为建立一种快速鉴别严重急性呼吸综合征冠状病毒2 (severe acute respiratory syndrome coronavirus 2,SARS-CoV-2)的5种主要变异株的Taq Man探针实时荧光定量PCR(real-time quantitative PCR, RT-qPCR)体系,基于SARS-CoV-2野生型及变异株alpha (N501Y、HV69-70del)、beta (E484K、K417N)、gamma (K417T、V1176F)、delta (L452R、T478K)和omicron (H655Y、N679K、P681H)序列设计特异性引物、探针,建立和优化一种鉴别新型冠状病毒(SARS-CoV-2) 5种主要变异株的Taq Man探针RT-qPCR方法,并进行该方法的特异性、敏感性、鉴别能力评价。该方法可准确区分出SARS-CoV-2野生型和突变型,与其他呼吸道病原体(n=21)无交叉,显示高特异性。该方法最低检测限为2×10;拷贝/mL,操作简单、快速、成本廉价,可用于监测SARS-CoV-2毒株的变异,精准指导疫情识别与防控。 相似文献

本研究尝试将触减 PCR与3’ cDNA末端快速扩增(rapid amplification of cDNA ends,RACE)技术进行结合,仅用一条特异性引物和一条通用引物,成功地实现了从3’末端cDNA库对鸡含锰超氧化物歧化酶(manganese-containing superoxide dismutase,MnSOD)全长cDNA的一步3’RACE快速构建。与常规使用的末端PCR或亚克隆方法相比,该法具有快速、省时、经济和特异性好的优点。Abstract: RACE(rapid amplification of cDNA ends) is a popular technique to rapidly obtain the full-length cDNA. After obtaining the 3’cDNA and 5’cDNA fragments with a overlapped region by 3’RACE and 5’RACE, the full-length cDNA could be generated by end-to-end PCR or subcloning. In this study, 3’RACE combined with touch-down PCR was successfully used for the rapid construction of full-length MnSOD cDNA of chickens. Compared with the conventional end-to-end PCR or subcloning, this method, called one-step 3’RACE, is fast, economical and highly specific. 应用实时荧光定量PCR法,构建了总甲烷菌实时荧光绝对定量PCR的标准品以及标准曲线,用于总甲烷菌的定量测定,所构建的质粒DNA浓度为160mg/L,标准曲线相关性为0.992,扩增效率为98.6%。研究表明,若尔盖花湖地区4月草地生态系统和湿地生态系统甲烷菌含量相当,湿地生态系统9月甲烷菌含量高于7月,7月甲烷菌含量高于4月,在7月和9月随着深度的增加甲烷菌含量也增加,而草地生态系统的甲烷菌含量并未表现出明显的季节和深度上的规律性。 相似文献

首次从长春花中克隆了Crlea(Crlea for Catharanthus roseus late embryogenesis abundant)的全长基因,采用荧光定量PCR方法对干旱胁迫下长春花叶片和根部Crlea基因的表达模式进行监测,结果表明,在0.5~8 h的胁迫时间中,叶片和根部的Crlea基因表现出相似的积累模式。长春花Crlea基因的表达随着胁迫时间的延长而表达增强。在叶片中,在6 h和8 h的干旱处理后,Crlea基因表达显著提高,分别是未处理材料的9.984和20.431倍。在根部, 在8 h的处理后,Crlea基因的表达量显著提高(2.831倍于对照)。初步结果表明Crlea基因的表达没有组织特异性,并且为干旱胁迫正调控。 相似文献

目的 探讨结核杆菌感染与恶性肿瘤的关系。方法 采用多聚酶链反应(PCR) 技术检测两种恶性肿瘤组织中结核杆菌DNA(TB- DNA) 。结果41 例恶性肿瘤组织中检出TB- DNA 阳性患者8 例,占19.5 % ,且TB-DNA 阳性患者肿瘤发病部位基本与结核病的好发部位一致。结论 结核杆菌感染可能与一些恶性肿瘤存在特殊相关性。 相似文献

