A well‐written application for funding in support of basic biological or biomedical research or individual training fellowship requires that the author perform several functions well. They must (i) identify an important topic, (ii) provide a brief but persuasive introduction to highlight its significance, (iii) identify one or two key questions that if answered would impact the field, (iv) present a series of logical experiments and convince the reader that the approaches are feasible, doable within a certain period of time and have the potential to answer the questions posed, and (v) include citations that demonstrate both scholarship and an appropriate command of the relevant literature and techniques involved in the proposed research study. In addition, preparation of any compelling application requires formal scientific writing and editing skills that are invaluable in any career. These are also all key components in a doctoral dissertation and encompass many of the skills that we expect graduate students to master. Almost 20 years ago, we began a grant writing course as a mechanism to train students in these specific skills. Here, we describe the use of this course in training of our graduate students as well as our experiences and lessons learned.  相似文献

The purpose of this study was to show that velocity-specific training may be implicated in modifications in the level of coactivation of agonist and antagonist muscles. Healthy males (n = 20) were randomly placed in to two groups: one group trained using concentric contractions (n = 12), the other was an untrained control group (n = 8). The training group underwent unilateral resistance training at a level of 35 (5)% of a one-repetition maximal contraction of the elbow flexors, executed at maximal angular velocity. Training sessions consisted of six sets of eight consecutive elbow flexions, three times per weak for a total of seven weeks. The velocity of the ballistic movements executed during training were measured using an optoelectronic measuring device (Elite), both at the beginning and at the end of the training period. Subjects were tested pre- and post-training during isokinetic maximal elbow flexions with constant angular torque (CAT) at 90 degrees (0 degrees = full extension), and at different velocities (60, 120, 180, 240 and 300 degrees x s(-1)) for concentric actions, and -60 and -30 degrees x s(-1) for eccentric and isometric contractions at 90 degrees. In order to verify the levels of activation of the agonist biceps brachii (BB) muscles and antagonist triceps brachii (TB) muscles during maximal voluntary activation, their myoelectrical activities were recorded and quantified as root mean square (RMS) amplitudes, between angles of 75 and 105 degrees . The results show that mean angular velocities between elbow angles of 75 and 105 degrees were similar before [302 (32) degrees x s(-1)] and after [312 (27) degrees x s(-1)] the training period. CAT significantly increased measures at angular velocities of 240 and 300 degrees x s(-1) by 18.7% and 23.5%, respectively. The RMS activity of BB agonist muscles was not significantly modified by training. Post-training normalized RMS amplitudes of TB antagonist muscles were inferior to those observed at pre-training, but values were only significantly different at 300 x s(-1). In conclusion, in this study we attempted to show that an increase of CAT to 240 and 300 degrees x s(-1), though velocity-specific training, may be due, in part, to a lowering of the level of coactivation.  相似文献

In this study, the influence of athletic training status and the contractile character of the active muscle on the magnitude of the pressor response (PR) to voluntary and electrically evoked isometric plantar flexion was investigated. Subjects were 10 sprint-trained athletes (sprint) (100-m, 200-m and 400-m) [mean (SD) age, 21 (2) years], 14 endurance trained athletes (distance) [22 (2) years] and 8 untrained men (control) [23 (3) years]. Twitch time to peak tension (TPT) in the sprint group [108 (7) ms] was significantly less (P<0.001) than that of the distance group [124 (10) ms]. During voluntary contraction, the mean change in systolic blood pressure (SBP), diastolic blood pressure (DBP) and heart rate (fc) was not significantly different between groups. During electrically evoked contractions, mean changes in SBP, DBP and fc were not significantly different between the sprint, distance and control groups. However, division of the sprint group into 400-m (sprint I) and 100/200-m athletes (sprint II) showed that an increase in DBP of 1.6 kPa (12 mm Hg) in sprint I was significantly less (P<0.05) than the 2.5 kPa (19 mm Hg) increase observed for both the distance and control groups. Prediction of the DBP response from our previously published relationship between TPT and DBP showed close agreement in all subject groups except sprint I; in these subjects the observed DBP response was only 55% of that predicted. Attenuation of the PR in the involuntary experiment suggests that some aspect of sprint training, but not endurance training, modifies the muscle afferent input to the PR in man.  相似文献

Innate immunomodulation via induction of innate memory is one mechanism to alter the host’s innate immune response to reduce or prevent disease. Microbial products modulate innate responses with immediate and lasting effects. Innate memory is characterized by enhanced (training) or depressed (tolerance) innate immune responses, including pro-inflammatory cytokine production, to secondary exposure following a priming event. To investigate the ability of β-glucans and bacillus Calmette-Guerin to induce innate training or tolerance in pig cells, porcine monocytes were cultured with priming agonist (β-glucans or bacillus Calmette-Guerin) then re-stimulated 5 d later with a heterologous microbial agonist to determine induction of innate memory. Priming with β-glucan from Saccharomyces cerevisiae depressed IL-1β and TNF-α cytokine responses to re-stimulation with LPS, indicative of a tolerized state. However, bacillus Calmette-Guerin priming induced a trained state in porcine monocytes, as LPS re-stimulation enhanced IL-1β and TNF-α gene expression and protein production. We present the first evidence of innate memory in pig monocytes, with bacillus Calmette-Guerin (training) or Saccharomyces cerevisiae β-glucan (tolerance). Induction of a trained or tolerized state in vitro is a first step to identify agonists to alter the innate immune system at the animal level with the intent of enhancing disease resistance.  相似文献

