一株多花黄精内生真菌的鉴别及其抗菌代谢产物   总被引:2,自引:0,他引:2  
【目的】药用植物内生真菌是一类重要的微生物资源,能代谢产生多种生物活性物质。本研究从浙江庆元百山祖自然保护区多花黄精(Polygonatum cyrtonema)分离获得1株具有抗菌活性的菌株zjqy610。【方法】通过形态和ITS rDNA序列分析,鉴定为变灰青霉(Penicillium canescens)。采用正相硅胶柱层析和凝胶(Sephadex LH-20)柱层析,以紫外光或碘蒸汽显迹,配合活性追踪等,从zjqy610发酵液中分离获得3个具抗真菌活性的化合物。【结果】通过质谱和核磁共振波谱技术分别将其结构鉴定为:乙基氧苯氨基亚胺乙酸(o-acetylbenzeneamidinocarboxylic acid)、灰黄霉素(griseofulvin)和[1,2-b]呋喃2-甲基3-羧甲基4-羟基-5-甲氧基萘(naphtho[1,2-b]furan-3-carboxylic acid,4-hydroxy-5-methoxy-2-methyl-)。抑菌活性测试表明,3个化合物对多种植物病原真菌具有抑制活性,其中化合物zjqy610D-4对番茄灰葡萄孢、圆形炭疽菌、泻根亚隔孢壳和核盘菌4种病原真菌的活性最强,半抑制浓度EC50分别为0.68、0.38、0.91和0.61mg/L。【结论】该化合物具有开发成农用抗生素的价值。  相似文献

The aim of two‐year‐long pot experiments was to investigate the influence of P. frequentans on the growth of over‐ground parts, the intensity of assimilation and transpiration of plants, as well as ths influence on the yield of potato bulbs. It was determined how the above fungus influenced the populaüon of G rostochiensis, the influence being characterized by the density change of cysts, eggs, and larvae in the soil. Experiments in vitro allowed to determine the wholesomeness of cysts, eggs, and larvae (J2) that were in direct contact with the mycelium ofP. frequentans.

It was stated that presence of P. frequentans intensified the development of plants and relevantly stimulated their growth, especially in the initial vegetation phase, as compared with the remaining combinations (Fig. 1). Positive effect of P. frequentans on the growth of potato yield was proved, in comparison with the control, and by co‐occurrence of the fungus with G. rostochiensis, also in comparison to the combination with nematodes only (Fig. 2).

Pot experiments confirmed the antagonistic effect of P. frequentans on the population of G. rostochiensis. Presence of P. frequentans in the ground caused namely a relevant diminution of density of cysts, eggs and larvae (Tab. II), as well as morphological changes by females, which was connected with size reduction of the cysts (Tab. III). Soil is presumed to be a major source of inoculum for Aspergillus flavus which contaminates cottonseed and produces the potent carcinogen, aflatoxin. Little is known about the mycoflora of the low desert soils of cotton fields where aflatoxin is a chronic problem. In this study, soils from cotton fields in southwestern Arizona and southeastern California were assayed for filamentous fungi. Forty-two taxa, predominantly in the genera Aspergillus, Penicillium and Fusarium, were isolated. To determine whether or not compounds produced by these fungi could be potential inhibitors of A. flavus, extracts of strains of each taxon were tested for their ability to inhibit growth of A. flavus. Twelve taxa produced compounds inhibitory to A. flavus, including several strains of Fusarium solani, Penicillium vinaceum and Aspergillus auricomus. The maximal carboxymethyl cellulase, filter paper (FP) cellulase and -glucosidase activities achieved byPenicillium janthinellum grown in a fermenter were 60, 5 and 9 U/ml, respectively. Adsorption for heavy metals via biomaterials such as fungal biomass presents a practical remediation technique for polluted water. Among all known filamentous fungi, Penicillium chrysogenum is widespread in nature and can serve as a biosorbent for heavy metals. In the current study, the ability of P. chrysogenum XJ-1 to remove copper (Cu2+) and chromium (Cr6+) from water was evaluated. The maximum biosorption capacity of XJ-1 for Cu2+ reached 42.83 ± 0.57 mg g?1 dry biomass at pH 5.0 after the equilibrium time of 1.5 h. The maximum biosorption capacity for Cr6+ at pH 3.0 reached 52.69 ± 1.68 mg g?1 dry biomass after the equilibrium time of 1.5 h. The biosorption data of XJ-1 biomass were well fitted to the Freundlich isotherm model and the pseudo-second-order Lagergren kinetic model. Laundry powder-treated and HCl-treated XJ-1 biomass significantly enhanced its adsorption capacity to Cu2+ and Cr6+, respectively. HCl and NaOH were suitable desorbents for Cu2+/Cr6+ loading biomass, respectively. Fourier transform infrared spectroscopy analyses revealed that hydroxyl, amine, and sulfonyl groups on the biosorbent contributed to binding Cu2+ and Cr6+ and that carbonyl and carboxyl groups were also vital binding sites of Cu2+. Scanning electron microscopy and energy-dispersive x-ray (SEM-EDX) analyses confirmed that considerable amounts of metals were precipitated on the cell surface of XJ-1. Our results suggested that XJ-1 might be used to purify multimetal-contaminated water. This low-cost and eco-friendly biomass of XJ-1 seems to have a broad use in the restoration of metal-contaminated water.  相似文献