Mycorrhizal colonization of Pinus muricata from resistant propagules after a stand-replacing wildfire 总被引:7,自引:5,他引:7
Colonization of mycorrhizal fungi was studied in a Pinus muricata forest on the coast in California, USA, burned by a stand-replacing wildfire in October 1995. Naturally established field seedlings of P. muricata were harvested 1 yr after the fire. The species composition of the mycorrhizal fungi on these field seedlings was dominated by Rhizopogon species, Wilcoxina mikolae and Tomentella sublilacina . Bioassays, set up with soil collected immediately after the fire, were used to determine which mycorrhizal species had colonized the burned area from resistant propagules. The P. muricata seedlings in these bioassays were dominated by suilloid and ascomycetous fungi, the same fungi which dominated the mycorrhizal flora of seedlings in pre-fire bioassays derived from the same forest site, suggesting that resistant propagules were the primary inoculum source for naturally establishing seedlings. Drying of post-fire soil for 1 month raised the number of bioassay seedlings associated with Rhizopogon olivaceotinctus , while the number of bioassay seedlings associated with Rhizopogon ochraceorubens was reduced. Fire appeared to have either stimulated or provided a competitive advantage to R. olivaceotinctus , which increased in abundance on the post-fire bioassay and field seedlings. Soil collected from the burned area was diluted with sterile soil in three different concentrations, and the number and frequency of mycorrhizal taxa on bioassay seedlings decreased with increased dilution. Although precise quantification was not possible, propagules of the Rhizopogon species were much more abundant than those of Tomentella or Wilcoxina species. Differences between the mycorrhizal associates of bioassay seedlings, naturally regenerated seedlings, and different inoculum sources are discussed. 相似文献

The limited population sizes used in many quantitative trait locus (QTL) detection experiments can lead to underestimation
of QTL number, overestimation of QTL effects, and failure to quantify QTL interactions. We used the barley/barley stripe rust
pathosystem to evaluate the effect of population size on the estimation of QTL parameters. We generated a large (n=409) population of doubled haploid lines derived from the cross of two inbred lines, BCD47 and Baronesse. This population
was evaluated for barley stripe rust severity in the Toluca Valley, Mexico, and in Washington State, USA, under field conditions.
BCD47 was the principal donor of resistance QTL alleles, but the susceptible parent also contributed some resistance alleles.
The major QTL, located on the long arm of chromosome 4H, close to the Mlo gene, accounted for up to 34% of the phenotypic variance. Subpopulations of different sizes were generated using three methods—resampling,
selective genotyping, and selective phenotyping—to evaluate the effect of population size on the estimation of QTL parameters.
In all cases, the number of QTL detected increased with population size. QTL with large effects were detected even in small
populations, but QTL with small effects were detected only by increasing population size. Selective genotyping and/or selective
phenotyping approaches could be effective strategies for reducing the costs associated with conducting QTL analysis in large
populations. The method of choice will depend on the relative costs of genotyping versus phenotyping.

Equilibrium analysis and phase synchronization of two coupled HR neurons with gap junction 总被引:1,自引:0,他引:1
The properties of equilibria and phase synchronization involving burst synchronization and spike synchronization of two electrically coupled HR neurons are studied in this paper. The findings reveal that in the non-delayed system the existence of equilibria can be turned into intersection of two odd functions, and two types of equilibria with symmetry and non-symmetry can be found. With the stability and bifurcation analysis, the bifurcations of equilibria are investigated. For the delayed system, the equilibria remain unchanged. However, the Hopf bifurcation point is drastically affected by time delay. For the phase synchronization, we focus on the synchronization transition from burst synchronization to spike synchronization in the non-delayed system and the effect of coupling strength and time delay on spike synchronization in delayed system. In addition, corresponding firing rhythms and spike synchronized regions are obtained in the two parameters plane. The results allow us to better understand the properties of equilibria, multi-time-scale properties of synchronization and temporal encoding scheme in neuronal systems. 相似文献
