The quinol-linked cytochrome bd oxidases are terminal oxidases in respiration. These oxidases harbor a low spin heme b(558) that donates electrons to a binuclear heme b(595)/heme d center. The reaction with O(2) and subsequent catalytic steps of the Escherichia coli cytochrome bd-I oxidase were investigated by means of ultra-fast freeze-quench trapping followed by EPR and UV-visible spectroscopy. After the initial binding of O(2), the O-O bond is heterolytically cleaved to yield a kinetically competent heme d oxoferryl porphyrin π-cation radical intermediate (compound I) magnetically interacting with heme b(595). Compound I accumulates to 0.75-0.85 per enzyme in agreement with its much higher rate of formation (~20,000 s(-1)) compared with its rate of decay (~1,900 s(-1)). Compound I is next converted to a short lived heme d oxoferryl intermediate (compound II) in a phase kinetically matched to the oxidation of heme b(558) before completion of the reaction. The results indicate that cytochrome bd oxidases like the heme-copper oxidases break the O-O bond in a single four-electron transfer without a peroxide intermediate. However, in cytochrome bd oxidases, the fourth electron is donated by the porphyrin moiety rather than by a nearby amino acid. The production of reactive oxygen species by the cytochrome bd oxidase was below the detection level of 1 per 1000 turnovers. We propose that the two classes of terminal oxidases have mechanistically converged to enzymes in which the O-O bond is broken in a single four-electron transfer reaction to safeguard the cell from the formation of reactive oxygen species.  相似文献

The title complexes were obtained as MIIM′II species [(bpy)2M(μ-abpy)M′(bpy)2](PF6)4, M,M′ = Ru or Os, using the new mononuclear precursor [(bpy)2Os(abpy)](PF6)2 for the osmium-containing dinuclear complexes. One-electron reduction produces radical complexes [(bpy)2M(μ-abpy)M′(bpy)2]3+ and [(bpy)2M(abpy)]+ with significant contributions from the metals, as evident from the EPR effects on successive replacement of ruthenium by osmium with its much higher spin-orbit coupling constant. The diruthenium and diosmium radical complexes were also studied by EPR at high-frequency (285 GHz), the latter shows an unusually large g anisotropy g1 − g3 = 0.25 in frozen solution. Further reduction was monitored by UV/Vis spectroelectrochemistry. Oxidation produced OsIII EPR signals for [(bpy)2Os(abpy)]3+ and [(bpy)2Os(μ-abpy)Ru(bpy)2]5+, indicating a RuIIOsIII species for the latter. The diosmium(III,II) and diruthenium(III,II) mixed-valent species remained EPR silent at 4 K, however, they exhibit weak inter-valence charge transfer (IVCT) bands at about 1460 nm. Whereas the cyclic voltammetric response towards reduction is only marginally different for the three dinuclear complexes, successive replacement of ruthenium by osmium causes the first oxidation potential to decrease. The much higher comproportionation constant Kc for the mixed valent diosmium(III,II) state (Kc > 1015) in comparison to the diruthenium(III,II) analogue with Kc = 1010 confirms the electron transfer alternative for the valence exchange mechanism, in contrast to the hole transfer established for analogous dinuclear complexes with the formally related diacylhydrazido(2−) bridging ligands.  相似文献

As an artificial ligand, a glycoconjugate polymer carrying carbohydrate moiety of lactosyl ceramide or globotriaosyl ceramide (Gb3) was synthesized. Gb3 is known as the receptor of Shiga toxin-producing Escherichia coli O157: H7. The preparation of the glycoconjugate polymer initially involves the construction of the carbohydrate moiety of Gb3 derivative which has n-pentenyl group as polymerizable group. In addition, the n-pentenyl group of the Gb3 derivative was modified and different polymerizable groups such as acrylamide group were introduced at ω-position of the aglycon. Radical polymerization of the synthesized glycosyl monomers with or without acrylamide proceeded smoothly in water using ammonium persulfate and N, N, N′, N′-tetramethylethylenediamine as usual initiator system and gave water-soluble glycoconjugate polymers having various polymer compositions. These polymers have the potential to neutralize Shiga toxin by reason of cluster effect and multivalency.  相似文献

