Determination of the pressure in the water-conducting vessels of intactNicotiana rustica L. plants showed that the pressure probe technique gave less-negative values than the Scholander-bomb method. Even though
absolute values of the order of −0.1 MPa could be directly recorded in the xylem by means of the pressure probe, pressures
between zero and atmospheric were also frequently found. The data obtained by the pressure probe for excised leaves showed
that the Scholander bomb apparently did not read the actual tension in the xylem vessles ofNicotiana plants. The possibility that the pressure probe gave false readings was excluded by several experimental controls. In addition,
cavitation and leaks either during the insertion of the microcapillary of the pressure probe, or else during the measurements
were easily recognized when they occurred because of the sudden increase of the absolute xylem tension to that of water vapour
or to atmospheric, respectively. Tension values of the same order could also be measured by means of the pressure probe in
the xylem vessels of pieces of stem cut from leaves and roots under water and clamped at both ends. The magnitude of the absolute
tension depended on the osmolarity of the bathing solution which was adjusted by addition of appropriate concentrations of
polyethylene glycol. Partial and uniform pressurisation of plant tissues or organs, or of entire plants (by means of the Scholander
bomb or of a hyperbaric chamber, respectively) and simultaneous recording of the xylem tension using the pressure probe showed
that a 1∶1 response in xylem pressure only occurred under a few circumstances. A 1∶1 response required that the xylem vessels
were in direct contact with an external water reservoir and/or that the tissue was (pre-)infiltrated with water. Corresponding
pressure-probe measurements in isolated vascular bundles ofPlantago major L. orP. lanceolata L. plants attached to a Hepp-type osmometer indicated that the magnitude of the tension in the xylem vessels was determined
by the external osmotic pressure of the reservoir. These and other experiments, as well as analysis of the data using classical
thermodynamics, indicated that the turgor and the internal osmotic pressure of the accessory cells along the xylem vessels
play an important role in the maintenance of a constant xylem tension. This conclusion is consistent with the cohesion theory.
In agreement with the literature (P.E. Weatherley, 1976, Philos. Trans. R. Soc. London Ser. B23, 435–444; 1982, Encyclopedia of plant physiology, vol. 12B, 79-109), it was found that the tension in the xylem of intact
plants under normal and elevated ambient pressure (as measured with the pressure probe) under quasi-stationary conditions
was independent of the transpiration rate over a large range, indicating that the conductance of the flow path must be flow-dependent. 相似文献

Cultures of Rhizosolenia formosa H. Peragallo were studied to assess whether or not physiological and optical characteristics of this large diatom were consistent with the ability to migrate vertically in the open ocean. Time-course experiments examined changes in chemical composition and buoyancy of R. formosa during nitrate (N)–replete growth, N starvation, and recovery. Cells could maintain unbalanced growth for at least 53 h after depletion of ambient nitrate. Increases in C:N and carbohydrate: protein ratios observed during N starvation reversed within 24 h of reintroduction of nitrate to culture medium. Buoyancy was related to nutrition: Upon N depletion, the percentage of positively buoyant cells decreased to 4% from 11% but reverted to 9% within 12 h of nitrate readdition. Cells took up nitrate in the dark. Nitrogen-specific uptake rates averaged 0.48 d?1; these rates were higher than N-specific growth rates (0. 15 d?1), indicating the potential for luxury consumption of nitrate, which can be stored for later use. Measurements of photosynthesis vs. irradiance, chlorophyll-specific absorption (aph*(λ)), and pigment composition showed that cells may be adapted for growth under a wide range of irradiances. Values of aph*(λ) were lower for N-depleted cells than for N-replete cells, and N-depleted cells had higher ratios of total carotenoids to chlorophyll a. Aggregation of chloroplasts was more pronounced in N-depleted cells. These are possibly photoprotective mechanisms that would be an advantage to N-depleted cells in surface waters. Compounds that absorb in the ultraviolet region were detected in N-replete cells but were absent in N-depleted cultures. Overall, these results have important implications for migrations of Rhizosolenia in nature. Cells may survive fairly long periods in N-depleted surface waters and will continue to take up carbon; then they can resume nitrate uptake and revert to positive buoyancy upon returning to deep, N-rich water. Uncoupled uptake of carbon and nitrogen during migrations of Rhizosolenia is a form of new production that may result in the net removal of carbon from oceanic surface waters. 相似文献

Five treatments (replication n=2) were applied to mesocosms in an oligotrophic lake (TP=6–10 µg 1-1) to assess the effects of fish on planktonic communities. The treatments were: (1) high fish (30 kg ha–1
Lepomis auritus, Linnaeus), (2) low fish (10 kg ha–1), (3) high removal of zooplankton, (4) low removal of zooplankton and (5) control. Total phosphorus, chlorophyll a, zooplankton biomass, and species richness decreased from high fish > low fish > control > low removal > high removal treatments. The fish treatments were dominated by crustacean zooplankton, while rotifers outnumbered the other zooplankters in the removal treatments. Calculations of zooplankton grazing rates suggested that clearance rates seldom exceeded 2% of the enclosure volume d–1 and were unlikely to have had much influence on phytoplankton biomass. Calculations from a phosphorus bioenergetics model revealed that when fish were present, their excretion rates were higher than the rates ascribed to zooplankton. Diet analysis showed that the fish derived most of their energy from the benthos and periphyton, and that fish excretion and egestion made significant contributions to the very oligotrophic pelagic phosphorus pool. In the absence of fish, zooplankton excretion was highest in the control treatments and lowest in the zooplankton removal treatments. Our results suggest that in oligotrophic systems, planktivorous fish can be significant sources of phosphorus and that fish and zooplankton induced nutrient cycling have significant impacts on planktonic community structure. 相似文献

This article describes the current status in the development of human monoclonal antibodies. Over the last ten years a lot of information about the human immune system has emerged. Combining these with the many new (bio-)technologies it is plausible that the long awaited breakthrough of this technology is close. This paper focuses on the classical cell-biological methods of achieving stable, antibody-producing human cell lines via cell fusion methods or virus derived transformations of human B-lymphocytes, as well as genetic engineering methods e.g. DNA libraries or phage display technology. The available in vitro immunization methods are critically reviewed and their impact on this topic is discussed. Increasingly electronic communication and a variety of electronic resources are accessible to a larger group of people within the scientific community. This paper outlines the range of resources that are available, and comments on their current and future value to the phycological community. Resources discussed include mailing lists and newsgroups. These are useful tools for rapid, informal, targeted communication, although the technology employed places limitations on the type and format of information which may be distributed. The World Wide Web (WWW) has the potential to overcome these limitations, the quality, complexity and value to the phycological community of the sites on the WWW are extremely variable, with some material being of dubious quality. However, it is possible to access high quality resources including culture collection catalogues, high quality images and microbial and molecular databases.
