病理性瘢痕中主要氧化酶和抗氧化酶活性测定   总被引:1,自引:0,他引:1  
采用化学比色法测定正常皮肤(8例)、增生性瘢痕(10例)及瘢痕疙瘩(10例)组织中黄嘌呤氧化酶(xanthine oxidase,XO)、铜锌超氧化物歧化酶(copper,zinc-superoxide dismutase,CuZn-SOD)、过氧化氢酶(catalase,CAT)、谷胱甘肽过氧化物酶(glutathione peroxidase,GPX)活性以及丙二醛(malonaldehyde,MDA)含量.结果表明:与正常皮肤比较,病理性瘢痕中XO和CuZn-SOD活性增加、CAT活性降低(P<0.05)而GPX活性不变,CAT/CuZn-SOD和GPX/CuZn-SOD活性比率下降(P<0.05),同时MDA含量升高(P<0.05).增生性瘢痕、瘢痕疙瘩之间比较均无差异.上述结果表明,在病理性瘢痕中,氧化酶XO,抗氧化酶CuZn-SOD、CAT以及GPX的活性改变可能是引起活性氧水平升高的原因之一,在抗氧化剂选择上,CAT可能较为合理.  相似文献

The present work quantifies hyaluronan (HA) during the late pregnancy and post-partum in order to provide a better understanding of the role of HA in the adaptations that occur in the pubic symphysis during this period. HA was quantified in situ (histochemically) and in interpubic tissue extracts by fluorimetric assay. Samples were taken from virgin mice and from pregnant animals at various stages of pregnancy: 12th-18th days into pregnancy, the day of delivery (D19) and the 3rd and 5th day post-partum. The quantitative fluorimetric analysis indicated a gradual increase of HA in the interpubic tissue throughout late pregnancy (2.4-14.6 microg/mg dry weight). This was followed by a decrease beginning on D19 (12.4 microg/mg), reaching close to virgin levels (2.2 microg/mg) on the 5th day post-partum. The same optical density changes could be seen in the HA staining. Furthermore, the histochemical analysis demonstrated the presence of HA both in the extracellular matrix of the tissue and within its cells. Such results indicate that the extracellular presence of HA may contribute to the transformation of the symphysis into a flexible structure. In addition, HA's intracellular presence (until the 18th day of pregnancy) may contribute to cellular proliferation. Finally, during parturition and on the 5th day post-partum, HA may contribute to the maintenance of the myofibroblastic phenotype of ligament cells, aiding the ligament involution after parturition.  相似文献

Debris slide occurrence on treed slopes of northeastern North America is still poorly documented, despite their abundance and their potential to change mountainous landscapes in short periods of time. To provide new information on their spatiotemporal dynamics, a study was undertaken in debris slide paths in the Wildlife Reserve of Port-Cartier-Sept-Iles, on the Québec North-Shore region of eastern Canada. Tree-ring dating of growth anomalies (impact scars and reaction wood) in nine debris slides allowed the identification of four debris slide events that occurred in 2003, 2006, 2008, and 2010. By comparison to other hillslope processes such as snow avalanches and debris flows, debris slides produce a very strong tree-ring signal. Therefore they do not require a large sample size considering also that they do not occur twice at the same place. The position of growth anomalies within individual tree rings allowed to determine the timing of the debris slide events: injuries located within a ring correspond to debris slides occurring during the growing season, whereas injuries located between the end of a ring and the beginning of the following ring were caused by debris slides occurring during the dormant season. The meteorological data indicate that a daily precipitation of 70 mm appears usually sufficient for the occurrence of debris slides.  相似文献

Abstract: We evaluated the use of naturally occurring nose scars to identify individual sea otters (Enhydra lutris) in Simpson Bay, Prince William Sound, Alaska, USA. We spent 520 hours over 103 days conducting photo-identification surveys from June to August 2002 and 2003. Altogether, we identified 114 individuals. The number of sightings per individual ranged from 1 to 26, with an average of 3.3. The maximum number of sightings of an individual within a single year was 19. We saw 54 otters (47%)on >1 day, with an average of 8.1 sightings per individual for those seen more than once. We identified 8 individuals (19% of those identified in 2002) in both years. Males and otters of undetermined sex that we first sighted in June had the highest re-sighting rates. We considered 45% of all individuals encountered identifiable from nose scars. Nose scars were present in 63% (n = 19) of males, 45% (n = 45) of females, and 40% (n = 49) of otters of undetermined sex. Our results are similar to the results of photo-identification studies of other marine mammals, suggesting that this technique may be a useful tool for the individual identification of sea otters as well.  相似文献

