采用RT-PCR和RACE技术,从一年生毛竹(Phyllostachys edulis)新鲜叶片中分离到3个捕光色素结合蛋白基因cab-PhE2、cab-PhE3和cab-PhE6,GenBank登记号分别为EF207230、EF405877和EF628209.序列分析表明,3个基因分别与其它植物如水稻、玉米、大麦等PSⅡ的Ihcb2、Ihcb1、Ihcb3基因有着较高的一致性,其编码的蛋白属于大量捕光天线.蛋白结构预测表明,3个基因编码的蛋白均由导肽和成熟蛋白组成,其中成熟蛋白的二级结构均包含4个α螺旋结构、3个类胡萝卜素结合位点,以及相应的叶绿素a、b结合位点.组织特异性表达检测表明,3个基因在叶片、叶鞘和幼茎中均有表达,且在叶片中最高,而在根中未检测到表达.在强光照射( 1500μmol·m-2·s-1)条件下,3个基因的表达量均呈下调趋势,不同基因间存在着一定的差异,其中cab-PhE3和cab-PhE2的表达丰度在光照4h内下降较快,至6 h cab-PhE2接近于零,而cab-PhE6经光照4h表达丰度仍为对照的70%以上,但之后2h后很快降至对照的5%以下.  相似文献

BACKGROUND AND AIMS: Xylem vessels containing gases (embolized) must be refilled with water if they are to resume transport of water through the plant, so refilling is of great importance for the maintenance of water balance in plants. However, the refilling process is poorly understood because of inadequate examination methods. Simultaneous measurements of plant anatomy and vessel refilling are essential to elucidate the mechanisms involved. In the present work, a new technique based on phase-contrast X-ray imaging is presented that visualizes, in vivo and in real time, both xylem anatomy and refilling of embolized vessels. METHODS: With the synchrotron X-ray micro-imaging technique, the refilling of xylem vessels of leaves and a stem of Phyllostachys bambusoides with water is demonstrated under different conditions. The technique employs phase contrast imaging of X-ray beams, which are transformed into visible light and are photographed by a charge coupled device camera. X-ray images were captured consecutively at every 0.5 s with an exposure time of 10 ms. KEY RESULTS: The interface (meniscus) between the water and gas phases in refilling the xylem vessels is displayed. During refilling, the rising menisci in embolized vessels showed repetitive flow, i.e. they temporarily stopped at the end walls of the vessel elements while gas bubbles were removed. The meniscus then passed through the end wall at a faster rate than the speed of flow in the main vessels. In the light, the speed of refilling in a specific vessel was slower than that in the dark, but this rate increased again after repeated periods in darkness. CONCLUSIONS: Real-time, non-destructive X-ray micro-imaging is an important, useful and novel technique to study the relationship between xylem structure and the refilling of embolized vessels in intact plants. It provides new insight into understanding the mechanisms of water transport and the refilling of embolized vessels, which are not understood well.  相似文献

Aim In this study, we evaluate the importance of the mean earlywood vessel size of oaks as a potential proxy for climate in mesic areas. Location The study was conducted in Switzerland at three forest sites dominated by oak (Quercus petraea and Q. pubescens). The three sites were in different climatic zones, varying mainly in terms of precipitation regime. Methods Three 50‐year‐long site chronologies of mean earlywood vessel size and tree‐ring widths were obtained at each site and related to monthly meteorological records in order to identify the main variables controlling growth. The responses of mean vessel size to climate were compared with those of the width variables to evaluate the potential climatic information recorded by the earlywood vessels. Results The results show that the mean vessel size has a different and stronger response to climate than ring‐width variables, although its common signal and year‐to‐year variability are lower. This response is better in particular at mesic sites, where it is linked to precipitation during spring, i.e. at the time of vessel formation, and is probably related to the occurrence of only a few processes controlling vessel growth, whereas radial increment is controlled by multiple and varying factors. Main conclusions The mean earlywood vessel size of oak appears to be a promising proxy for future climate reconstructions of mesic sites, where radial growth is not controlled by a single limiting factor.  相似文献

