目的优选妇宁康凝胶剂的基质配方。方法通过体外释药试验,采用HPLC法测定接收液中甘草酸含量,并计算甘草酸释药速率;以甘草酸释药速率为筛选指标,采用L9(3^4)正交试验对妇宁康凝胶剂的的基质配方进行优选。结果妇宁康凝胶剂的最佳基质配方为:卡伯姆2%,甘油5%,EDTA-2Na0.006%,丙二醇10%,三乙醇胺约1g。结论甘草酸体外释药符合Higuchi方程,配方中甘油和丙二醇的含量为主要影响因素,有极显著影响。 相似文献

The horse mussel Atrina zelandica (Gray) is a large, suspension feeding pinnid bivalve, common in coastal and estuarine areas of northern New Zealand. As a suspension feeder, Atrina is likely to be influenced by suspended sediment loads. We conducted a laboratory experiment to determine the effect of short-term elevations in turbidity levels, such as those commonly recorded during storms, on the physiological condition and clearance rates of Atrina. We also conducted a field survey and a 3-month transplant experiment at multiple sites along a gradient of increasing suspended sediment load in a New Zealand estuary. Laboratory clearance rates of Atrina declined above a threshold suspended sediment concentration, and Atrina physiological condition at the end of this experiment was lower in high cf. low turbidity treatments. Decreases in Atrina condition were detected after exposure to elevated levels for only 3 days. The field survey and transplant experiment provided empirical evidence of a strong, negative effect of increasing suspended sediment flux on the physiological condition of Atrina. We suggest that relationships between the physiological condition of suspension feeders and sediment settling flux could provide a link between sediment inputs, which commonly occur as a result of catchment runoff during rainfall events, and the ecological health of estuarine and shallow coastal areas. Our study also demonstrated that Atrina have a natural distribution limit controlled by suspended sediment load. Thus, there is potential for larger-scale functional and structural effects on benthic communities in estuarine and coastal areas with high rates of sedimentation. 相似文献

Endothelin-1 (ET) is known to stimulate mesangial cell (MC) proliferation, extracellular matrix (ECM) synthesis, and thereby contribute to the progression of glomerulonephritis (GN). To clarify the molecular and cellular mechanisms of how ET is involved in the development of glomerular sclerosis, we investigated the influence of ET on the MC-alpha1beta1 integrin-mediated collagen matrix reorganization using a collagen gel contraction assay. ET enhanced MC-alpha1beta1 integrin-mediated gel contraction in a dose-dependent manner. Addition of the endothelin A (ETA) receptor antagonist, BQ123, into collagen gels abolished ET-induced gel contraction by MC. Cell behavior involved in ET-induced gel contraction was investigated in combination with function-blocking anti-alpha1-integrin antibody. Migration and adhesion assays revealed that ET stimulated alpha1beta1 integrin-mediated MC migration but did not influence cell adhesion to type I collagen (collagen I). Integrin-function blocking studies using anti-alpha1 integrin antibody indicated that MC-alpha1beta1 integrin is required not only for collagen-dependent migration, but also for gel contraction. Zymography showed that ET increased MC matrix metalloproteinase-2 (MMP-2) activity in a dose-dependent manner during MC-induced gel contraction process. Finally, flow cytometry analysis indicated that ET did not affect the cell surface expression of the MC-alpha1beta1 integrin within the collagen gel. These data suggested that ET promotes collagen matrix reorganization through the enhancement of MC-alpha1beta1 integrin-dependent migration and MMP-2 activity. We therefore conclude that ET is a potential molecule inducing pathological collagen matrix remodeling observed in progressive GN. 相似文献

