Short-term metabolic and concomitant morphologic effects of streptozotocin diabetes on isoproterenol-induced myocardial infarction
was studied in Wistar rats, Of particular significance was the observation that myocardial infarction in concert with diabetes
brought about a distinctive exacerbation of the severity and complexity of the histopathological lesions. Of all the biochemical
parameters, serum glucose and free fatty acids registered maximum elevation and serum lactate and cardiac glycogen levels
a maximum reduction. Among the lipoproteins, an inverse relationship was found between high density lipoproteins and low density
and very low density lipoproteins; while high density lipoproteins, ratio of high density lipoprotein to low density lipoprotein
and the percentage of high density lipoprotein were decreased, there was a significant increase in low density lipoprotein
concentration and percentage values of low density and very low density lipoproteins. In diabetes, the B cell of the endocrine
pancreas depicted selective necrosis. Loss of insulin granules and wide-spread necrobiosis of cellular elements of the pancreatic
islets were observed, respectively, in myocardial infarction and in diabetes plus myocardial infarction combinations. Pathological
evidence of chemical-induced mild toxicity was present in the exocrine parenchyma. Mitotic features and the presence of centroacinar
cells in the damaged Langerhans’ islets supposedly formed the basis of regeneration of the tissue in diabetes, with or without

X-ray fluorescence spectrometry and atomic absorption spectrometry were used in a quantitative study of zinc, copper, and
magnesium in 71 postmortal human hearts. Samples were obtained from individuals who had demonstrated no previous clinical
or subsequent pathological findings of myocardial infarction and from victims of a recent or an old infarction. A significant
difference (p<0.001) in the elemental levels was observed between the noninfarct and the recent infarct groups. The noninfarct group had
higher cardiac levels of all three elements. However, the difference in elemental concentrations between the noninfarct and
the old-infarct groups was not significant. Cardiac levels of zinc (p<0.001) and copper (p<0.01) were significantly greater in the old-infarct group than in the recent-infarct group. Magnesium levels were higher
in the recent-and-old-infarct group than in the recent infarct group (p<0.01). It is possible that the elements are redistributed during myocardial infarction, and that uptake of these elements

The effect of chronic administration of -guanidinopropionic acid (GPA) on the protein profiling, energy metabolism and right ventricular (RV) function was studied in the rat heart during the weaning and adolescence period. GPA was given in tap water (1–1.5%) using pair drink controls. The feeding of animals with GPA solution for a six week period resulted in elevation of heart to body weight ratio due to body growth retardation. GPA accumulated in the myocardium up to 67.37 ± 5.3 moles.g dry weight and the tissue content of total creatine, phosphocreatine and ATP was significantly decreased to 15%, 9% and 65% of control values respectively. Total activity of creatine kinase (CK) was not changed, but the proportion of mitochondrial (Mi) CK isoenzyme was decreased; the percentage of MB isoenzyme of CK was significantly higher. GPA treatment resulted in an elevation of the content of cardiac collagenous proteins and decrease of non-collagenous proteins in the heart; in parallel, a decrease of the collagen I to collagen III ratio was detected. The function of the RV was assessed using an isolated perfused heart with RV performing pressure-volume work. As compared to pair-drink controls, RV function was significantly impaired the GPA group: at any given right atrial filling pressure, the RV systolic pressure and the rate of pressure development were decreased by almost a factor of two. Elevation of the RV diastolic pressure with increasing pulmonary artery diastolic pressure was also significantly steeper in the GPA group which also showed decrease of cardiac output, especially at high outflow resistance. It may be assumed that chronic administration of GPA deeply influenced metabolic parameters, protein profiles and contractile function of the developing heart. On the other hand, concentrations of glucose, total lipids and triglycerides in blood plasma were not affected. All these data confirm the concept that the CK system is of central importance both for heart function and for the regulation of normal growth of cardiac myocytes. 相似文献

Guanine nucleotide-binding regulatory proteins (G proteins) play a major role in the regulation of a number of physiological processes, such as stimulation or Inhibition of adenylate cyclase activity or gaiting of ionic channels. Myocardial ischemia could induce the changes in receptor-G protein signal transduction system in the heart. Therefore, this article will focus on the role and alterations of G proteins (especially, Gs and Gi) in myocardial ischemia. The Gi protein rapidly loses functional activity during very early myocardial ischemia. In contrast to Gi protein, the function of Gs protein during this phase has not been evaluated. Moreover, the changes in Gs protein after 30 min of ischemia are contradictory. However, the sensitization of the adenylate cyclase activity in the very early phase of acute ischemia is gradually replaced by a decrease in adenylate cyclase activity with prolonged ischemia. The decrease in the function and amount of Gs protein may be one of the factors that induce these changes. The function of Gs protein was also decreased in the canine hearts with ischemia and reperfusion. In contrast to ischemia and reperfusion, there are no significant alterations in G proteins and modulation of adenylate cyclase in the stunned myocardium. It has become increasingly evident that Gi protein may play an important role in the cardioprotective effects of preconditioning. When -adrenoceptor densities are reduced in chronic myocardial ischemia, decreased in the amount and function of Gi protein and increased amount of Gs protein may play the role in preservation of the adenylate cyclase activity. This study was carried out to assess manganese (Mn) status after an acute episode of myocardial infarction. Plasma and erythrocyte
Mn concentrations were measured from admission to hospital to day 15 postadmission in 21 patients suffering from acute myocardial
infarction and in three control groups. The determination of Mn in these biological fluids was performed by electrothermal
atomic absorption spectrometry. Plasma Mn was higher (p<0.01) and erythrocyte Mn was similar in the acute myocardial infarction group compared to healthy age-matched control group.
Plasma and erythrocyte Mn remained unchanged during the 2 wk after acute myocardial infarction and were not correlated to
enzyme activities. A decrease of erythrocyte Mn with age, expressed in nmol/L, was noted (p<0.02). These results suggest that plasma and erythrocyte Mn do not provide an indication of myocardial damage. Nonetheless,

