The present report describes intracellular signaling events elicited by gibberellin (GA) in barley aleurone, where the phytohormone induces an increase on phosphoinositides turnover in a time-dependent manner. A biphasic behavior for PtdInsP was demonstrated by changes in [32P]PtdInsP levels, the earlier phase peaking at 5 min and the second phase at 25 min while PtdInsP
2 turnover was rapid but transient, peaking only at 5 min. In contrast, the structural phospholipid PtdCho turnover was only affected after 40 min. In addition, GA stimulated the synthesis of all phospholipids. Incorporation of [32P]Pi in unstimulated aleurone showed that both synthesis and degradation of polyphosphoinositides were relatively faster than those of structural phospholipids. We discuss the possible reasons why the changes in synthesis and turnover of phosphoinositide are required for GA signaling pathway and why it may be a critical event in the control of secretion in aleurone. 相似文献

The composition of phospholipids from electric organ and from membranes enriched in acetylcholine receptors (AChRs) is analyzed in three elasmobranch fish (Torpedo marmorata, Torpedo californica, and Discopyge tschudii). Irrespective of their purity, AChR-containing membranes are similar to electric organ in lipid and fatty acid composition. The following characteristics are common to the three species: (a) Choline, ethanolamine, and serine glycerophospholipids account for 80-90% of the phospholipids. (b) Their major fatty acid constituents are monoenes, saturates, and long-chain (n-3) polyenes (especially docosahexaenoate). (c) A large proportion of the ethanolamine glycerophospholipids (30-50%) is made up by plasmenylethanolamine, which contains fewer polyenes than phosphatidylethanolamine per mole of lipid. (d) Polyphosphoinositides represent 20-30% of the inositides of electric organ. (e) Phosphatidylinositol and phosphatidate have large proportions of 20- and 22-carbon polyenes. (f) Diphosphatidylglycerol and triacylglycerols are rich in oleate but also contain long-chain polyenes. (g) Sphingomyelin has monoenes and saturates ranging from 14 to 26 carbons. Species-related variations are observed (a) in the ratios between some phospholipid classes and subclasses and (b) in the relative abundance of the major polyunsaturated acyl chains of phospholipids. Despite these differences, the average unsaturation and length of fatty acids in major phospholipid classes are similar for the three species. 相似文献

Alterations in brain phospholipid metabolism were observed after chronic ethanol administration for 16 days to developing rats. Animals were injected intraperitoneally with 32Pi 16 h prior to killing. Overall uptake of 32Pi by brain did not differ between the control and ethanol-treated groups, which were killed 2 h and 24 h after the last ethanol feeding. Except for an increase in the labeling of myelin after ethanol treatment, the amount of radioactivity recovered in the synaptosomal-mitochondrial and plasma membrane fractions of control and ethanol-treated groups was not different. Relative to the radioactivity of phosphatidylcholines, which indicated no change, there were increases (20-44%) in labeling of ethanolamine plasmalogens, phosphatidic acids, and phosphatidylinositols in cortical synaptosomes from the 2-h ethanol-treated group. In the plasma membrane fractions, however, increases (9-14%) in labeling of phosphatidylserines and phosphatidylinositols were observed in both 2- and 24-h ethanol-treated groups. In both membrane fractions, there was an obvious increase (44-86%) in labeling of polyphosphoinositides at 24 h after withdrawal from ethanol. Results thus indicate an adaptive increase in the biosynthesis of ethanolamine plasmalogen and brain acidic phospholipids due to chronic ethanol administration. Furthermore, the increase in labeling of polyphosphoinositides in the 24-h withdrawal group may reflect the hypoactivity associated with ethanol withdrawal. 相似文献
