Wheat is one of the most important cereal food crops in the world today. The productivity and quality of this crop is greatly affected by environmental conditions during grain filling. In this study, we have analyzed two genotypes of durum wheat, Blanqueta and Sula (traditional and a modern wheat respectively) in pre-industrial, current and future [CO2]. Plant growth and physiological parameters were analyzed during anthesis and grain filling in order to study the capacity of these plants to create new sinks and their role during the process of the acclimation of photosynthesis. It was observed that plants underwent photosynthetic acclimation at pre-industrial and future [CO2] (up and down-regulation respectively). However, the modern genotype averts the process of down-regulation by creating a new carbon sink (i.e. the spike). Here, we have shown the essential role that the spike plays as a new sink in order to avert the down-regulation of photosynthesis at future [CO2]. Moreover, we have demonstrated that at future [CO2] the growth response will depend on the ability of plants to develop new sinks or expand existing ones.  相似文献

The alkyl gallates are found in several natural and industrial products. In the latter products, these compounds are added mainly for preventing oxidation. In the present work, the potencies of methyl gallate, n‐propyl gallate, n‐pentyl gallate, and n‐octyl gallate as inhibitors of pyruvate carboxylation and lactate gluconeogenesis were evaluated. Experiments were done with isolated mitochondria and the isolated perfused rat liver. The potency of the gallic acid esters as inhibitors of pyruvate carboxylation in isolated mitochondria obeyed the following decreasing sequence: n‐octyl gallate > n‐pentyl gallate > n‐propyl gallate > methyl gallate. A similar sequence of decreasing potency for lactate gluconeogenesis inhibition in the perfused liver was found in terms of the portal venous concentration. Both actions correlate with the lipophilicity of the compounds. The effects are harmful at high concentrations. At appropriate concentrations, however, octyl gallate should act therapeutically because its inhibitory action on gluconeogenesis will contribute further to its proposed antihyperglycemic effects.  相似文献

Photosynthetic assimilation of CO2 in plants results in the balance between the photochemical energy developed by light in chloroplasts, and the consumption of that energy by the oxygenation processes, mainly the photorespiration in C3 plants. The analysis of classical biological models shows the difficulties to bring to fore the oxygenation rate due to the photorespiration pathway. As for other parameters, the most important key point is the estimation of the electron transport rate (ETR or J), i.e. the flux of biochemical energy, which is shared between the reductive and oxidative cycles of carbon. The only reliable method to quantify the linear electron flux responsible for the production of reductive energy is to directly measure the O2 evolution by 18O2 labelling and mass spectrometry. The hypothesis that the respective rates of reductive and oxidative cycles of carbon are only determined by the kinetic parameters of Rubisco, the respective concentrations of CO2 and O2 at the Rubisco site and the available electron transport rate, ultimately leads to propose new expressions of biochemical model equations. The modelling of 18O2 and 16O2 unidirectional fluxes in plants shows that a simple model can fit the photosynthetic and photorespiration exchanges for a wide range of environmental conditions. Its originality is to express the carboxylation and the oxygenation as a function of external gas concentrations, by the definition of a plant specificity factor Sp that mimics the internal reactions of Rubisco in plants. The difference between the specificity factors of plant (Sp) and of Rubisco (Sr) is directly related to the conductance values to CO2 transfer between the atmosphere and the Rubisco site. This clearly illustrates that the values and the variation of conductance are much more important, in higher C3 plants, than the small variations of the Rubisco specificity factor. The simple model systematically expresses the reciprocal variations of carboxylation and oxygenation exchanges illustrated by a “mirror effect”. It explains the protective sink effect of photorespiration, e.g. during water stress. The importance of the CO2 compensation point, in classical models, is reduced at the benefit of the crossing points Cx and Ox, concentration values where carboxylation and oxygenation are equal or where the gross O2 uptake is half of the gross O2 evolution. This concept is useful to illustrate the feedback effects of photorespiration in the atmosphere regulation. The constancy of Sp and of Cx for a great variation of P under several irradiance levels shows that the regulation of the conductance maintains constant the internal CO2 and the ratio of photorespiration to photosynthesis (PR/P). The maintenance of the ratio PR/P, in conditions of which PR could be reduced and the carboxylation increased, reinforces the hypothesis of a positive role of photorespiration and its involvement in the plant-atmosphere co-evolution.  相似文献

