Changes in rectal temperature were measured after the intracerebral microinjection of neurotensin (2.5 μg/0.5 μl) at 135 sites in the rat. At 63 of the 135 microinjection sites, the tridecapeptide produced a rapid onset of hypothermia ranging in magnitude from 0.8 to 2.3°C below the baseline rectal temperature. The drop in rectal temperature persisted for 2–4 hours following the microinjection. The greatest concentrations of neurotensin-sensitive sites were found in the medial preoptic region of the hypothalamus and in the periaqueductal gray area, both of which contain relatively large amounts of endogenous neurotensin. Other active sites were found in the ventral thalamus, the dorsomedial hypothalamus, and in the diagonal band of Broca. At no injection site did neurotensin evoke an increase in rectal temperature. These data support the proposition that neurotensin may act endogenously to mediate heat-loss mechanisms in the rat. The data provide further evidence indicating a potent neuromodulatory role for neurotensin.  相似文献

For researchers studying mysticere whales few methods for determining gender or for collecting biochemical and genetic information from unrestrained animals are available. The objective of this study was to develop a reliable method for collecting viable tissue samples for establishing continuous cell cultures from skin biopsies of free-ranging whales. A method to collect and process these samples is presented. Six of seven skin biopsies from gray whales were established in cell culture. Our results suggest that the viability of the samples is improved by (1) sterile processing in the field, (2) minimizing the time between collection and delivery to the cell culture facility, (3) reducing the concentration of antifungal agent, and (4) placing tissue explants under a coverslip. While the results reported in this paper are based on a small sample size, we believe that if the procedures are followed, they will increase the probability of successfully culturing cetacean tissue. Established cell lines can supply replenishable material from identified whales still living in the wild. These cultures can then be used for determination of sex from karyotypes, and for assessing genetic relationships of cetaceans from inherited protein, chromosomal and DNA polymorphisms. These much needed analytical tools can be used to determine familial and populational relationships, leading to a better understanding of mating systems, stock identification and effective population sizes of wild cetaceans.  相似文献

家兔用乌拉坦(700mg/kg)和氯醛糖(35mg/kg)静脉麻醉,用三碘季铵酚制动,在人工呼吸下进行实验。用电刺激下丘脑近中线区的方法诱发室性期前收缩(HVE)。中脑中央灰质(CG)内微量注射去甲肾上腺素(NA,4μg/2μl)对 HVE 有易化效应,使 HVE 次数增多。微量注射β-肾上腺素能受体阻断剂心得宁(2μg/2μl)使 HVE 次数明显减少,而α-肾上腺素能受体阻断剂酚妥拉明(2μg/2μl)则对 HVE 次数无明显减少作用。事先在 CG 内微量注射心得宁也可阻断 CG 内注入 NA 对 HVE 的易化效应。在 CG 内注射 NA 后,再在 CG 内微量注射吗啡,后者对 HVE 的抑制效应仍然存在。但在 GG 内注射 NA 可减少或消除刺激腓深神经对 HVE 的抑制作用。上述结果提示,在 CG 内β-受体的激活可增加 HVE 次数。刺激腓深神经对 HVE 的抑制作用可能部份是通过内源性吗啡样物质抑制 CG 内 NA 的释放而实现的。  相似文献

在大鼠中脑导水管周围灰质(PAG)注射吗啡10μg 可引起明显的镇痛作用,并有部位特异性。在脊髓蛛网膜下腔注射抗甲啡肽 IgG20μg 或抗强啡肽 IgG20μg,均能大部分对抗 PAG内注射吗啡引起的镇痛作用,这提示甲啡肽和强啡肽参与自 PAG 到脊随的下行抑制作用。本实验利用蛋白质 A-琼脂糖 CL-4B 亲和层析柱从血清中纯化 IgG,这种方法简单,提纯速度快,且 IgG 纯度高,为中枢微量注射抗体研究神经肽的生理功能提供了方便。  相似文献

Two pairs of harbor ( Phoca vitulina ) and three pairs of gray ( Halicboeruls grypus ) seals were exposed to one of three human handlers for 15 min, twice a day, for a total of six sessions. Following habituation to the familiar handler, animals were then exposed to a novel human for 7 min, and then retested for 7 min with the familiar human. In all cases, animals responded to the unfamiliar human with increased vigilant behavior, i. e., they spent more time oriented towards the unfamiliar handler during the first 2 min of the test session than during the same interval of either the final habituation session or the retest with the familiar human ( P = 0.03 in all cases). There was also a tendency for seals to contact the familiar handler with their noses more rapidly than the novel human ( P = 0.06). These results support the hypothesis that phocid seals are capable of discriminating between individual humans in their environment, setting the stage for human-based Pavlovian conditioning.  相似文献

