Checkpoint recovery upon completion of DNA repair allows the cell to return to normal cell cycle progression and is thus a crucial process that determines cell fate after DNA damage. We previously studied this process in Xenopus egg extracts and established Greatwall (Gwl) as an important regulator. Here we show that preactivated Gwl kinase can promote checkpoint recovery independently of cyclin-dependent kinase 1 (Cdk1) or Plx1 (Xenopus polo-like kinase 1), whereas depletion of Gwl from extracts exhibits no synergy with that of Plx1 in delaying checkpoint recovery, suggesting a distinct but related relationship between Gwl and Plx1. In further revealing their functional relationship, we found mutual dependence for activation of Gwl and Plx1 during checkpoint recovery, as well as their direct association. We characterized the protein association in detail and recapitulated it in vitro with purified proteins, which suggests direct interaction. Interestingly, Gwl interaction with Plx1 and its phosphorylation by Plx1 both increase at the stage of checkpoint recovery. More importantly, Plx1-mediated phosphorylation renders Gwl more efficient in promoting checkpoint recovery, suggesting a functional involvement of such regulation in the recovery process. Finally, we report an indirect regulatory mechanism involving Aurora A that may account for Gwl-dependent regulation of Plx1 during checkpoint recovery. Our results thus reveal novel mechanisms underlying the involvement of Gwl in checkpoint recovery, in particular, its functional relationship with Plx1, a well characterized regulator of checkpoint recovery. Coordinated interplays between Plx1 and Gwl are required for reactivation of these kinases from the G(2)/M DNA damage checkpoint and efficient checkpoint recovery.  相似文献

Polo-like kinase 1 (Plk1) plays key roles in many aspects of mitosis. We have previously shown that induction of p21Waf1 by p53 is responsible for protection of cells against adriamycin-induced polyploidy formation and mitotic catastrophe. Here we show that adriamycin treatment suppressed Plk1 expression in a p53- and p21Waf1-dependent manner. Ablation of p21Waf1 inhibited the adriamycin-induced p53 activation, and this inhibition was alleviated by knockdown of Plk1, suggesting that p21Waf1-dependent suppression of Plk1 expression is responsible for maintaining p53 activation during stress response. Plk1 associated with p53 and disrupted its interaction with target gene promoters in cells treated with adriamycin. Overexpression of Plk1 inhibited the p53-mediated prevention of caspase-independent mitotic death, but not polyploidy formation, in adriamycin-treated cells. Together our results indicate that suppression of Plk1 by p21Waf1 is responsible for p53-dependent protection against adriamycin-induced caspase-independent mitotic death.  相似文献

Protein kinase and phosphatase signaling cascade, coupled with other post-translational modifications, orchestrates temporal order of various events during cell division. Among the many mitotic kinases, Polo-like kinase 1 (PLK1) as a key regulator, participates in regulating mitosis from mitotic entry to cytokinesis. The advancement in optical reporter engineering and the recent development of specific chemical probes enable us to visualize spatiotemporal gradient of kinase activity at nano-scale. One of such tools is FRET-based optic sensor that allows us to delineate the PLK1 activity in space and time. In this review, we address the inter-relationships between PLK1 and other protein kinases/phosphatases, as well as the crosstalk between PLK1 phosphorylation and ubiquitination during cell division. In particular, we discuss the molecular mechanisms and steps underlying PLK1 kinase priming, activation and turn-off during cell division.  相似文献

Post-translational modifications of α-synuclein occur in the brain of patients affected by Parkinson''s disease and other α-synucleinopathies, as indicated by the accumulation of Lewy inclusions containing phosphorylated (at serine 129) and nitrated α-synuclein. Here we found that phospho-Ser 129 and nitrated α-synuclein are also formed within dopaminergic neurons of the monkey substantia nigra as a result of normal aging. Dopaminergic cell bodies immunoreactive for phospho-Ser 129 and nitrated α-synuclein were rarely seen in adult mature animals but became significantly more frequent in the substantia nigra of old primates. Dual labeling with antibodies against phospho-Ser 129 and nitrated α-synuclein revealed only limited colocalization and mostly stained distinct sub-populations of dopaminergic neurons. Age-related elevations of modified protein paralleled an increase in the number of neurons immunoreactive for unmodified α-synuclein, supporting a relationship between higher levels of normal protein and enhanced phosphorylation/nitration. Other mechanisms were also identified that likely contribute to α-synuclein modifications. In particular, increased expression of Polo-like kinase 2 within neurons of older animals could contribute to phospho-Ser 129 α-synuclein production. Data also indicate that a pro-oxidant environment characterizes older neurons and favors α-synuclein nitration. Aging is an unequivocal risk factor for human α-synucleinopathies. These findings are consistent with a mechanistic link between aging, α-synuclein abnormalities and enhanced vulnerability to neurodegenerative processes.  相似文献