4-Schiff base-7-benzyloxy-coumarins 5a(1)-5h(2) and its derivative 6 were designed and synthesized based on the 7-benzyloxy-coumarin structure as novel antioxidants. The in vitro antioxidant activities screening revealed that 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities of compounds 5b(1), 5d(1), 5f(1), 5f(2), 5g(1) and 5g(2), and 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulfonate) cation (ABTS(+)) radical scavenging activities of compounds 5a(1), 5b(1), 5c(1), 5c(2), 5d(1), 5e(1), 5e(2), 5f(2), 5g(1), 5g(2) and 5h(1) were better than that of the commercial antioxidant butylated hydroxytoluene (BHT), while the superoxide anion radical scavenging activities of 5a(2) and 5g(2) were stronger than that of the commercial antioxidant butylated hydroxyanisole (BHA), and the hydroxyl radical scavenging activity of 5e(1) was much better than that of the common antioxidant ascorbic acid.  相似文献

目的:探讨肝门部胆管癌术后口服胆汁患者的疗效。方法:采用病例对照法,回顾性分析2015年10月-2016年9月期间我科收治的肝门部胆管癌患者,术前行经皮肝穿刺胆道引流术(PTCD)并接受肝门部胆管癌根治术的患者89例,根据术后是否口服胆汁,将肝门部胆管癌患者分为两组,即口服胆汁组(51例)及对照组(38例)。所有患者均为术后3日开始进食。口服胆汁组:胆汁口服应遵循的条件为引流出的胆汁呈金黄色、无血性。术后第三天,胆汁满足口服的条件时,每6小时收集一次,经双层纱布过滤,胆汁中加入蜂蜜少量多次、随餐服下。对照组:除外口服胆汁,其余进食及治疗均相同。术后主要结局观察指标包括腹泻发生率、术后7天血清白蛋白水平,腹腔感染发生率;次要结局指标即住院时间。观察术后患者腹泻发生率、术后7天血清白蛋白水平,腹腔感染发生率和住院时间。结果:两组患者腹腔感染、腹泻发生及住院时间有统计学差异,p值均小于0.05,两组间白蛋白水平无统计学差异。结论:口服胆汁是一种有效、简便且经济的方法。术后腹腔感染并发症少,缩短住院时间,腹泻发生率较低。  相似文献

Dulcinoside (1), dulcisisoflavone (2), dulcisxanthone A (3) and sphaerobioside acetate (6) together with 22 known compounds were isolated from the green fruit of G. dulcis. Dulcisflavan (4), dulcisxanthone B (5) and isonormangostin (7) together with 22 known compounds were isolated from the ripe fruit. Compounds 6 and 7 were synthetic known compounds. Their structures were determined by spectroscopic methods. The radical scavenging and antibacterial activities of some of the compounds were investigated.  相似文献

Free radicals and other reactive species are involved in normal ovarian physiology. However, they are also highly reactive with complex cellular molecules (proteins, lipids, and DNA) and alter their functions leading to oxidative stress. Oxidative damage may play a prominent role in the development of disorders that considerably influence female fertility. Melatonin, because of its amphiphilic nature that allows for crossing morphophysiological barriers, is an effective antioxidant for protecting macromolecules against oxidative stress caused by reactive species. The balance between reactive oxygen species and antioxidants within the follicle seems to be critical to the function of the oocyte and granulosa cells and evidence has accumulated showing that melatonin is involved in the protection of these cells. Melatonin appears to have varied functions at different stages of follicle development, oocyte maturation, and luteal stage. Melatonin concentration in the growing follicle may be an important factor in avoiding atresia, because melatonin in the follicular fluid reduces apoptosis of critical cells. Melatonin also has protective actions during oocyte maturation reducing intrafollicular oxidative damage. An association between melatonin concentrations in follicular fluid and oocyte quality has been reported; this would allow a preovulatory follicle to fully develop and provide a competent oocyte for fertilization. The functional role of reactive species and the cytoprotective properties of melatonin on the ovary from oxidative damage are summarized in this brief review.  相似文献

