目的:检测脊神经切断大鼠背根节(DRG)神经元重复放电能力和钠电流的变化,并研究介导其电流变化的钠通道亚型的表达情况。方法:脊神经切断术后2~8d慢性痛大鼠模型背根节急性分离,对中等直径DRG神经元运用全细胞膜片钳技术记录神经元放电和钠电流的变化。对背根节神经元进行RT-PCR检测,分析其钠通道亚型的表达情况。结果:电流钳下,实验组DRG神经元在电流刺激下产生重复放电,而对照组神经元多诱发单个动作电位,电压钳记录发现实验组背根节神经元快钠电流和持续性钠电流幅值均明显大于对照组,PCR结果显示,Nav1.3、Nav1.7和Nav1.8通道亚型mRNA表达显著增高。结论:钠通道介导了脊神经受损模型的DRG神经元兴奋性增高,持续性钠电流可能通过调节阈下膜电位振荡的产生调节神经元兴奋性。  相似文献

Keratins modulate colonocyte electrolyte transport via protein mistargeting   总被引:6,自引:0,他引:6  
The function of intestinal keratins is unknown, although keratin 8 (K8)-null mice develop colitis, hyperplasia, diarrhea, and mistarget jejunal apical markers. We quantified the diarrhea in K8-null stool and examined its physiologic basis. Isolated crypt-units from K8-null and wild-type mice have similar viability. K8-null distal colon has normal tight junction permeability and paracellular transport but shows decreased short circuit current and net Na absorption associated with net Cl secretion, blunted intracellular Cl/HCO3-dependent pH regulation, hyperproliferation and enlarged goblet cells, partial loss of the membrane-proximal markers H,K-ATPase-beta and F-actin, increased and redistributed basolateral anion exchanger AE1/2 protein, and redistributed Na-transporter ENaC-gamma. Diarrhea and protein mistargeting are observed 1-2 d after birth while hyperproliferation/inflammation occurs later. The AE1/2 changes and altered intracellular pH regulation likely account, at least in part, for the ion transport defects and hyperproliferation. Therefore, colonic keratins have a novel function in regulating electrolyte transport, likely by targeting ion transporters to their cellular compartments.  相似文献

2009年全国其他感染性腹泻报告病例分析   总被引:2,自引:0,他引:2  
目的了解我国2009年其他感染性腹泻的流行特点和病原学信息,提出改善建议,为更好的控制其他感染性腹泻病的流行提供数据支持。方法收集中国疾病控制信息系统"疾病监测信息报告管理系统"中2009年其他感染性腹泻的病例报告,对其流行病学及病原学信息进行描述性统计分析。结果 2009年全国共报告655 965例其他感染性腹泻病例,其中≤5岁儿童分别占报告总病例的50.38%和死亡病例的70.0%。实验室确诊病例占报告病例数的5.04%,其中病毒性感染占92.79%,细菌性感染占6.92%。结论≤5岁的散居儿童应为其他感染性腹泻的重点监测人群。其他感染性腹泻报告病例中约有95%为临床诊断病例。我国其他感染性腹泻的病原学诊断率及病原学诊断结果的报告率亟待提高。  相似文献

As an important in vivo antioxidant, vitamin C is commonly used clinically to alleviate hypoxia-induced heart symptoms. To approach the protective mechanisms of vitamin C on hearts during hypoxia, we investigated the electrophysiological effects of vitamin C (1 mM, pretreated before hypoxia) on Na+ currents (including transient and persistent Na+ currents) in guinea pig ventricular myocytes during hypoxia by the whole-cell and single-channel patch-clamp techniques. Whole-cell recordings showed that the mean current density of I NaT in the hypoxia group decreased from the control value of 40.2142 ± 1.7735 to 27.1663 ± 1.8441 pA/pF and current density of I NaP increased from 0.3987 ± 0.0474 to 1.1854 ± 01994 pA/pF (n = 9, P < 0.05 vs. control) at 15 min. However, when vitamin C was administered before hypoxia as pretreatment, I NaT and I NaP varied moderately (mean current density of I NaT decreasing from 41.6038 ± 2.9762 to 34.6341 ± 1.9651 pA/pF and current density of I NaP increasing from 0.3843 ± 0.0636 to 0.6734 ± 0.1057 pA/pF; n = 9, P < 0.05 vs. hypoxia group). Single-channel recordings (cell-patched) showed that the mean open probability and open time of I NaP increased significantly in both groups at hypoxia 15 min. However, the increased current values of the hypoxia group were still marked at hypoxia 15 min (n = 9, P < 0.05 vs. vitamin C + hypoxia group). Our results indicate that vitamin C can attenuate the disturbed effects of hypoxia on Na+ currents (I NaT and I NaP) of cardiac myocytes in guinea pigs effectively.  相似文献