Aim In this study we examine fire history (i.e. c. 500 yr bp to present) of AraucariaNothofagus forests in the Andes cordillera of Chile. This is the first fire history developed from tree rings for an AraucariaNothofagus forest landscape. Location The fire history was determined for the Quillelhue watershed on the north side of Lanin volcano in Villarrica National Park, Chile. The long‐lived Araucaria araucana was commonly associated with Nothofagus pumilio and N. antarctica in more mesic and drier sites respectively. Methods Based on a combination of fire‐scar proxy records and forest stand ages, we reconstructed fire frequency, severity, and the spatial extent of burned areas for an c. 4000 ha study area. We used a composite fire chronology for the purpose of determining centennial‐scale changes in fire regimes and comparing the pre‐settlement (pre‐1883) and post‐settlement fire regimes. In addition, we contrasted Araucaria and Nothofagus species as fire‐scar recorders. Results In the study area, we dated a total of 144 fire‐scarred trees, representing 46 fire years from ad 1446 to the present. For the period from ad 1696 to 2000, using fire dates from Araucaria and Nothofagus species, the composite mean fire interval varied from 7 years for all fires to 62 years for widespread events (i.e. years in which ≥ 25% of recorder trees were scarred). Sensitivity to fire was different for Araucaria and Nothofagus species. More than 98% of the fires recorded by Nothofagus species occurred during the 1900s. The lack of evidence for older fire dates (pre‐1900) in Nothofagus species was due to their shorter longevity and greater susceptibility to being killed by more severe fires. Whereas the thin‐barked N. pumilio and N. antarctica are often destroyed in catastrophic fire events, large and thick‐barked Araucaria trees typically survive. The spatial extent of fires ranged from small patchy events to those that burned more than 40% of the entire landscape (c. > 1500 ha). Main conclusions Fire is the most important disturbance shaping the AraucariaNothofagus landscape in the Araucarian region. The forest landscape has been shaped by a mixed‐severity fire regime that includes surface and crown fires. High‐severity widespread events were relatively infrequent (e.g. 1827, 1909 and 1944) and primarily affected tall AraucariaN. pumilio forests and woodlands dominated by AraucariaN. antarctica. Although there is abundant evidence of the impact of Euro‐Chilean settlers on the area, the relative influence of this settlement on the temporal pattern of fire could only be tentatively established due to the relatively small number of pre‐1900 fire dates. An apparent increase in fire occurrence is evident in the fire record during Euro‐Chilean settlement (post‐1880s) compared with the Native American era, but it may also be the result of the destruction of evidence of older fires by more recent stand‐devastating fires (e.g. 1909 and 1944). Overall, the severe and widespread fires that burned in AraucariaNothofagus forests of this region in 2002, previously interpreted as an ecological novelty, are within the range of the historic fire regimes that have shaped this forested landscape.  相似文献

Proteoglycans were accurately localized in mouse pubic symphyseal tissues using the cuprolinic blue method. Specific glycosaminoglycans degradative enzymes, together with chondroitin sulfate and decorin antibodies, allowed the identification of glycosaminoglycans. Chondroitin sulfate proteoglycans were the main proteoglycans observed in hyaline cartilage, fibrocartilage, and dense connective tissue. Ultrastructurally, they were seen as electron-dense granules and filaments. The granules, rich in chondroitin sulfate chains, were exclusively found in hyaline cartilage, whereas filaments were present in cartilage, fibrocartilage, and dense connective tissue. The latter were classified by size and susceptibility to enzyme digestion into F1, F2 and F3 filaments: F1 filaments were small, thin, and collagen fibril-associated; F2 filaments were thick, heavily stained, and localized around individual collagen fibrils and between bundles of collagen fibrils; and F3 filaments were scattered throughout elastic fiber surfaces. Considering their localization, susceptibility to chondroitinase AC and immunohistochemical detection, the symphysial F1 filaments were found to be preferentially decorin substituted with chondroitin sulfate side chains. The F2 filaments were also susceptible to chondroitinase AC treatment, whereas F3 filaments could be digested by heparitinase.The data thus obtained on the localization and identification of pubic symphyseal proteoglycans in virgin mice may be useful in the study of structural modifications that occur throughout pregnancy.  相似文献