Several marine macroalgae representative of the Chlorophyceae, Rhodophyceae and Phaeophyceae were investigated for their potentials of photosynthesis and light independent (dark) carbon fixation. In addition, the activities of ribulose-1,5-bisphosphate carboxylase (RubP-C; EC 4.1.1.39) and of phosphoenolpyruvate carboxykinase (PEP-CK; EC 4.1.1.32) were studied. In contrast to the green and red algae investigated, all brown algae exhibited comparably high rates of dark fixation accounting for up to 20% of photosynthetic carbon uptake. These observations are confirmed by the activities of RubP-C and PEP-CK measured after extraction from different species and thallus regions. Dark fixed 14C was mainly recovered from aspartate, citrate, malate, glutamate, and alanine. Appreciable amounts of 14C were incorporated into insoluble (polymeric) constituents even after relatively short periods of dark fixation.  相似文献

Degradation of unsaturated hydrocarbons by methanogenic enrichment cultures   总被引:1,自引:0,他引:1  
Abstract The biodegradability of hydrocarbons under anaerobic conditions was studied in enrichment cultures using mineral media inoculated with sewage sludge or sediment samples of limnic and marine origin. No indication of methanogenic degradation was obtained with either n -hexane, n -hexadecane, n -heptadecane, 1-hexene, cis -2-hexene, trans -2-hexene, isoprene, 1-hexine, benzene, toluene, xylene, cyclohexene, cycloheptatriene, cyclopentadiene, styrene, naphthalene, azulene, or β-carotene. Squalene was incompletely converted to methane and carbon dioxide. Complete degradation was observed with 1-hexadecene. Methanogenic subcultures were maintained on 1-hexadecene and squalene. Both enrichments contained after several transfers Methanospirillum hungatei and Methanothrix soehngenii as prevalent methanogenic bacteria. Acetate (≤80 μ M) was the only intermediary product detected indicating that degradation proceeded via hydrogen-dependent syntrophic β-oxidations. Short rods on hexadecene and cocci on squalene were found to be associated with substrate degradation. The results indicate that terminal double bonds can be sufficient to allow methanogenic degradation of hydrocarbons whereas branching and terminal ring closures may significantly contribute to hydrocarbon stability in anoxic environments.  相似文献

Gas exchange characteristics, Pn-PAR and/or Pn-CO2 response parameters of rape (Brassica napus L) were studied in the field under different O3 concentrations (CF, 50 nl·L?1 and 100 nl·L?1) and fumigation regimes (constant concentration and dynamic varying concentrations) with a portable photosynthetic system (CIRAS-1). Results indicated: (1) Relatively to CF, higher ozone concentration decreased in Tr and increased in WUE under a constant concentration fumigation regime. In addition, an increase in Tr and decreases in Pn, Gs and WUE were found in the dynamic ozone exposure regime; (2) In the constant concentration exposure regimes, AQY, LSP and Pmax were markedly reduced and Rp and Γ were enhanced with an increase in ozone concentration (100 nl·L?1). There were significant differences in the parameters such as Rd, LCP, LSP, Pmax and CE between O3 exposure regimes; (3) Regardless of the exposure regimes which were imposed, the increase of O3 concentration induced significant decreases in Fv/Fo and Fv/Fm of the eighth leaves from the top canopy, but it had no effect on the fifth full-spread leaves. It can be concluded that the dynamic ozone exposure regime has greater detrimental effects on the photosynthesis of rape in spite of equal exposure dose, and thus is unfavorable for plant growth and the accumulation of dry matter.  相似文献

The tricarboxylic acid (TCA) cycle is the central hub of oxidative metabolism, running in the classic forward direction to provide carbon for biosynthesis and reducing agents for generation of ATP. Our metabolic tracer studies in melanoma cells showed that in hypoxic conditions the TCA cycle is largely disconnected from glycolysis. By studying the TCA branch point metabolites, acetyl CoA and citrate, as well as the metabolic endpoint glutamine and fatty acids, we developed a comprehensive picture of the rewiring of the TCA cycle that occurs in hypoxia. Hypoxic tumor cells maintain proliferation by running the TCA cycle in reverse. The source of carbon for acetyl CoA, citrate, and fatty acids switches from glucose in normoxia to glutamine in hypoxia. This hypoxic flux from glutamine into fatty acids is mediated by reductive carboxylation. This reductive carboxylation is catalyzed by two isocitrate dehydrogenases, IDH1 and IDH2. Their combined action is necessary and sufficient to effect the reverse TCA flux and maintain cellular viability.  相似文献

