Pollen and phytolith evidence for rice cultivation during the Neolithic at Longqiuzhuang, eastern Jianghuai, China 总被引:8,自引:0,他引:8
Phytolith and pollen analyses were carried out at the archaeological site at Longqiuzhuang in Gaoyou, Jiangsu, southern China. The results indicate that the key morphological phytolith types associated with cultivated rice (Oryza) are common in the Neolithic cultural layers at this site. The evidence strongly suggests that cultivated rice (mainlyO. japonica) was grown locally during the Neolithic. The archaeopalynological record provides information about the impact of human activity and, in particular, farming on the natural vegetation. The evergreen and deciduous broad-leaved forest was substantially altered, and herbaceous taxa, including ruderals, expanded. Based on the results from the phytolith and pollen analyses, two distinct phases of human activity have been recognized, namely (1) phase A (7000-6300 B.P., i.e. early Neolithic) a warm and humid period when arable farming, including rice cultivation, was pursued but the variation in the size of the carbonized rice grains was low, and (2) phase B (6300-5500 B.P., late Neolithic age), a period of relatively cold and/or arid climate when cultivated rice was of major importance and was morphologically similar to present-day rice. Environment, and in particular climate change in the late Neolithic, were important factors affecting the development of rice as a cultivated crop. Synopsis Spawning season, size at first reproduction, oocyte maturation, and fecundity ofAwaous guamensis, an amphidromous Hawaiian goby, were studied from June 1989 through May 1991 in the Wainiha River, Kau'ai, Hawai'i. Female fish larger than 73 mm standard length (SL) had mature gonads from August through December in 1989 and 1990. Gonadosomatic index (GSI) values for mature females ranged from 0.2 to 14.5 during the spawning season. Male fish larger than 64 mm SL had elevated GSI values from June 1989 through December 1989 and from August 1990 through December 1990. Mature sperm were found in two male fish collected in January and February. GSI values for mature males ranged from less than 0.01 to 4.0 in the spawning season. Size-frequency distributions of measurements of vitellogenic oocyte diameters and microscopic observations of oocytes indicated this species has group-synchronous oocyte development. Ovarian maturation stages examined over a 29-month period suggest that members of the stock spawned at different times within the spawning season, although mass spawning events have been documented for this species. Estimates of clutch sizes from nests measured in situ were comparable to estimates of potential fecundity from in vitro examination of ovaries, and indicated that female fish deposited an entire clutch during a spawning event. No evidence for multiple spawning by an individual fish in a single season was found. However, microscopic observations of brown bodies in some ovaries suggested that individual fish probably spawn more than once in a lifetime. 相似文献

Atlantic salmon populations are reported to be declining throughout its range, raising major management concerns. Variation in adult fish abundance may be due to variation in survival, growth, and timing of life history decisions. Given the complex life history, utilizing highly divergent habitats, the reasons for declines may be multiple and difficult to disentangle. Using recreational angling data of two sea age groups, one‐sea‐winter (1SW) and two‐sea‐winter (2SW) fish originated from the same smolt year class, we show that sea age at maturity of the returns has increased in 59 Norwegian rivers over the cohorts 1991–2005. By means of linear mixed‐effects models we found that the proportion of 1SW fish spawning in Norway has decreased concomitant with the increasing sea surface temperature experienced by the fish in autumn during their first year at sea. Furthermore, the decrease in the proportion of 1SW fish was influenced by freshwater conditions as measured by water discharge during summer months 1 year ahead of seaward migration. These results suggest that part of the variability in age at maturity can be explained by the large‐scale changes occurring in the north‐eastern Atlantic pelagic food web affecting postsmolt growth, and by differences in river conditions influencing presmolt growth rate and later upstream migration. 相似文献

A new method for assessing the effects of test compounds on Nicotiana alata pollen tubes in culture is described. Pollen tubes grow from a cluster of grains placed beneath a thin layer of gelled medium
in which test substances are incorporated and from which evaporation is prevented by a covering layer of oil. Pollen tubes
can grow to 8 mm in length in 24 h, which corresponds to about 25% of the maximum growth rate in styles. Growth is non-destructively
measured. The developmental stages reached by cultured tubes are similar to those of tubes growing in styles; growth changes
from being reserve-dependent to reserve-independent, callose plugs form, and the nucleus of the generative cell divides. Because
culture volumes are small (10–20 μl per replicate), the effects of known concentrations of microgram quantities of compounds
on the growth of pollen tubes can be tested.

