌氧乙醇杆菌发酵是生产纤维素乙醇的有效途径,而嗜热厌氧乙醇杆菌醛/醇脱氢酶(AdhE)是其乙醇代谢途径中的关键酶。以adhE启动子区域的DNA序列(TRRadhE)作为诱饵,通过制备核酸亲和层析柱,利用调控因子与DNA特异性结合的特点,得到调控因子PadhE-1。经过蛋白测序和NCBI数据库的比对,发现其N端序列与Thermoanaerobacter pseudethanolicus39E的LacI家族转录调控因子(GenBank accession No.YP_001665770)有90%的同源性。根据同源性设计引物,从嗜热厌氧乙醇杆菌JW200基因组中通过PCR扩增得到调控因子PadhE-1的基因,并克隆到表达载体pET-28a得到重组质粒pET-PadhE。转化大肠杆菌JM109(DE3),经IPTG诱导表达,Ni2+亲和柱纯化,得到重组表达的调控因子PadhE-1。体外凝胶阻滞实验证实PadhE-1能与TRRadhE特异性结合。这将有助于我们了解嗜热厌氧乙醇杆菌的乙醇代谢调控机理。  相似文献

有活力但不可培养(viable but non-culturable,VBNC)状态是细菌遭遇逆境时进入的一种特殊状态,该状态下的菌体在条件适宜时可复苏并恢复其致病性,被认为是细菌躲避不良环境的一种生存策略。VBNC状态菌体对人类医学和工农业生产具有巨大的潜在威胁,开展关于VBNC状态的检测及诱导、复苏及其机制研究可为减少或避免该状态细菌的危害提供理论基础。本文简要综述了细菌VBNC状态在诱导、复苏及致病性等方面的研究进展,并结合本实验室及国内外相关团队近年来在植物病原细菌VBNC状态研究中的结果,详细总结了VBNC状态细菌的形成和复苏机制,对植物病原细菌在环境胁迫下的存活机制、病害田间初侵染来源分析及VBNC状态菌体在病害循环中的作用等相关研究具有重要参考意义。  相似文献

Fatty alcohols are important components of a vast array of surfactants, lubricants, detergents, pharmaceuticals and cosmetics. We have engineered Saccharomyces cerevisiae to produce 1-hexadecanol by expressing a fatty acyl-CoA reductase (FAR) from barn owl (Tyto alba). In order to improve fatty alcohol production, we have manipulated both the structural genes and the regulatory genes in yeast lipid metabolism. The acetyl-CoA carboxylase gene (ACC1) was over-expressed, which improved 1-hexadecanol production by 56% (from 45 mg/L to 71 mg/L). Knocking out the negative regulator of the INO1 gene in phospholipid metabolism, RPD3, further enhanced 1-hexadecanol production by 98% (from 71 mg/L to 140 mg/L). The cytosolic acetyl-CoA supply was next engineered by expressing a heterologous ATP-dependent citrate lyase, which increased the production of 1-hexadecanol by an additional 136% (from 140 mg/L to 330 mg/L). Through fed-batch fermentation using resting cells, over 1.1 g/L 1-hexadecanol can be produced in glucose minimal medium, which represents the highest titer reported in yeast to date.  相似文献

Although better known as a pathogen of wheat stem bases, Fusarium pseudograminearum also causes Fusarium head blight. A natural isolate of F. pseudograminearum was identified that showed severely reduced virulence towards wheat heads and a map-based cloning approach was undertaken to identify the genetic basis of this phenotype. Using a population of 95 individuals, a single locus on chromosome 1 was shown to be responsible for the low virulence. Fine mapping narrowed the region to just five possible SNPs of which one was in the F. pseudograminearum homologue of velvet A. Knockout mutants of velvet A, which were non-pathogenic towards wheat, confirmed that velvet A regulates virulence in this pathogen. The mutation in velvet A was only found in a single field isolate and the origin of the mutation is unknown.  相似文献

The purpose of the research was to determine the effect of the foliar use of a growth regulator with the trade name of Tytanit, containing titanium ascorbate, on photosynthetic activity and chlorophyll content in Medicago × varia T. Martyn leaves. There were two kinds of plots: C – control series; Ti – plants treated with Tytanit, containing 8.5 g of titanium in 1 dm3. The following parameters were determined: maximum photosystem II efficiency (Fv/Fm) in a dark-adapted state, actual photosystem II efficiency (ΔF/Fm’) in a light-adapted state, photochemical quenching factor (QP), non-photochemical quenching factor (QN), and chlorophyll a and b content. The Fisher-Snedecor test was used to determine whether the impact of experimental factors was significant, and the HSD 0.05 value was calculated using Tukey’s test. Compared to control, the photosynthetic apparatus performance of alfalfa was positively affected by the regulator compared to control. Tytanit applied to plant leaves increased their photosynthetic activity as a result of an increase in the content of chlorophyll pigments. It was also found that periods of rainfall deficiency did not affect the beneficial effects of the regulator.  相似文献
