Formation of noncovalent complexes between tyrosinase from mushrooms and a cationic polyelectrolyte, polybrene (PB, poly (1,5-dimethyl-1,5-diazaundecamethyelene) bromide), was shown to activate and stabilize tyrosinase in water-ethanol mixtures. In the reaction of catechol oxidation in aqueous solutions, catalytic activity (k(cat)) of tyrosinase-PB complex ([PB]/[tyrosinase] molar ratio 100:1, per mole of polymer) in a wide range of pH was higher than that of free tyrosinase. In aqueous solutions and in water-ethanol mixtures at moderate concentrations of ethanol (10-40% v/v), the value of k(cat) of tyrosinase-PB complex exceeded the activity of free enzyme, from 1.2-2-fold, accompanied by the essential (up to 10-fold) increase in the value of the specificity constant (k(cat)/K(m)). The results are of practical importance for the construction of biocatalysts working successfully in polar organic media.  相似文献

After more than a decade of method development, cross-linking in combination with mass spectrometry and bioinformatics is finally coming of age. This technology now provides improved opportunities for modelling by mapping structural details of functional complexes in solution. The structure of proteins or protein complexes is ascertained by identifying amino acid pairs that are positioned in close proximity to each other. The validity of this technique has recently been benchmarked for large multi-protein complexes, by comparing cross-link data with that from a crystal structure of RNA polymerase II. Here, the specific nature of this cross-linking data will be discussed to assess the technical challenges and opportunities for model building. We believe that once remaining technological challenges of cross-linking/mass spectrometry have been addressed and cross-linking/mass spectrometry data has been incorporated into modelling algorithms it will quickly become an indispensable companion of protein and protein complex modelling and a corner-stone of integrated structural biology.  相似文献

Perfluorophenanthrene and decamethylferrocene cocrystallize as a molecular adduct in monoclinic space group P21/c with a = 8.842(2), b = 11.262(1), c = 30.695(8) Å, β = 95.89(2)°, V = 3040.3(8) Å3, Z = 4. The structure was refined to R = 0.0537 for 1567 observed reflections. The perfluoroarene is twisted and chiral; the crystal is a racemate, however.  相似文献

We report here the synthesis, characterization and in vitro antiamoebic activity of 5-nitrothiophene-2-carboxaldehyde thiosemicarbazones (TSC), 1–5, and their bidentate complexes [Ru(η4-C8H12)(TSC)Cl2] 1a–5a. The biological studies of these compounds were investigated against HK-9 strain of Entamoeba histolytica and the concentration causing 50% cell growth inhibition (IC50) was calculated in the micromolar range. The ligands exhibited antiamoebic activity in the range (2.05–5.29 μM). Screening results indicated that the potencies of the compounds increased by the incorporation of ruthenium(II) in the thiosemicarbazones. The complexes 1a–5a showed antiamoebic activity with an IC50 of 0.61–1.43 μM and were better inhibitors of growth of E. histolytica, based on IC50 values. The most promising among them is Ru(II) complex 2a having 1,2,3,4-tetrahydroquinoline as N4 substitution.  相似文献

Reactions of RhCl(cod)(THP) (cod = 1,5-cyclooctadiene; THP = P(CH2OH)3) with PMePh2 or PCyPh2 (Cy = cyclohexyl) in acetone/MeOH solution under H2 surprisingly form the complexes cismer-Rh(H)2Cl(PRPh2)3 (R = Me or Cy); both complexes are characterized by crystallography (the first structures in which the hydride ligands of such dihydrido-chloro-trisphosphine complexes have been located), and by detailed 1H and 31P NMR spectroscopy. The key role of the THP in the observed chemistry is discussed.  相似文献

Near-infrared absorption spectra of A2CoX4 (A = Cs, ethyl4N; X = Cl, Br) single crystals and from KBr pellets at low temperature are recorded by the Fourier transform technique. At 2 K a rich fine structure of v1(4A24T2) and v2(4A24T1) ligand field spectra is detected which can be assigned to low symmetry level splittings due to actual site symmetries superimposed by vibrational fine structure. Comparison of fundamental frequencies obtained from far-infrared spectra allows an assignment of all peaks measured in the vibronic spectra to vibrational modes of the MX4 complex. Zero-phonon bands are identified using the assistance of angular overlap calculations.  相似文献

We reported comprehensive screening for antigens (Ags) overexpressed on various carcinomas via isolation of human monoclonal antibodies (mAbs) that may be therapeutic in a previous paper (Proc. Natl. Acad. Sci. USA 105, 7287-7292, 2008). Twenty-one distinct Ags highly expressed on several carcinomas were identified and 356 mAbs with unique sequences turned out to bind to one of the 21 Ags. Among them CADM1/IGSF4 which had been originally referred to as tumor suppressor lung cancer 1 (TSLC1) was included. Therefore we examined the expression of CADM1 in lung cancers in this study. Eight different anti CADM1 mAbs were used for immunohistochemical analysis of 29 fresh lung cancer specimens. Staining patterns were categorized to six groups based on the extent of positive staining and the localization of stained portions. While overexpression of CADM1 was observed on the cell surface of adenocarcinomas at a high frequency, around 60%, positive stainings were rarely observed on that of other lung carcinomas including squamous cell carcinomas. Moreover, some clones among the eight mAbs gave different staining patterns from those by the other clones against the same fresh specimen, suggesting presence of variant forms of CADM1 differentiated by mAbs.  相似文献

Reactions of Sr(II) and Ba(II) chlorides with 2-hydroxybenzophenone and salicylaldehyde, hydroxyaromatic ketones or β-diketones in 1:1:1 molar ratios have resulted in the formation of mixed ligand complexes of the type [MLL′(H2O)2] (M = Sr(II) or Ba(II); HL = 2-hydroxybenzophenone and HL′ = salicylaldehyde, 2-hydroxyacetophenone, 2-hydroxypropiophenone, pentane-2,4-dione, 1-phenylbutane-1,3-dione or 1,3-diphenylpropane-1,3-dione). ZnCl2 reacts with 1,2,4-1H-triazole to afford Zn(trz)Cl. A spontaneous deprotonation of Htrz occurs. The crystal structure of Zn(trz)Cl has been solved. The compound crystallizes in the space group P21/n. The lattice parameters are a = 8.863(4), B = 9.762(4), C = 6.146(3) Å, β = 99.56(10)°, with Z = 4. The 1,2,4-triazolato bridges three zinc atoms through its three nitrogen atoms, affording a layered structure. The zinc atom is in an N3Cl tetrahedral coordination. The layers are not planar, but rather corrugated. The chlorine atoms point to either side of the layers, and play the role of spacers. The shortest interlayer ZnZn separation is 5.701 Å.  相似文献

The complex C60Pt[P(OPh)3]2 displays C60 ππ* intraligand bands in the UV-Vis region and a long-wavelength absorption at λmax = 770 nm which is assigned to a metal-to-ligand charge transfer (MLCT) transition from platinum to fullerene. The irradiation of the complex leads to the population of the reactive MLCT state and subsequently to the dissociation (C60Pt[P(OPh)3]2 → C60 + Pt[P(OPh)3]2) in the primary photochemical step. Product formation takes place by the interception of Pt[P(OPh)3]2 with suitable scavengers such as CHCl3 or O2.  相似文献
