Stretch of activated muscles leads to enhanced forces compared to isometric contractions at the same muscle length and the same level of activation. This so-called residual force enhancement (RFE) is thought to be a property of all muscles and preparations. However, observations concerning the existence, amount and duration of RFE are inconsistent, especially for voluntary activated large human muscles. Therefore, physiological relevance for daily activity is still questionable and the purpose of this study was to examine whether RFE is present in voluntary sub-maximal activated quadriceps femoris (QF). Seated in a rotational dynamometer with EMG attached to superficial parts of QF, 30 subjects performed isometric and isometric-eccentric-isometric contractions (20° stretch, ω = 60° s−1) at 30% and 60% of maximum voluntary activation (MVA) and contraction (MVC). To account for the complexity of the multi-headed QF, a compensation model based on physiological cross-sectional area and individual EMG-torque relations was used to interpret EMG data. For both levels of intensity and both feedback control strategies, ANOVA identified significant RFE (at the same level of activation) and reduced activation (at the same level of torque). Quercus coccifera L. is a Mediterranean sclerophyllous shrub with a high capacity to resist intense drought stress. Therefore, it could be
used in the study of physiological changes suffered by plants at very low water potentials. A remote sensing sensor was used
to measure continuously the physiological reflectance index (PRI; defined as the changes in reflectance at 531 nm with respect
to those at 570 nm; PRI = [(R531 − R570)/(R531 + R570)] at canopy level and under field conditions in an artificial carpet
of seedlings of Q. coccifera during a drought cycle. Correlations between leaf level-measured chlorophyll fluorescence parameters as well as the de-epoxidation
state of the xanthophyll cycle [(A + Z)/(V + A + Z)] and canopy level-measured PRI were reasonably good (R
2 = 0.57–0.63, P < 0.01), and quite interesting for water stress remote sensing purposes. The instrument’s temporal resolution allowed us
to follow the rapid response of PRI to changing photosynthetic active radiation, and to resolve, in response to cloud-induced
changes in light intensity, a fast and a slow PRI component. We report the disappearance of the rapid one under conditions
of intense drought in response to a sudden increase in light intensity. The underlying photoprotection mechanisms that Q. coccifera shows in response to intense drought stress periods seem to be related to the existence of a low intrathylakoid lumenal pH
at the end of the drought cycle. Under intense drought, these mechanisms allow this species to avoid oxidative damage, which
was evidenced by the maintenance of an unaltered photosynthetic pigment composition and constant photosystem II efficiency
in the mornings. It is concluded that, contrary to early reports, PRI is a sensible, indirect, non-destructive water stress
indicator, even in plants experiencing intense drought.

Performance of a clonal species in patchy environments: effects of environmental context on yield at local and whole-plant scales 总被引:1,自引:0,他引:1
Yield of the clonal plant Glechoma hederacea was compared at different spatial scales, in heterogeneous and homogeneous environments providing the same amount of nutrients.
For the heterogeneous treatments, environments were created with different patch sizes and different degrees of contrast in
nutrient concentration between patches of different quality. Total clone yield differed by almost 2.5-fold across treatments,
being highest in environments with large patches and high contrast, lowest in environments with small patches and high contrast,
and intermediate under homogeneous conditions. Compared with plants in homogeneous conditions, there were significant increases
or decreases in yield at all scales of measurement in many of the heterogeneous treatments. These effects on yield reflected
a combination of local responses to growing conditions and modification of these responses due to physiological integration
with other parts of the plant growing in contrasting conditions, supporting the proposal of de Kroon et al. (2005 New Phytol 166:73–82). The results show that plant yield at all scales is strongly dependent on environmental context, and
that maximum yield can only be realized under a limited range of heterogeneous conditions. 相似文献

目的对视网膜锥体细胞退行性变(retinal cone degeneration,RCD)大鼠的主要脏器及血液、生化指标进行检测。方法随机选择BN(Brown Norway)-RCDF2代大鼠,视网膜电图鉴定,取静脉血检测生化指标,并对重要脏器进行准确称量。结果此大鼠脏器质量在雌雄之间除体重、大脑和肾差异有显著性外(P〈0.05),其余均无差异;脏器系数脑干和小脑雌性显著大于雄性(P〈0.01);生化指标碱性磷酸酶、葡萄糖、白蛋白、尿素氮、总蛋白雌性高于雄性,总胆固醇、天冬氨酸转氨酶、丙氨酸转氨酶、甘油三酯雌性低于雄性,其中雌性碱性磷酸酶活性显著高于雄性;雌性离子钙浓度显著高于雄性。结论BN-RCD大鼠基础生理值和生化参数均在正常范围内,其中一些参数在性别之间有差异,但与正常SD大鼠相比,天冬氨酸转氨酶、丙氨酸转氨酶、肌酐、钾离子差异显著,葡萄糖、甘油三酯、离子钙有差异。RCD大鼠为遗传性视网膜疾病的研究提供了一种新的突变模式动物。 相似文献

