Summary A tumor model system of clones of myeloproliferative sarcoma virus (MPV)-transformed rat fibroblasts (NRK) with different growth properties and metastatic potential was studied. The relationship between metastatic behavior and composition of carbohydrate-binding proteins (lectins) was analyzed by affinity chromatography. The metastatic variant differs qualitatively from its parental clone in the presence of galactoside-binding proteins at apparent molecular weights of 80 kDa, 70 kDa, 22 kDa, 18 kDa and 16 kDa and of a fucose-binding protein at apparent molecular weight of 42 kDa. The -glucosyl-binding proteins at apparent molecular weights of 67 kDa and 53 kDa and a galactoside-binding protein of apparent molecular weight of 34 kDa, however, are not detectable in the metastatic variant in comparison to its parental clone. In this respect the parental clone shows closer resemblance to the clone 5–8#1 with different growth properties and low metastatic potential than to its own metastatic variant. Furthermore, only the parental clone has a melibiose- and a mannan-binding protein of an apparent molecular weight of 64 kDa and 14 kDa, respectively. Rosette formation as model system for intercellular interaction reveals differences in the inhibition pattern with sugar between the two clones 5–8#1 and 5–20#20, whereas the metastatic variant 5–20#20 (s) exhibits drastically reduced capability to form rosettes. Initial experiments demonstrate the feasibility of drug targeting to transformed fibroblasts via carbohydrate-binding proteins.  相似文献

Five strains of pink-pigmented, facultatively methylotrophic bacteria (PPFM) isolated from plant surfaces and grown in different carbon sources, were fixed, embedded and sectioned for examination with the electron microscope. The strains studied represented the two mainsub-groups, those that can use carbohydrates and those that can not use carbohydrates as a sole carbon and energy source. All of the isolates examined, produced crystalloid inclusions, internal membranous vesicles and internal membranous sheets although the number of cells with inclusions, varied with the carbon source and specific strain. Polybetahydroxybutyrate and polyphosphate bodies were observed in all strains, with all carbon sources used for growing cells which includes methanol, formate and glycerol. Isolates that could use glucose accumulated polyglucoside granules but not when other carbon sources were provided. The relationship of these inclusions to growth conditions is discussed.  相似文献

Summary Polyps of mature colonies of Hydractinia echinata obey the rule of distal transformation by regenerating heads but not stolons. However, this rule is not valid for young polyps as these regenerate stolons from proximal cut ends. Also, small cell aggregates and even small fragments excised from full-grown polyps are capable of stolon formation. Aggregates produced from dissociated cells undergo either distal or proximal transformation depending on their size, speed of head regeneration in the donor used for dissociation and the positional derivation of the cells. The latent capability of stolon formation is released under conditions that cause loss of morphogens and depletion of their sources. However, internal regulative processes can also lead to gradual proximal transformation: regenerating segments of polyps sometimes form heads at both ends and the distal pattern is duplicated. Subsequently, all sets of proximal structures, including stolons, are intercalated. In contrast to distal transformation, proximal transformation is a process the velocity of which declines with the age and size of the cell community.  相似文献

Chlorotetracycline inhibits the uncoupled oxidation of exogenous NADH by Jerusalem artichoke (Helianthus tuberosus L.) mitochondria extensively (over 80%) and rapidly (inhibition complete in 10 s) in the presence of added Ca2+. Half-maximal inhibition is observed at 15 μM chlorotetracycline in the presence of 2 mM Ca2+. The oxidation of succinate is only affected marginally by chlorotetracycline plus Ca2+. The inhibition of NADH oxidation and the fluorescence of CTC are well correlated. Mn2+ is the only other cation which shows an (increased) inhibition in the presence of chlorotetracycline. The inhibition by Ca2+ and chlorotetracycline disappears at acid pH, and the pH optimum in their presence is 6.4. The inhibition caused by other lipid-soluble Ca2+-chelators is not reversible or is enhanced by the addition of excess Ca2+. In contrast, inhibition caused by relatively water-soluble chelators is completely reversed by added Ca2+. It is suggested that a neutral 1:2 complex is formed between Ca2+ and chlorotetracycline which can substitute for Ca2+ bound at sites in the lipophilic phase of the inner mitochondrial membrane, which are essential for the activity of the external NADH dehydrogenase.  相似文献

The plant-communities from habitats of the metallophyte species Minuartia verna and Thlaspi alpestre (T. caerulescens) at sites disturbed and undisturbed by mining are described. Four communities were delineated by cluster and principal component analysis. Group 1 comprised species-poor communities on disturbed non-calcareous soils; group 2, relatively species-rich communities on disturbed calcareous soils; group 3, species-rich communities in the main on undisturbed calcareous soils. Group 4 consisted of species-rich communities with an alpine element, in damp habitats on base-rich soils derived from igneous rocks. Total and exchangeable elements As, Ba, Ca, Cd, Co, Cr, Cu, Fe, Mg, Mn, Ni, Pb and Zn were determined for the soils of these sites. Levels of soil Ca, Cd, Pb and Zn accounted for most of the variation along the first axis of the PCA and soil nutrient levels were probably the main predictor along the second.Abbreviations PCA Principal Component Analysis  相似文献

Summary Exactly 150 years after the publication of the so-called cell theory the role of the botanist Schleiden has been reconsidered. It is emphasized that the study of plant sexual reproduction systems has contributed considerably to the development of the general cell concept. Summary The molecular evolution of cytochrome c from angiosperms is compared to that from vertebrates. On the basis of a cladistic analysis from 26 plant species, compared to that from 27 vertebrate species, we find that although the vertebrate sequences yield reasonably well-defined minimal trees that are congruent with the biological tree, the plant sequences yield multiple minimal trees that are not only highly incongruent with each other, but none of which is congruent with any reasonably biological tree. That is, the plant sequence set is much more homoplastic than that of the animal. However, as judged by the relative rate test, the extent of divergence, and degree of functional constraint, cytochrome c evolution in plants does not appear to differ from that of vertebrates.  相似文献

Summary The rate of synonymous nucleotide substitution in nuclear genes of higher plants has been estimated. The rate varies among genes by a factor of up to two, in a manner that is not immediately explicable in terms of base composition or codon usage bias. The average rate, in both monocots and dicots, is about four times higher than that in chloroplast genes. This leads to an estimated absolute silent substitution rate of 6 × 10–9 substitutions per site per year that falls within the range of average rates (2–8 × 10–9) seen in different mammalian nuclear genomes.  相似文献

The greening of the upper part of the outerAllium cepa L. bulb scales, in particular along the vascular regions, is limited to the hypodermal cells in which typical leucoplasts are transformed to normal and functional chloroplasts. This process is light dependent and cannot afterwards be reversed or modified by darkness. 利用间接免疫荧光的细胞化学技术对番茄、黄瓜、菠菜、甜菜及小麦等不同抗寒性植物微管的冷稳性进行了比较研究。结果指出,不抗寒的喜温性植物番茄和黄瓜的气孔保卫细胞的微管在0℃—1℃冷处理3小时即解聚;属于中等抗寒性植物的菠菜和甜菜幼苗经秋季低温锻炼后,其气孔保卫细胞的微管在0℃和—5℃低温处理3小时,均不发生解聚;具有较强抗寒性的冬小麦品种农大139幼苗在2—3℃低温锻炼期间,微管结构保持完整,经过15天低温锻炼的幼苗在-8℃冰冻处理3小时,微管也不受破坏。这些结果表明,微管的冷稳性与植物的抗寒性成正相关。  相似文献
