目的:探讨琼脂糖凝胶的不同浓度对回收不同大小的酶切后质粒载体纯度的影响。方法:将2种质粒载体酶切,并经不同浓度的琼脂糖凝胶电泳分析,通过比较酶切前和酶切后载体的相对位置,研究胶浓度对回收酶切后载体纯度的影响。结果:不同浓度的琼脂糖凝胶中,酶切前和酶切后载体的相对位置会发生变化,对能否成功回收到纯度高的酶切后DNA片段有重要影响;质粒大小不同,胶浓度的影响也不同。结论:合适的胶浓度对于回收酶切后质粒载体具有重要意义,应选择合适的胶浓度回收酶切后质粒载体。  相似文献

在室内模拟生物净化槽中比较研究了氮磷对污水中藻类叶绿素含量和污水净化的影响。污水中磷含量均为7mg/L左右,氮含量分别为77.4、44.4、24.8和13.5mg/L,结果发现TN/TP=77.4/7.01mg/L组,污水经7d净化,藻类叶绿素含量最高,污水净化效果较好。四个实验组比较,叶绿素含量随TN/TP比例的上升而上升,呈显著正相关。  相似文献

骨代谢始终贯穿于动物的生命过程之中,而机体摄入的钙、磷水平和钙、磷比例又是骨代谢的重要影响因素。钙、磷摄入水平和比例的改变会使甲状旁腺激素(PTH)、降钙素(CT)和1α,25-双羟维生素D3[1,25-(OH)2D3]的水平产生变化,影响RANK/RANKL/OPG系统对骨细胞功能的调控,进而对骨代谢及骨组织形态产生影响。骨唾液酸蛋白(Bone Sialoprotein,BSP)是目前评价成骨细胞分化和骨骼矿化情况的新指标,研究BSP表达水平的变化规律,可以从分子水平解释钙、磷摄入水平和比例对骨组织形态影响的机理。而骨组织形态计量学(Bone Histomorphometry)则是研究钙、磷摄入水平和比例对骨组织形态影响的重要方法。  相似文献

参照美国临床实验室标准化委员会(NCCLS)推荐的《产孢丝状真菌的液基稀释法抗真菌药物敏感性试验方案》(M38-A),测定超临界CO2萃取的柠条锦鸡儿籽油对3种(每种15株)临床常见的皮肤癣菌的最低抑菌浓度(MIC);高效液相色谱法研究柠条锦鸡儿籽油对真菌细胞膜麦角甾醇生物合成的影响以探讨其作用机制。结果表明柠条锦鸡儿籽油对犬小孢子菌、须癣毛癣菌、红色毛癣菌的MIC范围分别为64~512μg/mL、32~512μg/mL、64~1024μg/mL;高效液相色谱法检测显示,与其生长对照菌相比,籽油作用的皮肤癣菌细胞膜上麦角甾醇含量明显降低,且具有剂量依赖性。柠条锦鸡儿籽油主要是通过影响真菌细胞膜上麦角甾醇的合成而发挥抗真菌作用。  相似文献

以三角褐指藻(Phaeodactylum tricornutum)为研究材料,设置了5个磷营养限制处理:磷营养分量分别设为f/2培养基的1/20(P1)、1/10(P2)、1/8(P3)、1/4(P4)、1/2(P5),以f/2为对照(Pck),在磷限制胁迫下培养10d,然后均以相同密度(2.5×105cells·mL-1)接种在f/2条件下恢复培养16d,测定了三角褐指藻在磷限制胁迫下和恢复培养阶段的生长状况。结果表明,三角褐指藻在受到磷限制胁迫后,细胞密度、蛋白质和可溶性糖含量都显著低于对照(p<0.05);恢复阶段,P1、P2和P3处理组的细胞密度、平均相对生长率及生物量在恢复培养的中前期显著高于对照(p<0.05),最高平均相对生长率分别为0.73、0.70、0.68d-1,显著高于对照(0.55d-1)(p<0.05);P4处理组的细胞密度和生物量与对照无显著差异;P5处理组的细胞密度和生物量在恢复培养的中前期显著低于对照(p<0.05);随着培养时间的推移,各处理组与对照之间的差异逐渐缩小,处理组和对照的细胞密度、生物量、蛋白质和可溶性糖含量等均无显著差异。  相似文献