摘要 目的:探讨腰背肌康复训练联合脊柱微调手法对腰椎间盘突出症(LDH)患者功能康复、血液流变学和生活质量的影响。方法:采用随机数字表法,将我院2020年3月~2022年3月期间收治的120例LDH患者分为对照组(60例,腰背肌康复训练)和观察组(60例,腰背肌康复训练联合脊柱微调手法)。观察两组患者临床疗效、功能康复效果、血液流变学和生活质量。结果:观察组的临床总有效率高于对照组(P<0.05)。观察组干预4周后日本骨科协会腰椎功能(JOA)评分高于对照组,Oswestry功能障碍指数(ODI)、视觉模拟评分法(VAS)评分低于对照组(P<0.05)。观察组干预4周后腰部屈曲腰部活动范围(ROM)、伸展ROM、测屈ROM和屈肌肌力、伸肌肌力高于对照组(P<0.05)。观察组干预4周后全血高切黏度、全血低切黏度、红细胞聚集指数低于对照组(P<0.05)。观察组干预4周后躯体功能、精神健康、活力、躯体疼痛、社会功能、总体健康、情感职能、生理机能维度评分高于对照组(P<0.05)。结论:腰背肌康复训练联合脊柱微调手法用于LDH患者干预中,可改善血液流变学,促进功能康复,有利于患者生活质量的提高。  相似文献

目的:观察生物反馈电刺激联合Kegel训练对产后盆底功能障碍性疾病(PFD)患者盆底功能电生理指标和生活质量的影响。方法:研究对象为2018年3月~2020年12月我院收治的80例产后PFD患者。采用双色球随机分组法将患者分为对照组(n=40)和研究组(n=40)。对照组给予Kegel训练,研究组给予生物反馈电刺激联合Kegel训练,两组均治疗8周。对比两组治疗8周后的疗效和尿失禁、盆底器官脱垂程度的改善情况。对比两组治疗前、治疗8周后的盆底功能电生理指标、日常生活质量和性生活质量。结果:治疗8周后,研究组的临床总有效率较对照组高(P<0.05)。治疗8周后,研究组I类肌纤维疲劳度、Ⅱ类肌纤维疲劳度、快肌最大肌电值及阴道动态压力均优于对照组(P<0.05)。研究组治疗8周后尿失禁、盆底器官脱垂程度的改善情况优于对照组(P<0.05)。治疗8周后,研究组盆底功能影响问卷简表(PIFQ-7)评分、盆腔器官脱垂-尿失禁性功能问卷(PISQ-12)评分均低于对照组(P<0.05)。结论:产后PFD患者采用生物反馈电刺激联合Kegel训练治疗疗效明确,可促进尿失禁、盆底器官脱垂程度情况及盆底功能改善,提高患者日常生活质量和性生活质量。  相似文献

摘要 目的:探讨视觉功能训练联合托吡卡胺滴眼液在治疗假性近视中的临床疗效。方法:选取中国人民解放军陆军特色医学中心眼科和重庆佑佑宝贝妇儿医院眼科2019年2月至2019年8月收治的400例假性近视患儿作为研究对象,将其随机分为对照组和观察组(n=200)。对照组给予滴眼液缓解眼疲劳治疗,观察组则采用视觉功能训练联合托吡卡胺滴眼液治疗。比较两组患儿治疗前及治疗2个月、6个月后裸眼视力、眼轴长度、屈光度、正相对调节量和视疲劳评分的变化。结果:治疗2个月和6个月后,观察组患儿裸眼视力分别为(0.75±0.05)和(0.81±0.04),较对照组显著上升(P<0.05);屈光度分别为(+0.05±0.50)和(+0.06±0.48),明显高于对照组(P<0.05);正相对调节量检测结果依次是(-1.98±0.07)和(-2.51±0.24),较对照组明显增加(P<0.05)。两组患儿视觉感知双眼视功能均有明显改善,其中观察组视觉功能恢复显著高于对照组(P<0.05)。视疲劳调查显示观察组患儿视疲劳评分分别为(6.90±0.89)分和(3.91±0.89)分,显著低于对照组(P<0.05)。结论:视觉功能训练联合托吡卡胺滴眼液治疗能有效提高患儿裸眼视力、改善屈光度、正相对调节量和视觉感知功能,从而减少视疲劳,预防近视。  相似文献

The study investigated the effect of high- and low-intensity exercise training on inflammatory reaction of blood and skeletal muscle in streptozotocin (STZ)-induced diabetic male Sprague-Dawley rats (243 ± 7 g, 8 weeks). The rats completed treadmill running in either high-intensity exercise (6 weeks of exercise training, acute bouts of exercise) or low-intensity exercise (6 weeks of exercise training). Non-running, sedentary rats served as controls. To induce diabetes mellitus, rats received a peritoneal injection of STZ (50 mg · kg−1). Rats were sacrificed immediately after an acute bout of exercise and 6 weeks of exercise training. Inflammatory factors were analyzed by ELISA and by immune blotting from the soleus and extensor digitorum longus muscles. In the serum, inflammatory cytokines (IL-1β, TNF-α, IL-6, IL-4) and reactive oxygen species (ROS) (nitric oxide and malondialdehyde) increased in diabetic rats. However, all exercise training groups displayed reduced inflammatory cytokines and reactive oxygen species. In skeletal muscles, low-intensity exercise training, but not high intensity exercise, reduced the levels of COX-2, iNOS, and MMP-2, which were otherwise markedly elevated in the presence of STZ. Moreover, the levels of GLUT-4 and MyoD were effectively increased by different exercise intensity and exercise duration. Low-intensity exercise training appeared most effective to reduce diabetes-related inflammation. However, high-intensity training also reduced inflammatory factors in tissue-specific muscles. The data implicate regular exercise in protecting against chronic inflammatory diseases, such as diabetes.  相似文献
