Environmental and developmental controls over the seasonal pattern of isoprene emission from aspen leaves 总被引:16,自引:0,他引:16

Isoprene emission from plants represents one of the principal biospheric controls over the oxidative capacity of the continental troposphere. In the study reported here, the seasonal pattern of isoprene emission, and its underlying determinants, were studied for aspen trees growing in the Rocky Mountains of Colorado. The springtime onset of isoprene emission was delayed for up to 4 weeks following leaf emergence, despite the presence of positive net photosynthesis rates. Maximum isoprene emission rates were reached approximately 6 weeks following leaf emergence. During this initial developmental phase, isoprene emission rates were negatively correlated with leaf nitrogen concentrations. During the autumnal decline in isoprene emission, rates were positively correlated with leaf nitrogen concentration. Given past studies that demonstrate a correlation between leaf nitrogen concentration and isoprene emission rate, we conclude that factors other than the amount of leaf nitrogen determine the early-season initiation of isoprene emission. The late-season decline in isoprene emission rate is interpreted as due to the autumnal breakdown of metabolic machinery and loss of leaf nitrogen. In potted aspen trees, leaves that emerged in February and developed under cool, springtime temperatures did not emit isoprene until 23 days after leaf emergence. Leaves that emrged in July and developed in hot, midsummer temperatures emitted isoprene within 6 days. Leaves that had emerged during the cool spring, and had grown for several weeks without emitting isoprene, could be induced to emit isoprene within 2 h of exposure to 32°C. Continued exposure to warm temperatures resulted in a progressive increase in the isoprene emission rate. Thus, temperature appears to be an important determinant of the early season induction of isoprene emission. The seasonal pattern of isoprene emission was examined in trees growing along an elevational gradient in the Colorado Front Range (1829–2896 m). Trees at different elevations exhibited staggered patterns of bud-break and initiation of photosynthesis and isoprene emission in concert with the staggered onset of warm, springtime temperatures. The springtime induction of isoprene emission could be predicted at each of the three sites as the time after bud break required for cumulative temperatures above 0°C to reach approximately 400 degree days. Seasonal temperature acclimation of isoprene emission rate and photosynthesis rate was not observed. The temperature dependence of isoprene emission rate between 20 and 35°C could be accurately predicted during spring and summer using a single algorithm that describes the Arrhenius relationship of enzyme activity. From these results, it is concluded that the early season pattern of isoprene emission is controlled by prevailing temperature and its interaction with developmental processes. The late-season pattern is determined by controls over leaf nitrogen concentration, especially the depletion of leaf nitrogen during senescence. Following early-season induction, isoprene emission rates correlate with photosynthesis rates. During the season there is little acclimation to temperature, so that seasonal modeling simplifies to a single temperature-response algorithm. 相似文献

We evaluated the efficacy of murine monoclonal antibodies (MAbs) targeted to the Aβ amyloid of Alzheimer's disease for development
of procedures for the in vivo identification of amyloid angiopathy (AA). MAbs to Aβ were prepared and screened for effectiveness
in visualizing AA and neuritic plaques in postmortem AD brain sections. They were assessed again after enzymatic cleavage
to produce Fab fragments and after labeling with technetium-99m (99mTc) using a diamide dimercaptide ligand system. Modified and radiolabeled Fab fragments retained activity and specificity
toward amyloid-laden blood vessels and neuritic plaques. A highly specific murine MAb, 10H3, was identified and characterized
that fulfills criteria necessary for the development of an in vivo diagnostic imaging agent. Toxicity studies in rats showed
the MAb to be safe. Biodistribution studies in mice demonstrated desirable properties for use as an imaging agent. Expansion
and adaptation of these strategies may provide the methods and materials for the noninvasive analysis of AA in living patients,

A family of highly dispersed repetitive elements, designated PREM-1, which are transcribed primarily during pollen development, has been identified in maize. Sequence data from six PREM-1-containing genomic clones suggest that the PREM-1 sequences are the LTRs of a family of putative retroelements. PREM-1 LTRs are estimated to be present in about 10 000 to 40 000 copies in the maize genome. Although related sequences have been detected in sorghum and crab grass, highly homologous sequences appear to be specific to the genus Zea (maize and teosinte). A diverse group of RNAs that contain portions of the PREM-1 sequence at their 3 ends are transcribed in pollen; highest levels appear in early uninucleate microspores. The PREM-1-containing cDNAs do not appear to code for protein products since stop codons are present in all three reading frames. The possible significance of expression of retroelements in the male gametophyte, in terms of transposition of DNA, is discussed. 相似文献

The present study illustrates clearly how pollen analysis may be applied to stratigraphy from a botanical point of view. Within a chronologically reliable frame (Middle Pliocene for the marine deposits on the basis of Foraminifera record; two subzones for continental deposits on the basis of Mammals record: Hautimagne for Terrats fauna, Sète for Serrat-d'en-Vacquer fauna), palynology provides a good stratigraphical boundary: the extinction of the Taxodiaceae. In a remblayage area, it is established that a continental level is not necessarily younger than a marine one unless they are superposed. Many profiles are replaced according to the «progradation of the pliocene gulf of Roussillon. The boundary between marine and continental deposits cuts through the chronological line of the Taxodiaceae extinction. The flora investigated (93 taxa) is the first known for the Pliocene of this area. The extinction of the Taxodiaceae in southern France has a climatic cause: the setting in of a mediterranean rhythm (dry summers). This extinction is much older than the one which took place in Italy (Tiberian boundary) and in the Netherlands (Reuverian-Pretiglian boundary). 相似文献