腹腔注射链脲佐霉素(65mg/kg)诱发Wistar大鼠糖尿病。糖尿病发病4周后,向饲料中加尼群地平(30mg·kg-1·d-1)。结果表明,糖尿病4周时大鼠心室舒张功能首先受损,8周后心室舒张和收缩功能均明显受累。尼群地平处理对糖尿病大鼠的心肌收缩性有一定的改善作用。提示尼群地平对大鼠糖尿病性心肌病有一定有益作用。 相似文献

摘要 目的:探讨丹参素注射液对急性心肌梗死大鼠的心室重构、心室功能及肢体导联与胸导联心电图参数的影响。方法:选择SD大鼠40只,将其鼠随机模型组、假手术组、硝酸甘油组、丹参注射液组。假手术组大鼠给予只在冠状动脉处穿针,不进行结扎,其余步骤同其余3组,其余3组均进行动物模型构建。假手术组、模型组大鼠均腹腔注射氯化钠注射液,硝酸甘油组腹腔注射硝酸甘油,丹参注射液组腹腔注射丹参注射液。对比4组大鼠的肢体导联与胸导联心电图参数,对比4组大鼠的血液流变学指标、左心室功能及左心室重构。结果:模型组的Ⅰ、Ⅱ、Ⅲ、aVL、aVF、V1、V2、V5、血浆粘度、纤维蛋白原、红细胞聚集指数、舒张末期室间隔厚度、左室舒张末期内径、左室收缩末期内径、左室舒张末期容积、左室收缩末期容积明显较假手术组、硝酸甘油组、丹参注射液组高,硝酸甘油、丹参注射液组以上指标明显较假手术组高,模型组的的左室舒张末期厚度、左室射血分数、左室短轴缩短率明显较假手术组、硝酸甘油组、丹参注射液组低,硝酸甘油、丹参注射液组的左室舒张末期厚度、左室射血分数、左室短轴缩短率明显较假手术组低。模型组的左心室重量指数、左心室截面直径明显较假手术组、硝酸甘油组、丹参注射液组高,硝酸甘油、丹参注射液组的左心室重量指数、左心室截面直径、梗死面积明显较假手术组高(P<0.05),硝酸甘油组与丹参注射液组以上指标对比无差异(P>0.05)。结论:丹参素注射液可改善急性心肌梗死大鼠的心室重构、左心室功能及肢体导联与胸导联心电图参数,可能与其可降低大鼠的血液流变学指标水平有关。 相似文献

S-2-(3 aminopropylamino) ethylphosphorothioic acid (WR-2721) shown to surpass radical scavenging thiols in their radioprotective efficacy in cancer-type diseases has been tested for its protective potential in the reperfused heart. We investigated the radical scavenger properties of the compound in a radical generating systemin vitro as well as in isolated rat hearts subjected to 30 min ischaemia and 30 min reperfusion with the electron-paramagnetic resonance spin trap technique. The action on high-energy phosphates is observed by means of phosphorus-31 nuclear magnetic resonance (NMR) spectroscopy while its influence on left ventricular systolic segmental length change (SSLC) during 60 min reperfusion following 60 min regional ischaemia was assessed with a fibreoptic system in anaesthetized open-chest rats. WR-2721 (0.1 mM) reduced the vascular concentration of radical adduct in isolated hearts by up to 78% (275±84% of pre-ischaemic baseline values vs 1260±413%, p<0.05) between 5 and 12.5 min reperfusion. This was accompanied by a reduction of the left ventricular end diastolic pressure to pre-ischaemic values at 30 min of reperfusion (9±6 mmHg vs 42±8 mmHg in the absence of WR-2721, p<0.02). An accelerated recovery of creatine phosphate levels (78±5% of pre-ischaemia values vs 41±5% within 60 min reperfusion; p<0.05) was observed under similar conditions with NMR-spectroscopy, although the post-ischaemic tissue content of adenosine triphosphate was not affected. The administration of WR-2721 (0.5 mmol/kg body weight) ledin situ to an accelerated restoration of contractile activity in the post-ligated left ventricular area reflected by the post-ischaemic recovery of SSLC (64±10% of pre-ischaemic values compared with 27±6% in control animals 60 min following reperfusion; p<0.02). The present data confirm an effective reduction in the exposure of the reperfused heart to endogenously released free radicals by WR-2721, a partial preservation of high-energy phosphates and an improvement in post-ischaemic contractility and encourage further investigation of such favourable action in injured myocardium. 相似文献
