Application of a suspension (log 8.0/ml) of Cryptococcus laurentii prior to harvest led to a reduction in Botrytis cinerea decay of strawberries stored at 4 or 20 °C, for 12 or 4 days, respectively. The frequency of spraying antagonist significantly influenced disease incidence in strawberry fruit. The best inhibition of disease was achieved when fruit sprayed C. laurentii with three applications that began 6 days prior to harvest, and the incidence of gray mold and natural decay treated with this method was 21% and 11%, compared with 88% and 62% in the control after storage at 20 °C for 4 days. A similar result occurred in the treated fruit after storage at 4 °C for 12 days. Dilation plate counts on Rose Bengal agar and scanning electron microscopy results showed that three applications with C. laurentii at 6, 3 and 0 days before harvest improved its ability to colonize the epidermis of strawberry fruit in the greenhouse and during storage compared to single application. In addition, antagonistic yeast spraying before harvest could reduce the weight loss, delay the decrease of the firmness and ascorbic acid, but had no significant effect on the contents of soluble solids, titratable acidity and fruit surface color. Activation of Ras into the GTP-binding, 'ON' state is a key switch in the neurotrophin-mediated neuronal survival and neurite outgrowth, in vitro as well as in vivo . In the current study we explored changes in GTP-Ras levels following facial nerve injury and the ensuing regeneration and the effects of perturbing these changes in vivo using synapsin-promoter mediated neuronal expression of constitutively active Val12H-Ras (synRas). Quantification of GTP-Ras and total Ras revealed a precipitous drop in the relative GTP-Ras levels in the axotomized facial motor nucleus, to 40% of normal levels at 2 days after cut, followed by a partial recovery to 50–65% at 4–28 days. On western blots, control and axotomized nuclei from synRas mutants showed a 2.2- and 2.5-fold elevation in GTP-Ras, respectively, compared with their wild type littermate controls ( p < 5%, anova , TUKEY post-hoc ), with the levels in the axotomized synRas nucleus slightly but not significantly above that in the uninjured littermate control ( p = 9.9%). Similar increase was also observed in the pERK but not pAKT targets of the Ras cascade. This moderate elevation of GTP-Ras strongly curtailed post-traumatic neuronal cell death (−65%), the influx of T-cells (−48%) as well as other parameters of neuroinflammatory response. Although synRas did not affect the speed of axonal regeneration or functional recovery it caused a very pronounced increase in central axonal sprouting. These current data emphasize the role of reduced active Ras, and by extension, the reduced overall level of retrograde neurotrophin signalling after axotomy, in mediating post-traumatic cell death and inflammation and in restricting the sprouting response. Moreover, the neuroprotective and central sprouting-enhancing effects of neuronal Val12H-Ras could help promote recovery in CNS injury. 相似文献

In onion ( Allium cepa L.) postponement of sprouting is necessary to achieve long term storage. We studied the factors determining sprouting during dry storage at 16°C. The period to visible sprouting depends on the length of the dormancy period, if present, and on the growth rate of the sprout. In the three cultivars tested, sprouts were initiated within 2 weeks after harvest indicating the absence of a real dormancy period. Sprout length increased linearly during storage. The mitotic activity of the apex decreased before harvest, was low at the transition from scale to leaf formation, and increased again when the sprout was initiated. From a few weeks before harvest, the initially high fructan content of the scales decreased, leading to a large increase in fructose. The sprout always contained enough carbohydrates for growth (between 50 and 60 mg g−1 dry weight, of which 30% was fructan). The activity of sucrose synthase (EC 2.4.1.13) increased as the sprout grew, indicating an increase in sink strength. Invertase (EC 3.2.1.26) was absent in all bulb organs, during the various developmental stages. Although carbohydrates and enzymes were available for fast sprouting, sprout growth was still linear instead of exponential during dry storage at temperatures favorable for growth (16°C). Preharvest sprouting (PHS) can be a problem in barley (Hordeum vulgare L.) especially malting barley, since rapid, uniform, and complete germination are critical. Information has been gained by
studying the genetics of dormancy (measured as germination percentage, GP). The objective of this study was to determine if
the quantitative trait loci (QTLs) discovered in previous research on dormancy are related to PHS. PHS was measured as sprout
score (SSc) based on visual sprouting in mist chamber-treated spikes and as alpha-amylase activity (AA) in kernels taken from
mist chamber-treated spikes that showed little or no visible sprouting. GP was also measured. All traits were measured at
0 and 14 days after physiological maturity. Evaluation of the spring six-row cross, Steptoe (dormant)/Morex (non-dormant)
doubled haploid mapping population grown in greenhouse and field environments revealed QTL regions for SSc, AA, and GP on
five, four, and six of the seven barley chromosomes, respectively. In total, seven and eight regions on five and six chromosomes
had effects ranging from 4 to 31% and 3 to 39% on PHS and dormancy, respectively. One chromosome 3H and three chromosome 5H
QTLs had the greatest effects. All PHS QTLs coincide with known dormancy QTLs, but some QTLs appear to be more important for

Following mitosis, specification and migration during embryogenesis, dopamine neurons of the mesencephalon undergo a postnatal naturally occurring cell death event that determines their final adult number, and a period of axonal growth that determines pattern and extent of target contacts. While a number of neurotrophic factors have been suggested to regulate these developmental events, little is known, especially in vivo , of the cell signaling pathways that mediate these effects. We have examined the possible role of Akt/Protein Kinase B by transduction of these neurons in vivo with adeno-associated viral vectors to express either a constitutively active or a dominant negative form of Akt/protein kinase B. We find that Akt regulates multiple features of the postnatal development of these neurons, including the magnitude of the apoptotic developmental cell death event, neuron size, and the extent of target innervation of the striatum. Given the diversity and magnitude of its effects, the regulation of the development of these neurons by Akt may have implications for the many psychiatric and neurologic diseases in which these neurons may play a role. 相似文献

Mortality, a critical parameter for population dynamics, is difficult to measure in long-lived trees or clonal herbaceous species because of the extremely low frequency of deaths. A model based on shoot recruitment would be helpful to estimate the population fate of species without a sufficient number of observed deaths. Existing matrix models are applicable to the dynamics of physiologically independent shoots, but not to physiologically dependent ones. We developed a shoot-dynamics model for plants with physiologically-dependent shoots, and used the model to estimate the effects of herbivory and conservation measures on the dynamics of a long-lived, shoot-sprouting shrub species, Paliurus ramosissimus (Rhamnaceae). Two populations of the endangered shrub have been severely damaged by herbivory by sika deer. The damage was mainly to new sprouting shoots. No deaths of individual plants were observed during an 8-year survey, and we could not estimate mortality. Thus, prediction of population dynamics based on births and deaths of individuals was impossible. Because P. ramosissimus is a shoot-sprouting species, we instead estimated the decline of individuals using a shoot-dynamics model. Using this model, we estimated the time to an 80 % decrease in shoot number per individual in the two populations at 37.8 and 37.2 years. These lengths suggest an immediate need for conservation measures to prevent herbivory even though no death of any individual was observed in the field survey. The estimated recovery times from the herbivory damage were 28.7 and 29.2 years if herbivory of new shoots is completely prevented by conservation measure. 相似文献
