Heterodera zeae, the corn cyst nematode, is redescribed and illustrated with comparative details and measurements of females, cysts, and larvae from Maryland, USA; and India. Scanning electron micrographs o f specimens from the United States are also presented. Luminol chemiluminescence was used to evaluate the scavenging of superoxide, hydroxyl and alkoxy radicals by four antioxidants: dipyridamole, diethyldithiocarbamic acid, (+)catechin, and ascorbic acid. Different concentrations of these compounds were compared with well-known oxygen radical scavengers in their capacity to inhibit the chemiluminescence produced in the reaction between luminol and specific oxygen radicals. Hydroxyl radicals were generated using the Fenton reaction and these produced chemiluminescence which was inhibited by diethyldithiocarbamate. Alkoxy radicals were generated using the reaction of tert-butyl hydroperoxide and ferrous ion and produced chemiluminescence which was inhibited equally by all of the compounds tested. For the determination of superoxide scavengers we describe a new, simple, economic, and rapid chemiluminescence method consisting of the reaction between luminol and horseradish peroxidase (HRP). With this method it was found that 40 nmol/l dipyridamole, 0.18 μmol/l ascorbic acid, 0.23 μmol/l (+)catechin, and 3 μmol/l diethyldithiocarbamic acid are equivalent to 3.9 ng/ml superoxide dismutase (specific scavenger of superoxide) in causing the same degree of chemiluminescence inhibition. These results not only indicated that the antioxidative properties of these compounds showed different degrees of effectiveness against a particular radical but also that they may exert their action against more than one radical. 相似文献

We present primer sequences for eight polymorphic microsatellite loci for the formicine ant Anoplolepis gracilipes, a serious pest species in South‐East Asia and Pacific islands and still spreading on all continents. Microsatellite loci were isolated with a highly efficient method of enrichment. The number of alleles ranged from two to 19 with an observed heterozygosity ranging from 0.842 to 1.0. Grasslands created by grass invasions into shrublands or woodlands followed by fire are now a dominant feature of many seasonally dry environments. In Hawaii Volcanoes National Park, introduced perennial grasses dominate grasslands created by fire in grass‐invaded woodlands. In a previous study, we found that net primary production in these grasslands is substantially lower than in unburned woodlands. Yet, our estimates of annual net nitrogen (N) mineralization showed higher rates in these savannas than in the unburned woodlands, rates that appear to greatly exceed annual N demand by the vegetation. We therefore hypothesized that N should not be limited to the plants growing in these sites. We tested this hypothesis with a 2‐yr fertilization experiment. At peak biomass, we found a 30 percent increase in live biomass in plots with N added and no increase in production with only phosphorus (P) added. N and P together were synergistic, suggesting that co‐limitation or P limitation becomes important when N is more available. Plants responded to added N by increasing individual leaf area and shoot length by over 50 percent. Tissue N was higher with added N; hence, biomass N was substantially higher. Tissue P concentrations declined with N addition but were elevated by P addition despite lack of a growth response to P alone. Overall, N limitation exists despite high annual rates of net N mineralization, and co‐limitation of production by P may occur when N is abundant. Here, asynchrony between plant nutrient demand and N availability may contribute to N limitation. 相似文献

Death receptors such as the 55 kDa tumor necrosis factor (TNF) receptor (TNF-R55) or Fas can initiate both apoptotic (caspase-dependent) and caspase-independent routes to programmed cell death (PCD). Here, we demonstrate for the first time that the single murine receptor for (TNF)-related apoptosis-inducing ligand (mTRAIL-R2) can induce a caspase-independent form of PCD with necrosis-like features in addition to apoptosis. Analysis of morphological and cellular features of caspase-independent PCD in response to TRAIL and TNF suggests that mTRAIL-R2 and TNF-R55 elicit caspase-independent PCD through similar pathways, although without participation of cathepsins. Cells overexpressing acid ceramidase (AC), an enzyme that metabolizes the sphingolipid ceramide, show enhanced survival from TRAIL-induced caspase-independent PCD but not from apoptosis, implicating a function of ceramide as a key mediator in caspase-independent PCD (but not apoptosis) induced by mTRAIL-R2. In concert with the enhanced resistance of AC-overexpressing cells against caspase-independent PCD induced by TNF, our results suggest that ceramide acts as a common mediator of caspase-independent PCD caused by death receptors such as mTRAIL-R2 and TNF-R55. 相似文献

