Homogenates or crude 300-16 000 g membrane fractions from Sinapis alba roots catalysed esterification of [4-14C]cholesterol with utilization of endogenous acyl sources. With acetone powder preparations cholesterol esterification was distinctly stimulated by a neutral lipid fraction isolated from S. alba roots. Among neutral lipids triacylglycerols were the most active in this process. Experiments with various acyl-labelled acylglycerols as acyl donors and non-labelled sterols as acceptors confirmed that triacylglycerols are directly utilized as the source of fatty acids for sterol esterification. Di- and mono-acylglycerols were much less effective.  相似文献

The effect of ouabain on the antiviral and anticellular activities of interferon and its components was tested in RSb cells. These cells were found to be inhibited in growth by both components. Ouabain, known to block the antiviral effect of interferon without affecting the growth inhibitory effect, depressed both effects of the antiviral component. The effect of the growth inhibitory component was unaltered by the drug. The results support earlier suggestions that two different mechanisms may be involved in the growth regulation by interferon.  相似文献

Evidence is presented for the ability of mouse epidermis to oxidize retinal by the action of a cytosolic enzyme with a pH optimum of about 8.5. The enzyme activity was slightly enhanced by the addition of either NAD or FAD. The reaction exhibited Michaelis-Menton kinetics with a km of 1.627 × 10?4M for retinal. Mouse epidermal cytosol was also capable of reducing retinal. This enzyme had an optimum pH of 6.0 and the activity of the undialyzed enzyme was enhanced 5- to 7-fold by the addition of reduced NAD or NADP. Possible roles of these enzymic activities in epidermis are discussed.  相似文献

Previous kinetic, isotopic studies have suggested that ‘insoluble’ phenolic esters may be precursors of lignin. Heretofore, the ‘insoluble’ esters have been detected by the chromatographic examinations of gross hydrolysis products of ethanol-insoluble resides and/or acetone powders. We have developed new methods for the isolation and purification of certain of the ethanol-insoluble, phenolic esters of Mentha arvensis. ‘Insoluble’ conjugates of caffeic, ferulic and p-coumaric acids were purified and were shown to be electro-phoretically and chromatographically homogeneous. These compounds were distinguished on the basis of their anionic mobility at pH 1·9. A second pool of caffeic acid was associated with a high MW fraction. Two acylated anthocyanins containing p-coumaric acid and caffeic acid were also obtained from acetone powders.  相似文献

The following sterols were found in the roots, stems, leaves, unripe and ripe fruits of Solanum dulcamara: cholesterol, sitosterol, stigmasterol, campesterol, brassicasterol, isofucosterol and 24-methylenecholesterol. The most abundant components are cholesterol, sitosterol and stigmasterol (77–84%). In all parts of the plant the sterols are present in the free form and as esters, glycosides and acylated glycosides. The total sterol content and the content of combined forms were determined photometrically. In the leaves 58% of the sterols were found in the form of glycoside (26%), acylated glycoside (29%) and ester (2%). In the roots only 25% of the sterol were found in combined form. In the other organs the ratio of free and combined sterols was intermediate. In all cases, the ester fraction was the least.  相似文献

Datura meteloides; plants were fed with tiglic acid-[-14C] via the roots and after 2 days the percentage incorporation into the alkaloids 3α-tigloyloxytropane, 3α,6β-ditigloyloxytropane, meteloidine and 3α,6β-ditigloyloxytropan-7β-ol were 15·2, 1·82, 2·2 and 1·8 respectively. 3α,6β-Ditigloyloxytropane was partially hydrolysed to 6β-hydroxy-3α-tigloyloxytropane which contained 58·1% of the radioactivity of the original base, whereas 3α,6β-ditigloyloxytropan-7β-ol gave meteloidine containing only 9·2% of the original activity. The results suggest that the di- and tri-hydroxytropanes may be formed by different routes.  相似文献

The effects of 12-O-tetradecanoylphorbol 13-acetate (TPA), a potent activator of protein kinase C, on high-affinity Na(+)-dependent glutamate transport were investigated in primary cultures of neurons and glial cells from rat brain cortex. Incubation of glial cells with TPA led to concentration- and time-dependent increases in the glutamate transport that could be completely suppressed by the addition of the protein kinase C (PKC) inhibitor 1-(5-isoquinolinylsulfonyl)-2-methylpiperazine. The TPA effects could be mimicked by oleoylacetylglycerol and by the diacylglycerol kinase inhibitor R59022. The effects of TPA were potentiated by the Ca2+ ionophore A23187. Under the chosen experimental conditions TPA had no effect on glutamate transport in neurons. We conclude that PKC activates the sodium-dependent high-affinity glutamate transport in glial cells and that it has dissimilar effects on neurons and glial cells.  相似文献

Fibroblasts are one of several cell types producing nerve growth factor (NGF) in neuronal targets. In previous studies we found that NGF production is up-regulated by 12-O-tetradecanoylphorbol 13-acetate (TPA) and serum, down-regulated by corticosterone, and unaffected by dibutyryl-cyclic AMP (db-cyclic AMP) in fibroblasts. As fibroblasts in vivo are likely to be exposed to regulatory effects by more than one of these agents at any given time, we examined the effects of combinations of them on NGF production using L929 fibroblasts as a model system. TPA and serum together stimulated NGF production 10-fold more than either agent alone. Corticosterone reduced NGF mRNA and NGF production to less than 10% of basal levels whether or not TPA or serum, or both, were present but not in the presence of the glucocorticoid antagonist RU486. Corticosterone did not increase the rate of NGF mRNA degradation. Forskolin and db-cyclic AMP prevented NGF mRNA induction by TPA and serum without changing basal levels. TPA induced c-fos and junB mRNAs transiently and preceding NGF mRNA induction but c-jun mRNA remained undetectable. Forskolin enhanced the induction of both junB and c-fos mRNA whereas corticosterone prolonged junB mRNA induction. Thus, TPA induction of NGF mRNA is modulated differentially by corticosterone and cyclic AMP. c-fos and junB may play a role in the underlying mechanisms.  相似文献

Two new sesquiterpene polyol esters with β-dihydroagarofuran skeleton were isolated from the root bark of Celastrus rosthornianus. Their structures were elucidated, mainly on the basis of spectral analyses, as 1 β-acetoxy-8β,9-dibenzoyloxy-6-hydroxy-2β(-methylbutanoyloxy)-β-dihydroagarofuran and 1β-acetoxy-9-benzoyloxy-8β-(β-furanocarbonyloxy)-6-hydroxy-2β(-methylbutanoyloxy)-β-dihydroagarofuran. The complete assignments of 13C NMR chemical shifts for both compounds on the basis of 1H-13C chemical-shift correlation spectrum were also carried out.  相似文献