Alkaline phosphatase (ALP), a homo‐dimeric enzyme has been widely used in various bioassays as disease markers and enzyme probes. Recent advancements of digital bioassay revolutionized ALP‐based diagnostic assays as seen in rapid growth of digital ELISA and the emerging multiplex profiling of single‐molecule ALP isomers. However, the intrinsic heterogeneity found among ALP molecules hampers the ALP‐based quantitative digital bioassays. This study aims quantitative analysis of single‐molecule activities of ALP from Escherichia coli and reveals the static heterogeneity in catalytic activity of ALP with two distinct populations: half‐active and fully‐active portions. Digital assays with serial buffer exchange uncovered single‐molecule Michaelis–Menten kinetics of ALP; half‐active molecules have halved values of the catalytic turnover rate, k cat, and the rate constant of productive binding, k on, of the fully active molecules. These findings suggest that half‐active ALP molecules are heterogenic dimers composed of inactive and active monomer units, while fully active ALP molecules comprise two active units. Static heterogeneity was also observed for ALP with other origins: calf intestine or shrimp, showing how the findings can be generalized across species. Cell‐free expression of ALP with disulfide bond enhancer and spiked zinc ion resulted in homogenous population of ALP of full activity, implying that inactive monomer units of ALP are deficient in correct disulfide bond formation and zinc ion coordination. These findings provide basis for further study on molecular mechanism and biogenesis of ALP, and also offer the way to prepare homogenous and active populations of ALP for highly quantitative and sensitive bioassays with ALP. 相似文献

Summary Eluates of stigmas of Brassica oleracea that were known to contain S locus-specific glycoproteins (SLSG) discriminated between self and cross pollen in vitro in three different media. Discrimination was equally evident in experiments that were the in vitro equivalents of reciprocal pollinations. In a TAPS-buffered medium, self eluates depressed pollen germination in a dose-dependent manner. TAPS medium allowed a bioassay of the effects of SLSG in eluates because it optimized germination in a way that eliminated the complicating features of the stimulatory substances in the eluates. Stigma eluates affected percentage pollen germination and optimum calcium concentrations in vitro whether or not SLSG were present in the eluates, but differently in different media, and depending on whether the eluates were cross or self with respect to the pollen tested. Thus, the effect of stigma eluates on the in vitro germination of pollen in Brassica depends on the balance of stimulatory versus inhibitory substances in the eluates. 相似文献

Because brain maturation in chickens is protracted and occurs well after the major developmental period of synaptogenesis, chicken forebrain is suitable to investigate whether the molecular mechanisms underlying memory consolidation are different in immature and mature animals. We have used antibodies and western blotting to analyze subcellular fractions from the intermediate medial mesopallium region of 14-day and 8-week chicken forebrain prepared 0, 45, and 120 min after learning a discriminative taste avoidance task. At both ages learning induced changes in the phosphorylation of the glutamate receptor subunit 1 at Ser831, the levels of calcium-calmodulin stimulated/dependent protein kinase II and the phosphorylation of calcium-calmodulin stimulated/dependent protein kinase II at Thr286 were observed only in the fraction enriched in post-synaptic densities. The changes were of the same type at the two ages but occurred faster in mature animals. The changes in extracellular signal regulated kinase and phosphorylated-extracellular signal regulated kinase were more complex with different subcellular fractions showing different patterns of change at the two ages. These results imply that the molecular changes induced by learning a behavioral task are faster in mature than immature brain and may involve a different balance of intracellular signaling pathways. 相似文献
