BACKGROUND AND AIMS: In spite of the impressive species diversity in the Asteraceae and their widespread appeal to many generalist pollinators, floral-nectary ultrastructure in the family has rarely been investigated. To redress this, a study using Echinacea purpurea, a plant of horticultural and nutraceutical value, was undertaken. Nectar secretion of disc florets was compared with floral nectary ultrastructure taking into account nectar's potential impact upon the reproductive success of this outcrossing species. METHODS: Micropipette collections of nectar in conjunction with refractometry were used to determine the volume and nectar-sugar quantities of disc florets throughout their phenology, from commencement of its production to cessation of secretion. Light, scanning-electron and transmission-electron microscopy were utilized to examine morphology, anatomy and ultrastructure of nectaries of the disc florets. KEY RESULTS: Florets were protandrous with nectar being secreted from anthesis until the third day of the pistillate phase. Nectar production per floret peaked on the first day of stigma receptivity, making the two innermost whorls of open florets most attractive to foraging visitors. Modified stomata were situated along the apical rim of the collar-like nectary, which surrounds the style base and sits on top of the inferior ovary. The floral nectary was supplied by phloem only, and both sieve elements and companion cells were found adjacent to the epidermis; the latter participated in the origin of some of the precursor cells that yielded these specialized cells of phloem. Companion cells possessed wall ingrowths (transfer cells). Lobed nuclei were a key feature of secretory parenchyma cells. CONCLUSIONS: The abundance of mitochondria suggests an eccrine mechanism of secretion, although dictyosomal vesicles may contribute to a granulocrine process. Phloem sap evidently is the main contributor of nectar carbohydrates. From the sieve elements and companion cells, an apoplastic route via intercellular spaces and cell walls, leading to the pores of modified stomata, is available. A symplastic pathway, via plasmodesmata connecting sieve elements to companion, parenchyma and epidermal cells, is also feasible. Uncollected nectar was reabsorbed, and the direct innervation of the nectary by sieve tubes potentially serves a second important route for nectar-sugar reclamation. Background and Aims
Floral development depends on photosynthetic products delivered by the phloem. Previous work suggested the path to the flower involved either the apoplast or the symplast. The objective of the present work was to determine the path and its mechanism of operation.Methods
Maize (Zea mays) plants were grown until pollination. For simplicity, florets were harvested before fertilization to ensure that all tissues were of maternal origin. Because sucrose from phloem is hydrolysed to glucose on its way to the floret, the tissues were imaged and analysed for glucose using an enzyme-based assay. Also, carboxyfluorescein diacetate was fed to the stems and similarly imaged and analysed.Key Results
The images of live sections revealed that phloem contents were released to the pedicel apoplast below the nucellus of the florets. Glucose or carboxyfluorescein were detected and could be washed out. For carboxyfluorescein, the plasma membranes of the phloem parenchyma appeared to control the release. After release, the nucellus absorbed apoplast glucose selectively, rejecting carboxyfluorescein.Conclusions
Despite the absence of an embryo, the apoplast below the nucellus was a depot for phloem contents, and the strictly symplast path is rejected. Because glucose and carboxyfluorescein were released non-selectively, the path to the floret resembled the one later when an embryo is present. The non-selective release indicates that turgor at phloem termini cannot balance the full osmotic potential of the phloem contents and would create a downward pressure gradient driving bulk flow toward the sink. Such a gradient was previously measured by Fisher and Cash-Clark in wheat. At the same time, selective absorption from the apoplast by the nucellar membranes would support full turgor in this tissue, isolating the embryo sac from the maternal plant. 羊草(Leymus chinensis(Trin.)Tzvel.)是一种多年生禾本科优良牧草。经过对东北草原3个固定样地连续12a,分别随机取样100个穗状花序的调查和测定,将羊草种群单穗数量性状的生态可塑性进行了统计分析,并且分别与不同生长发育阶段气候因子进行了相关分析。结果表明,羊草种群的单穗长度、小穗数和小花数的生态可塑性变化在样地内和样地间,以及年度内和年度间均达到了极显著水平。其中,均表现为年度内大于年度间,样地内大于样地间,样地间大于年度间。羊草种群单穗数量性状均与形成性状的生育期,及其以前各生育期不同阶段的光照时间、积温、降水量有着一定程度的相关关系。在整个冬性枝条生长发育期,较多的降水、较少的光照时间和较低的积温将对翌年幼穗分化及其生长有一定的促进作用,而较少的降水、较多的光照时间和较高的积温则有不利影响;从拔节到抽穗阶段较多的降水对幼穗分化具有一定的促进作用,但从返青到抽穗不同阶段较高的积温和较多的光照、以及返青初期较多的降水量则有着一定的不利影响。气候因子对羊草种群幼穗分化滞后效应的生态时间差可长达10个月。 相似文献

