The primary objective of this study was to construct an immune-related long noncoding RNAs (IRLs) classifier to precisely predict the prognosis and immunotherapy response of patients with thymic epithelial tumors (TET). Based on univariable Cox regression analysis and Lasso regression, six prognosis-related IRLs (AC004466.3, AC138207.2, AC148477.2, AL450270.1, HOXB-AS1 and SNHG8) were selected to build an IRL classifier. Importantly, results of qRT-PCR validated that higher expression levels of AC138207.2, AC148477.2, AL450270.1 and SNHG8 as well as lower expression levels of AC004466.3, and HOXB-AS1 in TETs samples compared with normal controls. The IRL classifier could effectively classify patients into the low-risk and high-risk groups based on the different survival parameters. In terms of predictive ability and clinical utility, the IRL classifier was superior to Masaoka staging system. Additionally, IRL classifier is significantly associated with immune cells infiltration (dendritic cells, activated CD4 memory T cells and tumor-infiltrating lymphocyte (TIL), T cell subsets in particular), immune microenvironment (immune score and immune checkpoint inhibitors) and immunogenicity (TMB) in TETs, which hints that IRL classifier is tightly correlated with immune characteristics and might guide more effective immunotherapy strategies for TETs patients. Encouragingly, according to TIDE algorithm, there were more immunotherapy responders in the low-risk IRL subgroup and the IRL score was robustly negatively linked to the immunotherapeutic response. To sum up, the IRL classifier was established, which can be used to predict the prognosis, immune infiltration status, immunotherapy response in TETs patients, and may facilitate personalized counseling for immunotherapy.  相似文献

The present study demonstrates that 3,4-dihydroxyphenylethylamine (DA, dopamine) prevents neurotensin (NT) stimulation of both prolactin (PRL) release and calcium influx by interacting with specific receptors that are functionally linked to calcium channels. As shown by the studies with dispersed cells from rat anterior pituitary, the pharmacology of the control of PRL release and calcium influx, both induced by NT, was found to be typical of a DAergic process. This was demonstrated by the order of potency of agonists in inhibiting PRL release and calcium influx (DA greater than epinephrine greater than norepinephrine much greater than isoproterenol); by the high affinity of antagonists such as haloperidol and fluphenazine for this process; and by the high degree of stereoselectivity of sulpiride. Specific D2 receptor agonists, such as bromocriptine and lisuride, and the specific D2 receptor antagonist (-)-sulpiride were found to be highly potent on the DA receptors negatively coupled with calcium channels and PRL release. DA was found to lack the capacity to change the influx of calcium induced by either the sodium channel activator veratridine or high extracellular potassium levels, thus indicating a specific action of this amine on calcium channels sensitive to NT. In a range of concentrations that are effective in inhibiting either the calcium influx or the PRL release, both induced by NT, DA did not alter the cyclic AMP generating system. 目的 探讨结核杆菌感染与恶性肿瘤的关系。方法 采用多聚酶链反应(PCR) 技术检测两种恶性肿瘤组织中结核杆菌DNA(TB- DNA) 。结果41 例恶性肿瘤组织中检出TB- DNA 阳性患者8 例,占19.5 % ,且TB-DNA 阳性患者肿瘤发病部位基本与结核病的好发部位一致。结论 结核杆菌感染可能与一些恶性肿瘤存在特殊相关性。  相似文献

目的探讨不同浓度碘对60日龄仔鼠垂体TSH细胞的影响。方法将断乳1个月的Wistar大鼠仔鼠,雌雄各半随机分为5组:NI组、10HI组、50HI组、100HI及LI组。饲养3个月的雌雄大鼠1∶1合笼交配产生仔鼠,断乳后的仔鼠喂养同上述大鼠。测定60日龄仔鼠垂体TSH细胞的体密度、强阳性细胞百分数及甲状腺的相对重量、血清甲状腺激素水平。结果50HI、100HI组仔鼠垂体TSH细胞的体密度、强阳性细胞百分数及血清T4明显高于NI组,LI组垂体TSH细胞的体密度明显高于NI组、血清T4明显低于NI组,各碘过量组甲状腺的相对重量与NI组相比没有差别,LI组仔鼠甲状腺的相对重量明显高于NI组。结论碘缺乏对甲状腺、垂体TSH细胞的影响要强于碘过量对甲状腺、垂体TSH细胞所造成的影响。  相似文献

肿瘤是21世纪威胁人类健康的主要疾患之一。临床上,实体瘤治疗仍以手术切除、放化疗和靶向治疗为主,但这些方法往往不能根除肿瘤病灶,易导致肿瘤复发和进展。肿瘤免疫治疗是利用人体的免疫系统,通过增强或恢复抗肿瘤免疫力实现控制和杀伤肿瘤的一种新的治疗模式。肿瘤免疫治疗能够在众多患者中产生持久反应,过继性免疫治疗和免疫检查点阻断剂治疗均可产生显著的抗原特异性免疫反应。肿瘤浸润淋巴细胞(TILs)是一种存在于肿瘤组织内部具有高度异质性的淋巴细胞,在宿主抗原特异性肿瘤免疫应答中发挥关键作用。最新研究表明,在肿瘤发生和治疗过程中,TILs的亚群组成和数量与患者预后密切相关;抗肿瘤的TILs介导的过继性免疫治疗方法已在多种实体瘤中取得了良好的疗效。文中就实体肿瘤中TILs的研究进展作一综述。  相似文献

