Cellulose biosynthesis is a vital but yet poorly understood biochemical process in Oomycetes. Here, we report the identification and characterization of the cellulose synthase genes (CesA) from Saprolegnia monoica. Southern blot experiments revealed the occurrence of three CesA homologues in this species and phylogenetic analyses confirmed that Oomycete CesAs form a clade of their own. All gene products contained the D,D,D,QXXRW signature of most processive glycosyltransferases, including cellulose synthases. However, their N-terminal ends exhibited Oomycete-specific domains, i.e. Pleckstrin Homology domains, or conserved domains of an unknown function together with additional putative transmembrane domains. Mycelial growth was inhibited in the presence of the cellulose biosynthesis inhibitors 2,6-dichlorobenzonitrile or Congo Red. This inhibition was accompanied by a higher expression of all CesA genes in the mycelium and increased in vitro glucan synthase activities. Altogether, our data strongly suggest a direct involvement of the identified CesA genes in cellulose biosynthesis.  相似文献

Red blood cells (RBC) are known to modulate T cell proliferation and function possibly through downregulation of oxidative stress. By examining parameters of activation, division, and cell death in vitro, we show evidence that the increase in survival afforded by RBC is due to the maintenance of the proliferative capacity of the activated T cells. We also show that the CD3+CD8+ T cell subset was preferentially expanded and rescued from apoptosis both in bulk peripheral blood lymphocyte cultures and with highly purified CD8+ T cells. The ability of RBC to induce survival of dividing T cells was not affected by blocking the CD58/CD2 interaction. Moreover, addition of hemoglobin, heme or protoporphyrin IX to cultures of activated T cells did not reproduce the effect of intact RBC. Considering that RBC circulate throughout the body, they could play a biological role in the modulation of T cell differentiation and survival in places of active cell division. Neither CD58 nor the heme compounds studied seem to play a direct relevant role in the modulation of T cell survival.  相似文献

In many bird species, males exhibit territorial aggression outside the breeding season, when testosterone concentrations are low and may not regulate territorial behaviors. The hormonal regulation of aggression at this time of year has only been studied in passerine birds. Here, we investigated the role of testosterone in the regulation of aggression in a non-passerine bird, the red grouse Lagopus lagopus scoticus. Male red grouse are aggressive in early spring when breeding starts, in autumn when they establish territories, and sporadically through much of the winter. We first describe seasonal variations in plasma testosterone concentrations and in the size of males' sexual ornaments, their red combs, which relates to aggressiveness. Testosterone concentrations and comb size were correlated. Both increased in autumn to a peak in October, and then increased again in spring, to a greater peak in early April. Secondly, we experimentally investigated the effects of testosterone, and of an anti-androgen (flutamide) used in combination with an aromatase inhibitor (ATD), on autumn territorial behavior. Males were treated with either empty implants, as controls (C-males), testosterone implants (T-males), or with flutamide and ATD implants (FA-males). One month after implanting, both T- and FA-males had higher concentrations of testosterone than C-males. Comb size, aggressive call rate, and response to playbacks of territorial call all significantly increased in T-males. However, the increase in testosterone in FA-males did not increase comb size or aggressive behavior. In the following spring, after the content of implants was used, FA-males had significantly lower testosterone than C-males, and had a reduced seasonal increase in comb size. The results suggest that testosterone plays a significant role in regulating red grouse aggressive behavior in autumn. However, the observation that flutamide and ATD treatment did not reduce territorial behavior, suggests that estradiol may also be involved in the regulation of non-breeding aggression.  相似文献

Glucose self-exchange flux (Jex) and net efflux (Jnet) in human red cells and ghosts were studied at 25°C and pH 7.2 by means of the combined use of the Millipore-Swinnex filtering method and the continuous flow tube method to show the dependence of time of storage after aspiration, ATP and insulin. In fresh cells (RBC), ghosts (G), ghosts with 2 mM ATP (G+), and cells stored at 4°C > 60 days (OC) both Jex and Jnet follow simple Michaelis-Menten kinetics where

Jexmax and Jnetmax (nmol · cm−2 · s−1), respectively, was: (RBC) 0.27 and 0.19, (G) 0.24 and 0.27, (G +) 0.23 and 0.24, (OC) 0.23 and 0.20. and (mM), respectively, was: (RBC) 7.5 and 1.3, (G) 4.8 and 14.2, (G +) 11.6 and 6.8, (OC) 3.8 and 9.0. In ghosts, the ATP-dependent fraction of the permeability shows a hyperbolic dependence on glucose concentrations lower than 80 mM. Insulin up to 1 μM had effect on neither Jex nor Jnet in RBC. Based on reported values of cytochalasin B binding sites the turnover rate per site in RBC appears to be as high as in maximally insulin-stimulated fat cells. Our results suggest that the number of transport sites remains constant, independent of age, ATP and insulin.  相似文献