By measuring the derivative absorption spectra of chloroplasts of different populations of Pinus armandi Franch., it was found that southern populations maintained higher absorption at 680 nm than at 670 nm, but some of the northern populations deviated the maximum absorption from 680 nm to 670 nm, which indicated that the activity of PS Ⅱ in some of the northern populations declined. Clear geographical differences also have been found in the positions of emission peaks of PS Ⅰ and PS Ⅱ in the fluorescence emission spectra at 77 K. Analysis of the fluorescence excitation spectra at 77 K revealed geographical changes in the absorption.status of Chl a. The β-glucuronidase (GUS) gene has been widely used as a reporter gene in the study of plant molecular biology and genetic engineering. One of the major reasons leading to the popularity of GUS-fusion system was the belief that there was no detectable intrinsic GUS activity in plant tissues. However, investigators have been troubled by the "false positive" results or "background" activities when GUS assays were performed. In the present experiment, histochemical observations of intrinsic GUS activity in various tissues and during pollen development of tobacco (Nicotiana tabacurn L. ) was carried out using 5-bromo-4- chloro-3-indolyl-β-D-glucuronic acid (X-gluc) as a substrate for overnight incubation of the treated tissues at 37℃. No detectable intrinsic GUS activity was found in seedling root, stem, leaf, anther wall and stigma of different stages, ovule, as well as isolated generative cell and embryo sac. During pollen development, two peaks of intrinsic GUS activity appeared, one, close to the microspore mitosis and the other from the full maturation of pollen lasting to the post-germination pollen tube stage, no or weak activity was found at other pollen developmental stages. GUS was located in the cytoplasm of the pollen. The pH value of staining solution strongly influenced the experimental results. Blue color was visualized at pH 5, even when 20% methanol or 0.2 mmol/L glucaric acid-l-4-1actone (GAL, a specific GUS inhibitor) were added. At pH 7, no detectable reaction was found at all. The aforementioned results indicate that when using tobacco pollen as the target of GUS gene transformation, the assay should be strictly controlled to neutral condition for avoiding false positive resuits. 相似文献

Pollen grains of 6 genera, including 36 species, of the Betulaceae, were examined
under LM, SEM and TEM.Their morphological characters are described and three types of
pollen are distinguished here. The evidence from pollen morphological characters does not
support Hutchinson's (1973)and Dahlgren's (1983)treatments which divided the Betulaceae
into two families: Corylaceae ( including Corylus, Ostryopsis, Carpinus and Ostrya) and
Betulaceae (including Alnus and Betula). The author agrees with Abbe's (1974)division of
the Betulaceae into three tribes with one modification that Ostryopsis is transferred from the
tribe Coryleae to the tribe Carpineae. The limits of the three tribes presented here are the same
as Kuprianova's (1963) treatment in which she divided the Betulaceae into three families. It is
more reasonable to treat Kuprianova's families as tribes. Pollen morphological characters of
the three tribes of the Betulaceae are described as follows:
1. Tribe Betuleae (including Alnus and Betula): Pollen grains are triporate or
stephanoporate and oblate, suboblate or spheroidal. Sometimes the exine has arci and the
ektexine thickens in the apertural region; the sculpture of exine is slightly rugulate under
scanning electron microscope (SEM) ; the exine structure is four-layered: tectum,
columellae, footlayer and endexine. The columellae are columellate, the endexine thickens in
the apertural region and sometimes it is separated from the footlayer to form vestibulum.
2. Tribe Coryleae (only Corylus): Pollen grains are triporate and suboblate. The exine
has no arci and the ektexine does not thicken in the apertural region; the sculpture of exine is
slightly rugulate under SEM; the exine structure is five-layered: granular layer, tectum,
columellae, footlayer and endexine. The tectum is vovered with a granular layer, the
columellae granular and the endexine thickens but is not separated from the footlayer in the
apertural region.
3. Tribe Carpineae (including Ostryopsis, Carpinus and Ostrya): Pollen grains are
mostly triporate and suboblate. The exine has no arci and the ektexine does not thicken in the
apertural region; the sculpture of exine is slightly rugulate under SEM; the exine structure is
five-layered which is the same as those of the Coryleae. The tectum is covered with a
granular layer, the columellae are columellate and the endexine is neither thickened nor separated from the footlayer in the apertural region. The operculum exists on the top of aperturemembrane. The pollen analysis of the Late Quaternary from South China, in particular, Yunnan and Guangdong provinces, made possible to restore the past distribution of Dacrydium in China. The chinese mainland flora today is totally devoid of Dacrydium. However, a single species (D. pierrei Hickel) is represented in the montane rainforests of Hainan Island (extreme south of China). The fossil pollen grains, morphologically comparable to that of D. pierrei, were discovered from the Tertiary and the Quaternary sediments in an extensive area of China. Even in the Last Interstadial of Wurm (40000–20000 Yr. B. P.), its distribution might extend to 22-24 degrees north latitude. The climatic deterioration and the environmental change during the Last Glacial (Late Wurm) had been a crucial factor when the flora of South China underwent drastic modification. It is in the same period that the present-day restriction of D. pierrei to Hainan Island took place. 相似文献