A novel freeze-quench instrument with a characteristic «dead-time» of 137±18 μs is reported. The prototype has several key features that distinguish it from conventional freeze-quench devices and provide a significant improvement in time resolution: (a) high operating pressures (up to 400 bar) result in a sample flow with high linear rates (up to 200 m s−1); (b) tangential micro-mixer with an operating volume of ∼1 nl yields short mixing times (up to 20 μs); (c) fast transport between the mixer and the cryomedium results in short reaction times: the ageing solution exits the mixer as a free-flowing jet, and the chemical reaction occurs “in-flight” on the way to the cryomedium; (d) a small jet diameter (∼20 μm) and a high jet velocity (∼200 m s−1) provide high sample-cooling rates, resulting in a short cryofixation time (up to 30 μs). The dynamic range of the freeze-quench device is between 130 μs and 15 ms. The novel tangential micro-mixer efficiently mixes viscous aqueous solutions, showing more than 95% mixing at η≤4 (equivalent to protein concentrations up to 250 mg ml−1), which makes it an excellent tool for the preparation of pre-steady state samples of concentrated protein solutions for spectroscopic structure analysis. The novel freeze-quench device is characterized using the reaction of binding of azide to metmyoglobin from horse heart. Reaction samples are analyzed using 77 K optical absorbance spectroscopy, and X-band EPR spectroscopy. A simple procedure of spectral analysis is reported that allows (a) to perform a quantitative analysis of the reaction kinetics and (b) to identify and characterize novel reaction intermediates. The reduction of dioxygen by the bo3-type quinol oxidase from Escherichia coli is assayed using the MHQ technique. In these pilot experiments, low-temperature optical absorbance measurements show the rapid oxidation of heme o3 in the first 137 μs of the reaction, accompanied by the formation of an oxo-ferryl species. X-band EPR spectroscopy shows that a short-living radical intermediate is formed during the oxidation of heme o3. The radical decays within ∼1 ms concomitant with the oxidation of heme b, and can be attributed to the PM reaction intermediate converting to the oxoferryl intermediate F. The general field of application of the freeze-quench methodology is discussed.  相似文献

Fourteen different erythrinaline alkaloids have been isolated from the flowers and pods of Erythrina lysistemon with four being reported for the first time in nature and five for the first time in this species and the rest having been re-isolated. The new compounds are (+)-11beta-hydroxyerysotramidine (1), (+)-11beta-methoxyerysotramidine (2), (+)-11beta-hydroxyerysotrine N-oxide (4) and (+)-11beta-hydroxyerysotrine (8). (+)-11alpha-Hydroxyerysotrine N-oxide (3), earlier misidentified as erythrartine N-oxide (beta-hydroxyerysotrine N-oxide 4), was also re-isolated along with four other alkaloids. Correct identification of compounds 4 and 8 was aided by the fact that the two sets of C-11 epimers 3, 4 and 8, 9 were both isolated in this study thus making it easier to identify and assign the individual epimers. (+)-Erythristemine (14) was found distributed in most of the plant parts investigated. Preliminary work on the crude chloroform/methanol (1:1) showed moderate toxicity to brine shrimp (LC50 23 ppm) and moderate (IC50 86 microg/ml) radical scavenging properties against stable 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical. The DPPH radical scavenging properties of the isolated compounds were assessed using TLC autographic and spectrophotometric assays whereupon only compounds 11 (1 microg; 90 microg/ml) and 12 (0.1 microg; 160 microg/ml) showed any notable activity. It appears the two compounds are slow reacting and do not reach steady state conditions within the standard half an hour time frame but only seemed to have reached steady state conditions after 4 h.  相似文献