从斜带石斑鱼垂体提取总。RNA,再取其50ng合成SMART cDNA。从所构建的垂体SMART cDNA质粒文库中筛选到生长激素/催乳素基因家族的2个成员的全长cDNA片段:生长激素(GH)基因全长为938bp,编码204个氨基酸;催乳素基因(PRI.)全长为1429bp,编码212个氨基酸。采用计算机软件Mega 2和CLUSTAL W1.64b对9种鱼的生长激素/催乳素基因家族的3个成员(GH、PRL和生长催乳素SL)的氨基酸序列进行系统分析,构建NJ分支系统树,对于序列中的插入/缺失位点则采用Pairaise Deletion,1000次自展(Bootstrap)分析计算各节点支持率。根据3个基因的氨基酸序列构建的系统树表明,石斑鱼与金头鲷、金鲈和牙鲆聚成一类,虹鳟与大马哈鱼聚成一类,鲫鱼与鲶鱼聚成一类,鳗鲡成另外一类。根据石斑鱼全长cDNA推断的氨基酸序列比较表明,SL相对GH和PRL有较高的保守性。石斑鱼的GH、PRL和SL的氨基酸同源性在24%~31%,但其C-端的氨基酸同源性较高,尤其是C-端的3个Cys是严格保守的。其中SL与GH的同源性(30.8%)高于与PRL的同源性(25.6%),GH和PRL的同源性最低(24.1%)。  相似文献

摘要 目的:探讨卵巢肿瘤良恶性病变的超声征象及其与卵巢癌临床分期、病理分级的相关性。方法:选取我院2020年8月到2023年8月收治的148例卵巢肿瘤患者进行回顾性分析,以手术病理及病理活检作为诊断金标准,将60例确诊为卵巢癌的患者纳入恶性组,将88例确诊为卵巢良性肿瘤的患者纳入良性组。对所有患者进行超声检查,分析其超声图像特征。随后分析60例卵巢癌患者不同临床分期和病理分级患者的超声诊断参数情况,并分析超声诊断相关参数与卵巢临床分期、病理分级的相关性。结果:超声对卵巢肿瘤良性诊断准确率为86.36 %(76/88)对恶性肿瘤诊断率为88.64 %(78/88);不同临床分期患者RI、PI、EDV、PSV对比差异显著,Ⅰ期患者RI、PI为(0.77±0.14)和(1.67±0.24)高于Ⅱ期(0.64±0.15)和(1.25±0.16)、Ⅲ期患者(0.52±0.17)和(0.96±0.16)、Ⅳ期(0.41±0.12)和(0.76±0.12),Ⅰ期患者EDV和PSV(8.63±1.27)cm/s和(16.53±2.53)cm/s低于Ⅱ期(10.25±1.68)cm/s和(18.44±1.58)cm/s、Ⅲ期(12.73±1.79)cm/s和(20.14±2.25)cm/s、Ⅳ期患者(15.51±1.12)cm/s和(23.06±1.98)cm/s(P<0.05);不同临床分期患者RI、PI、EDV、PSV对比差异显著,Ⅰ级患者RI、PI为(0.81±0.16)和(1.62±0.19)高于Ⅱ级(0.65±0.12)和(0.91±0.22)、Ⅲ级患者(0.47±0.17)和(0.67±0.13),Ⅰ级患者EDV和PSV(8.32±1.51)cm/s和(15.12±3.33)cm/s低于Ⅱ级(12.75.±1.14)cm/s和(21.31±3.14)cm/s、Ⅲ级患者(15.35±1.79)cm/s和(24.08±2.04)cm/s(P<0.05);Spearman相关分析结果表明:临床分期、病理分级与RI、PI呈负相关,与EDV、PSV呈正相关(P<0.05)。结论:超声对卵巢肿瘤的良恶性病变诊断具有重要指导价值,且与卵巢癌的临床分期、病理分级具有明显相关性,值得临床应用推广。  相似文献

Light-microscopic immunocytochemistry of ferret anterior pituitary revealed the localization of somatotropes in the pars distalis, but no immunoreactive cells were detected in the pars tuberalis. Ultrastructural studies by superimposition immunocytochemistry and immuno-electron microscopy, clucidated the morphological heterogeneity of these somatotropic cells. They were classified into 2 subtypes on the basis of size of the secretory granules. Type-I cells with small granules (mean diameter, 192 nm), were considered to be the immature somatotrop, while Type-II cells, with comparatively larger secretory granules (mean diameter, 257 nm), were considered to be the matured form of Type-I cells and the typical somatotropic cell-type, and were much more predominant than the Type-I cells. The fact that Type-II cells had a distinct Golgi zone and many mitochondria, while in Type-I cells the intracellular organelles were generally less developed, supports this suggestion. In addition to these two extreme subtypes, several intermediate forms were also encountered that may represent different transitional phases during the conversion of Type I to Type II. Protein A-gold immuno-electron microscopy illustrated the specific localization of growth hormone over the granules, with no labelling over any other cytoplasmic organelles of the 2 somatotrope subtypes.  相似文献
