BACKGROUND: Foot-and-mouth disease virus (FMDV) affects susceptible livestock animals and causes disastrous economic impact. Immunization with plasmid expressing VP1 that contains the major antigenic epitope(s) of FMDV as cytoplasmic protein (cVP1) failed to elicit full protection against FMDV challenge. MATERIALS AND METHODS: In this study, mice were immunized via electroporation with four cDNA expression vectors that were constructed to express VP1 of FMDV, as cytoplasmic (cVP1), secreted (sVP1), membrane-anchored (mVP1) or capsid precursor protein (P1), respectively, to evaluate whether expression of VP1 in specific subcellular compartment(s) would result in better immune responses. RESULTS: Electroporation enhanced immune responses to vectors expressing cVP1 or P1 and expedited the immune responses to vectors expressing sVP1 or mVP1. Immunization of mice via electroporation with mVP1 cDNA was better than sVP1 or cVP1 cDNA in eliciting neutralizing antibodies and viral clearance protection. Vaccination with P1 cDNA, nonetheless, yielded the best immune responses and protection among all four cDNAs that we tested. CONCLUSIONS: These results suggest that the antigenicity of a VP1 DNA vaccine can be significantly enhanced by altering the cellular localization of the VP1 antigen. Electroporation is a useful tool for enhancing the immune responses of vectors expressing VP1 or P1. By mimicking FMDV more closely than that of transgenic VP1 and eliciting immune responses favorably toward Th2, transgenic P1 may induce more neutralizing antibodies and better protection against FMDV challenge.  相似文献

Non-human primates are valuable models for biomedical research because of their similarities to human anatomy, immunology and physiology. Leishmaniasis, a disease caused by protozoan parasites, has a worldwide distribution and results in high morbidity and mortality. Availability of a non-human primate model of leishmaniasis would facilitate the study of different aspects of this disease and would accelerate the development of vaccines and new drugs. In this article, some interesting features of the vervet monkey (African Green monkey) model of human cutaneous and visceral leishmaniasis are discussed.  相似文献

Long-term retention of zooxanthellae in five different species belonging to two different nudibranch groups (Aeolidoidea and Dendronotoidea) was investigated. Specimens belonging to the species Phyllodesmium briareum, Phyllodesmium colemani, Phyllodesmium longicirrum, Pteraeolidia ianthina and Melibe engeli were cultivated for 70–270 days under various feeding conditions, and photosynthetic activity was analysed by taking pulse amplitude modulated (PAM) fluorometer measurements. All five species showed stable symbiosis and long-term retention of zooxanthellae. Interspecific differences are discussed. Morphological adaptations for housing zooxanthellae in the digestive glandular system were investigated and documented by histological means.  相似文献

Bordetella pertussis infection remains an important public health problem worldwide despite decades of routine vaccination. A key indicator of the impact of vaccination programmes is the inter-epidemic period, which is expected to increase with vaccine uptake if there is significant herd immunity. Based on empirical data from 64 countries across the five continents over the past 30–70 years, we document the observed relationship between the average inter-epidemic period, birth rate and vaccine coverage. We then use a mathematical model to explore the range of scenarios for duration of immunity and transmission resulting from repeat infections that are consistent with empirical evidence. Estimates of pertussis periodicity ranged between 2 and 4.6 years, with a strong association with susceptible recruitment rate, defined as birth rate × (1 − vaccine coverage). Periodicity increased by 1.27 years on average after the introduction of national vaccination programmes (95% CI: 1.13, 1.41 years), indicative of increased herd immunity. Mathematical models suggest that the observed patterns of pertussis periodicity are equally consistent with loss of immunity that is not as rapid as currently thought, or with negligible transmission generated by repeat infections. We conclude that both vaccine coverage and birth rate drive pertussis periodicity globally and that vaccination induces strong herd immunity effects. A better understanding of the role of repeat infections in pertussis transmission is critical to refine existing control strategies.  相似文献

