Adverse health risks from environmental agents are generally related to average (long-term) exposures. Because a given individual's contact with a pollutant is highly variable and dependent on activity patterns, local sources and exposure pathways, simple 'snapshot' measurements of surrounding environmental media may not accurately assign the exposure level. Furthermore, susceptibility to adverse effects from contaminants is considered highly variable in the population so that even similar environmental exposure levels may result in differential health outcomes in different individuals. The use of biomarker measurements coupled to knowledge of rates of uptake, metabolism and elimination has been suggested as a remedy for reducing this type of uncertainty. To demonstrate the utility of such an approach, we invoke results from a series of controlled human exposure tests and classical first-order rate kinetic calculations to estimate how well spot measurements of methyl tertiary butyl ether and the primary metabolite, tertiary butyl alcohol, can be expected to predict different hypothetical scenarios of previous exposures. We found that blood and breath biomarker measurements give similar results and that the biological damping effect of the metabolite production gives more stable estimates of previous exposure. We also explore the value of a potential urinary biomarker, 2-hydroxyisobutyrate suggested in the literature. We find that individual biomarker measurements are a valuable tool in reconstruction of previous exposures and that a simple pharmacokinetic model can identify the time frames over which an exogenous chemical and the related chemical biomarker are useful. These techniques could be applied to broader ranges of environmental contaminants to assess cumulative exposure risks if ADME (Absorption, Distribution, Metabolization and Excretion) is understood and systemic biomarkers can be measured. 相似文献

This article describes the integration of life-cycle assessment methods with a new input-output model of the world economy to analyze the environmental and economic implications of alternative future diets. The article reviews findings by industrial ecologists about the energy and land required for the production and consumption of alternative foods and diets in several European countries. It also reviews attributes of foods and diets identified by nutritionists as reducing the risks of obesity and major chronic diseases related to the diets of the affluent. The predominantly plant-based Mediterranean-type diet emerges as a dietary scenario that could satisfy both sets of concerns. The likely implications for agriculture and for farm policies of a shift toward this diet from the current average diet in the United States are discussed and shown to be substantial. The one-country studies reviewed in the article provide substantial insights into the potential ramifications of dietary change. Many of the limitations of these studies could be overcome by conducting the analysis in a global framework that represented the relationships among consumption, production, and trade and the physical constraints within which they operate. Analysis of the environmental and economic implications of alternative scenarios describing healthy diets can help stimulate more intensive dialogue, debate, and action among the interested parties; such analysis can both benefit from and contribute to initiatives such as the World Health Organization's global strategy on diet and health, which intends to enlist the support of governments, corporations, and civil society. 相似文献

In order to determine the effects of chronic exercise on the kidneys at different ages, young, adult, and old rats swam 1 hour either daily or twice a week for 10 weeks and then were killed along with unexercised controls. The kidneys were removed and sections were prepared for histometric analysis including planimetric measurements on camera lucida drawings of renal components and line sampling. With both degrees of exercise young rats showed lower kidney weight, fewer glomeruli and less medullary tissue than unexercised controls. In the adult group no significant differences were noted between exercised and unexercised rats. In old rats both degrees of exercise resulted in a loss of kidney weight and medullary and cortical mass, and a decrease in size of glomeruli while total number of glomeruli remained unchanged. Levels of Mo, As, Se, Fe, Cu, Zn, Ni, and Pb were determined in a vegetable commonly consumed in Pakistan. Samples were collected from three different sites (Ratokala, Phularwan, and Mailowal) supplied with three different water sources. Mo and Pb in water and Mo and As in C. sativum were higher than the suggested standards at the international level. Lower bioconcentration factor and pollution load index were seen at Site-I than at other sites. A positive association was found between the rhizosphere metals and those of the vegetable at all three sites. Enrichment factor at three sites ranged from 0.28 to 10.39. The highest value of daily intake of metals was found for Fe (0.245 mg/kg/day) and of the health risk index for As (70.41) at the wastewater inundated site. It is inferred that uptake of Mo, As, Cu, Ni, and Pb through C. sativum represents a high health danger to the individuals using this vegetable. 相似文献

In this research, carcinogenic and non-carcinogenic human health risks due to polycyclic aromatic hydrocarbons (PAHs) were investigated via three exposure pathways: accidental ingestion of soil, dermal contact of soils, and contaminated vegetable ingestion. To determine the contaminant concentrations in soil, samples were collected from areas adjacent to the Tehran oil refinery, located in Shahr-e-Ray city, Iran. Analyses of the samples indicated that the average of PAHs concentration in the soil samples were greater than clean-up level guidelines. Cancer risk of contaminants due to ingestion of cultivated vegetables that are sold in Tehran markets was significant in comparison with other exposure pathways. Moreover, the total cancer risk for 5th percentile, 95 upper confidence limit, and 95th percentile concentration of contaminants were 5.69E-04, 8.78E-02, and 2.13E-01, classifying the site as having a significant cancer risk potential. Furthermore, non-carcinogenic health risk analyses for the contaminants demonstrated hazard index of less than 1. Remediation of the soils is highly recommended to eliminate the potential cancer risks and prevent the contamination of the food chain for approximately 10 million Tehran residents. 相似文献