Purpose: Dosimetry of ionizing radiation quantifies the energy deposited by an incident beam to the medium. This study presents the relative response of two types of gel dosimeters describing their differences by estimating radiation chemical yields produced in water radiolysis.Methods: Two types of gel dosimeter were used, namely an acid ferrous ion solution infused with xylenol orange known as Fricke gel and a polymer gel based on acrylamide and N,N’-methylenebis(acrylamide) known as PAGAT. Samples were irradiated using two photon beam energies, one from a conventional X-ray tube operated at 44 kV and the other one from a LINAC operated at 6 MV. The dosimeters were analyzed by optical absorbance and magnetic resonance imaging. Additionally, the linear energy transfer of each beam was calculated using Monte Carlo simulations for further estimation of the radiation chemical yields produced during water radiolysis.Results: Obtained results for both gel dosimeters indicate that their response at 44 kV and 6 MV are different, regardless of the read-out technique. On average, the sensitivity at 44 kV was found to be 65 % of the response at 6 MV. The calculated radiation chemical yields are in agreement with the observed experimental results.Conclusions: The main reason for the difference in the response of the dosimeters may be related to the linear energy transfer of each photon beam, which varies the production of primary chemical species during water radiolysis. 相似文献

Summary Leek plants were preinoculated with a mixed inoculum ofGlomus caledonium, Glomus fasciculatum andGlomus sp., and transplanted to Dazomet disinfected and untreated field plots of moderate P deficiency. Successive harvests were
made until 99 days after transplanting. Preinoculated leeks attained marketable weights 25 days earlier than uninoculated
leeks from untreated soil and their final dry matter yields were 5.7 and 1.5 times as high as those of uninoculated leeks
from disinfected and untreated soil, respectively. Phosphorus concentration in preinoculated leeks remained highest for at
least 22 and 75 days after transplanting in untreated and disinfected soil, respectively. Preinoculation had a similar, although
smaller, influence on Cu and Zn concentrations. Infection levels produced by introduced and indigenous VA endophytes in leeks
reached plateaus of 90% and 40%, respectively, 47 days after transplanting. It is concluded that VAM is essential to leeks

The role of water molecules in assisting proton transfer (PT) is investigated for the proton-pumping protein ferredoxin I (FdI) from Azotobacter vinelandii. It was shown previously that individual water molecules can stabilize between Asp(15) and the buried [3Fe-4S](0) cluster and thus can potentially act as a proton relay in transferring H(+) from the protein to the μ(2) sulfur atom. Here, we generalize molecular mechanics with proton transfer to studying proton transfer reactions in the condensed phase. Both umbrella sampling simulations and electronic structure calculations suggest that the PT Asp(15)-COOH + H(2)O + [3Fe-4S](0) → Asp(15)-COO(-) + H(2)O + [3Fe-4S](0) H(+) is concerted, and no stable intermediate hydronium ion (H(3)O(+)) is expected. The free energy difference of 11.7 kcal/mol for the forward reaction is in good agreement with the experimental value (13.3 kcal/mol). For the reverse reaction (Asp(15)-COO(-) + H(2)O + [3Fe-4S](0)H(+) → Asp(15)-COOH + H(2)O + [3Fe-4S](0)), a larger barrier than for the forward reaction is correctly predicted, but it is quantitatively overestimated (23.1 kcal/mol from simulations versus 14.1 from experiment). Possible reasons for this discrepancy are discussed. Compared with the water-assisted process (ΔE ≈ 10 kcal/mol), water-unassisted proton transfer yields a considerably higher barrier of ΔE ≈ 35 kcal/mol. 相似文献

The field cricket Teleogryllus oceanicus has been introduced to Hawaii, where it is parasitized by an acoustically orienting parasitoid fly, Ormia ochracea. Previous work showed that call parameters from parasitized populations differ from those in unparasitized populations in a direction expected if selection by flies is occurring. Here we examined songs of males collected in the field and compare calling song characters of crickets later found to harbor parasitoid larvae with those of males free of parasitoids. The two groups differ significantly in several song characteristics, particularly the trill-like long chirp given at the beginning of each song. Males with longer long chirps containing shorter interpulse intervals are more likely to be parasitized, suggesting that the flies find such males more attractive. Depending on the traits females prefer, sexual selection may oppose natural selection in altering T. oceanicus song in parasitized populations. 相似文献