The feeding pattern of red foxes Vulpes vulpes, i.e., the composition of their diet and dietary responses to fluctuations in the abundance of common voles Microtus arvalis were compared in the periods before and after the limitation of anthropogenic food availability in a rural area in western Poland. During the winter seasons of 1996/97–2014/15, the diet of the foxes was estimated by analysing the stomach contents of individuals shot from November to March and the index of vole abundance was obtained by counting their burrow entrances. The major component of the fox diet over the entire study period was anthropogenic food (mainly, the remains of farm animals) that showed both the highest average proportion in the stomach content volume (53.4%) and the highest average frequency of occurrence in the stomachs (62.7%), whereas small mammals were the second important component (27.5% and 54.2%, respectively). The frequency of anthropogenic food decreased and the frequencies of wild ungulate carrion and other wild vertebrates (mainly birds) increased after the limitation of anthropogenic resources. The observed fluctuations in the index of vole abundance resulted in positive changes in the small mammal frequency and negative changes in the anthropogenic food frequency, but the course of these relationships did not significantly differ between the two periods. The frequency of hares in fox stomachs did not change significantly between these periods, but recently the occurrence of hares in the diet of foxes increased with vole abundance. This study indicated that the reduction of anthropogenic food availability for carnivorous mammals may result in their switching to other prey species and potentially may have a negative influence on the demography of such prey species.  相似文献

An artificial neural network (ANN) model was used to predict removal efficiency of Lanaset Red (LR) G on walnut husk (WH). This adsorbent was characterized by FTIR-ATR. Effects of particle size, adsorbent dose, initial pH value, dye concentration, and contact time were investigated to optimize sorption process. Operating variables were used as the inputs to the constructed neural network to predict the dye uptake at any time as an output. Commonly used pseudo second-order model was fitted to the experimental data to compare with ANN model. According to error analyses and determination of coefficients, ANN was the more appropriate model to describe this sorption process. Results of ANN indicated that pH was the most efficient parameter (43%), followed by initial dye concentration (40%) for sorption of LR G on WH.  相似文献

The potential for local adaptation between pathogens and their hosts has generated strong theoretical and empirical interest with evidence both for and against local adaptation reported for a range of systems. We use the Linum marginale-Melampsora lini plant-pathogen system and a hierarchical spatial structure to investigate patterns of local adaptation within a metapopulation characterised by epidemic dynamics and frequent extinction of pathogen populations. Based on large sample sizes and comprehensive cross-inoculation trials, our analyses demonstrate strong local adaptation by Melampsora to its host populations, with this effect being greatest at regional scales, as predicted from the broader spatial scales at which M. lini disperses relative to L. marginale. However, there was no consistent trend for more distant pathogen populations to perform more poorly. Our results further show how the coevolutionary interaction between hosts and pathogens can be influenced by local structure such that resistant hosts select for generally virulent pathogens, while susceptible hosts select for more avirulent pathogens. Empirically, local adaptation has generally been tested in two contrasting ways: (1) pathogen performance on sympatric versus allopatric hosts; and (2) sympatric versus allopatric pathogens on a given host population. In situations where no host population is more resistant or susceptible than others when averaged across pathogen populations (and likewise, no pathogen population is more virulent or avirulent than others), results from these tests should generally be congruent. We argue that this is unlikely to be the case in the metapopulation situations that predominate in natural host-pathogen interactions, thus requiring tests that control simultaneously for variation in plant and pathogen populations.  相似文献

Protein misfolding and aggregation are involved in a number of the so-called "conformational" diseases (e.g., transmissible spongiform encephalopathies and Alzheimer disease). The development of rational strategies to interfere with aggregation is a potential therapeutic approach that requires complete knowledge of the aggregation process. We studied the aggregation of ovalbumin in beta-sheet conformation using mainly the spectral changes in the spectra of two dyes (Congo Red and pinacyanol) caused by the aggregates. We assumed a linear model of polymerization that fit to the experimental data. The critical aggregation constant, concentration of half-aggregation, nucleation parameter, growth parameter, and number of aggregation and free energy changes (total and per residue) were determined as aggregation-related parameters. Beta-Ovalbumin aggregates in a cooperative way. Moreover, the differences between such parameters obtained with Congo Red and pinacyanol suggest that each dye interacts with the protein in its own way.  相似文献

This review describes aspects of chicken natural history and cross-disciplinary biological value. The diversity of extant genetic variants available to researchers is reviewed along with institutional stock locations for North America. This paper presents a novel multiscale methodology for quantitative analysis of pulmonary gas exchange. The process of oxygen uptake in the lungs is a complex multiscale process, characterized by multiple time and length scales which are coupled nonlinearly through the processes of diffusion, convection and reaction, and the overall oxygen uptake is significantly influenced by the transport and reaction rate processes at the small-scales. Based on the separation of length scales, we characterize these disparate scales by three representative ones, namely micro (red blood cell), meso (capillary and alveolus) and macro (lung). We start with the fundamental convection-diffusion-reaction (CDR) equation that quantifies transport and reaction rates at each scale and apply spatial averaging techniques to reduce the dimensionality of these models. The resultant low-dimensional models embed each scale hierarchically within the other while retaining the important parameters of the small-scales in the averaged equations, and drastically reduce the computational efforts involved in solving them. We use our multiscale model for pulmonary gas exchange to quantify the oxygen uptake abnormalities in patients with hepatopulmonary syndrome (HPS), a disease which is characterized by coupled abnormalities in multiple length scales. Based on our multiscale modeling, we suggest a strategy to stratify patients with HPS into two categories--those who are oxygen-responsive and those who are oxygen non-responsive with intractable hypoxemia.  相似文献