Although filamentous microorganisms are widely used in industrial fermentation processes, their growth and differentation are not yet fully understood, because their biomass is structured, and therefore difficult to descrbie and to quantify. This lack of appropriate tools can hinder the optimization and control of the fermentation. A quantitative image analysis method was therefore developed for characterizing the physiology of the penicillin-producing mold Penicillium chrysogenum. This method is based on a differntial staining procedure showing six physiological states: growing material, three differentiated states characterzied by an increasing granulation, a highly vacuolized state, and dead segments having lost their cytoplasm. The image analysis software, with versions written for monochrome and color images, consisted of a semiautomatic binary mask computation step and a fully automatic segmentation step based on a fuzzy classification. The pectinases produced byPenicillium frequentans were subjected to pectin- or sodium polypectate-PAGE. The fungus secreted one endo- and one exo-polygalacturonase during the first 10 h of incubation, either in media supplemented with pectin or in the absence of carbohydrate source. After 17 h of cultivation, another two exo-and three endo-polygalacturonases were detected in cultures supplemented with pectin. The results indicate that two constitutive polygalacturonases are secreted initially, followed by the synthesis of other inducible pectinases, in a sequential production of the pectinolytic complex. Glucose oxidase overproducing mutants of Penicillium variabile (P16)   总被引:1,自引:0,他引:1  
Abstract Conidia of Penicillium variabile P16 were subjected to mutagenesis and selection for glucose oxidase production on media containing o -dianisidine. Studies of the relationship between dose of UV irradiation and conidial survival and frequency of mutation showed that the best frequency of positive mutation (17%) was obtained in correspondence to a conidial survival of 52%. Out of 54 overproducing mutants tested in shaken flasks, M-80.10 showed the highest level of glucose oxidase activity (127% higher than the wild-type). M-80.10 mutant, transferred every 15 days to fresh medium and tested monthly for 8 months, appeared stable. The time course of growth and enzyme production by the mutant M-80.10 showed an increase of the glucose oxidase activity in the culture medium up to 19 U ml−1 after 96 h of fermentation.  相似文献

Several strains of the genusPenicillium isolated from swabs from uranium miners working environment were screened for production of known mycotoxins; and embryotoxicity of chloroform extracts from the isolates was investigated in chick embryos.Penicillium aurantiogriseum was found to produce chaetoglobosin A. ED50 of this mycotoxin for 2-, 3- and 4-day-old chick embryos was found 0.040 (0.031–0.052), 0.074 (0.051–0.107), and 0.180 (0.113–0.229) µg/embryo, respectively. The effect was purely embryolethal with no signs of teratogenicity recorded. This is the first report on the isolation of chaetoglobosin A from the genusPenicillium.  相似文献
