刺芹侧耳是一种典型的具四极性交配型系统的担子菌,通常需要一对可亲和的单核菌丝体才能够形成稳定的双核菌丝体。本研究将刺芹侧耳单核菌株181(A1B1)中A和B交配型基因HD1和HD2以及信息素前体基因PEphb3.1和PEphb3.3克隆后,通过PEG介导的方式敲入到可亲和的单核菌株183(A2B2)中,获得了22个阳性转化子。运用荧光显微镜对其中11个具有锁状联合的转化子进行了详细观察,描述了锁状联合和细胞核相。研究结果为进一步开展交配型基因的遗传操作研究提供了方法学上的借鉴和参考。本文还探讨了同核双核体的概念和应用价值。 相似文献

苏云金芽胞杆菌幕虫亚种的伴胞晶体在预芽胞外壁内侧形成,呈现晶体芽胞粘连的现象。根据已发表的cry26Aa1和cry28Aa1基因序列设计引物,从苏云金芽胞杆菌幕虫亚种T02中扩增得到cry26Aa和cry28Aa基因,通过穿梭载体将这两个基因分别和同时转化到苏云金芽胞杆菌无晶体突变株BMB171后,透射电镜下可在芽胞外壁内侧和外侧同时观察到伴胞晶体,而单独表达时可在芽胞外壁外侧观察到伴胞晶体。结果表明,伴胞晶体在芽胞外壁内侧表达不单独依赖于启动子的时空调控,可能还受到晶体蛋白相互作用的影响。 相似文献

Summary The development of pit-pairs between differentiating xylem cells has been examined by transmission electron microscopy in young shoots ofSorbus torminalis. In some vessel-to-tracheid pits, as well as in previously studied intertracheid pits, a thickening of the pit membrane containing branched plasmodesmata was observed. A secondary wall-like cap was deposited over the thickening prior to cytoplasmic autolysis; some plasmodesmata, parallel to the plane of section, appeared to perforate the cap. At the end of the cell maturation stage, the central part of the primary wall thickening was hydrolysed, while the cap, including plasmodesmata remnants, appeared unaltered. In half-bordered pit-pairs between a parenchyma cell and a vessel or a tracheid, similar structures could be observed beside the conducting elements. When the vessel or tracheid matured, sealing of the pit membrane plasmodesmata resulted from the formation of a protective layer on the parenchyma-side rather than from the deposition of a cap on the conducting cell-side. These observations provide the first information on the presence of symplasmic connections in pits between differentiating vessels and neighbouring xylem cells. InS. torminalis, xylem differentiation is probably highly coordinated within a symplasmic domain; the persistence of such connections may account for the lack of specialization ofSorbus wood. 相似文献

Summary Pit cells, on which almost no further contributions have been presented since the first report by Wisse et al. (1976), are described in detail in the rat liver. These cells show several characteristic features: 1) rod-cored vesicles, a new type of vesicular inclusion observed first in our study; 2) electron-dense granules, which we consider to arise from multivesicular bodies by the accumulation of dense material; and 3) well-developed pseudopodia. Although these features clearly differentiate pit cells from conventional lymphocytes, these two cell types display similarities (i) in a number of ultrastructural features, (ii) in the pattern of their intralobular distribution, and (iii) in their presence in the spleen and peripheral blood. 相似文献

Orbilia querci, a new nematode-trapping fungus, was found on rotten wood of Quercus sp. in Huai-rou County, Beijing, China. It is characterized by having a tear-shaped spore body in the cylindrical ascospore. Pure culture was obtained from the ascospores. Conidiophores were simple or occasionally branched, bearing a single conidium on the tip. Conidia were spindle-shaped, mostly with 3-septa. Nematodes were captured by means of adhesive stalked knobs. The adhesive knobs were produced frequently on nutritional agar plates even in the absence of challenging nematodes. Its anamorph is placed in Dactylellina and named as D. querci. The sequence divergence of the ITS1 region between the fungus and the other knob-forming species tested was 23.8-33.4%, supporting O. querci as a distinct species. This is the first report of the connection between a knob-forming nematophagous hyphomycete and an Orbilia teleomorph. 相似文献