Fifty-four isolates of Sclerotinia sclerotiorum from Ranunculus acris and other natural hosts were applied as mycelial infested kibbled wheat onto 6 month-old R. acris plants in two glasshouse screening experiments. Most isolates (90%) did not differ in their pathogenicity towards R. acris. One isolate, S. sclerotiorum G45, was selected based on its ability to cause severe disease and suppress regeneration of R. acris. A field experiment was conducted to determine the efficacy of S. sclerotiorum (G45) against R. acris in infested dairy pastures in the Takaka Valley, Golden Bay, New Zealand. Isolate G45 was formulated as a wettable powder and was applied as a slurry at 20 and 40 ml/plant in December 1995. After 10 weeks, regeneration from the crown of treated plants was apparent and a second application of S. sclerotiorum was made in February 1996. Best control of R. acris was obtained when the plants were inoculated in full flower in December. At the first time of treatment, the 40 ml application of S. sclerotiorum slurry reduced the total dry weight of R. acris by an average of 57%. The second application had no effect on total dry weight, possibly because moisture levels were not sufficient for S. sclerotiorum infection. This study confirmed S. sclerotiorum to be an aggressive pathogen of R. acris under both glasshouse and field conditions. As a result, this pathogen has potential as a mycoherbicide for R. acris. Further experiments are required to explore ways of enhancing the efficacy of S. sclerotiorum against R. acris by manipulation of the host, pathogen and environment. 相似文献

Helicoverpa armigera single nucleocapsid nucleopolyhedrovirus (HaSNPV) has been developed as a commercial biopesticide to control the cotton bollworm, H. armigera, in China. The major limitation to a broader application of this virus has been the relative long time to incapacitate the target insect. Two HaSNPV recombinants with improved insecticidal properties were released in bollworm-infested cotton. One recombinant (HaCXW1) lacked the ecdysteroid UDP-glucosyltransferase (egt) gene and in another recombinant (HaCXW2), an insect-selective scorpion toxin (AaIT) gene replaced the egt gene. In a cotton field situation H. armigera larvae treated with either HaCXW1 or HaCXW2 were killed faster than larvae in HaSNPV-wt treated plots. Second instar H. armigera larvae, which were collected from HaCXW1 and HaCXW2 treated plots and further reared on artificial diet, showed reduced ST(50) values of 15.3 and 26.3%, respectively, as compared to larvae collected from HaSNPV-wt treated plots. The reduction in consumed leaf area of field collected larvae infected with HaCXW1 and HaCXW2 was approximated 50 and 63%, respectively, as compared to HaSNPV-wt infected larvae at 108 h after treatment. These results suggest that in a cotton field situation the recombinants will be more effective control agents of the cotton bollworm than wild-type HaSNPV. 相似文献

As global climate is warming and the nitrogen cycle accelerates, plants are likely to respond not only by shifting community composition, but also by adjusting traits such as tissue chemistry. We subjected a widespread wetland plant, Phragmites australis, to increased nitrate supply and elevated temperature in enclosures that were established in a littoral permanently submerged freshwater marsh. The nitrogen (N) and phosphorus (P) concentrations in green leaves ranged from 11.4 to 13.8 mg N and from 1.5 to 2.0 mg P g−1 dry mass. While the N concentration changed little in brown litter, the P concentration decreased to 0.53–0.65 mg P g−1 litter dry mass. Neither experimental warming of the water and sediment surface, nor nitrate enrichment during the growing season affected nitrogen or phosphorus concentrations in green leaves. Concentrations of the two major structural carbon compounds in plant litter, cellulose and lignin, were also unaffected, ranging from 32.1 to 34.2% of dry mass for cellulose and from 16.3 to 17.7% of dry mass for lignin. Warming, however, significantly increased the nitrogen concentration of fully brown leaf litter. Thus, temperature appears to be more important than the supply of dissolved N in the water, especially in affecting leaf litter N concentrations in P. australis, even when only water but not air temperature is increased. This result may have implications for decomposition processes and decomposer food webs, which both depend on the quality of plant litter. 相似文献