水稻颖花的开闭受多种因素的影响。35~40℃的温度,含>5%CO_2的气体和饱和CO_2的水溶液能显著促进开颖;低温、呼吸抑制剂抑制颖开闭。湿度对颖开闭过程没有影响。开颖前数日的浆片细胞中含有淀粉粒,开颖时淀粉粒已消失。开颖后,浆片细胞开始自溶,细胞的解体物通过导管向小穗轴撤离。根据对温度和呼吸抑制剂的反应,可把水稻颖开闭过程分为三个阶段:1.与呼吸有关的临开颖阶段,2.与呼吸无关的开颖阶段,3.与呼吸有关的闭颖阶段。 相似文献

Evolutionary shifts among radiate, disciform and discoid flowerheads have occurred repeatedly in a number of major lineages across the Asteraceae phylogeny; such transitions may also appear within evolutionarily young groups. Although several studies have demonstrated that CYC2 genes partake in regulating floral morphogenesis in Asteraceae, the evolution of capitulum forms within a recently diverging lineage has remained poorly understood. Here, we study the molecular regulation of the shift from a radiate to a disciform capitulum within the Chrysanthemum group. This is a recently radiating group mainly comprising two genera, Chrysanthemum and Ajania, that are phylogenetically intermingled but distinct in flowerhead morphology: Chrysanthemum spp. with radiate capitula and Ajania spp. with disciform capitula. We found that the morphogenesis of zygomorphy in the marginal floret in Ajania was disrupted soon after floral primordium emergence; CYC2g, one of the CYC2 copies that was expressed prominently in the ray floret of Chrysanthemum was not expressed in flowerheads of Ajania. Weakening the expression of ClCYC2g in Chrysanthemum lavandulifolium led to the gradual transition of a ray flower toward the disc-like form. Molecular evolutionary analyses indicated that the disciform capitulum might have evolved only once, approximately 8 Mya, arising from dysfunction of the CYC2g orthologs. A 20-nt deletion, including a putative TATA-box of the Ajania-type CYC2g promoter, appeared to inhibit the expression of the gene. Considering the divergent habitats of Chrysanthemum and Ajania, we propose that the shift from radiate to disciform capitulum must have been related to changes in pollination strategies under selective pressure. 相似文献

The effect of exogenous ethephon on cambial activity, xylem production and ray population in young shoots of Leucaena leucocephala was investigated anatomically. The application of ethephon showed a diphasic effect on cambial activity and xylogenesis in a dose dependent manner. Lower concentration of ethephon enhanced cambial activity while high concentrations reduced cambial cell divisions and daughter-cell differentiation. High ethephon concentration also resulted in shorter vessel elements, thick walled fibers and phenolic accumulation in ray cells and vessel elements, whereas low concentration allowed elongation of fibers and vessel elements. The density of rays increased significantly with increase in ethylene concentration. The evaluation of longitudinal sections of cambial zone in ethephon treated plants showed high frequency of transformation of fusiform initials into ray initials through divisions and segmentation, resulting in high ray frequency in both xylem and phloem. The present study demonstrates that ethylene plays an important role in regulating secondary vascular tissue composition by reducing the population of fusiform initials in the cambium. 相似文献

Yield studies show that increases in grain yield are always accompanied by an increase in grain number and, hence, further increases in yield potential may require additional improvements in grain number. The improvement of modern durum wheat was mainly based on the introduction of semidwarf genes. A 2‐year field drought stress experiment, concerning two different genotype groups (landraces vs modern cultivars), was carried out under a rainout shelter in order (a) to assess the effect of water deficit on floret dynamics and grain number determination, (b) to explore the relationship between plant water status with grain number per spike and its components (i.e., spikelets per spike, fertile florets per spikelet and grain set) and (c) to quantify the importance of several plant traits in determining the final number of grains per spike and fertile florets per spike when the main source of variation is water availability. Compared to control (well irrigated), grain number per spike was reduced, depending on year, genotype and water availability level, by 12.4–58.7% and this reduction was evident almost in all spikelet positions along the spike. Although there were some doubts in the past about the increased sensitivity of semidwarf cultivars to drought stress, they were not confirmed from our results. In most of the cases, the variation in plant water status (by means of water potential index [WPI]) during floret primordia phase (FPP) explained most of the variance in grain number per spike, fertile florets per spikelet, grain set and fertile spikelets per spike. In general, increasing water stress intensity decreased grain number per spike by an average rate of 13.5 and 9.4 grains per 0.2 MPa decrease in WPI, in modern cultivars and landraces, respectively. However, seasonal and genotypic effects were evident by modifying the slopes of the linear regressions between WPI and the studied plant traits. Commonality analysis revealed that the number of fertile florets per spikelet was the best predictor of grain number per spike, indicating that there is still much space for further improvement for this trait in landraces. However, this trait has been clearly improved in modern cultivars, especially at the basal and central spikelets. Although the number of spikelets per spike was the best unique predictor of the number of grains per spike in modern cultivars, grain set presented the highest total effect. 相似文献