The Tyr corner is a conformation in which a tyrosine (residue “Y”) near the beginning or end of an antiparallel β-strand makes an H bond from its side-chain OH group to the backbone NH and/or CO of residue Y – 3, Y – 4, or Y – 5 in the nearby connection. The most common “classic” case is a Δ4 Tyr corner (more than 40 examples listed), in which the H bond is to residue Y – 4 and the Tyr x1 is near ?60°. Y – 2 is almost always a glycine, whose left-handed β or very extended β conformation helps the backbone curve around the Tyr ring. Residue Y – 3 is in polyproline II conformation (often Pro), and residue Y – 5 is usually a hydrophobic (often Leu) that packs next to the Tyr ring. The consensus sequence, then, is LxPGxY, where the first x (the H-bonding position) is hydrophilic. Residues Y and Y – 2 both form narrow pairs of β-sheet H-bonds with the neighboring strand, Δ5 Tyr corners have a 1-residue insertion between the Gly and the Tyr, forming a β-bulge. One protein family has a Δ4 corner formed by a His rather than a Tyr, and several examples use Trp in place of Tyr. For almost all these cases, the protein or domain is a Greek key β-barrel structure, the Tyr corner ends a Greek key connection, and it is well-conserved in related proteins. Most low-twist Greek key β-barrels have 1 Tyr corner. “Reverse” Δ4 Tyr corners (H bonded to Y + 4) and other variants are described, all less common and less conserved. It seems likely that the more classic Tyr corners (Δ4, Δ5, and Δ3 Tyr, Trp, or His) contribute to the stability of a Greek key connection over a hairpin connection, and also that they may aid in the process of folding up Greek key structures. 相似文献

Balsam poplar seeds are short‐lived and require moist seedbeds soon after they are released to germinate. In addition to sexual reproduction, balsam poplar stands can regenerate clonally by root suckering. The origin of stands will in turn affect their genetic structure and root system architecture, which are poorly understood for upland forest stands. Three stands were hydraulically excavated in Quebec (moist) and Alberta (dry) to determine the origin of trees and to characterize root systems with respect to presence of parental roots and root grafts connections. Clones were identified using single‐nucleotide polymorphism (SNPs), and all stems, roots and root grafts were aged using dendrochronology techniques. All 82 excavated trees were of sucker origin, and four of the six stands contained a single clone. Parental root connections were found between 22% and 25% of excavated trees, and 53% and 48% of trees were linked with a root graft between the same or different clones, in Alberta and Quebec, respectively. Mean distance between trees connected by parental root was significantly lower than the distance between unconnected trees (0.47 ± 0.25 m vs. 3.14 ± 0.15 m and 1.55 ± 0.27 m vs. 4.25 ± 0.13 m) in Alberta and in Quebec, respectively. The excavations also revealed many dead stumps with live roots, maintained through root connections with live trees. This research highlights that balsam poplar growing in upland stands is a clonal species that can maintain relatively high genotypic diversity, with frequent root connections between trees at maturity. Maintaining an extensive root system through root connections increases the chances of a clone surviving when the above ground tree is dead and may also enhance the resilience of balsam poplar stands after disturbance. 相似文献

Summary Direct evidence for monosynaptic connections between filiform hair sensory axons and giant interneurons (GIs) in the first instar cockroach, Periplaneta americana, was obtained using intracellular recording and HRP injection followed by electron microscopy. GIs 1–6 all receive monosynaptic input from at least one filiform afferent axon. GI1, GI2 and GI5 receive input only from the medial (M) axon, while GI3, GI4 and GI6 receive input from both M and lateral (L) axons. The dendrites of GI3 and GI6 which are contralateral to the cell bodies receive input from both axons whereas the smaller ipsilateral dendritic fields have synapses only from the L axon. GI5 has M axon input only onto its contralateral dendrites. In 50% of preparations GI7 receives weak input from the ipsilateral L axon. There is no obvious relationship between the morphology of the giant interneurons and the pattern of input they receive from the filiform afferents.Abbreviations

自1987—1990,在长江以南九省(区)采集52个香菇品系,43株制作了孢子印用以检测交配型,又以检测A.B因子位点的等位基因方法测定了担孢子的基因型,保藏了以A_xB_x(或A_xB_y……)标记的标准单核菌丝体,这是建立中国香菇种质库的第一步,将为香菇杂交育种提供有效亲本。结果表明:每一菌株的四个交配型呈随机分配。具有四个交配型的菌株百分率是79%,有二个交配型的香菇菌株百分率是21%。有9个菌株的单核菌丝的基因型从A_1B_1……A_(18)B_(18)不同,A.B因子等位基因差异明显,这些菌株显示了A.B因子的很低的地区性重复频率。本项研究中建立了区分同宗A同宗B以及四个交配型的试验方法,遵循着核移动的规律,这个方法能够重复。 相似文献