There has been extensive recent interest in the concepts of behavioral types, behavioral syndromes, and personalities in nonhuman animal species. Evidence for behavioral types now exists from a wide range of taxa, from mollusks to mammals. However, marine mammals are poorly represented in this literature. Here, we describe an in‐field experimental test of behavioral types in breeding gray seals, using a remotely controlled vehicle to deliver a standardized test stimulus to target individuals. We report on the design and implementation of this test and on the behavioral responses of individuals. Analysis of behavioral responses from both males and females revealed consistent individual differences across tests, suggesting that this is a practical and viable technique for determining individual variation in behavioral type in the field. Despite extensive literature on behavioral types, studies of behavioral types in wild populations remain rare. It is, therefore, important to develop ways to identify and quantify the existence of behavioral types in natural populations, because only by doing this, can we hope to ascertain the ecological and evolutionary relevance of behavioral types.  相似文献

Western gray whales ( Eschrichtius robustus ) are critically endangered and anthropogenic threats, such as entanglement in fishing gear and collisions with vessels, may be acting to limit recovery of the population. Thus, examining the magnitude of such anthropogenic interactions using a scar-based approach is warranted. A multi-year (1995–2005) photo-identification study of western gray whales on their feeding ground off northeastern Sakhalin Island, Russia, has resulted in a large data set of digital and film images of 150 individuals. These images were reviewed and scored for anthropogenic scarring by recording the presence of visible scars resulting from fishing gear entanglement and vessel collisions in 21 defined body regions. In total, 20.0% ( n = 30) of whales identified during the study period had detectable anthropogenic scarring, with 18.7% ( n = 28) determined to have been previously entangled in fishing gear at least once and 2.0% ( n = 3) to have survived at least one vessel collision. These estimates are likely to be conservative given the nature of the photo-identification data set, but indicate that male and female western gray whales are subject to anthropogenic interactions. Future studies designed to systematically estimate the frequency and rates of anthropogenic events are needed and would have direct conservation and management implications.  相似文献

We conducted shore-based sighting surveys to estimate the number of northbound migrating gray whale calves passing Piedras Blancas, California, for seven consecutive years (1994–2000). In addition, we conducted aerial surveys to determine offshore distribution of the migration in 1994 and 1995, measured day/night migration rates with thermal sensors in 1994–1996, and maintained concurrent replicate watches near the peak of each migration to estimate the proportion of the cow/calf pairs missed by the standard watch team. During good weather, we counted 325, 194, 407, 501, 440, 141, and 96 calves during 1994–2000, respectively. Correcting these counts for periods not on watch and for calves missed, produced final estimates of 945 calves (SE = 68.21) for 1994, 619 calves (SE = 67.19) for 1995, 1,146 calves (SE = 70.67) for 1996, 1,431 calves (SE = 82.02) for 1997, 1,388 calves (SE = 91.84) for 1998, 427 calves (SE = 41.10) for 1999, and 279 calves (SE = 34.79) for 2000. Calf production indices (calf estimate/total population estimate) are 4.2%, 2.7%, 4.8%, 5.8%, 5.5%, 1.7%, and 1.1% for the years 1994–2000, respectively. Fluctuations in calf production over this time period were positively correlated with the length of time that primary feeding habitat was free of seasonal ice during the previous year.  相似文献

通过平板抑菌试验和孢子萌发试验研究了芳樟醇对灰葡萄孢的生长抑制作用,并通过盆栽试验进一步验证了芳樟醇对番茄灰霉病的防控效果。结果表明: 芳樟醇能够显著抑制灰葡萄孢菌丝的生长,半最大效应浓度(EC50)值为0.581 mL·L-1。孢子萌发试验中,芳樟醇能够有效抑制灰葡萄孢孢子的萌发,并表现出浓度依赖性。芳樟醇处理提高了灰葡萄孢菌菌丝体的相对电导率和丙二醛(MDA)含量,说明芳樟醇可引起氧化损伤效应导致灰葡萄孢菌的膜系统被破坏;芳樟醇处理后灰葡萄孢菌中的超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)活性较对照组分别下降了27.4%、68.9%和26.0%,说明芳樟醇抑制了灰葡萄孢菌体内的抗氧化系统。盆栽试验结果显示,芳樟醇处理的病斑直径较对照组显著降低;番茄叶片中的SOD、CAT、POD、多酚氧化酶和苯丙氨酸解氨酶活性均显著高于对照组;而MDA含量下降了41.5%,说明芳樟醇可减轻灰葡萄孢菌对番茄植株造成的氧化损伤以提高植物抗病性。综上,芳樟醇对灰葡萄孢的生长有显著抑制作用并对番茄灰霉病有较好的防治效果,研究结果可为开发新型植物源抑菌剂防控番茄灰霉病提供理论依据。  相似文献