Na+-channel activity recorded in cell-attached patches from entorhinal cortex neurons in the absence of gating-modifying drugs was examined to determine the possible occurrence of substate openings. Brief sojourns to subconductance levels were occasionally observed within prolonged (“persistent”) burst openings. Subconductance occurrence and amplitude were determined following two distinct, complementary approaches: (1) direct visual inspection and (2) automated detection by application of a method that exploits the current variance of fixed-width tracing segments to sort amplitude estimations. The two approaches led to comparable results. At least six subconductance levels in addition to the full open state were revealed, with amplitudes that were approximately 20%, 30%, 40%, 50%, 60% and 75% that of full openings. The global probability of subconductance opening occurrence within a burst as well as the probability of observing one particular subconductance level within a burst showed no clear dependence upon membrane potential in the −40 to +10 mV range. Open- and closed-time distributions of substate openings could either be similar to those observed in burst full openings or show distinct patterns. Low-amplitude late openings were also observed in isolation, separately from full-size openings. These openings corresponded to conductance levels very similar to those of the substates observed within full-size burst openings; therefore, they were interpreted as isolated subconductance openings. Early, transient openings responsible for the fast-inactivating whole-cell Na+-current component also manifested distinct conductance levels, the two most prominent of which were in an approximate 75:100 amplitude ratio. Interestingly, the 75% conductance level observed among early openings occurred much more frequently than in “persistent” burst openings. We conclude that pharmacologically untreated Na+ channels from native neurons generate substate openings that may influence differently the multiple gating modes displayed by these channels. The BB cell line derived from the brain tissue of a barramundi (Lates calcarifer) that survived nervous necrosis virus (NNV) infection is persistently infected with NNV. To elucidate whether interferon (IFN) plays a role in the mechanism of NNV-persistent infection in BB cell line, a virus-negative control cell line was obtained by treating BB cells with NNV-specific rabbit antiserum for 5 subcultures. After the treatment, NNV titer or RNA or capsid protein was no longer detected in the cured BB (cBB) cells. Expression of Mx gene, encoding a type I IFN-inducible antiviral protein, was found in BB cells and cBB cells following NNV infection, but not in NNV-free cBB cells. Moreover, expression of Mx gene and antiviral activity against NNV were induced in cBB cells by the treatment with MAb-neutralized BB cell supernatant. Furthermore, NNV persistent infection was induced again in cBB cell culture if multiplicity of infection (MOI) was low (< or = 1). These experimental results indicated that IFN-like cytokines existed in the culture supernatant of BB cells, and IFN-induced response played an important role in protecting the majority of cells from virus lytic infection and regulating NNV persistence in the BB cell line.  相似文献

Escherichia coli (E. coli) that produces adhesin F18 is the main pathogen responsible for porcine post-weaning diarrhea and edema disease. The receptor for E. coli F18 has not been described in pigs, however the alpha (1,2)-fucosyltransferase (FUT1) gene on chromosome 6 has been proposed as a candidate. The objective of this study, therefore, was to investigate the relationship between FUT1 gene expression and E. coli F18 receptor in Sutai pigs of different ages (8-, 18-, 30- and 35-day-old). FUT1 gene expression was detected in 11 pig tissues with the highest level in lung, and expressed consistently at the four time points. In most tissues, FUT1 gene expression levels decreased from days 8 to 18, then continually increased on days 30 and 35, with expression around weaning time higher than that on day 8. Gene ontology and pathway analysis showed that FUT1 was involved in 32 biological processes, mainly those integral to the membrane, or involved in glycosylation, as well as regulation of binding, interestingly participating in three pathways related to glycosphingolipid biosynthesis. From this analysis and the high linkage disequilibrium between the FUT1 gene and the E. coli F18 receptor locus, we can speculate that higher expression of the FUT1 gene in small intestine is beneficial to the formation of receptors to the E. coli F18 strain and is related to the sensitivity to the pathogen.  相似文献

目的对婴幼儿腹泻A组轮状病毒的三种不同检测方法进行评价。方法同时采用胶体金、琼脂糖凝胶电泳、逆转录PCR(RT-PCR)对浙江萧山医院1周320份腹泻患儿的标本进行轮状病毒检测,然后进行方法学比较。结果三种方法的阳性检出率分别为36.3%、48.1%和42.8%,经卡方检验三种方法的检出率之间差异均存在统计学意义(P〈0.05);以琼脂糖凝胶电泳为检测的最终标准,RT-PCR的敏感性为88.9%,特异性达100%;胶体金法的敏感性71.4%,特异性达96.4%。结论胶体金法检测轮状病毒的敏感性和特异性能够满足临床上对轮状病毒快速筛检的要求;PT-PCR法的敏感性和特异性均最高,既可用于临床检测,又是轮状病毒血清G、基因P分型的重要手段,在对无菌体液和环境样本中轮状病毒的检测具有不可比拟的优势。  相似文献
