This study attempted to eradicate Apple stem pitting virus (ASPV) and Apple stem grooving virus (ASGV) from virus‐infected in vitro shoots of apple rootstocks ‘M9’ and ‘M26’ using shoot tip culture and cryopreservation. In shoot tip culture, shoot tips (0.2 mm in length) containing two leaf primordia failed to show shoot regrowth. Although shoot regrowth rate was the highest in the largest shoot tips (1.0 mm in length) containing four leaf primordia, none of the regenerated shoots was virus‐free. Shoot tips (0.5 mm in length) containing two and three leaf primordia produced 100% and 10% of ASPV‐free shoots, respectively, while those (1.0 mm) containing four leaf primordia were not able to eradicate ASPV. ASGV could not be eradicated by shoot tip culture, regardless of the size of the shoot tips tested. In cryopreservation, shoot tips (0.5 mm in length) containing two leaf primordia did not resume shoot growth. Although 1.0‐mm and 1.5‐mm shoot tips gave similarly high ASPV‐free frequencies, the latter had much higher shoot regrowth rate than the former. Very similar results of shoot regrowth and virus eradication by shoot tip culture and cryopreservation were observed in both ‘M9’ and ‘M26’. Histological observations showed that only cells in upper part of apical dome and in leaf primordia 1–3 survived, while other cells were damaged or killed, in shoot tips following cryopreservation. Virus immunolocalization found ASPV was not detected in upper part of apical dome and leaf primordia 1 and 2, but was present in lower part of apical dome, and in leaf primordium 4 and more developed tissues in all samples tested. ASPV was also detected in leaf primordium 3 in about 16.7% and 13.3% samples tested in ‘M9’ and ‘M26’. ASGV was observed in apical dome and leaf primordia 1–6, leaving only a few top layers of cells in apical dome free of the virus. Sporangia of three isolates of Phytophthora ramorum representing three different clonal lineages were subjected to relative humidity (RH) levels between 80 and 100% for exposure periods ranging from 1 to 24 h at 20°C in darkness. Plastic containers (21.5 × 14.5 × 5 cm) were used as humidity chambers with 130 ml of glycerine solution added to each container. Glycerine concentrations corresponded to 100, 95, 90, 85 and 80% RH based on refractive index measurements. Sporangia suspensions were pipeted onto nitrile mesh squares (1.5 × 1.5 cm, 15 micron pore size) which were placed in the humidity chambers and incubated at 20°C in darkness. Following exposure periods of 1, 2, 4, 8, 12 and 24 h, mesh squares were inverted onto Petri dishes of selective medium and sporangia germination assessed after 24 and 48 h. At 100% RH, we observed a mean value of 88% germination after 1 h exposure declining to 18% germination following 24 h incubation. At 95% RH, a steeper decline in germination was noted, with means ranging from 79% at 1 h to less than 1% at 24 h exposure. At 90% RH, no germination was noted after 8 or more h exposure, and values were 57%, 22% and 3% germination for the 1, 2 and 4 h exposures, respectively. Germination was only observed at 1 h exposure for both the 85% RH treatment (52% germination) and the 80% RH treatment (38% germination). The three isolates responded similarly over the range of RH values tested. The germination response of P. ramorum sporangia to RH values between 80% and 100% was comparable to that reported for other Phytophthora species. Knowledge of conditions that affect P. ramorum sporangia germination can shed light on pathogenesis and epidemic potential and lead to improved control recommendations. 相似文献

该实验以辣椒疫霉菌(Phytophthora capsici)致病性菌株NJ01和‘苏椒5号’辣椒幼苗为研究对象,通过肉桂醛对辣椒疫霉菌的体外抑菌作用、室内侵染效果以及对辣椒幼苗防卫反应的调控作用,揭示肉桂醛对增强辣椒疫霉病抗性的作用机制。结果表明:(1)肉桂醛对实验所用致病性菌株NJ01的抑制中浓度(EC50)值为0.81mmol/L;肉桂醛处理可导致NJ01菌丝严重皱缩、畸形、断裂;PI染色显示NJ01菌丝出现明显的细胞死亡现象。(2)单独NJ01菌株接种的辣椒植株表现出明显病症(茎基部变黑褐色、干枯萎缩,植株倒伏,叶片脱落,生物量下降);而肉桂醛处理2h后接种NJ01菌株的辣椒植株长势良好,无明显病症,鲜重和叶绿素含量显著上升至对照水平。(3)与单独接种NJ01菌株处理相比,肉桂醛处理2h后接种NJ01菌株的辣椒植株体内抗氧化酶(CAT、SOD、POD)活性显著上升,抗氧化物质(GSH和ASA)含量显著增加。研究认为,肉桂醛可能通过抑制辣椒疫霉菌的生长及其对辣椒植株的侵染能力,同时调控辣椒植株防卫反应,进而提高辣椒对疫病抗性。 相似文献