To provide a reference for wetlands elsewhere we analysed soil nutrients and the vegetation of floodplains and fens in the relatively undisturbed Biebrza-valley, Poland. Additionally, by studying sites along a water-table gradient, and by comparing pairs of mown and unmown sites, we aimed with exploring long-term effects of drainage and annual hay-removal on nutrient availabilities and vegetation response. In undrained fens and floodplains, N mineralization went slowly (0–30 kg N ha−1 year−1) but it increased strongly with decreasing water table (up to 120 kg N ha−1 year−1). Soil N, P and K pools were small in the undisturbed mires. Drainage had caused a shift from fen to meadow species and the disappearance of bryophytes. Biomass of vascular plants increased with increasing N mineralization and soil P. Annual hay-removal tended to have reduced N mineralization and soil K pools, but it had increased soil P. Moreover, N concentrations in vascular plants were not affected, but P and K concentrations and therefore N:P and N:K ratios tended to be changed. Annual hay-removal had induced a shift from P to K limitation in the severely drained fen, and from P to N limitation in the floodplain. The low nutrient availabilities and productivity of the undisturbed Biebrza mires illustrate the vulnerability of such mires to eutrophication in Poland and elsewhere. In nutrient-enriched areas, hay removal may prevent productivity increase of the vegetation, but also may severely alter N:P:K stoichiometry, induce K-limitation at drained sites, and alter vegetation structure and composition.  相似文献

The surfactin can inhibit proliferation and induce apoptosis in cancer cells. Moreover, surfactin can induce cell death in human breast cancer MCF-7 cells through mitochondrial pathway. However, the molecular mechanism involved in this pathway remains to be elucidated. Here, the reactive oxygen species (ROS) and Ca2+ on mitochondria permeability transition pore (MPTP) activity, and MCF-7 cell apoptosis which induced by surfactin were investigated. It is found that surfactin evoked mitochondrial ROS generation, and the surfactin-induced cell death was prevented by N-acetylcysteine (NAC, an inhibitor of ROS). An increasing cytoplasmic Ca2+ concentration was detected in surfactin-induced MCF-7 apoptosis, which was inhibited by 1,2-bis (2-aminophenoxy) ethane-N,N,N′,N′-tetraacetic acid (BAPTA-AM, a chelator of calcium). In addition, the relationship between ROS generation and the increase of cytoplasm Ca2+ was determined. The results showed that surfactin initially induced the ROS formation, leading to the MPTP opening accompanied with the collapse of mitochondrial membrane potential (ΔΨm). Then the cytoplasmic Ca2+ concentration increased in virtue of the changes of mitochondrial permeability, which was prevented by BAPTA-AM. Besides, cytochrome c (cyt c) was released from mitochondria to cytoplasm through the MPTP and activated caspase-9, eventually induced apoptosis. In summary, surfactin has notable anti-tumor effect on MCF-7 cells, however, there was no obvious cytotoxicity on normal cells.  相似文献

The contribution of sediment release to the phosphorus budget of hypereutrophic Onondaga Lake was determined through laboratory measurements made on intact cores. Rates ranged from 9–21 mg P m–2 d–1 with a mean of 13 mg P m–2 d–1, values similar to those observed in other lakes of comparable trophic state. There was no statistically significant trend in rates in time (July versus September) or in space (location along the major N/S axis of the lake). Rates of sediment phosphorus release measured in the laboratory compared favorably with the observed rate of soluble reactive phosphorus accumulation in the lake's hypolimnion. The sediments are the second largest source of phosphorus for Onondaga Lake, contributing 24% of the overall phosphorus load to the system.  相似文献

The simultaneous effect of oxygen availability and carbon source concentration on yeast lipid and carotenoid production has never been studied before. In this work, a Doehlert distribution design was used to study the simultaneous effect of carbon concentration and oxygen availability on Rhodosporidium toruloides NCYC 921 carotenoid and lipid production. A cheap industrial byproduct was used as carbon source (carob pulp syrup). A total sugar concentration of 106.3 g/L and a medium volume of 0.120 L induced the highest total carotenoid and total fatty acid productivities (4.60 μg/Lh and 0.029 g/Lh, respectively). Flow cytometry was used to assess yeast stress response under different cultivation conditions. The highest proportion of cells with permeabilised membrane (>20%) was induced when the cultivations were carried out at the highest sugar concentration studied (130.0 g/L) or when the culture reached the minimum final medium pH (4.60). The results showed that the total sugar concentration had a positive influence on the yeast biomass and carotenoid content, while the oxygen availability had little influence on the biomass concentration, but had a slight positive influence on the carotenoid content.