Polo-like kinase 1 (PLK1) plays a critical role during multiple stages of cell cycle progression and is involved in the development and metastasis of malignant tumours. The protein contains a regulatory polo-box domain (PBD) that can recognise and bind to a wide variety of phosphorylated substrates. Here, a systematic amino acid preference profile of phosphopeptide interaction with PLK1 PBD domain was created based on the crystal structures of the domain in complex with its natural phosphopeptide ligands. With the profile we were able to explore the structural basis and energetic landscape of phosphopeptide binding to the domain. Moreover, in addition to domain peptide-binding pocket we also examined the intermolecular interaction between the N-terminal region of phosphopeptide and a newly discovered crystal packing site of the domain. All the harvested information was successfully integrated to guide the structural design and optimisation of phosphopeptide ligands with improved affinity and selectivity for the domain, which were then confirmed in vitro by fluorescence anisotropy assays. Structural analysis and energetic analysis revealed that the phosphopeptide ligand can be divided into three functionally independent sections: a N-terminal flexible tail, a C-terminal unbinding tail and a central binding region. The N-terminal tail possesses a high flexibility that can roll over on the surface of PBD crystal packing site, while the C-terminal tail points out of the peptide-binding pocket of the domain and no substantial interaction can be observed between them. In contrast, the central region binds tightly to the domain pocket and exhibits modest conformational change over different ligands.  相似文献

Aurora kinases play an essential role in mitosis and cell cycle regulation. In recent years Aurora kinases have proved popular cancer targets and many inhibitors have been developed. The majority of these clinical candidates are multi-targeted, rendering them inappropriate as tools for studying Aurora kinase mediated signaling. Here we report discovery of a highly selective inhibitor of Aurora kinases A, B and C, with potent cellular activity and minimal off-target activity (PLK4). The X-ray co-crystal structure of Aurora A in complex with compound 2 is reported, and provides insights into the structural determinants of ligand binding and selectivity.  相似文献

The Polo-like kinase (PLK) in Trypanosoma brucei plays multiple roles in basal body segregation, flagellum attachment, and cytokinesis. However, the mechanistic role of TbPLK remains elusive, mainly because most of its substrates are not known. Here, we report a new substrate of TbPLK, SPBB1, and its essential roles in T. brucei. SPBB1 was identified through yeast two-hybrid screening with the kinase-dead TbPLK as the bait. It interacts with TbPLK in vitro and in vivo, and is phosphorylated by TbPLK in vitro. SPBB1 localizes to both the mature basal body and the probasal body throughout the cell cycle, and co-localizes with TbPLK at the basal body during early cell cycle stages. RNAi against SPBB1 in procyclic trypanosomes inhibited basal body segregation, disrupted the new flagellum attachment zone filament, detached the new flagellum, and caused defective cytokinesis. Moreover, RNAi of SPBB1 confined TbPLK at the basal body and the bilobe structure, resulting in constitutive phosphorylation of TbCentrin2 at the bilobe. Altogether, these results identified a basal body protein as a TbPLK substrate and its essential role in promoting basal body segregation and flagellum attachment zone filament assembly for flagellum adhesion and cytokinesis initiation.  相似文献

以高表达Polo样激酶1(Polo-like kinase 1,PLK1)的宫颈癌细胞系HeLa细胞为模型,观察针对PLK1基因的短发夹状RNA(short hairpin RNA,shRNA)对其凋亡和增殖的影响.设计并合成了针对PLK1的shRNA,将其导入构建的携带增强型绿色荧光蛋白(EGFP)的RNAi(RNA interference)表达载体中.通过 RT-PCR和Western 印迹分别检测HeLa细胞PLK1基因和蛋白水平的表达,以流式细胞仪和PI-Hochest双染法测细胞凋亡,MTT法检测细胞的增殖水平.成功构建了携带EGFP的RNAi表达载体pEGFP-H1.转染shRNA后,HeLa细胞PLK1的表达降低至30%.与对照组和空载体转染组相比,shRNA转染组的HeLa细胞凋亡率明显增加,其增殖活性则明显降低.本课题构建的RNAi表达载体便于观察靶基因的转染情况,且不影响H1启动子的体内转录.PLK1的基因沉默能明显增加HeLa细胞的凋亡,抑制该细胞的增殖,有可能为未来肿瘤的治疗找到新的靶点和有效途径.  相似文献

A series of new peptidomimetics targeting the polo-box domain (PBD) of polo-like kinase 1 (Plk1) was identified based on the potent and selective pentapeptide Plk1 PBD inhibitor PLHSpT. Unnatural amino acid residues were introduced to the newly designed compound and the N-terminal substituent of the peptidomimetic was investigated. The optimized compound 9 inhibited the Plk1 PBD with IC50 of 0.267 μM and showed almost no inhibition to Plk2 PBD or Plk3 PBD at 100 μM. Biolayer interferometry studies demonstrated that compound 9 showed potent binding affinity to Plk1 with a Kd value of 0.164 μM, while no Kd were detected against Plk2 and Plk3. Compound 9 showed improved stability in rat plasma compared to PLHSpT. Binding mode analysis was performed and in agreement with the observed experimental results. There are only two natural amino acids remained in the chemical structure of 9. This study may provide new information for further research on Plk1 PBD inhibitors.  相似文献