Whole‐genome sequencing‐based bulked segregant analysis (BSA) for mapping quantitative trait loci (QTL) provides an efficient alternative approach to conventional QTL analysis as it significantly reduces the scale and cost of analysis with comparable power to QTL detection using full mapping population. We tested the application of next‐generation sequencing (NGS)‐based BSA approach for mapping QTLs for ascochyta blight resistance in chickpea using two recombinant inbred line populations CPR‐01 and CPR‐02. Eleven QTLs in CPR‐01 and six QTLs in CPR‐02 populations were mapped on chromosomes Ca1, Ca2, Ca4, Ca6 and Ca7. The QTLs identified in CPR‐01 using conventional biparental mapping approach were used to compare the efficiency of NGS‐based BSA in detecting QTLs for ascochyta blight resistance. The QTLs on chromosomes Ca1, Ca4, Ca6 and Ca7 overlapped with the QTLs previously detected in CPR‐01 using conventional QTL mapping method. The QTLs on chromosome Ca4 were detected in both populations and overlapped with the previously reported QTLs indicating conserved region for ascochyta blight resistance across different chickpea genotypes. Six candidate genes in the QTL regions identified using NGS‐based BSA on chromosomes Ca2 and Ca4 were validated for their association with ascochyta blight resistance in the CPR‐02 population. Fusarium graminearum is the main causal agent of Fusarium head blight (FHB) of small grain cereals, but the importance of weeds in the FHB disease cycle and the establishment of F. graminearum in agroecosystems are still not fully understood. The objective of this study was to determine the potential role of weeds present within cereal crop rotations as alternative hosts. F. graminearum was isolated from different organs of asymptomatic weeds sampled from six fields with cereal-crop rotations in Lithuania for two consecutive years (2015 and 2016). The fungi were identified using morphological and molecular methods. Out of 57 weed species that were investigated, 41 (71.9%) harboured F. graminearum isolates. Twenty five weed species were identified as new, previously undocumented, hosts. The majority (73.3%) of the isolates of F. graminearum from this study belonged to the 15ADON genotype while a smaller proportion (23.4%) belonged to the 3ADON genotype. All F. graminearum isolates that were assessed induced FHB symptoms on artificially inoculated spring wheat tested in the field. 相似文献

Electrophysiological and behavioral responses of the wingless tea aphid, Toxoptera aurantii (Boyer), to 14 synthetic volatiles identified from tea shoots, their partial (GLV mixture) and full (ACB mixture) blends, and fresh young tea leaves, buds, tender stems, adult tea leaves and tea aphid-damaged young leaves (ADYL) were studied by using an electroantennography (EAG) and a four-arm olfactometer. ACB elicited the largest EAG responses. Major volatile components, Z-3-hexen-1-ol, E-2-hexenal, n-hexanol, methyl salicylate and benzylalcohol, from the tea shoots were strongly EAG active. All the 4 tested tea shoot tissues also elicited significant EAG responses, with the young tea leaves being the strongest, followed by buds, tender stems and adult tea leaves. Surprisingly, ADYL elicited a weakly negative EAG response. In the olfactory assays, the fresh and tender tea leaves, as well as the individual major volatile components, e.g. Z-3-hexenyl acetate, methyl salicylate, E-2-hexen-1-ol and Z-3-hexen-1-ol, from the tender shoots (EAG-active) were all attractive. This result might indicate that the wingless tea aphids may use tea shoot volatiles as kairomone to find their optimal feeding sites, e.g. fresh tender tea shoots. 相似文献