该研究采用典型抽样法,以成都云顶山5种人工林——柏木 枫杨林(BF)、银杏 楠木林(YN)、光皮梾木 香樟林(GZ)、枫杨 桤木林(FQ)、柏木林(CB)为研究对象,分析不同人工林的林下植物组成与多样性特征,并确定影响林下植物多样性的主导林分因子,为当地人工林经营管理提供理论依据。结果表明:(1)研究区共记录林下植物168种,隶属于62科130属;5种人工林灌木层与草本层的科属种数均以GZ最多。(2)5种不同人工林灌木层与草本层的优势种数分别为7、4、7、6、4种和5、4、9、9、10种,数量都较少。(3)5种人工林的Shannon Wiener多样性指数(H)、Simpson优势度指数(H′)、物种丰富度指数(D)、Pielou均匀度指数(Jsw) 均基本表现为草本层>灌木层,BF、GZ灌木层的D略高;灌木层的H、H′、D值均以GZ最大,但不同人工林的Jsw差异不显著;草本层的H、H′、D、Jsw均基本呈现CB>FQ>GZ>BF>YN趋势,GZ的D值略高于FQ。(4)6个林分因子对灌木层4个物种多样性指数的影响均无显著差异;林分平均树高、平均枝下高、平均胸径、平均冠幅和林分密度是影响草本层H、D的主要因子,但各林分因子对草本层H′、Jsw的影响差异不显著。研究认为,林分结构对林下草本层物种多样性的影响更大,平均树高、平均枝下高、平均胸径、平均冠幅、林分密度对草本层多样性有显著影响。 相似文献

Metal coordinating propertiesof DOC (dissolved organic carbon), and henceits influence on heavy metal release andmineral weathering, depend on the compositionand properties of DOC. Tree species producelitter with different chemical composition anddegradability, and these differences mightinfluence the composition and reactivity of DOCin soil solutions. Accordingly, analysis ofcomposition and reactivity of DOC in soilsolution samples collected by centrifugationfrom 16 forest soil O horizons from the fourtree species beech (Fagus sylvatica L.),oak (Quercus robur L.), grand fir (Abies grandis Lindl.), and Norway spruce (Picea abies (L.) Karst.) on two clayey and twosandy soils were carried out. The compositionand properties of DOC were determined bycapillary zone electrophoresis, acid-basetitration, Cu ion titration, total elementalanalysis, IR and UV spectroscopy, and metalrelease assays. Concentrations of DOC rangedfrom 20 to 163 mM with pH ranging from 4.6 to7.3. Norway spruce produced the highest DOCconcentration, and the lowest pH. Carbon inlow-molecular-weight aliphatic carboxylic acids(LACA) accounted for less than 6% of DOC withformic and acetic acids as the most abundantLACAs. The DOC was cation exchanged and protonsaturated to obtain comparable forms of DOC.Titratable carboxylic acid and phenolic groupswere in the range 51 to 82 and 20 to64 mmol·mol–1 C, respectively, with fewerphenolic groups in grand fir DOC as the onlysignificant difference. Infrared spectra offreeze-dried DOC samples suggest low contentsof aromatic C in the DOC especially from grandfir stands. Stability constants, log K of Cu-DOC complexes, determined by Cu ion titrationof DOC samples with fitting of the data to atwo-site binding model, were in the range 5.63to 6.21 for the strong binding sites and 3.58to 4.10 for the weak sites, but with nosignificant effects of tree species or site.Freeze-dried DOC samples were found to consistof 41 to 45% C, 38 to 49% O, 4.4 to 5.4% Hand 1.2 to 2.0% N and C/N ratios in the range26 to 42. Reactivity of DOC in terms of releaserates of Cd, Cu and Fe cations from a soilsample (flow cell experiments) showed nosignificant differences among DOC samples fromdifferent tree species and soil types.Apparently, only minor differences occur inchemical composition and reactivity ofequivalent concentrations of DOC in forestfloor soil solutions irrespective of origin,i.e. four tree species and two soil types. Soilsolution pH and the concentration of DOCproduced by various tree species are thecritical parameters when distinguishing amongtree species in relation to heavy metal releaseand mineral weathering. 相似文献

The Argentine ant (Linepithema humile) is a damaging invasive species that has become established in many Mediterranean-type ecosystems worldwide. To identify likely sources of introduced populations we examined the relationships among native Linepithema populations from Argentina and Brazil and introduced populations of L. humile using mitochondrial cytochrome b sequence data and nuclear microsatellite allele frequencies. The mitochondrial phylogeny revealed that the populations in Brazil were only distantly related to both the introduced populations and the native populations in Argentina, and confirmed that populations in Brazil, previously identified as L. humile, are likely a different species. The microsatellite-based analysis provided resolution among native and introduced populations of L. humile that could not be resolved using the mitochondrial sequences. In the native range, colonies that were geographically close to one another tended to be genetically similar, whereas more distant colonies were genetically different. Most samples from the introduced range were genetically similar, although some exceptions were noted. Most introduced populations were similar to native populations from the southern Rio Parana and were particularly similar to a population from Rosario, Argentina. These findings implicate populations from the southern Rio Parana as the most likely source of introduced populations. Moreover, these data suggest that current efforts to identify natural enemies of the Argentine ant for biological control should focus on native populations in the southern Rio Parana watershed. 相似文献

The Clytra subfasciata species group is proposed, to comprise Clytra subfasciata Lacordaire, 1848, C. hajeki Medvedev &; Kantner, 2002 and C. kadleci sp. n. (Yemen). The species of the group are characterised by the peculiar shape of the aedeagus, with a dorsal plate separated from the rest of aedeagus by a deep narrow slit.