通过设置临时标准地对龙栖山、武夷山和天宝岩3个国家级自然保护区的3种毛竹混交林群落类型结构进行调查与统计分析.结果表明,在物种数量上从多到少依次为:武夷山国家级自然保护区的毛竹 少叶黄杞(Phyllostachys pubescences Mazel ex H. Delehaie Engelhardtia fenzelii Merr.)混交林(68科125属230种),龙栖山国家级自然保护区的毛竹 南方红豆杉[Taxus chinensis (Pilger) Rehd. var. mairei (Lemee et Lévl.) Cheng et L. K. Fu]混交林(69科135属194种)和天宝岩国家级自然保护区的毛竹 长苞铁杉(Tsuga longibracteata Cheng)混交林(41科71属105种).3个毛竹混交林群落结构分明,毛竹在混交林群落乔木层中占绝对优势的地位.乔木层、灌木层和草本层层次清晰,还具有层间层.毛竹 南方红豆杉群落和毛竹 长苞铁杉群落乔木层可分为3个亚层,而毛竹 少叶黄杞群落乔木层可分为2个亚层.毛竹 少叶黄杞群落的灌木层物种数和草本层盖度均比其他2个群落高.  相似文献

华东天目山脉、黄山山脉、大别山脉竹类植物较为丰富,分别有10属/75种(含种下等级)、8/49、7/30.通过对属、种的分布型分析,前二者竹类植物区系属于典型的华东区系,起源的时间不迟于早第三纪的始新世;而大别山脉虽仍属华东区系的范畴,因渗入了不少华中区系的成分,已具有某些经向过渡性特征,且起源的时间较晚.这3个山脉的竹类植物区系均具有显著的亚热带性质.天目山脉不仅是刚竹属Phyllostachys和短穗竹属Brachystachyum的分化变异中心,也极有可能就是它们的起源中心,但它们自此地起源后的散布方式有所不同,刚竹属在我国亚热带为四周放射状顺序渐进,扩散的范围颇广;短穗竹属存在东西南北4条散布路线,但向各方扩散的距离相差甚远.这些山脉与华东其他地区竹类植物区系联系密切,与华中竹类植物区系也有一定的联系,但与华南、西南等地竹类植物区系联系微弱.从地理位置和区系组成来看,黄山山脉的竹类植物区系来源于天目山脉,大别山脉的竹类植物区系又来源于黄山山脉,前者都是在后者扩展的基础上融合了其他区系的一些成分后发展而成,大别山脉又是北方秦岭以东广大丘陵平原地区竹类植物区系的发祥地.  相似文献

Floral syndromes and pollination of three species of Phyllostachys bamboos were studied in Central China in 1999 and 2000. All were protogynous. Stigmas were receptive and had pollen deposited on them 2 days before anther dehiscence. The period of anthesis in the three bamboos was 3 days. Individual pollen grains of the three species were similar in size (30–40 μm in diameter) and had features typical of wind-pollinated plants. The ratios of pollen to ovules (p/o ratio) in P . nidularia , P . heteroclada and P . nuda were 6500, 12 700 and 33 000, respectively. Mean pollen loads on each flower (one ovule) of these three species were 7.3, 8.8 and 9.4 grains, respectively. Pollen transfer in P . heteroclada and P . nuda depended on wind, and no flower visitors were seen in the field. However, in P . nidularia , Apis cerana Fab. was a frequent pollen collector observed from 1200 to 1330 h. The visits undertaken by thousands of honeybees resulted in a large number of pollen grains being released from the anthers in a short time (10–15 min) in one day, which accelerated and synchronized the release of pollen from the anthers that seemed to enhance the chance of pollination. Given that honeybees played an indirect role in pollen transfer this could partly explain the low p/o ratio in P . nidularia . Reviewing the literature, we found insect visits to flowers of bamboos were not infrequent phenomena. Abstract. Age and size structure of saplings of Picea abies, Pinus sylvestris and Betula pubescens were examined in a 26-yr old forest fire area in a Picea abies-Vaccinium myrtillus forest in northern Sweden. Picea, which is a shade-tolerant species, had its maximum regeneration prior to the shade-intolerant Pinus. The shift from Picea to Pinus regeneration in the late 1970s, did not seem to be related to variations in summer temperature. Instead, it is suggested that Picea established in the shade created by dead trees, and, that increased reindeer browsing of Betula in combination with a simultaneous thinning of the tree layer, favoured Pinus recruitment. These regeneration patterns do not confirm conventional views of post-fire succession in Sweden.  相似文献

Two novel chitin-binding peptides, designated Pp-AMP 1 and Pp-AMP 2, which had antimicrobial activity against pathogenic bacteria and fungi, were purified from Japanese bamboo shoots (Phyllostachys pubescens) by a simple procedure based on chitin affinity chromatography. They had the common structural features of the plant defensin family, but they could not be grouped in any type of that family. They showed a high degree of homology to mistletoe toxins.  相似文献