The marine environment is commonly used for the deliberate disposal of industrial, mining and metabolic wastes. Managers will benefit from experimental work that identifies ways of reducing environmental impacts by varying the frequency and intensity of toxicant release. Using a field dosing technique we investigated the effects of three frequencies of pulse copper pollution event, and two intensities of copper dose, on developing assemblages of sessile marine invertebrates. The resulting impacts could then be compared in assemblages exposed to the same amount of toxicant via different disturbance regimes. The experiment was replicated simultaneously at three sites within Port Philip Bay, Victoria, Australia. Pulse pollution events altered assemblage composition through a direct negative effect on densities of large solitary ascidians. In response to the removal of the spatially dominant solitary ascidians, there were increases in recruitment of many different phyla, and in the abundance of older individuals of some serpulid and bryozoan taxa. Biodiversity, as measured by total species number and Shannon's H′, did not reflect the dramatic structural changes apparent within assemblages. If pulse pollution events had either a negative or positive effect on a species' density, then that effect was accentuated by increasing the intensity (strength) or the frequency of the pollution disturbance. Within populations, however, adult mortality might benefit new recruits of the same species through the freeing up of settlement space. In this case, the effects of copper pulses were evident as changes in the population size structure rather than the overall density of an organism. There were variable responses to manipulating the output rate of the toxicant between sites that appear to be driven by the rate of recovery of the dominant space occupiers. At one site there was a negligible effect of disturbance intensity and impacts could be minimized through reducing only the frequency of toxicant release. At two other sites both the intensity and frequency of disturbance determined the pollution effect and minimizing impacts was feasible only through an overall reduction in the amount of toxicant released. The management and reduction of pollution impacts through the control of toxicant release will require site- and season-specific modifications.  相似文献

ABSTRACT In west-central Texas, USA, abatement efforts for the gray fox (Urocyon cinereoargenteus) rabies epizootic illustrate the difficulties inherent in large-scale management of wildlife disease. The rabies epizootic has been managed through a cooperative oral rabies vaccination program (ORV) since 1996. Millions of edible baits containing a rabies vaccine have been distributed annually in a 16-km to 24-km zone around the perimeter of the epizootic, which encompasses a geographic area >4 × 105 km2. The ORV program successfully halted expansion of the epizootic into metropolitan areas but has not achieved the ultimate goal of eradication. Rabies activity in gray fox continues to occur periodically outside the ORV zone, preventing ORV zone contraction and dissipation of the epizootic. We employed a landscape-genetic approach to assess gray fox population structure and dispersal in the affected area, with the aim of assisting rabies management efforts. No unique genetic clusters or population boundaries were detected. Instead, foxes were weakly structured over the entire region in an isolation by distance pattern. Local subpopulations appeared to be genetically non-independent over distances >30 km, implying that long-distance movements or dispersal may have been common in the region. We concluded that gray foxes in west-central Texas have a high potential for long-distance rabies virus trafficking. Thus, a 16-km to 24-km ORV zone may be too narrow to contain the fox rabies epizootic. Continued expansion of the ORV zone, although costly, may be critical to the long-term goal of eliminating the Texas fox rabies virus variant from the United States.  相似文献

Examination of variables in the vaccination of mice against cutaneous leishmaniasis using living avirulent cloned lines and killed promastigotes of Leishmania major. International Journal for Parasitology15: 677–684. In vaccination experiments not involving adjuvants, genetically-susceptible mice were injected with living avirulent cloned promastigotes of Leishmania major or killed promastigotes prior to cutaneous challenge with virulent cloned promastigotes. Emphasis was placed on aspects that may contribute to marked variability between experiments and between laboratories in vaccination of mice against cutaneous leishmaniasis. One variable, the challenge promastigotes, was shown to be important in that cloned virulent parasites (V121) were less pathogenic in terms of rate of cutaneous lesion development, than the parental isolate LRC-L137 when low doses of promastigotes were used and particularly when harvested from the log phase of culture. It is likely that avirulent parasites in mixed isolates can increase the rate of lesion development after cutaneous deposition. As reported previously, intraperitoneal, and more particularly intravenous injections of living avirulent cloned parasites (A12) increase resistance in mice. Most importantly, a difference has been demonstrated in the vaccinating efficacy of killed promastigotes of various isolates injected intravenously. This implies that certain isolates of L. major (e.g. the “Moshkovsky strain”) express “host-protective antigens” at higher levels, or in a qualitatively different manner, than other isolates (e.g. LRC-L137). The finding will greatly facilitate the identification of vaccine antigens in this system using immunochemical and gene cloning approaches.  相似文献

The lysate of erythrocytes infected with Babesia bovis was tested for proteinases using an electrophoretic method in which substrate was included in the acrylamide matrix. Two babesial proteinases, which seemingly exist in both free and complexed forms, were detected. One of the proteinases was prepared by chromatography and preparative electrophoresis and used to vaccinate four splenectomized calves. The latter, along with a group of control splenectromized calves, were challenged with a strain of B. Bovis from which the proteinase was obtained. All the control calves died whereas only one of the vaccinates died. The protection was evident as a suppression of parasitaemia.  相似文献
