Homopurine deoxyribonucleoside phosphorothioates, as short as hexanucleotides and possessing all internucleotide linkages of RP configuration, form a triple helix with two RNA or 2'-OMe-RNA strands, with Watson-Crick and Hoogsteen complementarity. Melting temperature and fluorescence quenching experiments strongly suggest that the Hoogsteen RNA strand is parallel to the homopurine [RP-PS]-oligomer. Remarkably, these triplexes are thermally more stable than complexes formed by unmodified homopurine DNA molecules of the same sequence. The triplexes formed by phosphorothioate DNA dodecamers containing 4-6 dG residues are thermally stable at pH 7.4, although their stability increases significantly at pH 5.3. FTIR measurements suggest participation of the C2-carbonyl group of the pyrimidines in the stabilization of the triplex structure. Formation of triple-helix complexes with exogenously delivered PS-oligos may become useful for the reduction of RNA accessibility in vivo and, hence, selective suppression/inhibition of the translation process.  相似文献

树木死亡在森林动态中发挥着重要作用, 环境因子是影响树木死亡的重要因素。为了阐明喀斯特常绿落叶阔叶林死亡个体空间分布格局及其与环境的相关性, 本研究以木论25 ha森林动态监测样地的两次木本植物调查数据为基础, 分析了死亡个体的物种组成、径级结构、分布格局及其与生境的关联。结果表明, 样地内共有死亡个体17,306株, 隶属57科130属194种; 其中, 死亡率排名前三的科和属分别为八角枫科、蔷薇科、大风子科以及山麻杆属(Alchornea)、火棘属(Pyracantha)、八角枫属(Alangium); 在物种水平上, 山麻杆(Alchornea trewioides)、毛桐(Mallotus barbatus)、野桐(Mallotus japonicus)等死亡率较高。样地内死亡个体的平均胸径为3.83 cm, 最大47.11 cm; 径级分布呈倒“J”型。从分布格局来看, 样地内大部分死亡个体在0-50 m的尺度上呈聚集分布, 部分大径级树木死亡个体趋向于随机分布。生境关联分析发现, 个体死亡率与山顶、陡坡、缓坡、洼地呈正关联的物种数分别为41种、13种、41种和38种, 呈负关联的物种数分别为38种、67种、33种和10种, 与各生境无关联的物种数分别为4种、3种、9种和35种; 多度排名前20的物种其个体死亡率均与4种生境类型呈现出正关联或者负关联; 不同径级个体死亡率都与山顶生境呈正关联, 与缓坡生境呈负关联, 其中5 cm ≤ DBH < 10 cm和10 cm ≤ DBH < 15 cm个体死亡率均与洼地生境呈正关联; DBH ≥ 15 cm个体死亡率与陡坡生境呈正关联, 与洼地生境呈负关联。在喀斯特亚热带常绿落叶阔叶林中, 死亡个体空间分布格局总体表现为聚集分布, 树木死亡受生境作用的影响较大。  相似文献

Liquid nitrogen (LN) storage is recommended for conserving plants with seeds for which standard seed banking is not feasible, but there are few empirical reports confirming its long-term effectiveness. In this study we evaluated in vitro viability and ex vitro growth of embryo axes of a species that is short-lived in seed bank conditions (Juglans nigra) and two recalcitrant seeded species (Aesculus hippocastanum and A. glabra) that were stored 11–23 years in LN. Viability of J. nigra axes did not decrease significantly after 23 years and produced normal-appearing plantlets. Similarly, viability of A. hippocastanum axes did not decrease after 23 years. However, A. glabra axes showed a significant decline in viability after 23 years, from 80% when freshly harvested to 33%. These results demonstrate that LN storage can preserve embryo axes of J. nigra and Aesculus sp. for over two decades, providing a workable conservation tool for these and similar species.  相似文献

Many plant species exhibit strong association with topographic habitats at local scales. However, the historical biogeographic and physiological drivers of habitat specialization are still poorly understood, and there is a need for relatively easy‐to‐measure predictors of species habitat niche breadth. Here, we explore whether species geographic range, climatic envelope, or intraspecific variability in leaf traits is related to the degree of habitat specialization in a hyperdiverse tropical tree community in Amazonian Ecuador. Contrary to our expectations, we find no effect of the size of species geographic ranges, the diversity of climate a species experiences across its range, or intraspecific variability in leaf traits in predicting topographic habitat association in the ~300 most common tropical tree species in a 25‐ha tropical forest plot. In addition, there was no phylogenetic signal to habitat specialization. We conclude that species geographic range size, climatic niche breadth, and intraspecific variability in leaf traits fail to capture the habitat specialization patterns observed in this highly diverse tropical forest.  相似文献

Understanding the genetic causes of neurodegenerative disease (ND) can be useful for their prevention and treatment. Among the genetic variations responsible for ND, heritable germline variants have been discovered in genome-wide association studies (GWAS), and nonheritable somatic mutations have been discovered in sequencing projects. Distinguishing the important initiating genes in ND and comparing the importance of heritable and nonheritable genetic variants for treating ND are important challenges. In this study, we analysed GWAS results, somatic mutations and drug targets of ND from large databanks by performing directed network-based analysis considering a randomised network hypothesis testing procedure. A disease-associated biological network was created in the context of the functional interactome, and the nonrandom topological characteristics of directed-edge classes were interpreted. Hierarchical network analysis indicated that drug targets tend to lie upstream of somatic mutations and germline variants. Furthermore, using directed path length information and biological explanations, we provide information on the most important genes in these created node classes and their associated drugs. Finally, we identified nine germline variants overlapping with drug targets for ND, seven somatic mutations close to drug targets from the hierarchical network analysis and six crucial genes in controlling other genes from the network analysis. Based on these findings, some drugs have been proposed for treating ND via drug repurposing. Our results provide new insights into the therapeutic actionability of GWAS results and somatic mutations for ND. The interesting properties of each node class and the existing relationships between them can broaden our knowledge of ND.  相似文献

The European black poplar (Populus nigra L.) has been used as a germplasm resource for the breeding of new poplar varieties around the world. The identification and screening of its high nitrogen use efficiency genotypes could enable the breeding of new resource-efficient poplar varieties. The accessions were screened using MALDI-TOF MS genotyping technology for ammonium transporter (AMT) and nitrate transporters (NRT) genes against phenotypic data for seedling height and ground diameter traits, in both low and high nitrogen environments. Allele re-sequencing of seven genes related to root development was carried out using the minisequencing method. By cluster analysis, 101 accessions of black poplar were divided into 4 populations, and it was concluded that Central Europe is the origin of the evolution of low-nitrogen and highefficiency populations of European black poplar. Association study between SNP typing and seedling height and ground diameter traits showed that there were significant correlations between four SNP loci and growth traits under the contrasting N levels. We found that SNP3 and SNP4 in the PttAMT1;3 gene were significantly associated with seedling height traits, and that SNP2 and SNP7 in the PttAMT1;2 and PttAMT1;5 genes, respectively, were significantly associated with ground diameter traits. Thus, considerable allelic diversity is present within the candidate genes studied and can be utilized to develop functional markers to select for poplars with improved growth under N stress conditions.  相似文献