Seedlings (70-d-old) of two tall fescue (Festuca arundinacea Schreb.) genotypes, heat-tolerant Jaguar 3 and heat-sensitive TF 66, were exposed to a high temperature stress of 35/30 °C (day/night) for 20 d and both light-saturated and CO2-saturated leaf stomatal conductance decreased, especially in TF 66. Higher reductions of quantum efficiency, carboxylation efficiency and maximum photochemical efficiency of photosystem 2 in dark adapted leaves (measured as Fv/Fm) occurred in TF 66 than in Jaguar 3. High temperature stress increased photorespiration in the two plants, but more in TF 66. Moreover, high temperature stress also reduced the growth, chlorophyll content and caused cell membrane injuries in the two cultivars, the changes were again more pronounced in TF 66 than in Jaguar 3.  相似文献

Recently, several studies reported that the optimum temperature for the initial slope [IS(Ci)] of the light-saturated photosynthetic rate (A) versus intercellular CO2 concentration (Ci) curve changed, depending on the growth temperature. However, few studies compare IS(Ci) with ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) properties. Here, we assessed Rubisco activation state and in vitro Rubisco kinetics, the main determinants of IS(Ci), in spinach leaves grown at 30/25 [high temperature (HT)] and 15/10 degrees C [low temperature (LT)]. We measured Rubisco activation state and A at a CO2 concentration of 360 microL L(-1) (A360) at various temperatures. In both HT and LT leaves, the Rubisco activation state decreased with increasing temperatures above the optimum temperatures for A360, while the activation state remained high at lower temperatures. To compare Rubisco characteristics, temperature dependences of the maximum rate of ribulose 1,5-bisphosphate (RuBP) carboxylation (Vcmax), specificity factor (Sc/o) and thermal stability were examined. We also examined Vcmax, and thermal stability in the leaves that were transferred from HT to LT conditions and were subsequently kept under LT conditions for 2 weeks (HL). Rubisco purified from HT, LT and HL leaves are called HT, LT and HL Rubisco, respectively. Thermal stabilities of LT and HL Rubisco were similar and lower than that of HT Rubisco. Both Vcmax and Sc/o in LT Rubisco were higher than those of HT Rubisco at low temperatures, while these were lower at high temperatures. Vcmax in HL Rubisco were similar to those of LT Rubisco at low temperatures, and to those of HT Rubisco at high temperatures. The predicted photosynthetic rates, taking account of the Rubisco kinetics and the Rubisco activation state, agreed well with A360 in both HT and LT leaves. This study suggests that photosynthetic performance is largely determined by the Rubisco kinetics at low temperature and by Rubisco Kinetics and the Rubisco activation state at high temperature.  相似文献

The regulation of photosynthesis through changes in light absorption, photochemistry, and carboxylation efficiency has been studied in plants grown in different environments. Iron deficiency was induced in sugar beet (Beta vulgaris L.) by growing plants hydroponically in controlled growth chambers in the absence of Fe in the nutrient solution. Pear (Pyrus communis L.) and peach (Prunus persica L. Batsch) trees were grown in field conditions on calcareous soils, in orchards with Fe deficiency-chlorosis. Gas exchange parameters were measured in situ with actual ambient conditions. Iron deficiency decreased photosynthetic and transpiration rates, instantaneous transpiration efficiencies and stomatal conductances, and increased sub-stomatal CO2 concentrations in the three species investigated. Photosynthesis versus CO2 sub-stomatal concentration response curves and chlorophyll fluorescence quenching analysis revealed a non-stomatal limitation of photosynthetic rates under Fe deficiency in the three species investigated. Light absorption, photosystem II, and Rubisco carboxylation efficiencies were down-regulated in response to Fe deficiency in a coordinated manner, optimizing the use of the remaining photosynthetic pigments, electron transport carriers, and Rubisco.  相似文献