Cardiac hypertrophy induced by exercise is associated with less cardiac fibrosis and better systolic and diastolic function, suggesting that the adaptive mechanisms may exist in exercise-induced hypertrophy. To identify molecular mechanisms by which exercise training stimulates this favorable phenotype, a proteomic approach was employed to detect rat cardiac proteins that were differentially expressed or modified after exercise training. Sixteen male Sprague–Dawley rats were divided into trained (T) and control(C). T rats underwent eight weeks of swimming training seven days/week, using a high intensity protocol. Hearts were used to generate 2-D electrophoretic proteome maps. Training significantly altered 23 protein spot intensities (P < 0.05), including proteins associated with the mitochondria oxidative metabolism, such as prohibitin, malate dehydrogenase, short-chain acyl-CoA dehydrogenase, triosephosphate isomerase, electron transfer flavoprotein subunit beta, ndufa10 protein, ATP synthase subunit alpha and isocitrate dehydrogenase [NAD] subunit. Additionally, Prohibitin was increased in the exercise-induced hearts. Cytoskeletal, signal pathway, stress and oxidative proteins also increased within T groups. These results strongly support the notion that the observed changes in the expression of energy metabolism proteins resulted in a potential increase in the capacity to synthesise ATP, probably via mitochondrial oxidative metabolism. The observed changes in the expression of these metabolic and structural proteins induced by training may beneficially influence heart metabolism, stress response and signalling paths, and therefore improve the overall cardiac function. 相似文献

Water stress is a major limitation for plant survival and growth. Several physiological and antioxidative mechanisms are involved
in the adaptation to water stress by plants. In this experiment, tea cultivars (TV-1, TV-20, TV-29 and TV-30) were subjected
to drought stress by withholding water for 20 days followed by rehydration. An experiment was thus performed to test and compare
the effect of dehydration and rehydration in growing seedlings of tea cultivars. The effect of drought stress and post stress
rehydration was measured by studying the reactive oxygen species (ROS) metabolism in tea. Water stress decreased nonenzymic
antioxidants like ascorbate and glutathione contents with differential responses of enzymic antioxidants in selected clones
of Camellia sinensis indicating an oxidative stress situation. This was also apparent from increased lipid peroxidation, O2
− and H2O2 content in water stress imposed plants. But the oxidative damage was not permanent as the plants recovered after rehydration.
Comparatively less decrease in antioxidants, higher activities of POX, GR, CAT with higher phenolic contents suggested better
drought tolerance of TV-1, which was also visible from the recovery study, where it showed lower ROS level and higher recovery
of antioxidant property in response to rehydration, thus proving its better recovery potential. On the other hand, highest
H2O2 and lipid peroxidation with decrease in phenolic content during stress in TV-29 suggested its sensitivity to drought. The
antioxidant efficiency and biochemical tolerance in response to drought stress thus observed in the tested clones of Camellia sinensis can be arranged in the order as TV-30 > TV-1 > TV-29 > TV-20. 相似文献

We present an upgrade to the giant unilamellar vesicle (GUV) electroformation method allowing easy GUV production in different buffers and with various membrane compositions. Our experimental results reveal that lipid deposits obtained from aqueous liposome or proteoliposome dispersions are highly efficient for GUV electroformation. This is related to the ability of such dispersions to produce readily well-oriented membrane stacks. Furthermore, we present a protocol for GUV electroformation in various aqueous media, including electrolyte-containing buffers at characteristic concentrations of biological fluids. This work unlocks historical barriers to GUV applications in scientific fields like biology, biochemistry, or biophysics where membrane composition, as well as its aqueous environment, should be adapted to biological significance. 相似文献

Two contrasting sea buckthorn (Hippophae rhamnoides L.) populations from low and high altitude regions were employed to investigate the effects of prevailing and enhanced ultraviolet-B (UV-B) radiation on plant growth and physiological properties under a UV-B-enhanced/exclusion system. The experimental design included three UV-B regimes, including excluded (-UVB), near-ambient (NA) and enhanced UV-B (+UVB) radiation. Compared with the control (-UVB), NA caused the formation of smaller but thicker plant leaves in both sea buckthorn populations, paralleled with significant increments of carotenoids and UV-absorbing compounds as well as improved water economy. NA also induced more biomass partition from shoot to root, but CO(2) assimilation rate (A), photosynthetic area and biomass accumulation were unaffected. The low-altitude population seemed sensitive to +UVB, as indicated by the decreases in total biomass, A and ascorbic acid content (Asa, an antioxidant) compared with NA. However, little +UVB effect occurred on the high-altitude population, and we suggest that the higher tolerance of this population could be associated with its specific morphological and physiological characteristics, such as small but thick leaves and high-level of Asa content, as well as its greater physiological modification in response to NA, e.g., increases in protective compounds (carotenoids and UV-absorbing compounds) and improvement in water economy, in comparison to the low-altitude population, which form an effective adaptation strategy to enhanced UV-B stress. 相似文献

Variation within species and inter-species comparison of seed dormancy and germination of four annual Lamium species 总被引:1,自引:1,他引:0
In an ecological context, knowledge of intra-species variation in dormancy and germination is necessary both for practical and theoretical reasons. We used four or five seed batches (replicates) of four closely related annuals co-occurring in arable fields in Sweden: Lamium amplexicaule, L. confertum, L. hybridum and L. purpureum. Seeds used for experiments stemmed from plants cultivated on two sites, each site harbouring one population of each species, thereby ensuring similar environmental history of seeds. Seeds were tested for germination when fresh and after three different pre-treatments (cold or warm stratification, or dry storage) for up to 24 weeks. Seeds were also sown outdoors. Despite substantial intra-species variation, there were clear differences between species. The general seed dormancy pattern, i.e. which environmental circumstances that affect dormancy, was similar for all species; dormancy reduction occurred during warm stratification or dry storage. Even though the response to warm stratification indicates a winter annual pattern, successful plants in Sweden were mostly spring emerged. Germination in autumn occurred, but plants survived winters poorly. Consequently, as cold stratification did not reduce dormancy, strong dormancy in combination with dormancy reduction during dry periods might explain spring germination. It is hypothesised that local adaptations occur through changes mainly in dormancy strength, i.e. how much effort is needed to reduce dormancy. Strong dormancy restricts the part of each seed batch that germinate during autumn, and thus reduces the risk of winter mortality, in Sweden. 相似文献
